Viral Metagenomic Analysis of Bat Fly Pupae.

Bats are important reservoirs of zoonotic viruses, yet the role of their obligate ectoparasites, particularly bat flies, in viral maintenance and transmission remains poorly understood. Although viral sequences have been detected in adult bat flies, vertical transmission to their offspring has not been investigated. Here, we present the first metagenomic analysis of bat fly pupae to assess transgenerational viral persistence. Eighty pupae associated with a large Miniopterus schreibersii colony in Hungary were analyzed by next-generation sequencing. We generated more than 69 million reads and o

Ecdysis triggering hormone is essential for larva-pupa-adult transformation in Leptinotarsa decemlineata.

In Drosophila melanogaster, ecdysis triggering hormone (ETH) is the key factor triggering ecdysis behaviour and promoting trachea clearance. However, whether ETH plays the dual roles in non-dipteran insects is unknown. In this survey, we found that Ldeth mRNA levels were positively correlated with circulating 20-hydroxyecdysone (20E) titers in Leptinotarsa decemlineata. Ingestion of an ecdysteroid agonist halofenozide or 20E stimulated the transcription of Ldeth, whereas RNA interference (RNAi) of ecdysteroidogenesis (LdPTTH or LdSHD) or 20E signalling (LdEcR, LdUSP or LdFTZ-F1) genes inhibite

Natural infection of a holometabolous insect with gregarines and their detection in pupae.

Gregarines are symbionts of the gut or other body cavities associated with numerous insect species. Knowledge about natural gregarine infection levels and across metamorphosis in holometabolous hosts is sparse. Individuals of the mustard leaf beetle (Phaedon cochleariae, Coleoptera: Chrysomelidae) were collected from eight populations and dissected to estimate the prevalence and load of infection with gregarines. In the laboratory, the effect of larval density (single individuals versus groups of five) on subsequent larval gametocyst load and adult infection with Gregarina cochlearium was test

Anti-Herpes Simplex Virus Efficacy of Silk Cocoon, Silkworm Pupa and Non-Sericin Extracts.

Herpes simplex virus (HSV) infections are prevalent worldwide and are the cause of life- threatening diseases. Standard treatment with antiviral drugs, such as acyclovir, could prevent serious complications; however, resistance has been reported specifically among immunocompromised patients. Therefore, the development of an alternative approach is needed. The silk cocoon derived from silkworm, Bombyx mori, has been recognized for its broad-spectrum biological activity, including antiviral activity; however, its effects against HSV infection are unknown. In this study, we investigated the inhib

Origin of insect wings, metamorphosis, pupae and the maj or insect orders.

Although most discussions on the origin and evolution of insect wings and metamorphosis have assumed that the ancestors of winged insects were terrestrial, it now seems possible that they were actually aquatic. Changing the basic assumptions affects our interpretations of the origin of metamorphosis and our understanding of insect diversity. It is argued that the ancestors of winged insects were similar to primitive mayflies, developing from aquatic larvae into terrestrial adults, and that metamorphosis originated as an inevitable consequence of an amphibiotic life cycle. It is suggested that

[Determination of 50 antibiotic residues in drone pupa powder by liquid chromatography-tandem mass spectrometry].

A method was established for the determination of 50 kinds of antibiotic residues (macrolides, quinolones, sulfonamides, tetracyclines, nitroimidazoles, lincomycin and chloramphenicol) in drone pupa powder by liquid chromatography-tandem mass spectrometry (LC-MS/MS). The samples were extracted with perchloric acid and lead acetate solution, and the protein was precipitated. The extraction solution was adjusted to pH 8 using dipotassium hydrogen phosphate, then the solution was purified by solid phase extraction (SPE) and analyzed by LC-MS/MS. The target compounds in the drone pupa powder were

Ultrastructure of the Eggs, Larvae, and Pupae of Hyphantria cunea (Lepidoptera: Erebidae).

