The purple membrane of Halobacterium halobium: a new system for light energy conversion.

Patches of a distinctly different structure from the rest of the cell membrane occur in Halobacterium halobium. The isolated patches are called the purple membrane. It derives its colour from a retinal-protein complex, bacteriorhodopsin, which is the only protein species in this membrane serving a phototransducing function. Light energy is converted by a photochemical cycle going on continuously under illumination and accompanied by a cyclic release and uptake of protons. In the intact cell, this cycle operates as a vectorial process and therefore builds up an electrochemical gradient across t

Extraction and purification of purple membrane for photochromic thin film development: application in photoelectrochemical investigation.

Purple membrane (PM) has been extracted and purified from archaebacteria for thin film development. The purified purple membrane is isolated in 1 % polyvinyl alcohol solution for making thin film within gelatin and organically modified silicate matrices. For thin film within gelatin matrix, homogenized purple membrane suspension is mixed with 8 % gelatin and poured into a specially designed block with desired thickness of spacer having hydrophobicity followed by gelatinization of the same over home-made thermostatic control unit at 38 °C. The gelatinized matrix is then allowed to dry under con

Investigations into the effect of acid on the spectral and kinetic properties of purple membrane from Halobacterium halobium.

The formation and reversal of the acid species of purple membrane generated below pH 4.00 (22 degrees C) is studied together with the photochemical cycle over the pH range 6.40--3.20. The buffering capacity of the membrane reaches a peak at pH 4.30, indicating the possibility of a conformational change taking place. The generation of the new spectral species can take place in the dark and is unaffected by the addition of reducing agents. Kinetic parameters measured indicate that the group being titrated below pH 4.00 could be the same as that protonated in the formation of intermediate O. The

Thermal motions and function of bacteriorhodopsin in purple membranes: effects of temperature and hydration studied by neutron scattering.

The internal dynamics of bacteriorhodopsin, the light-driven proton pump in the purple membrane of Halobacterium halobium, has been studied by inelastic neutron scattering for various conditions of temperature and hydration. Light activation can take place when the membrane is vibrating harmonically. The ability of the protein to functionally relax and complete the photocycle initiated by the absorption of a photon, however, is strongly correlated with the onset of low-frequency, large-amplitude anharmonic atomic motions in the membrane. For a normally hydrated sample, this occurs at about 230

Rapid long-range proton diffusion along the surface of the purple membrane and delayed proton transfer into the bulk.

The pH-indicator dye fluorescein was covalently bound to the surface of the purple membrane at position 72 on the extracellular side of bacteriorhopsin and at positions 101, 105, 160, or 231 on the cytoplasmic side by reacting bromomethylfluorescein with the sulfhydryl groups of cysteines introduced by site-directed mutagenesis. At position 72, on the extracellular surface, the light-induced proton release was detected 71 +/- 4 microseconds after the flash (conditions: pH 7.3, 22 degrees C, and 150 mM KCl). On the cytoplasmic side with the dye at positions 101, 105, and 160, the corresponding

Photoelectric response of purple membrane fragments adsorbed on a lipid monolayer supported by mercury and characterization of the resulting interphase.

Purple membrane (PM) fragments were adsorbed on a dioleoylphosphatidylcholine (DOPC) monolayer supported by mercury to investigate the kinetics of light-driven proton transport by bacteriorhodopsin (bR). PM fragments were also adsorbed on a mercury-supported triethyleneoxythiol (TET) monolayer. On both monolayers, the light-on current exhibits a finite, potential dependent stationary component that decreases linearly with a positive shift in the applied potential. The light-on and light-off capacitive photocurrents were interpreted on the basis of a simple equivalent circuit, which accounts fo

Molecular Interactions Between Bacteriorhodopsin Protein Embedded Within Purple Membrane and the Surfaces of Polyvinyl Alcohol (PVA) and Graphene.

Bacteriorhodopsin (bR), the light-driven proton pump of Halobacterium salinarum, is a promising candidate for bioelectronic and optoelectronic applications. Its utility in hybrid systems depends on how surrounding materials influence structural stability and photocycle dynamics. In this work, all-atom molecular dynamics simulations are conducted under isothermal-isobaric conditions to evaluate bR in three environments: its native purple membrane, adsorption onto graphene sheets, and incorporation within a polyvinyl alcohol (PVA) matrix. Across all systems, bR displayed preferential adsorption

Precise Fabrication of Graphite-Like Material Directly on a Biological Membrane Enabled by Ethanol Ice Resist.

