Protein glycosylation is a very common post-translational modification seen in all branches of biology. The functional roles for protein glycosylation are many and varied, essential in eukaryotes but seemingly dispensable in bacteria. One group of bacteria where protein glycosylation has been looked at for at least 50 years are the actinobacteria, a large and diverse group of bacteria which include well know pathogens like Mycobacteria tuberculosis, Corynebacterium diphtheriae, and well know species important in biotechnology like Streptomyces lividans and Corynebacterium glutamicum. Actinobac

Regulation of SIRT1 activity is vital to energy homeostasis and plays important roles in many diseases. We previously showed that insulin triggers the epigenetic regulator DBC1 to prime SIRT1 for repression by the multifunctional trafficking protein PACS-2. Here, we show that liver DBC1/PACS-2 regulates the diurnal inhibition of SIRT1, which is critically important for insulin-dependent switch in fuel metabolism from fat to glucose oxidation. We present the x-ray structure of the DBC1 S1-like domain that binds SIRT1 and an NMR characterization of how the SIRT1 N-terminal region engages DBC1. T

BACKGROUND: Sepsis originates from the host's dysregulated response to pathogens, and its pathophysiological mechanisms are extremely complex. Recent research has found that post-translational modifications (PTMs) can regulate gene transcription without altering the genetic sequence, thereby playing a key role in the occurrence and development of sepsis.
OBJECTIVE: This review aims to systematically categorize the main types of PTMs and elucidate their roles in the pathogenesis of sepsis, thereby providing new perspectives for a deeper understanding of the complex pathophysiological processes

Spinal muscular atrophy (SMA) is an autosomal recessive disease characterized by a progressive loss of the spinal motoneurons. The SMA-determining gene has been termed survival motor neuron (SMN) and is deleted or mutated in over 98% of patients. The encoded gene product is a protein expressed as different isoforms. In particular, we showed that the rat SMN cDNA produces two isoforms with M(r) of 32 and 35kDa, both localized in nuclear coiled bodies, but the 32kDa form is also cytoplasmic, whereas the 35kDa form is also microsomal. To determine the molecular relationship between these two isof

Antiphospholipid antibody syndrome (APS) is a systemic autoimmune disease characterized by arterial and venous thrombosis and/or pregnancy morbidity. The incidence is around five new cases/100,000 persons per year and the prevalence is around 40-50 cases/100,000. The prevalence is higher (about 30%) among patients with systemic lupus erythematosus. APS is associated with circulating antiphospholipid antibodies (aPLs), a heterogeneous group of autoantibodies directed against negatively charged molecules and a combination of protein-complexed phospholipids. The predominant protein antigen in thi

Rheumatoid arthritis (RA) is a chronic autoimmune disease that significantly compromises patient quality of life due to its high prevalence and risk of disability. While its etiology remains incompletely understood, increasing evidence highlights the critical involvement of epigenetic mechanisms, particularly post-translational modifications (PTMs), in RA pathogenesis. Advances in proteomics have identified various PTMs-including phosphorylation, methylation, acetylation, ubiquitination, glycosylation, lactylation, as well as citrullination and carbamylation-as key regulators of inflammation,

Amyloid : the international journal of experimental and clinical investigation : the official journal of the International Society of Amyloidosis2021eng
AA amyloidosis is a disease caused by extracellular deposition of insoluble β-pleated sheet fibrils composed of amyloid A (AA) protein, an amino (N)-terminal fragment of serum amyloid A (SAA). The deposits disrupt tissue structure and compromise organ function. Although the disease is systemic, deposition in kidney glomeruli is the most common manifestation. The leading cause of AA amyloidosis is sustained or recurrent inflammation accompanied by elevated levels of SAA. Factors determining the conversion of SAA to AA amyloid fibrils have yet to be fully resolved. Herein, we present liquid chro

Nephrolithiasis is a prevalent urological disorder worldwide, whose pathogenesis involves a complex network of crystal formation, cellular injury, and microenvironmental dysregulation. As a critical mechanism for regulating cellular functions, protein post-translational modifications (PTMs) have been increasingly implicated in multiple facets of kidney stone formation, including crystal-cell interactions, oxidative stress responses, and inflammatory signaling pathways. This review systematically synthesizes the biochemical foundations of PTMs, the molecular microenvironment of nephrolithiasis,

