Fluorescence Anisotropy of Riboflavin vis-à-vis Protochlorophyllide in Lemon Juice.

A violet laser of wavelength 405 nm is used to excite steady-state fluorescence in the juice of a lemon. Prominent wavelength peaks appear at 520 and 670 nm due to emissions from riboflavin (vitamin B2) and protochlorophyllide (pchlide), the precursor of chlorophyll [Formula: see text], respectively. Using a polariser for excitation with vertically polarised light, and another one in parallel as well as in perpendicular positions to this one in the emission-path, counts at the two peaks in the high-resolution spectrometer are noted. Fluorescence anisotropies calculated from the obtained intens

Gossypium hirsutum gene of unknown function Gohir.A03G0737001 encodes a potential Chaperone-like Protein of protochlorophyllide oxidoreductase (CPP1).

A gene of unknown function identified in Gossypium hirsutum , Gohir.A03G0737001.1, was studied using sequence and bioinformatic tools. The encoded protein (referred to here as GhCPP1-A0A1U8HKT6) was predicted to function as a Chaperone-like protein of protochlorophyllide oxidoreductase (CPP1), which is involved with initiation of photochemical reactions of chlorophyll biosynthesis. Sequence analysis indicates it is embedded in the chloroplast envelope membrane through four transmembrane regions and contains a J-like domain that is structurally similar to the J domain of DnaJ/Hsp40 "holdase" ch

Multiplicity of different cell- and organ-specific import routes for the NADPH-protochlorophyllide oxidoreductases A and B in plastids of Arabidopsis seedlings.

The NADPH-dependent protochlorophyllide (Pchlide) oxidoreductase (POR) is a photoenzyme that requires light for its catalytic activity and uses Pchlide itself as a photoreceptor. In Arabidopsis there are three PORs denoted PORA, PORB and PORC. The PORA and PORB genes are strongly expressed early in seedling development. In contrast to PORB the import of PORA into plastids of cotyledons is substrate-dependent and organ-specific. These differences in the import reactions between PORA and PORB most likely are due to different import mechanisms that are responsible for the uptake of these proteins

Differential expression of genes encoding the light-dependent and light-independent enzymes for protochlorophyllide reduction during development in loblolly pine.

The expression patterns of the two distinct subfamilies of genes (designated porA and porB) encoding the light-dependent NADPH:protochlorophyllide oxidoreductases (PORs) in loblolly pine (Pinus taeda L.) were examined. Transcripts arising from both gene subfamilies were shown to be present at high levels in the cotyledons of dark-grown pine seedlings and to a lesser extent in their stems. Exposure of dark-grown seedlings to light resulted in increased levels of both porA and porB transcripts, as well as increased levels of mRNAs encoding other photosynthesis-related gene products, suggesting t

Regulation of protochlorophyllide synthesis genes in darkness by class I TCP transcription factors TCP14 and TCP15 prevents photodamage after illumination in Arabidopsis.

De-etiolation, which takes place when seedlings germinated in the dark are first exposed to light, involves profound morphological and physiological changes, among them the synthesis of chlorophylls from the highly reactive precursor protochlorophyllide (Pchlide). As excess Pchlide accumulation can cause cellular damage upon illumination, the entire chlorophyll synthesis pathway is subjected to tight transcriptional control. Here, we report that the activity of the class I TCP transcription factors (TCPs) TCP14 and TCP15 is required to repress genes involved in Pchlide synthesis in the dark, t

The miR396-GRFs Module Mediates the Prevention of Photo-oxidative Damage by Brassinosteroids during Seedling De-Etiolation in Arabidopsis.

The switch from dark- to light-mediated development is critical for the survival and growth of seedlings, but the underlying regulatory mechanisms are incomplete. Here, we show that the steroids phytohormone brassinosteroids play crucial roles during this developmental transition by regulating chlorophyll biosynthesis to promote greening of etiolated seedlings upon light exposure. Etiolated seedlings of the brassinosteroids-deficient det2-1 (de-etiolated2) mutant accumulated excess protochlorophyllide, resulting in photo-oxidative damage upon exposure to light. Conversely, the gain-of-function

Effects of iron deficiency and exogenous sucrose on the intermediates of chlorophyll biosynthesis in Malus halliana.

