Osteopontin is induced by nitric oxide in RAW 264.7 cells.

Nitric oxide (NO) produced by macrophages is thought to contribute to various pathological conditions. Osteopontin (OPN) is a phosphorylated glycoprotein produced principally by macrophages. OPN inhibits inducible nitric oxide synthase (iNOS), which generates large amounts of NO production. However, the relationship between NO and endogenous OPN in activated macrophages has not yet been elucidated. We therefore examined expression of endogenous iNOS and OPN in a murine macrophage cell line, RAW 264.7 cells, by treating the cells with lipopolysaccharide (LPS) and interferon-gamma (IFN-gamma). T

Thiram modulates pro-inflammatory mediators in RAW 264.7 murine macrophage cells.

Thiram (TMTD) is a widely used dithiocarbamate pesticide and fungicide and is one of potent contact allergens. In the light of known properties, thiram is also considered to be used as an inhibitor of inflammation. To investigate whether known pro-oxidative properties of thiram might be involved in immunogenic mechanisms, we carried out an in vitro study aimed at analysis of reactive oxygen species (ROS) generation, activation of NF-κB, expression of iNOS and COX-2, production of NO, PGE2 and IL-1β in murine macrophage cells (RAW 264.7). The cells were treated by thiram alone (0.5 µg/mL; 2 μM

Inhibitory effects of spermidine on lipopolysaccharide-induced inflammation in RAW 264.7 cells.

Spermidine (SPD) is a naturally occurring polyamine with anti-inflammatory and antioxidant properties. Given the pivotal role of macrophages in inflammatory pathogenesis (such as rheumatoid arthritis and autoimmune encephalomyelitis), the present study hypothesized that SPD exerts its anti-inflammatory effects by suppressing macrophage polarization. Briefly, RAW 264.7 cells were activated using lipopolysaccharide (LPS) and were treated with a vehicle control (complete DMEM) or varying SPD concentrations. The ratio of M1 and M2 macrophages in each group was determined using flow cytometry. Infl

Acute transcriptomic changes in murine RAW 264.7 cells following pseudorabies virus infection.

Next-generation sequencing enables the evaluation of gene expression changes resulting from virus-host interactions at the RNA level. Pseudorabies virus (PRV) causes substantial economic loss in the swine industry. Recent research has revealed that PRV can be transmitted to and infect humans as well. To identify physiopathological and pathological responses post-PRV infection, we characterized transcriptomic changes in the murine RAW 264.7 cell line over the course of 36 h. In total, 156, 153, and 190 differentially expressed genes were identified at 2 h, 12 h, and 36 h, respectively. Seven di

ERK inhibits osteoclast differentiation in RAW 264.7 cells through the osteoprotegerin-mediated autophagy.

Osteoclasts (OCs) are bone-resorbing cells derived from the monocyte/macrophage lineage. The extracellular signal-regulated kinase (ERK) pathway controls cellular responses such as proliferation, differentiation, and survival, including those of OCs. In the present study, ERK inhibitors reduced the proliferation of bone marrow-derived macrophages (BMMs) and RAW 264.7 cells. However, ERK inhibitors decreased OC differentiation in BMMs but increased it in RAW 264.7 cells. ERK downregulation using small interfering RNA transfection also increased the OC differentiation and the expression of recep

Engineered nanotopography on electrospun PLLA microfibers modifies RAW 264.7 cell response.

In this study, we created a new method of electrospinning capable of controlling the surface structure of individual fibers (fiber nanotopography). The nanotopographical features were created by a phase separation in the fibers as they formed. To control the phase separation, a nonsolvent (a chemical insoluble with the polymer) was added to an electrospinning solution containing poly-l-lactic acid (PLLA) and chloroform. The nanotopography of electrospun fibers in the PLLA/chloroform solution was smooth. However, adding a small weight (<2% of total solution) of a single nonsolvent (water, ethan

Anti-inflammatory effect of polydeoxyribonucleotide on zoledronic acid-pretreated and lipopolysaccharide-stimulated RAW 264.7 cells.

