Quillaja bark (soapbark)--induced asthma.

A 24-yr-old man developed sensitization to Quillaja bark (soapbark) dust at his work place. Within 3 mo of being employed in a factory processing Quallaja bark to produce saponin, he experienced asthma symptoms while handling the bark but only nasal symptoms on being exposed to the purified saponin. Bronchial provocation using Quillaja bark dust resulted in immediate bronchoconstriction as well as faintness, diffuse erythema, and hypotension. Radioallergosorbent test (RAST) using the pulverized crude Quillaja bark was markedly positive, and cross-reactivity between gum acacia and gum tragacant

Occupational immediate type allergy to soapnut and quillaja bark.

A 58-year-old non-atopic chemical worker complained about work-related asthma and rhinoconjunctivitis about 4 years after exposure to quillaja bark and soapnut. Bronchial hyperresponsiveness was demonstrated after withdrawal of medication for 12 hours. Skin prick tests with extracts from quillaja bark and soapnut from the workplace were positive, but ImmunoCAP was positive only with quillaja bark, probably due to the low protein content of the extract from soapnut. Sensitizations to quillaja bark and soapnut, but not to saponin were demonstrated by immunoblot. An inhalation test with a dosimet

The effects of graded levels of quillaja saponins on the productivity of commercial laying hens.

A study was conducted to determine the effects of feeding graded levels of quillaja saponins (Clarity Q®, Huvepharma, Antwerp, Belgium) on the performance and productivity of commercial layers. Three thousand H&N layer pullets were given one of 4 levels (0, 125, 250 and 500 mg/kg) of quillaja saponins in feed and their growth performance was monitored throughout a 5-to-15-week rearing period in a completely randomized design. At 16 weeks, pullets were moved to a laying facility where they continued to consume the same treatment feeds through 60 weeks of age. At 21 weeks, collection of egg prod

Protective vaccination against murine visceral leishmaniasis using aldehyde-containing Quillaja saponaria sapogenins.

The presence of aldehyde groups at C-23 and C-24 of the triterpen aglycon moiety was disclosed in 1H NMR spectra of both the Riedel de Haen saponin (R) (delta 9.336) and Quillaja saponaria QuilA saponin (delta 9.348). The sign of the C-28 acylated linked moiety (delta 176) was present in both saponins, while the delta 171 at C-28 (carboxy group) corresponding to the deacylated saponin, was only detected in the QuilA preparation, indicating 50% of hydrolysis of the ester moiety, probably due to the storage in aqueous solution. The normoterpen moiety was present in both saponins (signals at delt

Structure and immunological activity of QS-21 variant from Quillaja saponaria aerial biomass.

QS-21 is a triterpenoid saponin adjuvant component widely used in human vaccines, but its commercial supply is limited because it is sourced almost exclusively from the bark of mature Quillaja saponaria trees in Chile. In this study, we report the identification, isolation, as well as structural and immunological characterization of QS-21-Rhamnose (QS-21-Rha), a naturally occurring structural variant that predominates in the leaves and twigs of young Q. saponaria shrubs cultivated in California. Analytical profiling showed that QS-21-Rha represents more than 95% of QS-21 variants in the aerial

Quillaja brasiliensis saponin-based nanoparticulate adjuvants are capable of triggering early immune responses.

Commercially available saponins are extracted from Quillaja saponaria barks, being Quil A® the most widely used. Nanoparticulate immunostimulating complexes (ISCOMs or ISCOMATRIX) formulated with these, are able to stimulate strong humoral and cellular immune responses. Recently, we formulated novel ISCOMs replacing QuilA® by QB-90 (IQB-90), a Quillaja brasiliensis leaf-extracted saponin fraction, and reported that IQB-90 improved antigen uptake, and induced systemic and mucosal antibody production, and T-cell responses. However, its mechanism of action remains unclear. In this study we provid

ISCOM-like Nanoparticles Formulated with Quillaja brasiliensis Saponins Are Promising Adjuvants for Seasonal Influenza Vaccines.

Vaccination is the most effective public health intervention to prevent influenza infections, which are responsible for an important burden of respiratory illnesses and deaths each year. Currently, licensed influenza vaccines are mostly split inactivated, although in order to achieve higher efficacy rates, some influenza vaccines contain adjuvants. Although split-inactivated vaccines induce mostly humoral responses, tailoring mucosal and cellular immune responses is crucial for preventing influenza infections. Quillaja brasiliensis saponin-based adjuvants, including ISCOM-like nanoparticles fo

Temporal and stimulus-specific transcriptional responses of maize to a biostimulant mixture of quillaja extract and fulvic acid.

Biostimulant-induced responses in plants are dynamic, involving rapid signaling events followed by slower, downstream transcriptional and metabolic adjustments. To characterize the temporal molecular responses of maize to two biostimulant formulations, including fulvic acid (BLG) and mixture of fulvic acid and Quillaja extract (BLGQ), RNA sequencing was performed on leaf tissues collected at two time points representing the early (3 h) and later (27 h) transcriptional responses following biostimulant application. Differential gene expression, functional enrichment, and transcription factor ana

Immunization against leishmaniasis by PLGA nanospheres loaded with an experimental autoclaved Leishmania major (ALM) and Quillaja saponins.

Immune responses against the Leishmania antigens are not sufficient to protect against a leishmania challenge. Therefore these antigens need to be potentiated by various adjuvants and delivery systems. In this study, Poly (d,l-lactide-co-glycolide (PLGA) nanospheres as antigen delivery system and Quillaja saponins (QS) as immunoadjuvant have been used to enhance the immune response against autoclaved Leishmania major (ALM). PLGA nanospheres were prepared by a double-emulsion (W/O/W) technique. Particulate characteristics were studied by scanning electron microscopy and particle size analysis.