Malus halliana is an iron (Fe)-efficient apple rootstock growing in calcareous soil that shows obvious 'greenness' traits during Fe deficiency. Recent studies have shown that exogenous sugars can be involved in abiotic stress. To identify the key regulatory steps of chlorophyll (Chl) biosynthesis in M. halliana under Fe deficiency and to verify whether exogenous sucrose (Suc) is involved in Fe deficiency stress, we determined the contents of the Chl precursor and the expression of several Chl biosynthetic genes in M. halliana. The results showed that Fe deficiency caused a significant increase

Manganese Deficiency Suppresses Growth and Photosynthetic Processes but Causes an Increase in the Expression of Photosynthetic Genes in Scots Pine Seedlings.

Manganese deficiency is a serious plant nutritional disorder, resulting in the loss of crop productivity in many parts of the world. Despite the progress made in the study of angiosperms, the demand for Mn in gymnosperms and the physiological responses to Mn deficiency remain unexplored. We studied the influence of Mn deficiency for 24 weeks on Pinus sylvestris L. seedling growth, ion homeostasis, pigment contents, lipid peroxidation, chlorophyll fluorescence indices and the transcript levels of photosynthetic genes and genes involved in chlorophyll biosynthesis. It was shown that Mn-deficient

Nitrogen deficiency hinders etioplast development in stems of dark-grown pea (Pisum sativum) shoot cultures.

The effects of nitrogen (N) deprivation were studied in etiolated pea plants (Pisum sativum cv. Zsuzsi) grown in shoot cultures. The average shoot lengths decreased and the stems significantly altered considering their pigment contents, 77 K fluorescence spectra and ultrastructural properties. The protochlorophyllide (Pchlide) content and the relative contribution of the 654-655 nm emitting flash-photoactive Pchlide form significantly decreased. The etioplast inner membrane structure characteristically changed: N deprivation correlated with a decrease in the size and number of prolamellar bodi

[Photoluminescence of singlet oxygen in solutions of chlorophylls and pheophytins].

With the use of mechanical phosphoroscopes the photoinduced luminescence of singlet oxygen (1270 nm) have been found in air saturated solutions of chlorophyll a, bacteriochlorophyll a, protochlorophyll and pheophytins in CCl4, CS2, and freon 112. The excitation spectra of the luminescence coincide with the absorption spectra of the pigments. The relative quantum yields of the luminescence are determined, the data are used for calculation the probablities of intersystem crossing in pigment molecules. All the pigments are shown to quench 1O2, the rate constants of the quenching are measured. The

YGL46 encoding ferredoxin-like protein interacts with FTR and CAO to regulate chlorophyll synthesis and leaf development in maize.

Chlorophyll is crucial in photosynthesis, enabling plants to absorb light energy for photosynthesis. Here, we identified a yellow-green leaf mutant ygl46 derived from natural variation. Phenotypic characterization exhibited pale leaves. In the mutant, chlorophyll accumulation was significantly reduced, especially chlorophyll b. The mutated phenotype is governed by a single recessive gene. The mutation site was mapped to a 270-kb physical interval between the Indel markers ID0270 and ID0302 on chromosome 5 using an F2 mapping population. Structures of LPOR-Chlide complexes reveal the structural basis of membrane remodeling and photocatalysis.

Light-dependent protochlorophyllide oxidoreductase (LPOR) is a light-driven enzyme in flowering plants. It is involved in chlorophyll biosynthesis while also reorganizing membrane lipids into the cubic membrane network that supports chloroplast development. However, the structural basis of these two activities and their relationship have remained unclear. Here, cryo-electron microscopy of chlorophyllide-bound LPOR oligomers reveals nine distinct assembly states, including helical filaments, stacked rings and segmented strings of dimers. We find that strings of LPOR dimers reshape lipid bilayer

5-Aminolevulinic acid and hydrogen sulphide alleviate chilling stress in pepper (Capsicum annuum L.) seedlings by enhancing chlorophyll synthesis pathway.

5- Aminolevulinic acid (ALA) as a precursor in chlorophyll (Chl) synthesis and hydrogen sulphide (H2S) as a gas signalling molecules can alleviate various abiotic stresses by enhancing photosynthesis. However, little is known about their mechanisms ameliorating photosynthesis under chilling stress, or interactions of ALA and H2S in Chl synthesis. In this study, we explored the effects of exogenous ALA and H2S on chilling stress-induced photosynthesis damage in pepper (Capsicum annuum L.) seedlings. Chilling inhibited the photosynthetic capacity of pepper seedlings, ALA or H2S treatment alone c

Isolation and in vitro characterization of BchE, the cobalamin-dependent anaerobic magnesium protoporphyrin IX monomethylester cyclase.

