Pseudomonas syringae pv. syringae Associated With Mango Trees, a Particular Pathogen Within the "Hodgepodge" of the Pseudomonas syringae Complex.

The Pseudomonas syringae complex comprises different genetic groups that include strains from both agricultural and environmental habitats. This complex group has been used for decades as a "hodgepodge," including many taxonomically related species. More than 60 pathovars of P. syringae have been described based on distinct host ranges and disease symptoms they cause. These pathovars cause disease relying on an array of virulence mechanisms. However, P. syringae pv. syringae (Pss) is the most polyphagous bacterium in the P. syringae complex, based on its wide host range, that primarily affects

Genetic characterization of pseudomonas syringae pv. syringae strains from stone fruits in california.

Strains of Pseudomonas syringae pv. syringae were isolated from healthy and diseased stone fruit tissues sampled from 43 orchard sites in California in 1995 and 1996. These strains, together with P. syringae strains from other hosts and pathovars, were tested for pathogenicity and the presence of the syrB and syrC genes and were genetically characterized by using enterobacterial repetitive intergenic consensus (ERIC) primers and PCR. All 89 strains of P. syringae pv. syringae tested were moderately to highly pathogenic on Lovell peach seedlings regardless of the host of origin, while strains o

Identification and Genetic Characterization of Pseudomonas syringae pv. syringae from Sweet Cherry in Turkey.

Pseudomonas syringae pv. syringae, which causes bacterial canker, is the most polyphagous bacterium in the P. syringae complex because of its broad host range. This pathogen is considered the major bacterial disease in cherry orchards. In this study, several samples were collected from infected sweet cherry (Prunus avium L.) trees in different locations of the Marmara region in Turkey between 2016 and 2018. Sixty-three isolates were identified as P. syringae pv. syringae by pathogenicity, LOPAT, GATTa, and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry tests. Tota

Genomic correlates of tailocin sensitivity in Pseudomonas syringae.

Phage-derived bacteriocins, also referred to as tailocins, are structures encoded by bacterial genomes and deployed into the extracellular environment to kill sensitive cells. Tailocins display great potential as agricultural antimicrobials due to their durability, efficiency, and specificity of killing with prophylactic application demonstrated to prevent infection by multiple phytopathogens. Previous reports suggest that tailocins of Pseudomonas syringae interact with sugar moieties in the lipopolysaccharide (LPS) to target sensitive cells. However, it remains unclear how genetic and genomic

A Large Tn7-like Transposon Confers Hyper-Resistance to Copper in Pseudomonas syringae pv. syringae.

Copper resistance mechanisms provide an important adaptive advantage to plant pathogenic bacteria under exposure to copper treatments. Copper resistance determinants have been described in Pseudomonas syringae pv. syringae (Pss) strains isolated from mango intimately associated with 62 kb plasmids belonging to the pPT23A family (PFP). It has been previously described that the indiscriminate use of copper-based compounds promotes the selection of copper resistant bacterial strains and constitutes a selective pressure in the evolution of copper resistance determinants. Hence, we have explored in

Identification and Characterization of Pseudomonas syringae pv. syringae, a Causative Bacterium of Apple Canker in Korea.

In the present investigation, bacterial isolates from infected apple trees causing apple canker during winter were studied in the northern Gyeongbuk Province, Korea. The pathogen was identified as Pseudomonas syringae pv. syringae (Pss) through various physiological and biochemical characterization assays such as BIOLOG, gas chromatography of fatty acid methyl esters, and 16S rRNA. Bioassays for the production of phytotoxins were positive for syringopeptin and syringomycin against Bacillus megaterium and Geotrichum candidum, respectively. The polymerase chain reaction (PCR) method enabled the

Bio-integration of rhizobacteria and biogenic silver nanoparticles against Pseudomonas syringae pv. syringae causing plum bacterial canker.

BACKGROUND: Plum is a nutritionally and medicinally valuable fruit with high economic value; however, in temperate regions, its yield continuously suffers from bacterial canker caused by Pseudomonas syringae pathovars. RESULTS: To bridge the research gap, a study was conducted to optimize the use of rhizobacteria and biogenic silver nanoparticles (AgNPs) for controlling bacterial canker in plums. Rhizobacteria were isolated from plum orchards and identified (through 16 S rRNA gene sequencing, phylogenetic analysis using MEGA) and further evaluated for plant growth-promoting characteristics (s

Antimicrobial Activity of LysX and LysP Endolysins Against Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. juglandis.

