Identification of Pyrus single nucleotide polymorphisms (SNPs) and evaluation for genetic mapping in European pear and interspecific Pyrus hybrids.

We have used new generation sequencing (NGS) technologies to identify single nucleotide polymorphism (SNP) markers from three European pear (Pyrus communis L.) cultivars and subsequently developed a subset of 1096 pear SNPs into high throughput markers by combining them with the set of 7692 apple SNPs on the IRSC apple Infinium® II 8K array. We then evaluated this apple and pear Infinium® II 9K SNP array for large-scale genotyping in pear across several species, using both pear and apple SNPs. The segregating populations employed for array validation included a segregating population of Europe

Fatty acid profile and in vitro biological properties of two Rosacea species (Pyrus glabra and Pyrus syriaca), grown as wild in Iran.

The high demands for the consumption of edible oils have caused scientists to struggle in assessing wild plants as a new source of seed oils. Therefore, in this study, the oil yield, fatty acid and tocopherol compositions, antioxidant and antibacterial activities of the oils obtained from Iran's two endemic plants (Pyrus glabra and Pyrus syriaca) were investigated. The obtained oil yields from the P. glabra and P. syriaca seeds were 33 ± 0.51 and 26 ± 0.28 w/w%, respectively. Oleic acid (C18:1) with the amount of 49.51 ± 1.05% was the major fatty acid in the P. glabra oil, while the main fatty

Molecular characterization of intergeneric hybrids between Malus and Pyrus.

Apple (Malus) and pear (Pyrus) are economically important fruit crops well known for their unique textures, flavours, and nutritional qualities. Both genera are characterised by a distinct pattern of secondary metabolites, which directly affect not only resistance to certain diseases, but also have significant impacts on the flavour and nutritional value of the fruit. The identical chromosome numbers, similar genome size, and their recent divergence date, together with DNA markers have shown that apple and pear genomes are highly co-linear. This study utilized comparative genomic approaches, i

Genome-wide analysis of the HDAC gene family in Pyrus betulifolia and functional validation of PcHDAC5 and PcHDT1.2 in Pyrus calleryana seed dormancy release.

Histone deacetylation, catalyzed by histone deacetylases (HDACs), is a core epigenetic regulatory mechanism in plants. However, no systematic characterization of the HDAC gene family has been reported in pear (Pyrus spp.), a globally important temperate fruit crop. In this study, we performed genome-wide identification and functional characterization of HDAC genes in pear, with a specific focus on their regulatory roles in seed dormancy release. We identified 22 PbHDAC genes from the genome of Pyrus betulifolia Bunge, which were classified into three conserved subfamilies. Evolutionary analysi

Integration of volatile and non-volatile metabolites and the transcriptome reveals the formation mechanisms of differential aroma compounds between Pyrus communis and Pyrus pyrifolia cultivars.

INTRODUCTION: Aroma compounds are important flavor components in pear fruit. Among cultivated pears, fruits from Pyrus communis (hereafter referred to as P. communis) cultivars are famous for their abundant aroma, while the fruits of most Pyrus pyrifolia (hereafter referred to as P. pyrifolia) cultivars lack aroma compounds. A comparative study on the formation of differential aroma compounds between the two species could provide a theoretical foundation for improving the aroma quality of P. pyrifolia cultivars. However, there is a lack of systematic research on this subject. METHODS: An anal

The draft genome sequence of European pear (Pyrus communis L. 'Bartlett').

We present a draft assembly of the genome of European pear (Pyrus communis) 'Bartlett'. Our assembly was developed employing second generation sequencing technology (Roche 454), from single-end, 2 kb, and 7 kb insert paired-end reads using Newbler (version 2.7). It contains 142,083 scaffolds greater than 499 bases (maximum scaffold length of 1.2 Mb) and covers a total of 577.3 Mb, representing most of the expected 600 Mb Pyrus genome. A total of 829,823 putative single nucleotide polymorphisms (SNPs) were detected using re-sequencing of 'Louise Bonne de Jersey' and 'Old Home'. A total of 2,279

Antioxidant response and quality of sunburn Beurré D'Anjou pears (Pyrus communis L.).

Sunburn is a physiological fruit disorder induced by exposure to excessive solar radiation. This disorder leads to significant losses in the yield of marketable fruits by negatively affecting quality parameters such as maturity and external color of the fruits. The purpose of this work was to characterize the physiological and biochemical aspects related to oxidative metabolism in Beurré D'Anjou pear fruit with different sunburn levels. Fruits were collected and classified into three sunburn levels at harvest: no sunburn (S0), mild sunburn (S1), and moderate sunburn (S2). On sunburned area, th

Seedless formation of a landrace 'Sanenashi' (Pyrus L.) collected from Northern Tohoku, Japan.

'Sanenashi' is a landrace of Pyrus ussuriensis var. aromatica (Iwateyamanashi) with seedless fruit originating from northern Tohoku, Japan. To determine the mechanism of seedless fruit formation, we compared the number of styles in the pistil, fruit, seed set and the pollen tube growth between 'Sanenashi' and the Japanese pear cultivar 'Kosui'. Morphological variations such as short or browning pistils were observed in 64.2% of 'Sanenashi' and 5.9% of 'Kosui'. The initial fruit set rate of 'Sanenashi' was 48.3% at 4 weeks after cross-pollination with pear, and there were no fruit sets with pea

Anti-Obesity Effects of Pear Extract (Pyrus pyrifolia): Clinical Implications and Therapeutic Potential.

Mitigating obesity is a pivotal strategy for addressing metabolic diseases, including cardiovascular diseases, liver diseases, and diabetes. Although pear extract exhibits various pharmacological effects in regenerative diseases, such as skin, nerve, and adipose tissue dysfunction, by eliminating excessive reactive oxygen species, its exact clinical role in obesity has not been well studied. In this study, we investigated the efficacy and safety of pear extract (UPhenonⓇ), a dietary ingredient extracted from unripe pear (Pyrus pyrifolia), in Korean participants with obesity. The participants u

Transcriptome Analysis of Pyrus betulaefolia Seedling Root Responses to Short-Term Potassium Deficiency.

Potassium (K) plays a crucial role in multiple physiological and developmental processes in plants. Its deficiency is a common abiotic stress that inhibits plant growth and reduces crop productivity. A better understanding of the mechanisms involved in plant responses to low K could help to improve the efficiency of K use in plants. However, such responses remain poorly characterized in fruit tree species such as pears (Pyrus sp). We analyzed the physiological and transcriptome responses of a commonly used pear rootstock, Pyrus betulaefolia, to K-deficiency stress (0 mM). Potassium deprivation

Fast loading ester fluorescent Ca2+ and pH indicators into pollen of Pyrus pyrifolia.

Loading of Ca(2+)-sensitive fluorescent probes into plant cells is an essential step to measure activities of free Ca(2+) ions in cytoplasm with a fluorescent imaging technique. Fluo-3 is one of the most suitable Ca(2+) indicators for CLSM. We loaded pollen with fluo-3/AM at three different temperatures. Fluo-3/AM was successfully loaded into pollen at both low (4°C) and high (37°C) temperatures. However, high loading temperature was best suited for pollen, because germination rate of pollen and growth of pollen tubes were relatively little impaired and loading time was shortened. Moreover, Ca