Quillaja brasiliensis saponins induce robust humoral and cellular responses in a bovine viral diarrhea virus vaccine in mice.

A saponin fraction extracted from Quillaja brasiliensis leaves (QB-90) and a semi-purified aqueous extract (AE) were evaluated as adjuvants in a bovine viral diarrhea virus (BVDV) vaccine in mice. Animals were immunized on days 0 and 14 with antigen plus either QB-90 or AE or an oil-adjuvanted vaccine. Two-weeks after boosting, antibodies were measured by ELISA; cellular immunity was evaluated by DTH, lymphoproliferation, cytokine release and single cell IFN-γ production. Serum anti-BVDV IgG, IgG1 and IgG2b were significantly increased in QB-90- and AE-adjuvanted vaccines. A robust DTH respons

Intranasal Delivery of Quillaja brasiliensis Saponin-Based Nanoadjuvants Improve Humoral Immune Response of Influenza Vaccine in Aged Mice.

Increasing the effectiveness of vaccines against respiratory viruses is particularly relevant for the elderly, since they are prone to develop serious infections due to comorbidities and the senescence of the immune system. The addition of saponin-based adjuvants is an interesting strategy to increase the effectiveness of vaccines. We have previously shown that ISCOM matrices from Q. brasiliensis (IMXQB) are a safe and potent adjuvant. In this study, we evaluated the use of IMXQB as an adjuvant for the seasonal trivalent influenza vaccine (TIV) in an aged mice model. Herein, we show that subcu

Enhanced performance of nanocomposite drilling fluids with synthesized TiO2 Quillaja saponin Cr nanoparticles and their impact on rheology and filtration.

This study addresses critical challenges in the formulation of advanced drilling fluids for high-pressure high-temperature (HPHT) environments by investigating TiO2@Quillaja Saponin (QS)/Cr nanocomposites as a multifunctional additive. The primary objective was to enhance rheological and filtration properties through optimized nanoparticle-polymer interactions. TiO2@QS/Cr was synthesized and incorporated into drilling fluids at varying concentrations (50-1000 ppm) to evaluate viscosity, yield point, and filtration behavior under simulated drilling conditions. Rheological experiments showed tha

Alternative inactivated poliovirus vaccines adjuvanted with Quillaja brasiliensis or Quil-a saponins are equally effective in inducing specific immune responses.

Inactivated polio vaccines (IPV) have an important role at the final stages of poliomyelitis eradication programs, reducing the risks associated with the use of attenuated polio vaccine (OPV). An affordable option to enhance vaccine immunogenicity and reduce costs of IPV may be the use of an effective and renewable adjuvant. In the present study, the adjuvant activity of aqueous extract (AE) and saponin fraction QB-90 from Quillaja brasiliensis using poliovirus antigen as model were analyzed and compared to a preparation adjuvanted with Quil-A, a well-known saponin-based commercial adjuvant. The characterization of a rhamnosylated triterpenic saponin from Quillaja saponaria Mol. reveals novel adjuvant, antibacterial, antiviral, antifungal and antiproliferative properties.

Quillaja: is an abundant source of triterpenic saponins, glycosides with immunostimulant properties, currently employed as vaccine adjuvants. Although current research and commercial applications have focused on the most abundant saponins in Quillaja extracts (QS-7, QS-17, QS-18 and QS-21), the vast chemical variety in Quillaja saponins remains largely unexplored. The discovery of new saponins suited as vaccine adjuvants would open the door to a wider supply of adjuvants in the future. This paper describes the isolation and chemical characterization of Saponin FR, a new saponin fraction purifi

Leaf saponins of Quillaja brasiliensis enhance long-term specific immune responses and promote dose-sparing effect in BVDV experimental vaccines.

Saponin-based adjuvants are promising adjuvants that enhance both humoral and T-cell-mediated immunity. One of the most used natural products as vaccine adjuvants are Quillaja saponaria bark saponins and its fraction named Quil A®. Despite that, its use has been restricted for human use due to safety issues. As an alternative, our group has been studying the congener species Quillaja brasiliensis saponins and its performance as vaccine adjuvants, which have shown to trigger humoral and cellular immune responses comparable to Quil A® but with milder side effects. Here, we studied a semi purifie

Inclusion of Quillaja Saponin Clarity Q Manages Growth Performance, Immune Response, and Nutrient Transport of Broilers during Subclinical Necrotic Enteritis.

Necrotic enteritis (NE) is an intestinal disease that results in poor performance, inefficient nutrient absorption, and has a devastating economic impact on poultry production. This study evaluated the effects of a saponin-based product (Clarity Q, CQ) during an NE challenge. A total of 1200 male chicks were randomly assigned to four dietary treatments (10 pens/treatment; 30 birds/pen): treatment 1 (NC), a non-medicated corn-soybean basal diet; treatment 2 (PC), NC + 50 g/metric ton (MT) of bacitracin methylene disalicylate (BMD); and treatments 3 (CQ15) and 4 (CQ30), NC + 15 and 30 g/MT, resp

A rabies vaccine adjuvanted with saponins from leaves of the soap tree (Quillaja brasiliensis) induces specific immune responses and protects against lethal challenge.

Quillaja brasiliensis (Quillajaceae) is a saponin producing species native from southern Brazil and Uruguay. Its saponins are remarkably similar to those of Q. saponaria, which provides most of the saponins used as immunoadjuvants in vaccines. The immunostimulating capacities of aqueous extract (AE) and purified saponin fraction (QB-90) obtained from leaves of Q. brasiliensis were favorably comparable to those of a commercial saponin-based adjuvant preparation (Quil-A) in experimental vaccines against bovine herpesvirus type 1 and 5, poliovirus and bovine viral diarrhea virus in mice model.
