Upregulation of ATP-purinergic P2×2 receptors in the cochlea over-amplifies hearing sensitivity leading to hyperacusis and attenuation by antagonists.

Hearing hypersensitivity (hyperacusis) is a common hearing stress and can cause many psychological diseases, e.g., anxiety, learning disabilities, and attention-deficit/hyperactivity disorder (ADHD). Here, we report an unexpected finding that the upregulation of P2×2 ATP-purinergic receptors in the cochlea links to hyperacusis generation. We found that P2×2 expression in the cochlea but not in auditory centers was upregulated in the hyperacusis generated by Cx26 deficiency. Overexpression of P2×2 in the cochlea also caused hyperacusis. Conversely, downregulation of P2×2 expression or administr

Ectopic purinergic sensitivity develops at sites of chronic nerve constriction injury in rat.

The possibility that ectopic purinergic sensitivity develops following peripheral nerve injury was investigated in chronic constriction injury (CCI). Spontaneous firing of A-fibers originated from the injury site or from sensory endings of afferents in the contralateral sciatic nerve. ATP injected intravenously excited most of the injured fibers whereas none of the contralateral afferents responded to ATP. The ATP-induced effect was blocked by the P2 receptor antagonist reactive blue 2, but not the P1 receptor antagonist aminophylline. Neither the alpha-adrenoreceptor antagonist phentolamine n

Autocrine regulation of wound healing by ATP release and P2Y2 receptor activation.

AIMS: Application of exogenous nucleotides can modulate wound healing via the activation of purinergic receptors. However, evidence for the release of endogenous nucleotides and the subsequent activation of purinergic receptors in this process has not been well defined. Therefore, the current study aimed to investigate wound-mediated nucleotide release and autocrine purinergic signalling during HaCaT keratinocyte wound closure following scratch injury. MAIN METHODS: An in vitro scratch wound apparatus was employed to study wound healing over 24-h in the presence of modulators of ATP release,

Medicinal chemistry of P2 and adenosine receptors: Common scaffolds adapted for multiple targets.

Prof. Geoffrey Burnstock originated the concept of purinergic signaling. He demonstrated the interactions and biological roles of ionotropic P2X and metabotropic P2Y receptors. This review paper traces the historical origins of many currently used antagonists and agonists for P2 receptors, as well as adenosine receptors, in early attempts to identify ligands for these receptors - prior to the use of chemical libraries for screening. Rather than presenting a general review of current purinergic ligands, we focus on common chemical scaffolds (privileged scaffolds) that can be adapted for multipl

The platelet P2 receptors in arterial thrombosis.

ADP and ATP play a crucial role in hemostasis and thrombosis and their receptors are potential targets for antithrombotic drugs. The ATP-gated channel P2X1 and the two G protein-coupled P2Y1 and P2Y12 ADP receptors selectively contribute to platelet aggregation. Due to its central role in the formation and stabilization of a thrombus, the P2Y12 receptor is a well established target of antithrombotic drugs like clopidogrel which has proved efficacious in many clinical trials and experimental models of thrombosis. Competitive P2Y12 antagonists have also been shown to be effective in experimental

Synthesis of uracil nucleotide analogs with a modified, acyclic ribose moiety as P2Y(2) receptor antagonists.

A series of new uracil nucleotide analogs (monophosphates, triphosphates, and phosphonates) was synthesized, in which the ribose moiety was replaced by acyclic chains, including branched or linear alkyl or dialkylether linkers. 1-omega-Bromoalkyluracil derivatives (2) were converted to the corresponding alcohols by treatment with sodium hydroxide and subsequently phosphorylated using phosphorus oxychloride followed by hydrolysis to yield the monophosphates, or by coupling with diphosphate to form the triphosphates. Reaction of 2 with triethyl phosphite followed by deprotection with trimethylsi

Nucleotides in the eye: focus on functional aspects and therapeutic perspectives.

The presence and activity of nucleotides and dinucleotides in the physiology of most, if not all, organisms, from bacteria to humans, have been recognized by the scientific community, and the eye is no exception. Nucleotides in the dynamic fluids interact with many ocular structures, such as the tears and aqueous humor. Moreover, high concentrations of nucleotides in these secretions may reflect disease states such as dry eye and glaucoma. Apart from the nucleotide concentration in these fluids, P2 purinergic receptors have been described on the ocular surface (cornea and conjunctiva), anterio

Identification, characterization, and inhibition of the platelet ADP receptors.

Adenosine diphosphate (ADP) plays a crucial role in hemostasis and thrombosis, and its receptors are potential targets for antithrombotic drugs. Two G-protein-coupled P2 receptors contribute to platelet aggregation: the P2Y1 receptor initiates aggregation through mobilization of calcium stores, whereas the P2Y12 receptor coupled to adenylyl cyclase inhibition is essential for a full aggregation response to ADP and the stabilization of aggregates. The latter is defective in certain patients with a selective congenital deficiency of aggregation to ADP. It is also the target of the antithrombotic

P2-purinergic activation of phosphoinositide turnover is potentiated by A1-receptor stimulation in thyroid cells.

We recently discovered that in FRTL-5 cells the P1-purinergic agonist PIA (phenylisopropyladenosine) markedly enhanced P2-purinergic agonist-induced responses in an IAP (islet-activating protein or pertussis toxin)-sensitive manner. In this study we tested PIA and other P1 agonists for their permissive effects on GTP (a P2 agonist)-induced inositol phosphate production and arachidonate release and found that the order of potency was PIA = CHA (cyclohexyladenosine) greater than NECA (N-ethylcarboxamidoadenosine) = CADO (chloradenosine). The P1 agonists also caused an inhibition of thyrotropin-i

Expression of purinergic receptors on microglia in the animal model of choroidal neovascularisation.