Bisphosphonates are generally used as therapeutic agents for bone diseases. However, previous reports on bisphosphonates-related osteonecrosis of the jaw (BRONJ) demonstrated that inflammation triggers and worsens the disease. Recently, polydeoxynucleotide (PDRN), an A2A receptor agonist, has been suggested for the treatment of various diseases and broadly studied for its anti-inflammatory effect. The present study aimed to measure the effect of PDRN on macrophage cells treated with zoledronic acid (ZA) and lipopolysaccharide (LPS). Macrophage cells were cultured with ZA for 24 h, following wh

Dissociated ROS production and ceramide generation in sulfasalazine-induced cell death in Raw 264.7 cells.

Sulfasalazine (SSZ) is a drug used in inflammatory bowel disease, whose precise mechanism of action remains to be clarified. Here, we report that incubation of Raw 264.7 cells with SSZ but not salicylates [acetylsalicylic acid (ASA), 4-aminosalicylic acid (4-ASA), and 5-ASA] causes a mixed apoptotic and necrotic form of cell death. In contrast to its metabolites, sulfapyridine and 5-ASA, SSZ exposure in Raw 264.7 cells resulted in a threefold increase in ceramide generation, as well as a robust production of reactive oxygen species (ROS). However, inhibition of ceramide production by fumonisin

Citosol (thiamylal sodium) triggers apoptosis and affects gene expressions of murine leukemia RAW 264.7 cells.

Citosol (thiamylal sodium) is one of generally used anesthetic-sedative agents for clinical patients, and it has not been reported to show induction of cytotoxic effects in cancer cells, especially in mice leukemia RAW 264.7 cells in vitro. In the present study, we investigated the cytotoxic effects of citosol on mice leukemic RAW 264.7 cells, including the effects on protein and gene expression levels which are determined by Western blotting and DNA microarray methods, respectively. Results indicated that citosol induced cell morphological changes, cytotoxic effect, and induction of apoptosis

Targeted metabolomic profiling of RAW 264.7 cells infected with Brucella canis identifies time-dependent metabolic changes.

Canine brucellosis caused by Brucella canis is gaining public-health relevance as companion-dog ownership and close human-animal contact increase. Yet mechanistic insight into B. canis host responses remains limited at the pathway level. To address this gap, we profiled infection-associated metabolic responses in RAW 264.7 cells across multiple post-infection time points using targeted metabolomics (CE-MS). We observed a coherent, time-ordered pattern with the largest shifts at 24 h: arginine, hydroxyproline, choline, cysteine, β-alanine, and glycerol-3-phosphate showed pronounced decreases, a

Synergistic Activation of NO and Cytokine Production by TLR3 With TLR5 or TLR7 Agonists in RAW 264.7 Cells.

Macrophages are key components of the innate immune system, recognizing pathogen-associated molecular patterns (PAMPs) via Toll-like receptors (TLRs) to initiate immune responses. This study investigated the individual and combinatorial effects of TLR3 Poly(I:C), TLR5 (Flagellin), and TLR7 (Imiquimod) ligands on nitric oxide (NO) production and pro-inflammatory cytokine expression in RAW 264.7 mouse macrophage cells. Our results demonstrate that all three individual TLR agonists induced NO production and cytokine expression. Notably, co-stimulation with Poly(I:C) and imiquimod led to a signifi

Riboflavin deprivation inhibits macrophage viability and activity - a study on the RAW 264.7 cell line.

Riboflavin, or vitamin B2, as a precursor of the coenzymes FAD and FMN, has an indirect influence on many metabolic processes and determines the proper functioning of several systems, including the immune system. In the human population, plasma riboflavin concentration varies from 3·1 nM (in a moderate deficiency, e.g. in pregnant women) to 10·4 nM (in healthy adults) and 300 nM (in cases of riboflavin supplementation). The purpose of the present study was to investigate the effects of riboflavin concentration on the activity and viability of macrophages, i.e. on one of the immunocompetent cel