The radical S-adenosylmethionine (SAM) superfamily comprises more than 800,000 enzymes that use [Fe4S4] clusters to initiate radical chemistry that mediates an exceptionally broad range of chemical transformations. Within this superfamily, cobalamin (Cbl)-dependent radical SAM enzymes constitute a major subclass predominantly associated with methylation reactions. However, several notable members catalyze nonmethylase reactions, for which the mechanistic role of Cbl is poorly understood. Bacteriochlorophyll biosynthesis enzyme BchE is a Cbl-dependent radical SAM enzyme that catalyzes a six-ele

Psb28 protein is involved in the biogenesis of the photosystem II inner antenna CP47 (PsbB) in the cyanobacterium Synechocystis sp. PCC 6803.

The role of the Psb28 protein in the structure and function of the photosystem II (PSII) complex has been studied in the cyanobacterium Synechocystis sp. PCC 6803. The protein was localized in the membrane fraction and, whereas most of the protein was detected as an unassembled protein, a small portion was found in the PSII core complex lacking the CP43 antenna (RC47). The association of Psb28 with RC47 was further confirmed by preferential isolation of RC47 from the strain containing a histidine-tagged derivative of Psb28 using nickel-affinity chromatography. However, the affinity-purified fr

LCAA, a novel factor required for magnesium protoporphyrin monomethylester cyclase accumulation and feedback control of aminolevulinic acid biosynthesis in tobacco.

Low Chlorophyll Accumulation A (LCAA) antisense plants were obtained from a screen for genes whose partial down-regulation results in a strong chlorophyll deficiency in tobacco (Nicotiana tabacum). The LCAA mutants are affected in a plastid-localized protein of unknown function, which is conserved in cyanobacteria and all photosynthetic eukaryotes. They suffer from drastically reduced light-harvesting complex (LHC) contents, while the accumulation of all other photosynthetic complexes per leaf area is less affected. As the disturbed accumulation of LHC proteins could be either attributable to

Screening and characterization of long noncoding RNAs involved in the albinism of Ananas comosus var. bracteatus leaves.

Long noncoding RNAs (lncRNAs) have been reported to play key regulatory roles in plant growth, development, and biotic and abiotic stress physiology. Revealing the mechanism of lncRNA regulation in the albino portions of leaves is important for understanding the development of chimeric leaves in Ananas comosus var. bracteatus. In this study, a total of 3,543 candidate lncRNAs were identified, among which 1,451 were differentially expressed between completely green (CGr) and completely white (CWh) leaves. LncRNAs tend to have shorter transcripts, lower expression levels, and greater expression

Mg protoporphyrin monomethylester cyclase deficiency and effects on tetrapyrrole metabolism in different light conditions.

Mg protoporphyrin monomethylester (MgProtoME) cyclase catalyzes isocyclic ring formation to form divinyl protochlorophyllide. The CHL27 protein is part of the cyclase complex. Deficiency of CHL27 has been previously reported to compromise photosynthesis and nuclear gene expression. In a comprehensive analysis of different CHL27 antisense tobacco lines grown under different light conditions, the physiological consequences of gradually reduced CHL27 expression on the tetrapyrrole biosynthetic pathway were explored. Excessive amounts of MgProtoME, the substrate of the cyclase reaction, accumulate

Chloroplastic protein PORC undergoes heat-induced condensation and enhances thermotolerance in Arabidopsis.

Chloroplast stress granules (cpSGs) are emerging as dynamic suborganellar condensates that play a crucial role in mediating stress response in plants. In this study, we demonstrate that the chlorophyll biosynthesis enzyme protochlorophyllide oxidoreductase C (PORC) localizes to cpSGs in response to acute and prolonged heat stress in Arabidopsis (Arabidopsis thaliana). While PORC promoter activity was developmentally regulated and remained unresponsive to heat, PORC protein re-localized from a diffuse chloroplast distribution into punctate structures under elevated temperatures. This condensati

Evolution of Ycf54-independent chlorophyll biosynthesis in cyanobacteria.

Chlorophylls (Chls) are essential cofactors for photosynthesis. One of the least understood steps of Chl biosynthesis is formation of the fifth (E) ring, where the red substrate, magnesium protoporphyrin IX monomethyl ester, is converted to the green product, 3,8-divinyl protochlorophyllide a In oxygenic phototrophs, this reaction is catalyzed by an oxygen-dependent cyclase, consisting of a catalytic subunit (AcsF/CycI) and an auxiliary protein, Ycf54. Deletion of Ycf54 impairs cyclase activity and results in severe Chl deficiency, but its exact role is not clear. Here, we used a Δycf54 mutant