To investigate the effect of P2 receptor on microglia and its inhibitor PPADS on choroidal neovascularization. Forty CX3CR1GFP/+ mice were randomly divided into 8 groups. In addition to the normal group, the rest of groups were receiving laser treatment. The retina and choroid from the second, third, fourth and fifth group of mice were taken in the 1, 4, 7, 14 days after laser treatment. The mice in the sixth and seventh group received intravitreal injection of 2 µl PPADS or PBS respectively immediately after laser treatment. The mice in the eighth group received topical application of PPADS o

P2Y14 receptor in the nervous system: Pharmacology, mechanisms, and therapeutic potential.

The P2Y14 receptor, a nucleotide sugar-sensing G protein-coupled receptor activated primarily by UDP-glucose (UDP-G), has emerged as an important regulator of purinergic signaling in both peripheral and central tissues. Distinct from other P2Y receptors, P2Y14 links extracellular metabolic cues to immune activation, chemotaxis, and inflammatory gene expression. Recent evidence demonstrates that P2Y14 is widely expressed in the nervous system and contributes to neuroinflammatory responses, glial activation, and maladaptive neuronal plasticity associated with neuropathic pain and other neurologi

Purinergic P2 receptors as targets for novel analgesics.

Following hints in the early literature about adenosine 5'-triphosphate (ATP) injections producing pain, an ion-channel nucleotide receptor was cloned in 1995, P2X3 subtype, which was shown to be localized predominantly on small nociceptive sensory nerves. Since then, there has been an increasing number of papers exploring the role of P2X3 homomultimer and P2X2/3 heteromultimer receptors on sensory nerves in a wide range of organs, including skin, tongue, tooth pulp, intestine, bladder, and ureter that mediate the initiation of pain. Purinergic mechanosensory transduction has been proposed for

Inhibitory effects of metals on ATP-induced current through P2X7 receptor in NG108-15 cells.

We investigated the effects of heavy metal ions on the ATP-induced nonselective cation current through P2X7 receptor (I(NS x P2X7)) in NG108-15 cells using the whole-cell patch-clamp technique. Cu2+ inhibited the I(NS x P2X7) most potently among the metal ions investigated. Other metals such as Ni2+, Cd2+, Zn2+ and Co2+ also inhibited the I(NS x P2X7) in concentration-dependent manners. The order of potency was Cu2+ >> Ni2+ > Cd2+ > Zn2+ > Co2+ with IC50 values of 16 nM, 0.79 microM, 1.2 microM, 3.0 microM and 4.6 microM, respectively. Fe3+ (10 and 100 microM) and Mn2+ (10 microM) did not affe

P2Y1 receptor modulation of the pre-Bötzinger complex inspiratory rhythm generating network in vitro.

ATP is released during hypoxia from the ventrolateral medulla (VLM) and activates purinergic P2 receptors (P2Rs) at unknown loci to offset the secondary hypoxic depression of breathing. In this study, we used rhythmically active medullary slices from neonatal rat to map, in relation to anatomical and molecular markers of the pre-Bötzinger complex (preBötC) (a proposed site of rhythm generation), the effects of ATP on respiratory rhythm and identify the P2R subtypes responsible for these actions. Unilateral microinjections of ATP in a three-dimensional grid within the VLM revealed a "hotspot" w

Investigation of reported small molecule P2Y6 receptor antagonists.

Interest in the P2Y6 receptor (P2Y6R) has increased substantially over the past decade, driven by evidence that pharmacological inhibition is beneficial in neurodegenerative disease models, inflammatory pathologies, and cancer. However, therapeutic targeting of the P2Y6R has remained a challenge due to a lack of available drug-like molecules. In 2023 and 2024, benzimidazole and quinoline chemotypes respectively, were reported as potent P2Y6R antagonists with pharmacological relevance in an inflammatory bowel disease model. In the present study positive and negative controls from each chemotype

Functional interaction between purinergic system and opioid withdrawal: in vitro evidence.

The effects exerted by P1 and P2 as well as by A1 and A2 agonists and antagonists purinoceptor on the acute opiate withdrawal induced by morphine were investigated in vitro. Following a 4 min in vitro exposure to morphine, the guinea-pig isolated ileum exhibited a strong contracture after the addition of naloxone. The P1 purinoceptor agonist, adenosine, was able dose-dependently to reduce morphine withdrawal whereas alpha,beta-methylene ATP (APCPP), a P2 purinoceptor agonist, increased morphine withdrawal. The P2Y2 receptor sensitizes mouse bladder sensory neurons and facilitates purinergic currents.

Sensitization of bladder afferents is an underlying contributor to the development and maintenance of painful bladder syndrome/interstitial cystitis. Extracellular purines and pyrimidines (e.g., ATP and UTP), released during bladder distension or from damaged cells after tissue insult, are thought to play an important role in bladder physiological and pathological states by actions at ionotropic P2X and metabotropic P2Y receptors. In the present study, we examined the ability of P2Y receptors to sensitize and modulate P2X-mediated responses in mouse bladder sensory neurons. UTP (a P2Y(2) and P

Receptor-mediated release of inositol phosphates in the cochlear and vestibular sensory epithelia of the rat.

Various neurotransmitters, hormones and other modulators involved in intercellular communication exert their biological action at receptors coupled to phospholipase C (PLC). This enzyme catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PtdInsP2) to inositol 1,4,5-trisphosphate (InsP3) and 1,2-diacylglycerol (DG) which act as second messengers. In the organ of Corti of the guinea pig, the InsP3 second messenger system is linked to muscarinic cholinergic and P2y purinergic receptors. However, nothing is known about the InsP3 second messenger system in the vestibule. In this stud
