Iron-Catalyzed C-H Arylphosphorylation of Quinoxalines.

A one-pot strategy for iron-catalyzed C2,3-H arylphosphorylation of electron-deficient quinoxalines with phosphines and aryl compounds is reported. The proposed method features the use of non-noble metal catalysts, the capacity of utilizing multiple aryl compounds as substrates, the simultaneous formation of C-P and C-C bonds in one pot, the simplicity of its operation, the mildness of the reaction conditions, and its compatibility with a wide range of substrates. Moreover, it offers a practical route for direct access to 2-aryl-3-phosphino N-heteroarenes, a class of potential cyclometalated C

Quinoxaline and quinoxaline-1,4-di-N-oxides: An emerging class of antimycobacterials.

Tuberculosis (TB) is a highly dreaded, infectious, chronic, airborne disease affecting more than two million people all around the world, with more than eight million cases every calendar year. TB is the second leading infectious cause of death after HIV/AIDS. Over the past few decades, numerous efforts have been undertaken to develop new anti-TB agents. The current frontline therapy for TB consists of administering three or more different drugs (usually isoniazid, rifampin, pyrazinamide, and ethambutol) over an extended period of time. But these drugs will take 6-12 months to cure TB, along w

Electrochemical synthesis of hydroxyalkylated quinoxalines via C-H functionalization.

An efficient electrochemical C-H hydroxyalkylation of quinoxalines with aldehydes and ketones has been developed in an undivided cell. A variety of aliphatic and aromatic aldehydes as well as ketones have been examined, affording the corresponding hydroxyalkylated quinoxalines in good yields. Mechanism investigations indicate that the hydroxyalkylation is achieved through cathodic reduction to produce ketyl radicals and heteroaryl radical anions, followed by radical-radical cross-coupling.

Regioselective Synthesis of Trisubstituted Quinoxalines Mediated by Hypervalent Iodine Reagents.

A facile and regioselective synthesis of quinoxalines, an important motif in medicinal chemistry and materials sciences, was developed. Despite their prospective utility, the regioselective preparation of trisubstituted quinoxalines has not been previously established. In the reported system, hypervalent iodine reagents catalyzed the annulation between α-iminoethanones and o-phenylenediamines in a chemo/regioselective manner to afford trisubstituted quinoxalines. Excellent regioselectivities (6:1 to 1:0) were achieved using [bis(trifluoroacetoxy)iodo]benzene and [bis(trifluoroacetoxy)iodo]pent

Emergence of pyrido quinoxalines as new family of antimalarial agents.

A series of novel N-alkyl dihydro pyrido quinoxaline derivatives were synthesized using Gould-Jacobs reaction and evaluated their antimalarial activity in vitro against chloroquine sensitive (3D7) and drug resistant (Dd2) strains of Plasmodium falciparum. Among the compounds tested, 10 compounds were more potent than their structural standard analog ciprofloxacin, including 2 derivatives 5e and 5h, which showed 3.3-7.4 times more potency than ciprofloxacin against both the parasite strains. The results are encouraging and a lead molecule may emerge which is useful alone or in combination thera

Current status of quinoxaline and quinoxaline 1,4-di-N-oxides derivatives as potential antiparasitic agents.

Parasitic diseases are a public health problem, especially in developing countries where millions of people are affected every year. Current treatments have several drawbacks: emerging resistance to the existing drugs, lack of efficacy, and toxic side effects. Therefore, new antiparasitic drugs are urgently needed to treat and control diseases that affect human health, such as malaria, Chagas disease, leishmaniasis, amebiasis, giardiasis schistosomiasis, and filariasis, among others. Quinoxaline is a compound containing a benzene ring and a pyrazine ring. The oxidation of both pyrazine ring ni

Design, Synthesis, and Pharmacological Characterization of Substituted Quinoxalines as NOP Receptor Ligands.

INTRODUCTION: The goal of this work was the discovery of moderate-efficacy, selective, partial agonists of the nociceptin/orphanin-FQ peptide (NOP) receptor. METHODS: A novel collection of quinoxaline-containing molecules was designed, synthesized, and assayed against NOP and mu-opioid receptors in vitro to establish the relationship between structural modification and pharmacological activity. The lead molecule identified was further assessed in vivo to determine brain penetration, analgesic effects in rodent models of pain, and motor deficits. RESULTS: This SAR approach revealed that the p

Antiplasmodial 2-thiophenoxy-3-trichloromethyl quinoxalines target the apicoplast of Plasmodium falciparum.

The identification of a plant-like Achille's Heel relict, i.e. the apicoplast, that is essential for Plasmodium spp., the causative agent of malaria lead to an attractive drug target for new antimalarials with original mechanism of action. Although it is not photosynthetic, the apicoplast retains several anabolic pathways that are indispensable for the parasite. Based on previously identified antiplasmodial hit-molecules belonging to the 2-trichloromethylquinazoline and 3-trichloromethylquinoxaline series, we report herein an antiplasmodial Structure-Activity Relationships (SAR) study at posit

Photoinduced Synthesis of Bisphosphinated Quinoxalines via Radical Cyclization of o-Diisocyanoarenes with Diphosphines.

The cycloaddition reaction of o-diisocyanoarenes with interelement compounds under light is a very important reaction system to clarify whether this reaction proceeds by radical cyclization or by aza-Bergman cyclization. In this study, a series of diphosphines with phosphorus-phosphorus single bonds were selected as interelement compounds, and their cycloaddition reactions with o-diisocyanoarenes under light were investigated in detail to achieve a novel photoinduced synthesis of bisphosphinated quinoxalines via the radical cyclization pathway. In addition, the photoinduced reaction of diphosp

Quinoxaline derivatives as potential antitrypanosomal and antileishmanial agents.

Continuous efforts have been made to discover new drugs for the treatment of Chagas' disease, human African trypanosomiasis, and leishmaniasis. We have previously reported the synthesis and antileishmanial and antitrypanosomal (Y strain) properties of 2,3-disubstituted quinoxalines. Considering their promising antiparasitic potential, the present study was conducted to expand our search and take advantage of high-throughput assays to investigate the effects of quinoxaline derivatives against Leishmania donovani, Trypanosoma brucei, and Trypanosoma cruzi (Tulahuen strain). These compounds were

Quinoxaline derivatives: novel and selective butyrylcholinesterase inhibitors.

Alzheimer's disease (AD) is a progressive brain disorder which occurs due to lower levels of acetylcholine (ACh) neurotransmitters, and results in a gradual decline in memory and other cognitive processes. Acetycholinesterase (AChE) and butyrylcholinesterase (BChE) are considered to be primary regulators of the ACh levels in the brain. Evidence shows that AChE activity decreases in AD, while activity of BChE does not change or even elevate in advanced AD, which suggests a key involvement of BChE in ACh hydrolysis during AD symptoms. Therefore, inhibiting the activity of BChE may be an effectiv

Design, synthesis, and anti-colorectal cancer activity of quinoxalines with improved drug-like properties.

Colorectal cancer (CRC) is strongly associated with aberrant activation of the PI3K/Akt/mTOR pathway, in which PI3Kα represents a clinically validated therapeutic target. Herein, we report the rational design, synthesis, and biological evaluation of quinoxaline derivatives as potent PI3Kα selective inhibitors derived from the lead compound 18a, which was developed in our previous study and suffered from poor drug-like properties. Structure-guided modification led to the identification of compound 8b, which exhibited remarkably improved solubility and oral bioavailability, and it showed nano-mo