Post-translational modifications (PTMs) play a pivotal role in the activity and stability of proteins and can, therefore, significantly affect the quality of protein- or cell-based biological therapeutic products. Accordingly, PTMs are often evaluated as critical quality attributes, which require thorough characterization and evaluation throughout product development. Continued progress in understanding and characterizing protein PTMs is important to enable the distribution of safe and effective biologic medicines. Currently, a wide range of analytical techniques are employed for the detection

BACKGROUND: The foetal testes produce the androgens necessary to masculinise the developing embryo and support the maturation of germ cells, that will eventually develop into sperm, thus ensuring future reproductive capacity. The testes develop from the bi-potential gonads in a highly orchestrated process resulting in the differentiation of a complex tissue with multiple cellular lineages. While recent transcriptomic and chromatin-based analyses of human foetal testes have provided an unprecedented level of insight into signalling pathways activated during this process, proteomic studies of th

Sepsis is a common and highly fatal condition in intensive care units and is one of the leading causes of death worldwide. As a clinically complex syndrome with intricate pathophysiology, early identification and assessment of sepsis remain challenging. Deeper insights at the molecular level are crucial for understanding the complex pathophysiological mechanisms, discovering new biomarkers, and improving prognosis. Post-translational modifications (PTMs) refer to the attachment of specific chemical groups to amino acid side chains through covalent, enzymatic, or non-enzymatic means, greatly ex

Post-translational modifications (PTMs) play significant roles in regulating protein structure, activity and function, and they are closely involved in various pathologies. Therefore, the identification of associated PTMs is the foundation of in-depth research on related biological mechanisms, disease treatments and drug design. Due to the high cost and time consumption of high-throughput sequencing techniques, developing machine learning-based predictors has been considered an effective approach to rapidly recognize potential modified sites. However, the imbalanced distribution of true and fa

The extracellular matrix (ECM) of bone and dentin contains several non-collagenous proteins. One category of non-collagenous protein is termed the SIBLING (Small Integrin-Binding LIgand, N-linked Glycoprotein) family, that includes osteopontin (OPN), bone sialoprotein (BSP), dentin matrix protein 1 (DMP1), dentin sialophosphoprotein (DSPP), and matrix extracellular phosphoglycoprotein (MEPE). These polyanionic SIBLING proteins are believed to play key biological roles in the mineralization of bone and dentin. Although the specific mechanisms involved in controlling bone and dentin formation ar

Ultraviolet (UV) irradiation leads to acute photodamage, photoaging and skin cancers. Post-translational modifications (PTMs) represent a key regulatory layer by modulating protein function, localization and interactions under UV stress. This review discusses the mechanistic involvement of PTMs in UV-induced skin damage and summarizes emerging PTM-related photoprotective strategies. Phosphorylation is a central mediator of UV-induced signal transduction, driving activation of the epidermal growth factor receptor (EGFR) and its downstream signalling pathways. Acetylation exerts distinct regulat

Pentosinane is a structurally complex nonenzymatic post-translational modification of proteins believed to be present in all living things. It falls into the category of advanced glycation end products (AGEs) and is structurally related to the other AGEs pentosidine and glucosepane. Although pentosidine and glucosepane have been widely studied for their role in wide-ranging conditions (e.g., diabetes mellitus, Alzheimer's disease, and human aging), relatively little is known about pentosinane. Interestingly, previous reports have suggested that pentosidine may derive from pentosinane. The (pat

Alpha-1-antitrypsin (AAT), an acute-phase protein encoded by the SERPINA1 gene, is a member of the serine protease inhibitor (SERPIN) superfamily. Its primary function is to protect tissues from enzymes released during inflammation, such as neutrophil elastase and proteinase 3. In addition to its antiprotease activity, AAT interacts with numerous other substances and has various functions, mainly arising from the conformational flexibility of normal variants of AAT. Therefore, AAT has diverse biological functions and plays a role in various pathophysiological processes. This review discusses m

Lung cancer remains the second most prevalent malignancy worldwide and is characterized by persistently high incidence and mortality rates. As the disease progresses, most patients develop immune evasion and metastatic dissemination, which represent major threats to overall survival. The advent of targeted therapies and immunotherapies has fundamentally reshaped the clinical management of lung cancer; however, therapeutic resistance and limited durability of response remain critical challenges. Lactylation has recently emerged not only as a novel post-translational modification but also as a p