Hyphantria cunea is a widespread pest that causes significant issues in forestry and agriculture, with a far-reaching global impact. However, ultramorphological studies of H. cunea, especially in terms of ultramorphological changes, are far from in-depth. Therefore, we investigated the ultrastructure of its eggs, larvae, and pupae using scanning electron microscopy, focusing on ultrastructural changes in larvae across different instars. The eggs have rough surfaces, and the micropylar region consists of rosette cells. The larval head chaetotaxy is symmetrically arranged, and there are three ty

Sound production by hawkmoth larvae and pupae through abdominal spiracles.

Some insect species produce defensive sounds across developmental stages. To test whether the characteristics, organs and mechanisms of sound production differ across stages within a single species, we examined how larvae and pupae of Phyllosphingia dissimilis (Lepidoptera: Sphingidae) produce sounds in response to simulated predator attacks via tactile stimulation. Laboratory experiments showed that final-instar larvae produce sound pulses (dominant frequency: 9.2-12.9 kHz; sound pressure level: 50.2-62.4 dB SPL) by expelling air through the pair of eighth abdominal (A8) spiracles, whereas pu

Silkworm pupa protects against hypoxic lung injury by activating the Nrf2 pathway and inhibiting ferroptosis.

BACKGROUND: Hypoxic lung injury (HLI) remains a significant clinical challenge, primarily characterized by oxidative damage and vascular remodeling. Recent studies have identified ferroptosis as a key contributor to hypoxia-related organ injury, and its inhibition represents a potential therapeutic strategy for HLI. Silkworm pupa (SW) contains diverse bioactive compounds with well-established anti-inflammatory, antioxidant, and anti-apoptotic properties. However, whether SW exerts protective effects against hypoxic lung injury through the regulation of ferroptosis remains unknown. METHODS: In

Allergenic characterization of Bomb m 4, a 30-kDa Bombyx mori lipoprotein 6 from silkworm pupa.

BACKGROUND: Silkworm pupa (SWP) food anaphylaxis has been described frequently in Asian countries. However, false-positive reactions by skin pricks and serum IgE (sIgE) tests to the extract complicate diagnosis, requiring identification of clinically relevant major allergens. OBJECTIVES: In this study, we characterized a novel SWP allergen, Bomb m 4, a 30-kDa lipoprotein, and evaluated its diagnostic sensitivity. METHODS: Bomb m 4 was identified by a proteomic analysis. This recombinant (r)Bomb m 4 was overexpressed in Escherichia coli, and the IgE reactivity by ELISA was compared with other

Uncovering the Molecular Mechanism of Anti-Allergic Activity of Silkworm Pupa-Grown Cordyceps militaris Fruit Body.

Cordyceps militaris has been widely used as an herbal drug and tonic food in East Asia and has also been recently studied in the West because of its various pharmacological activities such as antitumoral, anti-inflammatory and immunomodulatory effects. In this study, we examined the molecular mechanism underlying the anti-allergic activity of ethanol extract prepared from silkworm pupa-cultivated Cordyceps militaris fruit bodies in activated mast cells. Our results showed that ethanol extract treatment significantly inhibited the release of [Formula: see text]-hexosaminidase (a degranulation m

Mispatterning in the ommatidia of Apis mellifera pupae treated with a juvenile hormone analogue.

To further understand the function of morphogenetic hormones in honeybee eye differentiation, the alterations in ommatidial patterning induced by pyriproxyfen, a juvenile hormone (JH) analogue, were studied by scanning and transmission electron microscopy. Prepupae of prospective honeybee workers were treated with pyriproxyfen and the effects on ommatidial differentiation were described at the end of the pupal development. The results show that the entire ommatidia, i.e., the dioptric as well as the receptor systems, were affected by the JH analogue. The wave of ommatidial differentiation, whi

Mounting Drosophila pupae for laser ablation and live imaging of the dorsal thorax.

This protocol describes the preparation of Drosophilamelanogaster pupae for laser ablation and live imaging of the notum (dorsal thorax). Because the pupa is stationary, it can be continuously live imaged for multiple days if desired, making it ideal for studying wound signaling and repair, from before laser ablation through wound closure. In this protocol, we demonstrate the processes of staging, partially dissecting, mounting, wounding, and live imaging the pupal notum, with the wounding occurring during the live imaging process. For complete details on the use and execution of this protocol
