Targeting proliferating cell nuclear antigen (PCNA) for cancer therapy.

Proliferating cell nuclear antigen (PCNA) is an essential scaffold protein in many cellular processes. It is best known for its role as a DNA sliding clamp and processivity factor during DNA replication, which has been extensively reviewed by others. However, the importance of PCNA extends beyond its DNA-associated functions in DNA replication, chromatin remodelling, DNA repair and DNA damage tolerance (DDT), as new non-canonical roles of PCNA in the cytosol have recently been identified. These include roles in the regulation of immune evasion, apoptosis, metabolism, and cellular signalling. T

Proliferating cell nuclear antigen in the assessment of Barrett's mucosa.

Proliferating cell nuclear antigen (PCNA) expression was assessed in oesophageal epithelium from 70 patients with Barrett's oesophagus. PCNA counts were expressed in the form of a PCNA index. There were 24 patients with dysplastic epithelium, nine with regenerative atypia, 18 with benign histology and 19 with adenocarcinoma arising in Barrett's epithelium. The PCNA index of malignant tissue was significantly different from that of benign (P < 0.001) and dysplastic (P < 0.05) specimens. PCNA indices in histologically normal Barrett's epithelium adjacent to a tumour were significantly different

Proliferating cell nuclear antigen in human placenta and trophoblastic disease.

Expression of proliferating cell nuclear antigen (PCNA), an auxiliary factor for the DNA polymerase delta, is closely related to cell proliferation. Using the monoclonal anti-PCNA antibody 19F4, we examined the distribution of PCNA in formalin-fixed, paraffin-embedded sections of mature and first-trimester placentas, 8 hydatidiform moles, and 7 choriocarcinomas. In normal placentas there was strong expression of PCNA in cytotrophoblastic nuclei, while the syncytium and the amnionic epithelium were PCNA unreactive. Staining of stromal cells was variable. These results are in complete agreement

Proliferating cell nuclear antigen (PCNA) in common epidermal lesions. An immunohistochemical study of proliferating cell populations.

A commercially available antibody to proliferating cell nuclear antigen was used to characterize and compare proliferating cell populations in paraffin sections of benign, premalignant, and malignant lesions of human epidermis using routine immunohistochemical techniques. Three patterns emerged. An ordered pattern was found in prurigo nodularis and keratoacanthoma, wherein moderately and strongly positive nuclei were distributed in a continuous, basal-suprabasal layer of relatively uniform thickness. There was graded loss and ultimate extinction of PCNA staining in progressively more superfici

Proliferating cell nuclear antigen is required for DNA excision repair.

Fractionation of extracts from human cell lines allows nucleotide excision repair of damaged DNA to be resolved into discrete incision and polymerization stages. Generation of incised intermediates depends on the XP-A protein, a polypeptide that recognizes sites of damaged DNA, and on the human single-stranded DNA-binding protein HSSB. The proliferating cell nuclear antigen (PCNA) is required for the DNA synthesis that converts the nicked intermediates to completed repair events. This need for PCNA implies that repair synthesis is carried out by DNA polymerase delta or epsilon. The ability to

Prognostic value of proliferating cell nuclear antigen in parotid gland cancer.

Although cell proliferation is related to tumour aggressiveness and prognosis, there are few studies describing the expression of proliferative markers in salivary gland cancer. Our aim was to assess the long-term prognostic value of the proliferating cell nuclear antigen (PCNA) in a large group of histologically different salivary gland cancers. We analysed the expression of PCNA in 159 patients with parotid gland cancer by means of immunohistochemistry. The mean follow-up time was 56.6 months. A high expression of PCNA showed a significant correlation to the patients' pathological lymph node

The evaluation of salivary gland tumours using proliferating cell nuclear antigen.

In order to assess its discriminating and prognostic value, we studied the immunoreactivity for proliferating cell nuclear antigen (PCNA) in tissue from 52 human salivary gland tumours using the murine monoclonal antibody PC10. The PCNA percentage count, namely, the average number of positive nuclei counted per 100 randomly selected tumour cells was recorded for each tumour. Anaplastic carcinoma was used as a positive control and histologically 'normal' salivary gland and tonsil served as a negative control. A PCNA count of 30 per cent was postulated to predict malignancy within a given saliva

Proliferative potential of meningiomas evaluated by proliferating cell nuclear antigen expression.

Meningiomas are principally benign in nature. Some meningiomas, however, grow fast or recur even after total removal. The biological behavior of meningiomas often can not be predicted from conventional histopathological studies. A monoclonal antibody against proliferating cell nuclear antigen (PCNA) was used to investigate the usefulness of the PCNA index as a parameter to estimate the proliferative activity of meningiomas. Fifty-two meningiomas were examined. The mean PCNA index of recurrent meningiomas (3.37 +/- 0.92%) was significantly higher than that of non-recurrent meningiomas (1.12 +/-

Prognostic significance of proliferating cell nuclear antigen expression in colorectal cancer.

BACKGROUND: Recognition of a biologic marker in colorectal cancer tissue that correlates with recurrence and poor survival would offer a rationale for planning aggressive adjuvant therapy. This study assessed the prognostic significance of proliferation activity in cancer cells and nonneoplastic epithelial cells in patients with colorectal cancer, using proliferating cell nuclear antigen (PCNA) immunohistochemical analysis. METHODS: A mouse monoclonal antibody that reacted with PCNA was used to measure proliferation indexes in neoplastic and nonneoplastic colonic tissues of two sex-matched an

Pentobarbital suppresses breast cancer proliferation by downregulating proliferating cell nuclear antigen.

