Revisiting rabbit models for keratoconus: A long-term study on collagenase-induced disease progression.

Keratoconus (KC) is a degenerative disorder resulting from the degradation of the stromal collagen fibril network in the cornea, leading to its thinning and conical deformation. Various studies have established animal models of KC by using the collagenase type II enzyme to gain a better understanding of the pathogenesis, however, long-term monitoring or follow-up of the models have not been reported so far. This study evaluates the long-term stability of collagenase type II-induced KC in a rabbit model. Six New Zealand rabbits were divided into 4 study groups with 3 eyes per group. The groups

Differentiating primary dry eye disease from ocular neuropathic pain: implications for symptom management.

CLINICAL RELEVANCE: Eyecare practitioners' management of ocular surface disease is essential in managing increasing dry eye disease (DED) presentations including ocular neuropathic pain (ONP). Topical Proxymetacaine offers a simple, readily available and practical method of detecting ONP in practice and can be used to differentiate ONP from DED by eyecare practitioners, when accompanied with an anterior segment examination. BACKGROUND: Differentiating DED from ONP presents a significant opportunity to eyecare providers, allowing appropriate treatment choices for more adequate symptom control,

Bilateral topical anaesthetic abuse keratopathy.

A woman in her late 30s presented with severe pain, photophobia and redness in both eyes (BE) for 3 weeks. She was diagnosed and on treatment elsewhere for presumed bilateral herpes simplex virus (HSV) keratitis, with oral antivirals and topical steroids for 4 months. Examination showed bilateral near-total corneal epithelial defects with hypertrophic edges and diffuse stromal oedema. Detailed history uncovered frequent use of topical proparacaine hydrochloride solution (0.5%) for severe pain. She underwent bilateral amniotic membrane transplantation (AMT) with tarsorrhaphy, resulting in rapid

Impact of topical anaesthetic eye drops on corneal thickness and intraocular pressure measurements in broiler chickens: implications for ophthalmological evaluation.

1. Ophthalmological examinations in poultry are essential for monitoring overall health and welfare. These are commonly assessed using intraocular pressure (IOP) and corneal thickness (CT). This study examined the effects of 0.5% proparacaine HCl topical anaesthetics on IOP and CT across various corneal regions in broiler chickens.2. Ninety male broilers were housed under controlled conditions and underwent ophthalmic examinations, including CT and IOP. Baseline measurements were recorded for both eyes, after which 0.5% proparacaine HCl drops were instilled in the right eyes, while the left ey

The effect of Tityus serrulatus scorpion toxin gamma on Na channels in neuroblastoma cells.

The effects of highly purified toxin gamma from the venom of the scorpion Tityus serrulatus (TiTx gamma) on nerve membrane ionic channels have been investigated using the suction electrodes voltage clamp technique on neuroblastoma cells. The amplitude of the normally voltage-dependent Na current is reversible reduced by approximately 50% after 15-105 nM TiTx gamma, whereas even the highest toxin concentrations have no significant effect on the outward K current in the presence of tetrodotoxin. TiTx gamma causes a transient inward current to appear at membrane potentials between -70 and -40 mV,

Eledoisin and lacrimal secretion in the rabbit.

Eledoisin has been tried as a possible treatment for dry eye based on the hypothesis that it pharmacologically stimulates tear secretion when topically applied to the eye. To determine if topically applied eledoisin pharmacologically stimulates orbital lacrimal secretion, the orbital lacrimal gland excretory duct of normal rabbits was cannulated, and eledoisin was applied topically with and without prior administration of proparacaine. To determine if topically applied eledoisin stimulated accessory lacrimal gland secretion, isotonic buffer with and without eledoisin was tested in a rabbit mod

Topical proparacaine abuse resulting in evisceration.

A 72-year-old man was admitted to our clinic because of pain in the right eye. Corneal oedema, peripheral anterior synechiae formation and intraocular lens were determined in the right eye. The left eye was normal except for nuclear sclerosis. Intraocular pressure was 35 mm Hg in the right eye and 14 mm Hg in the left eye. The patient was diagnosed as having bullous keratopathy and glaucoma. He was treated with antiglaucoma drugs and artificial tears as an outpatient. Persistent keratopathy was observed at follow-up, despite adequate therapy. In the detailed anamnesis of the patient, we discov

Use of 7,7,8,8-tetracyanoquinodimethane for spectrophotometric determination of certain local anaesthetics and procainamide hydrochloride.

A simple and rapid spectrophotometric procedure has been developed for the determination of four local anaesthetics containing a free primary amine moiety and of procainamide hydrochloride as the drug substances and in dosage forms. The method is based on the reaction of the drug with 7,7,8,8-tetracyanoquinodimethane in alkaline solution to produce yellow products. The chromogen was measured at 473 nm. The effects of several variables on colour development were established. Job's plots of absorbance versus molar ratio of drug to reagent indicated a 1:1 ratio for all the drugs studied. Results

A proposed mechanism for increased tear-film osmolarity in contact lens wearers.

Contact lenses decrease corneal sensitivity and increase tear-film osmolarity. To determine whether the decrease in corneal sensitivity is responsible for the increase in tear-film osmolarity, we studied tear-film osmolarity in normal rabbits and rabbits with surgically induced keratoconjunctivitis sicca before and after the instillation of 0.5% proparacaine. Instillation of proparacaine resulted in a significant increase in tear-film osmolarity. Decreased corneal sensitivity, with a resultant decrease in tear secretory rates, is the most likely cause for increased tear-film osmolarity in wear

Morphological changes of the corneal epithelium after application of topical anaesthetic ointments.

The effect of topical anaesthetic ointments (4% cocaine, 4% xylocaine, 0.5% proparacaine, 0.2% oxybuprocaine) on the corneal epithelium of rabbits was examined using a scanning electron microscope. Even after a single application, the more toxic effect of cocaine compared to the other topical anaesthetics was evident. Cocaine caused disruption of both the plasma membrane and the cytoplasm. After a single application, the other preparations caused a marked decrease in the microvilli and microplicae, disruption of the intercellular spaces and the prominence of the cell nucleus which under normal