Pseudomonas syringae pv. syringae and Xanthomonas arboricola pv. juglandis are the causal agents of bacterial canker in cherry and walnut blight, respectively, which cause significant production losses worldwide. These diseases have traditionally been controlled by copper-based agrochemicals and, more recently, antibiotics. However, the prolonged use of these compounds has led to the emergence of resistant bacterial strains. The search for new, efficient, and environmentally friendly biocontrol alternatives has intensified. Phages are promising candidates due to their ability to specifically i

Natural products for biocontrol of Pseudomonas syringae: mechanisms and applications.

Pseudomonas syringae functions as a model phytopathogen causing numerous crop diseases, resulting in substantial economic losses in global agriculture. Presently, management of P. syringae predominantly depends on chemical pesticides; however, their prolonged application has contributed to escalating resistance and environmental contamination, highlighting urgent requirement for sustainable biological control approaches. In this review, we examine recent advances in the utilization and mechanistic understanding of natural products derived from plants, animals, and microorganisms for the contro

Bacteriophages Control Epiphytic Pseudomonas syringae Populations in Highbush Blueberry Leaves.

The Pseudomonas syringae complex (Psc) is a group of globally distributed phytopathogens responsible for substantial agricultural losses. Although bacteriophage-based biocontrol has shown promise against Psc, no studies have examined phages targeting blueberry-tropic Psc lineages. Here, we isolated phages infecting Psc strains from diseased highbush blueberry (Vaccinium corymbosum), and evaluated their suitability for biocontrol using a multi-stage screening pipeline incorporating host-range analysis, comparative genomics, environmental stability testing, in vitro antibacterial efficacy assays

The Pseudomonas syringae effector protein HopZ1a suppresses effector-triggered immunity.

*The Pseudomonas syringae pv syringae type III effector HopZ1a is a member of the HopZ effector family of cysteine-proteases that triggers immunity in Arabidopsis. This immunity is dependent on HopZ1a cysteine-protease activity, and independent of known resistance genes. We have previously shown that HopZ1a-triggered immunity is partially additive to that triggered by AvrRpt2. These partially additive effects could be caused by at least two mechanisms: their signalling pathways share a common element(s), or one effector interferes with the response triggered by the other. *Here, we investigate

Loop-mediated isothermal amplification: a rapid molecular technique for early diagnosis of Pseudomonas syringae pv. syringae of stone fruits.

BACKGROUND: Pathogenic bacteria cause significant economic damages in agriculture. The detection of such bacteria is considered as a continual interest for plant pathologists to prevent disease dissemination. Pseudomonas syringae pv. syringae is one of the most important bacterial pathogens infecting yield and quality of stone fruits throughout the world. Biochemical assays such as a LOPAT and GATTa are common methods to detect this pathogen. Serological tests and culturing on King's B selective medium also used to isolate this bacterium. Spatiotemporal metabolomics of tobacco leaves in response to Pseudomonas syringae infection.

Tobacco (Nicotiana tabacum L.) is a major economic crop and a model for plant-pathogen interactions, yet the spatiotemporal dynamics of defense metabolism during infection remain poorly characterized. Here, we used MALDI-MSI-based spatial metabolomics to systematically profile tobacco leaves during Pseudomonas syringae infection. Multidimensional analysis of 1,399 annotated metabolites revealed distinct spatiotemporal regulation patterns. Temporally, early infection (12 h postinfection (hpi)) was characterized by increased organic acids and terpenoids, followed by a mid-stage shift toward phen

Different biosynthetic pathways to fosfomycin in Pseudomonas syringae and Streptomyces species.