Ice lithography holds the potential to bridge cryogenic electron microscopy and electron-beam lithography and achieve direct high-precision functionalization of fragile biomaterials. Here we demonstrate that 5 keV electron irradiation of ethanol ice creates a material, patterned with <100 nm resolution, that is stable in the solid phase under ambient conditions. Employing the purple membrane from Halobacterium salinarum as a test target, we additionally show that the fabrication process results in minimal biomaterial mass loss. Ketene, an unstable intermediate, was identified in the irradiated

Automated setpoint adjustment for biological contact mode atomic force microscopy imaging.

Contact mode atomic force microscopy (AFM) is the most frequently used AFM imaging mode in biology. It is about 5-10 times faster than oscillating mode imaging (in conventional AFM setups), and provides topographs of biological samples with sub-molecular resolution and at a high signal-to-noise ratio. Unfortunately, contact mode imaging is sensitive to the applied force and intrinsic force drift: inappropriate force applied by the AFM tip damages the soft biological samples. We present a methodology that automatically searches for and maintains high resolution imaging forces. We found that the

Gram-negative-staining Bacillaceae with thick cell wall and monoderm architecture uncover evolutionary diversity and challenge Gram-based classification.

Gram staining has guided microbiology for over a century by coloring cells purple or pink, a read-out thought to distinguish monoderms (single membrane, thick peptidoglycan) from diderms (inner and outer membranes with thin wall). Here we show this rule fails repeatedly across Bacillaceae lineages historically deemed "Gram-positive". By combining light and transmission-electron microscopy, antibiotic-sensitivity assays and comparative genomics across 57 strains, we identify "Gram-negative-staining monoderms" lacking outer membrane yet retaining thick peptidoglycan walls. These bacteria lack li

Effect of metal ion exchange on the photocurrent response from bacteriorhodopsin on tin oxide electrodes.

The transient photocurrent response from bacteriorhodopsin (bR) on tin oxide electrodes was strongly influenced by metal ions bound to bR molecules. The photocurrent polarity reversal pH, which corresponded to the pH value for the reversal of the proton release/uptake sequence in the bR photocycle, of cation-substituted purple membrane (PM) was shifted to lower pH with the increase in the cation affinities to carboxyl groups and a close correlation was noted between the two values. This suggests that the metal ion present in the extracellular region of a bR molecule modulates the pK(a) of prot

Potato purple top phytoplasma infection induces autophagy-associated lipid dynamics that support pathogen proliferation.

Phytoplasmas are unculturable, phloem-restricted bacterial pathogens responsible for devastating diseases in crops and ornamentals worldwide. Their mechanism for nutrient acquisition from host plants remains largely unknown. This study demonstrated that infection with potato purple top phytoplasma induced extensive remodeling of lipid metabolism in tomato plants, closely linked to autophagy activation. Western blot and confocal analyses revealed increased ATG8 lipidation and autophagosome formation at endoplasmic reticulum stress sites, alongside the redistribution of lipid droplets toward phy

Identification of the Biomarkers and Pathological Process of Heterotopic Ossification: Weighted Gene Co-Expression Network Analysis.

Heterotopic ossification (HO) is the formation of abnormal mature lamellar bone in extra-skeletal sites, including soft tissues and joints, which result in high rates of disability. The understanding of the mechanism of HO is insufficient. The aim of this study was to explore biomarkers and pathological processes in HO+ samples. The gene expression profile GSE94683 was downloaded from the Gene Expression Omnibus database. Sixteen samples from nine HO- and seven HO+ subjects were analyzed. After data preprocessing, 3,529 genes were obtained for weighted gene co-expression network analysis. High

Biotechnology of Anoxygenic Phototrophic Bacteria.

Anoxygenic phototrophic bacteria are a diverse collection of organisms that are defined by their ability to grow using energy from light without evolving oxygen. The dominant groups are purple sulfur bacteria, purple nonsulfur bacteria, green sulfur bacteria, and green and red filamentous anoxygenic phototrophic bacteria. They represent several bacterial phyla but they all have bacteriochlorophylls and carotenoids and photochemical reaction centers which generate ATP and cellular reductants used for CO2 fixation. They typically have an anaerobic lifestyle in the light, although some grow aerob

Dot enzyme immunosorbent assay for the serodiagnosis of typhoid fever.

A nitrocellulose membrane strip dotted with a specific 50 kDa outer membrane protein of Salmonella typhi was applied for the serodiagnosis of typhoid fever. Using horseradish peroxidase conjugated IgM and IgG antibodies with 4-chloronaphthol as substrate, antibodies in typhoid patients were clearly visualised as bluish purple dots while sera from patients with non-typhoid fevers gave negative results. The detection of specific IgM and IgG antibodies in typhoid patients suggest either recent or current infection. Combined with the high specificity, reliability and rapidity of the test, the dot
