[Serological characteristic of lipopolysaccharides of Ralstonia solanacearum].

By immunochemical investigations of eight strains of Ralstonia solanacearum six strains were attributed to four serogroups. Two of them are formed by pairs of R. solanacearum strains 4 and 526; 758 and 7954; two others are represented by single strains--TX1 Ta TS3, correspondingly. Antigenic structure of R. solanacearum 7954 O-polysaccharide unites antigenic epitopes of R. solanacearum strains 4, 35, 526, 749, however the absence of cross-reactivity does not permit uniting them into the same group. The latter, and also the fact that the antiserum to R. solanacearum 749 in the reaction with LPS

Diversity of Ralstonia solanacearum Infecting Eggplant in the Philippines.

ABSTRACT The diversity of Ralstonia solanacearum strains isolated from eggplant (Solanum melongena) grown in five provinces of the Philippine island group of Luzon was assessed using a recently described hierarchical system. All strains keyed to race 1, biovar 3 or 4. Phylotype-specific multiplex polymerase chain reaction (PCR) indicated that, like most other strains of Asian origin, all the strains in our Philippine collection belong to phylotype I. Taxometric and phylogenetic analyses of partial endoglucanase gene sequences of strains from this collection and those previously deposited into

Characterization of filamentous bacteriophage PE226 infecting Ralstonia solanacearum strains.

AIMS: The aim of this study was to isolate and characterize new bacteriophages that infect a wide range of plant pathogenic Ralstonia solanacearum strains. METHODS AND RESULTS: Fifteen bacteriophages were isolated from pepper, tomato and tobacco plant rhizospheres infected with R. solanacearum. A host specificity analysis of the isolated phages using nine strains of R. solanacearum indicated great phage diversity in a single soil. Two phages, PE226 and TM227, showed clear plaques on all nine bacterial hosts tested and were virtually identical in morphology and genome. PE226, an Inovirus, is a

Deoxymikanolide adversely altered physiology and ultrastructure of Ralstonia solanacearum.

Deoxymikanolide (DEO) was isolated from Mikania micrantha Bunge and identified as a novel antibacterial compound previously. However, the mode of antimicrobial mechanism of DEO was not clear but hypothesized to affect the morphology and physiology of Ralstonia solanacearum cells. In this study, we confirmed our hypothesis via transmission electron microscopy (TEM) observation and comprehensive physiological analyses, including electric conductivity, glycan and phosphorus metabolism, activities of antioxidant enzymes (catalase, peroxidase, and superoxide dismutase), intrabacterial reactive oxyg

Protein O-linked glycosylation in the plant pathogen Ralstonia solanacearum.

Ralstonia solanacearum is one of the most lethal phytopathogens in the world. Due to its broad host range, it can cause wilting disease in many plant species of economic interest. In this work, we identified the O-oligosaccharyltransferase (O-OTase) responsible for protein O-glycosylation in R. solanacearum. An analysis of the glycoproteome revealed that 20 proteins, including type IV pilins are substrates of this general glycosylation system. Although multiple glycan forms were identified, the majority of the glycopeptides were modified with a pentasaccharide composed of HexNAc-(Pen)-dHex(3),

Ralstonia solanacearum AmpD is required for wild-type bacterial wilt virulence.

Abstract A gene resembling enterobacterial ampD was identified in the bacterial wilt pathogen, Ralstonia solanacearum. The gene lies 13 bp 3' of pehSR, a two-component positive regulator of virulence factors such as plant cell wall-degrading polygalacturonases and bacterial motility. AmpD, an N-acetylmuramyl-l-alanine amidase, degrades and recycles bacterial cell wall components and also plays a role in the induction of beta-lactamase, which confers ampicillin resistance. AmpD is probably not involved in beta-lactamase regulation in R. solanacearum, because the species produces no detectable b

Metabolic Analysis Reveals Alkaloid-Mediated Resistance of Tobacco against Ralstonia solanacearum.

Tobacco (Nicotiana tabacum L.) is a globally important industrial crop, yet its productivity is severely threatened by bacterial wilt caused by R. solanacearum. In this study, integrative metabolomics and transcriptomics profiling of tobacco plants infected with R. solanacearum identified 571 differentially accumulated metabolites (DAMs), peaking at 96 h postinoculation (hpi). Among 88 alkaloid DAMs, six plumerane alkaloids (indole, indole-3-acetic acid (IAA), 3-indoleacetonitrile (IAN), methoxyindoleacetic acid (MIAA), N-acetyl-5-hydroxytryptamine, and N-acetylisatin) showed the most pronounc

4-Aminobenzoic acid attenuates Ralstonia solanacearum virulence by suppressing quorum sensing system.

Quorum sensing (QS) is widely involved in the regulation of the biological functions and pathogenic processes of bacteria in a cell density-dependent manner. The anthranilic acid signal is directly sensed by the receptor RaaR, a transcription regulator, to control the expression level of downstream target genes in Ralstonia solanacearum, thus affecting the QS signals generation and the bacterial virulence. Here, we reported that 4-Aminobenzoic acid (PABA), a structural analogue of anthranilic acid, not only significantly inhibited the production of extracellular polysaccharides (EPS), biofilm

Pleiotropic Phenotypes of the Tomato diageotropica Mutant Enable Resistance to Ralstonia solanacearum.