Fosfomycin is a wide-spectrum antibiotic that is used clinically to treat acute cystitis in the United States. The compound is produced by several strains of streptomycetes and pseudomonads. We sequenced the biosynthetic gene cluster responsible for fosfomycin production in Pseudomonas syringae PB-5123. Surprisingly, the biosynthetic pathway in this organism is very different from that in Streptomyces fradiae and Streptomyces wedmorensis. The pathways share the first and last steps, involving conversion of phosphoenolpyruvate to phosphonopyruvate (PnPy) and 2-hydroxypropylphosphonate (2-HPP) t

Structure and Biosynthesis of Mangotoxin Produced by the Plant Pathogen Pseudomonas syringae.

Pathogenic bacteria interfere with plant metabolism through specialized metabolites and mechanisms. Mangotoxin was identified in 2003 from Pseudomonas syringae pv. syringae, which causes devastating diseases globally in various crops, including mango. Mangotoxin was found to inhibit arginine biosynthesis in tomato and Escherichia coli; however, its chemical structure remained unknown. We determined the structure of mangotoxin from the native bacterial producer P. syringae to be an alanine-citrulline dipeptide that contains an internal alkyne on citrulline. We showed that mangotoxin also inhibi

[Studies on the variability of the phaseolotoxin production by Pseudomonas syringae pv. phaseolicola].

Isolation of bacteria from a field of bean plants (Phaseolus vulgaris L.) with conspicuous symptoms of halo blight disease resulted in 123 bacterial strains from which 57 were identified as Pseudomonas syringae pv. phaseolicola . At 18 degrees C the phaseolotoxin production of the isolated strains differs widely in submerse culture. Only few strains produce high amounts of phaseolotoxin being comparable with those of the reference strain 1321, while most of the strains show a low capability of phaseolotoxin production. Furthermore, we proved the stability of phaseolotoxin production of 29 stra

Syringotoxin Biosynthetic Gene Cluster Required for Antiamoebal Defense in Pseudomonas syringae.

Syringotoxins are chlorinated cyclic lipopeptides from Pseudomonas syringae that were structurally characterized more than three decades ago. The corresponding biosynthetic gene cluster required for their production, however, has not been experimentally assigned. Based on targeted genetics, metabolite profiling, and structure analysis, we identify a divergent syringomycin-type nonribosomal peptide synthetase gene cluster in P. syringae DSM 1242 and show that psyt is the syringotoxin biosynthetic gene cluster. Deletion of psyt abolishes syringotoxin production and renders the mutant susceptible

Pseudomonas syringae pv. actinidiae: a re-emerging, multi-faceted, pandemic pathogen.

UNLABELLED: Pseudomonas syringae pv. actinidiae is the causal agent of bacterial canker of green-fleshed kiwifruit (Actinidia deliciosa) and yellow-fleshed kiwifruit (A. chinensis). A recent, sudden, re-emerging wave of this disease has occurred, almost contemporaneously, in all of the main areas of kiwifruit production in the world, suggesting that it can be considered as a pandemic disease. Recent in-depth genetic studies performed on P. syringae pv. actinidiae strains have revealed that this pathovar is composed of four genetically different populations which, to different extents, can infe

Abscisic Acid-Cytokinin Antagonism Modulates Resistance Against Pseudomonas syringae in Tobacco.

Phytohormones are known as essential regulators of plant defenses, with ethylene, jasmonic acid, and salicylic acid as the central immunity backbone, while other phytohormones have been demonstrated to interact with this. Only recently, a function of the classic phytohormone cytokinin in plant immunity has been described in Arabidopsis, rice, and tobacco. Although interactions of cytokinins with salicylic acid and auxin have been indicated, the complete network of cytokinin interactions with other immunity-relevant phytohormones is not yet understood. Therefore, we studied the interaction of k

Kiwifruit Xylem Sap Promotes Pseudomonas syringae pv. actinidiae Proliferation and Vascular Colonization.

The xylem has traditionally been considered a nutrient-poor environment, yet it is associated with systemic colonization by Pseudomonas syringae pv. actinidiae (Psa), the causal agent of kiwifruit bacterial canker. To clarify how Psa proliferates within this niche, we combined histopathology, metabolomics, bacterial growth kinetics, and gene-expression analyses. Psa exhibited a proliferation-during-migration pattern, reaching approximately 109 CFU·g-1 while traversing the vasculature at 0.24 cm·h-1 under condition of 16°C and 70% relative humidity, causing irreversible vascular necrosis.
