Carcinogenicity of bracken fern (Pteridium esculentum) in New Zealand.

Bracken fern (Pteridium esculentum) was harvested from two sites LB and TB one of which (TB) was on a central North Island New Zealand farm where bovine enzootic haematuria (BEH) was known to occur. The fern was dried, ground and incorporated (25% w/w) into a pelleted diet and fed to female rats for a total of 162 days. Fifteen weeks later when the rats were autopsied it was found that numerous tumours, mainly of the ileum and urinary bladder were present in the animals fed the bracken fern from the TB site. Neoplasms were found in 85% of rats from the TB group compared with 11% in the LB grou

Toxicological Activities of Pteridium aquilinum Rhizomes and Fiddleheads in HPV16-Transgenic Mice.

Pteridium aquilinum is a globally distributed plant species, highly adaptable to various environments and historically significant as a food source for both animals and humans. This study evaluated the in vivo effects of aqueous extracts from Pteridium aquilinum rhizomes and freeze-dried fiddleheads in a transgenic mouse model of human papillomavirus type 16 (HPV16)-induced cancer. Rhizome extract was administered in drinking water at concentrations of 0.0125, 0.025, and 0.05 g/mL for 28 days across six experimental groups (n = 5): G1 (wild-type, control), G2 (wild-type, 0.05 g/mL), G3 (HPV, c

Pteridium aquilinum (L.) Kuhn-A Review of Its Toxicology, Pharmacology, and Phytochemistry.

Pteridium aquilinum (L.) Kuhn, known as bracken fern, is considered a poisonous plant due to its toxic substances. This species contains toxic substances and enzymes: thiaminase and an anti-thiamine substance, which cause thiamine deficiency syndrome. Prunasin induces acute cyanide poisoning. Ptaquiloside causes haematuria, retinal atrophy, immunodeficiency, and lymphoproliferative disorders. It also induces carcinogenesis in livestock, and in animals and human cell lines. Ptaquiloside has been found in the milk of cattle, goats, and sheep that grazed on P. aquilinum in pastures. Ptaquiloside

Ptaquiloside from bracken (Pteridium spp.) promotes oral carcinogenesis initiated by HPV16 in transgenic mice.

Bracken (Pteridium spp.) is a common weed that is consumed as food especially in Asia, and is suspected of promoting carcinogenesis induced by papillomaviruses in the digestive and urinary systems. This is particularly worrying because the incidence of head-and-neck cancers associated with the human papillomavirus (HPV) is rapidly increasing, and HPV co-carcinogens urgently need to be identified. This study tested the hypothesis that two bracken compounds, ptaquiloside and rutin, are able to promote head-and-neck and bladder carcinogenesis in HPV16-transgenic mice. Expression of HPV16 E6 and E

Kinetics of the natural evolution of hydrogen cyanide in plants in neotropical Pteridium arachnoideum and its ecological significance.

The time-dependent natural release of hydrogen cyanide (HCN) was studied quantitatively using young croziers of the neotropical bracken fern Pteridium arachnoideum. HCN production was quantified in crushed tissue using a flow reactor at 30.0 +/- 0.1 degrees C. Released HCN was carried into appropriate traps with a moist air flow. Aliquots were drawn from the traps at fixed time intervals, and the HCN concentration was evaluated spectroscopically. All available prunasin (Pru), the only cyanogenic glycoside present, underwent decomposition into HCN in less than 1200 min. Fiddleheads (N = 76) con

Ptaquiloside from bracken (Pteridium spp.) inhibits tumour-infiltrating CD8+ T cells in HPV-16 transgenic mice.

Bracken is a fern with worldwide distribution. Exposure to bracken toxins such as ptaquiloside is hypothesized to increase the risk of papillomavirus-related cancers of the upper digestive tract. Ptaquiloside is thought to be an immunosupressor, thus allowing for the development of viral lesions. We have used a human papillomavirus type 16-transgenic (K14-HPV16) mouse model to study the effects of ptaquiloside on tumour-infiltrating CD8+ T lymphocytes, which are critical players in anti-tumour immunity. HPV16+/- mice received ptaquiloside (0.5 mg/mouse/week) for 10 weeks. These were then eutha

Hematuria and urinary bladder lesions compatible with bracken fern (Pteridium aquilinum) intoxication in captive fallow deer (Dama dama).

Hemangiosarcoma, hemangioma, transitional cell carcinoma, and chronic cystitis were diagnosed in the urinary bladder of six captive fallow deer (Dama dama). Hematuria and thin body condition were observed in the advanced cases. These findings were compatible with chronic enzootic hematuria and were suspected to have been induced by chronic ingestion of bracken fern (Pteridium aquilinum) present on the premises. These lesions were similar to those described in bovine enzootic hematuria in cattle. Hemangiosarcoma metastases, hydronephrosis, and renal carcinoma were also associated in some cases.

DNA damage and apoptosis induced by Pteridium aquilinum aqueous extract in the oral cell lines HSG and OSCC-3.