Pentobarbital (PB), a barbiturate anesthetic, is widely used in many clinical treatments, including management of seizures and preoperative sedation. However, its potential role in cancer therapy remains underexplored. The inhibitory effects of PB on breast cancer proliferation were investigated in this study. Breast cancer cell lines MDA-MB-231 and MCF-7 were used to evaluate the effects of PB on cell proliferation. Proliferating cell nuclear antigen (PCNA) overexpression and knockdown models were established to assess its role in PB-mediated proliferation inhibition. Cell proliferation was m

Proliferating cell nuclear antigen (PCNA) expression in gestational trophoblastic diseases (GTD).

Gestational Trophoblastic Disease is an abnormal condition of the placenta, the incidence of which is very high in the state of Kerala, India. The proliferative rate of molar placentas in comparison with the normal placentas of comparable gestational age group was done in order to find out its role in the prognosis of this tumor by assessing the expression of PCNA in trophoblasts. PCNA expression was evaluated in 149 trophoblastic tumors and 96 normal placental tissue. The percentage of positive cells was significantly increased in molar placentas of the 1st trimester in comparison to the norm

Expression of proliferating cell nuclear antigen in ameloblastomas and odontogenic cysts.

The identification of the proliferative activity in tumours may be useful to predict the biological behaviour of different lesions. Proliferating cell nuclear antigen (PCNA) has been used for the evaluation of the proliferative ability of many lesions. In this study 22 ameloblastomas (4 follicular, 5 plexiform, 4 acanthomatous, 5 unicystic, 4 recurrent), 12 odontogenic keratocysts (OKC), 8 dentigerous cysts (DC), and 12 radicular cysts (RC) were analysed. PCNA+ cells were present in all cyst types but the OKC contained the highest number of PCNA+ cells. In OKC the location of PCNA+ cells was m

Expression of p63 and Proliferating Cell Nuclear Antigen in Oral Submucous Fibrosis.

AIM: Oral squamous cell carcinoma (OSCC) may be preceded by potentially malignant disorders such as oral submucous fibrosis (OSMF). p63 can detect genetic changes in OSMF and it facilitates early detection of malignant transformation, whereas proliferating cell nuclear antigen (PCNA) is a marker of proliferation and may prove to be a useful objective indicator of the biological behavior of various tumors. The aim of this study was to investigate the expression and pattern of the intensity of p63 protein and PCNA in normal oral mucosa and OSMF using immunohistochemistry (IHC) and to correlate t

FOS-Like Antigen 1 Regulates Proliferating Cell Nuclear Antigen Expression and Pancreatic Cancer Cell Proliferation and Migration.

BACKGROUND: Pancreatic cancer ranks among the leading causes of cancer-related death worldwide, and novel therapeutic strategies are urgently needed for advanced or metastatic disease states. Notably, targeted therapy remains a particularly promising approach. The transcription factor FOS-like antigen 1 (FOSL1) is overexpressed in refractory pancreatic cancers and drives tumor progression, correlating with unfavorable clinical outcomes. Thus, this study aimed to investigate the molecular mechanisms through which FOSL1 and proliferating cell nuclear antigen (PCNA) promote pancreatic cancer prog

Cytosolic proliferating cell nuclear antigen (PCNA) orchestrates neutrophil hyperactivation in COVID-19.

Neutrophils are central mediators of the hyperinflammatory response in severe SARS-CoV-2 infection. We report elevated cytosolic levels of proliferating cell nuclear antigen (PCNA) in neutrophils from patients with severe and critical COVID-19, correlating with enhanced NADPH oxidase-dependent reactive oxygen species (ROS) generation and neutrophil extracellular trap (NET) formation. Using T2AA, a small-molecule inhibitor of the PCNA scaffold, we demonstrate potent suppression of NADPH oxidase activation and NET release, particularly in response to SARS-CoV-2 RNA. Mechanistically, we identify

Complex roles for proliferating cell nuclear antigen in restricting human cytomegalovirus replication.

DNA viruses at once elicit and commandeer host pathways, including DNA repair pathways, for virus replication. Despite encoding its own DNA polymerase and processivity factor, human cytomegalovirus (HCMV) recruits the cellular processivity factor, proliferating cell nuclear antigen (PCNA) and specialized host DNA polymerases involved in translesion synthesis (TLS) to replication compartments (RCs) where viral DNA (vDNA) is synthesized. While the recruitment of TLS polymerases is important for viral genome stability, the role of PCNA is poorly understood. PCNA function in DNA repair is regulate

Ubiquitinated proliferating cell nuclear antigen activates translesion DNA polymerases eta and REV1.

In response to DNA damage, the Rad6/Rad18 ubiquitin-conjugating complex monoubiquitinates the replication clamp proliferating cell nuclear antigen (PCNA) at Lys-164. Although ubiquitination of PCNA is recognized as an essential step in initiating postreplication repair, the mechanistic relevance of this modification has remained elusive. Here, we describe a robust in vitro system that ubiquitinates yeast PCNA specifically on Lys-164. Significantly, only those PCNA clamps that are appropriately loaded around effector DNA by its loader, replication factor C, are ubiquitinated. This observation s
