2,3,8-Trisubstituted Quinolines with Antimalarial Activity.

Combination therapy drugs are considered a fundamental way to control malaria as it mimimizes the risk of emergence of resistance to the individual partner drugs. Consequently, this type of therapy constitutes a driving force for the discovery of new drugs with different modes of action, since this will provide options for combining different drugs to achieve the optimum antimalarial treatment. In this context, a 2,3,8-trisubstitued quinoline compound was found in a high throughput screen (HTS) to show an excellent inhibition of P. falciparum NF54 (IC50 = 22 nM) and low cytotoxicity. We perfor

Copper-Catalyzed Selective 1,2-Reduction of Quinolines.

Selective reduction of heteroaromatic compounds to partially hydrogenated, dearomatized molecules is a tantalizing task. A well-defined, air stable, and pyridine-based Cu(II) NNN pincer complex is developed, which catalyzes selective 1,2-reduction of quinolines. The unstable 1,2-hydrogenated quinolines are transformed into the corresponding amides and isolated in good yields. Mechanistic studies confirmed that the reaction proceeds via molecular species and the formation of an unexpected copper bimetallic intermediate, which facilitates the catalysis through metal-ligand cooperation.

Quinolines interfere with heme-mediated activation of artemisinins.

Artemisinin-based combination therapies (ACTs) remain the mainstay of treatment for Plasmodium falciparum malaria, despite reports of ACT treatment failure. ACTs consist of an artemisinin and a longer-lived partner drug, which is often a quinoline. Given that heme is central to the mechanism of action of artemisinins and some quinolines, we hypothesized that these antimalarials would exhibit strong drug-drug interactions. Previous studies using standard 48 h or 72 h assays identified additive to mildly antagonistic interactions between artemisinins and quinolines. Here, we sought to re-evaluat

Antiprotozoal Activities of Tetrazole-quinolines with Aminopiperidine Linker.

BACKGROUND: Human African Trypanosomiasis (HAT, sleeping sickness) and Malaria both are insect vectored tropical diseases. Only a couple of drugs is able to cure HAT, but all of them are toxic, prone to resistance and require parenteral administration. Malaria is responsible for high morbidity and mortality in humans. It is one of the global killers of children. Wide-spread drug resistance against traditional therapeutics which were once highly effective makes them almost useless. Therefore new drugs against both diseases are urgently needed. OBJECTIVE: Recently, we reported the synthesis and

Asymmetric Allylic Dearomatization of Quinolines via Dynamic Ligand Exchange.

The strong coordination of quinolines to transition metals typically interferes with chiral catalysts, restricting their use in asymmetric synthesis. Here, we harness this challenging property as a stereocontrol element through the integration of dynamic ligand exchange with designed hydrogen-bonding interactions between a chiral phosphine ligand and the quinoline substrate. This creates a confined microenvironment that stabilizes the chiral palladium complex and directs a highly enantioselective allylic amination/alkynylation cascade. This approach enables the first asymmetric allylic dearoma

Single-Atom Fe-Catalyzed Acceptorless Dehydrogenative Coupling to Quinolines.

A single-atom iron catalyst was found to exhibit exceptional reactivity in acceptorless dehydrogenative coupling for quinoline synthesis, outperforming known homogeneous and nanocatalyst systems. Detailed characterizations, including aberration-corrected HAADF-STEM, XANES, and EXAFS, jointly confirmed the presence of atomically dispersed iron centers. Various functionalized quinolines were efficiently synthesized from different amino alcohols and a range of ketones or alcohols. The iron single-atom catalyst achieved a turnover number (TON) of up to 105, far exceeding the results of current hom

Multi-Substituted Quinolines as HIV-1 Integrase Allosteric Inhibitors.

Allosteric HIV-1 integrase (IN) inhibitors, or ALLINIs, are a new class of antiviral agents that bind at the dimer interface of the IN, away from the enzymatic catalytic site and block viral replication by triggering an aberrant multimerization of the viral enzyme. To further our understanding of the important binding features of multi-substituted quinoline-based ALLINIs, we have examined the IN multimerization and antiviral properties of substitution patterns at the 6 or 8 position. We found that the binding properties of these ALLINIs are negatively impacted by the presence of bulky substitu

Stereoselective Reduction of Alkynes: Synthesis of 4-Organoselenyl Quinolines.

This study describes the reaction of 2-amino arylalkynyl ketones with organoselenolates to form (Z)-vinyl selenides, which lead to 4-organoselenyl quinolines via an intramolecular condensation. Using the optimized reaction conditions, the generality of this cyclization was studied with various arylalkynyl ketones and diorganyl diselenides. The study of the reaction mechanisms led to the isolation and identification of a vinyl selenide, which was the key intermediate for this cyclization. To expand the structural diversity and to demonstrate the applicability of the 4-organoselenyl quinolines p

Bioisosteric ferrocenyl-containing quinolines with antiplasmodial and antitrichomonal properties.

Bioisosteric ferrocenyl-containing quinolines and ferrocenylamines containing organosilanes and their carbon analogues, were prepared and fully characterised. The molecular structures of two ferrocenyl-containing quinolines, determined using single-crystal X-ray diffraction, revealed that the compounds crystallise in a folded conformation. The compounds were screened for their antiplasmodial activity against the chloroquine-sensitive (NF54) and CQ-resistant (Dd2) strains of P. falciparum, as well as for their cytotoxicity against Chinese Hamster Ovarian (CHO) cells. The ferrocenyl-containing q

Quinolines: the role of substitution site in antileishmanial activity.

Leishmaniasis is one of the most widespread parasitic diseases in the world, primarily affecting the poorest and most vulnerable populations. The development of new therapeutic agents that are more efficient, safe, and selective remains a challenge. The quinoline framework emerges as a privileged scaffold for this purpose. This mini-review comprehensively analyses advancements from the last two decades on 2-, 3-, 6-, and 8-substituted quinolines, as well as polysubstituted analogues, as potential antileishmanial agents, focusing on how the position and nature of substituents influence their ac

Chemoselective Heterogeneous Hydrogenation of Sulfur Containing Quinolines under Mild Conditions.

Sulfur, alongside oxygen and nitrogen, holds a prominent position as one of the key heteroatoms in nature and medicinal chemistry. Its significance stems from its ability to adopt different oxidation states, rendering it valuable as both a polarity handle and a hydrogen bond donor/acceptor. Nevertheless, the poisonous nature of its free electron pairs makes sulfur containing substrates inaccessible for many catalytic protocols. Strong and (at low temperatures) irreversible chemisorption to the catalyst's surface is in particular detrimental for heterogeneous catalysts, possessing only few cata

Quinolines attenuate PAF-induced pulmonary pressor responses and edema formation.

In the present study we have investigated the mechanisms of pulmonary edema caused by platelet-activating factor (PAF) in isolated rat lungs as well as in mice in vivo. In blood-free perfused and ventilated rat lungs, PAF increased lung weight by 0.59 +/- 0.18 g. The cyclooxygenase inhibitor aspirin (500 microM) blocked this response by one-third, and the quinolines quinine (330 microM), quinidine (100 microM), and chloroquine (100 microM) by two-thirds. Lipoxygenase inhibition (10 microM AA861) alone or in combination with thromboxane receptor antagonism (10 microM SQ29548) had no effect on P