BACKGROUND: Bracken fern (Pteridium aquilinum) has been consumed by humans and animals for centuries. However, its consumption is associated with a high incidence of cancer in the upper digestory tract of different species. Although the oral cavity is the first site of contact with ingested toxic substances, the interaction of bracken fern composites with oral cell lines has not yet been studied. METHODS: In order to study the biological responses of oral cells exposed to bracken fern, we evaluated the genotoxic and cytotoxic effects of a bracken fern aqueous extract in oral cell lines. Neoplastic and non-neoplastic lesions in the upper digestive tract associated with the ingestion of Pteridium esculentum subsp. arachnoideum by 3 water buffalo in Brazil.

Upper digestive tract neoplasms associated with ingestion of Pteridium spp. (bracken fern) have been documented in cattle but not in buffalo. Here, we describe upper digestive tract lesions associated with the ingestion of P. esculentum subsp. arachnoideum in 3 water buffalo (Bubalus bubalis) in Brazil. Gross inspection revealed proliferative and ulcerative lesions in the mucosa of the base of the tongue, oropharynx, and esophagus in all 3 cases. Proliferative lesions were well-demarcated, white-to-brown, exophytic nodules on the lingual and oropharyngeal mucosa (papillomas, cases 1-3); a well

Fern-synthesized nanoparticles in the fight against malaria: LC/MS analysis of Pteridium aquilinum leaf extract and biosynthesis of silver nanoparticles with high mosquitocidal and antiplasmodial activity.

Malaria remains a major public health problem due to the emergence and spread of Plasmodium falciparum strains resistant to chloroquine. There is an urgent need to investigate new and effective sources of antimalarial drugs. This research proposed a novel method of fern-mediated synthesis of silver nanoparticles (AgNP) using a cheap plant extract of Pteridium aquilinum, acting as a reducing and capping agent. AgNP were characterized by UV-vis spectrophotometry, Fourier transform infrared (FTIR) spectroscopy, energy-dispersive X-ray spectroscopy (EDX), and X-ray diffraction (XRD). Phytochemical

Bracken-fern extracts induce cell cycle arrest and apoptosis in certain cancer cell lines.

Bracken fern [Pteridium aquilinem (L.) kuhn (Dennstaedtiaceae)] is one of the most common species on the planet. It has been consumed by humans and animals for centuries. Use by some human groups is because they believe bracken fern is good for health as plant medicine. However, it is also one of the few known plants that can cause tumors in farm animals. Many interested groups have focused their attention on bracken fern because of these interesting features. In order to evaluate the biological effects of exposure to this plant in cellular level, human cancer cell lines were treated with the

Drying Temperature and Storage Duration for the Retention of Bioactive Compounds of Selected Wild Edible Plants From Ethiopia.

Foods rich in bioactive compounds significantly prevent chronic diseases; however, these compounds are susceptible to drying temperature and storage duration because they lose some of their functionalities. This study aimed to determine effective drying temperatures and storage durations for better retention of bioactive compounds of three Ethiopian wild edible plants: Mussaenda arcuata, Celosia trigyna, and Pteridium aquilinum. The collected samples were oven-dried at 60°C, 65°C, and 70°C in a convective oven dryer to equilibrium moisture content and cooled to room temperature and analyzed fo

Prolonged detection of BPV2 in urine and blood of papillomatosis-affected cattle not grazing bracken fern.

BPV-2 infection can cause bladder infections in cattle that, when associated with bracken fern consumption, can progress to cancerous bladder tumors and also present as bovine enzootic hematuria (BEH). This study aimed to evaluate the prolonged natural BPV-2 infection in the blood and urine of cattle, excluding bracken fern consumption. Thirteen Girolando papillomatosis-affected cattle with no bracken fern contact history were monitored for 20 months. Blood, urine, and wart samples were collected for BPV-2 detection and clinical laboratory analyses. All animals showed the presence of BPV-2 in

Diagnosis of Enzootic Bovine Hematuria: Comparative performance of endoscopy and ultrasound in naturally occurring cases.

Ultrasound (US) and cystoscopy techniques assist in the diagnosis of diseases; however, in ruminant clinics, these methods are underutilized. Both have advantages and disadvantages that must be evaluated regarding the diagnosis of Enzootic Bovine Hematuria (EBH), a disease associated with the chronic consumption of Pteridium spp. due to the presence of ptaquiloside and dienone, compounds capable of reacting with nitrogenous bases when in aqueous solution, generating hematopoietic and carcinogenic effects on the bladder mucosa. Bracken Fern Carcinogen, Ptaquiloside, Forms a Guanine O6-Adduct in DNA.

Bracken fern (Pteridium sp.) is a viable and vigorous plant with invasive potential, ingestion of which causes chronic illness and cancers in farm animals. Bracken is a suspected human carcinogen, and exposure can result from ingestion of bracken-contaminated water, dairy products, or meat derived from livestock grazing on bracken fern. Bracken is also consumed in the diets of some communities. Ptaquiloside (PTQ), a known bracken carcinogen, is an illudane-type glycoside that forms a highly reactive electrophile, PTQ dienone, known to produce N7-guanine and N3-adenine adducts in DNA.