Quantitative disease resistance (QDR) is the most common form of disease resistance in crops, but it is challenging to understand at the cellular level due to the involvement of many genes and biological processes. Ralstonia solanacearum, the causal agent of bacterial wilt disease, is a destructive plant pathogen of Solanaceous species that is best controlled by quantitatively resistant varieties, but few QDR genes are known. We previously found that a tomato auxin pathway mutant known as diageotropica (dgt) has enhanced resistance to R. solanacearum. Here, we show that, as with wild-type quan

Effect and mechanism of NaHS on tobacco bacterial wilt caused by Ralstonia solanacearum.

Since its discovery as a third unique gaseous signal molecule, hydrogen sulfide (H2S) has been extensively employed to resist stress and control pathogens. Nevertheless, whether H2S can prevent tobacco bacterial wilt is unknown yet. We evaluated the impacts of the H2S donor sodium hydrosulfide (NaHS) on the antibacterial activity, morphology, biofilm, and transcriptome of R. solanacearum to understand the effect and mechanism of NaHS on tobacco bacterial wilt. In vitro, NaHS significantly inhibited the growth of Ralstonia solanacearum and obviously altered its cell morphology. Additionally, Na

Identification of Xanthones from the Mangosteen Pericarp that Inhibit the Growth of Ralstonia solanacearum.

Bacterial wilt caused by Ralstonia solanacearum is one of the most destructive bacterial diseases in agriculture. There is no effective control method, although chemical pesticides are used to prevent this disease, but they may lead to serious problems of environmental pollution. Natural products from plants can be rich and environmentally friendly sources for a broad spectrum biological control of bacteria. This study focuses on the pericarp of mangosteen (Garcinia mangostana) using bioactivity-guided analysis of different fractions and liquid chromatography-mass spectrometry combined with mu

Ralstonia solanacearum Alters Root Developmental Programmes in Auxin-Dependent and -Independent Manners.

Microbial pathogens and other parasites can modify the development of their hosts, either as a target or a side effect of their virulence activities. The plant-pathogenic bacterium Ralstonia solanacearum, causal agent of the devastating bacterial wilt disease, is a soilborne microbe that invades host plants through their roots and later proliferates in xylem vessels. In this work, we studied the early stages of R. solanacearum infection in the model plant Arabidopsis thaliana, using an in vitro infection system. In addition to the previously reported inhibition of primary root length and incre

Transcriptomics and overexpression analyses reveal StERF87 confers resistance to Ralstonia solanacearum in potato.

StERF87 directly activates StPR1a via GCC-box binding and integrates SA/ET signaling to enhance bacterial wilt resistance. Potato (Solanum tuberosum) is an important food crop worldwide, yet its yield is severely constrained by bacterial wilt caused by Ralstonia solanacearum. We used RNA-Seq to study gene expression in 'Z1076-1' at 0, 1, and 2 days post-inoculation (dpi) with R. solanacearum (10⁶ CFU mL⁻1). We identified 6663 differentially expressed genes at 1 dpi and 7390 at 2 dpi. Calcium signaling and MAPK cascade genes were upregulated at 1 dpi. PR protein and ROS-related genes showed str

Targeting flagellar-mediated virulence: The lipopeptide PW-1 suppresses pathogenicity in Ralstonia solanacearum.

Bacterial wilt caused by Ralstonia solanacearum is a devastating soil-borne disease, calling for novel green control strategies. This study reports that the natural lipopeptide PW-1 controls this disease through an "anti-virulence" mechanism without inhibiting pathogen growth. Phenotypic analysis showed that PW-1 specifically impaired bacterial motility. Transcriptome sequencing revealed that PW-1 coordinately downregulated flagellar biosynthesis-related genes, and transmission electron microscopy confirmed the loss of flagellar filaments in PW-1-treated cells. Using GFP-tagged bacteria, we de

Metabolic adaptation of Ralstonia solanacearum during plant infection: a methionine biosynthesis case study.

MetE and MetH are two distinct enzymes that catalyze a similar biochemical reaction during the last step of methionine biosynthesis, MetH being a cobalamin-dependent enzyme whereas MetE activity is cobalamin-independent. In this work, we show that the last step of methionine synthesis in the plant pathogen Ralstonia solanacearum is under the transcriptional control of the master pathogenicity regulator HrpG. This control is exerted essentially on metE expression through the intermediate regulator MetR. Expression of metE is strongly and specifically induced in the presence of plant cells in a

A Ralstonia solanacearum effector RipAD perturbs aquaporin NbPIP oligomeric status to suppress plant immunity.

Bacterial wilt caused by Ralstonia solanacearum is a devastating disease that affects a wide range of crops, including tobacco. The pathogen promotes infection by secreting type III effector (T3E) proteins into host plants to facilitate pathogenicity; however, the molecular mechanisms for many of these effectors remain elusive. Here, we characterize RipAD, a conserved T3E that significantly contributes to R. solanacearum virulence in Nicotiana tabacum and Nicotiana benthamiana. We demonstrate that RipAD dampens early immune responses, including pathogen-associated molecular pattern-triggered r

Fabrication of pH-Sensitive Tetramycin Releasing Gel and Its Antibacterial Bioactivity against Ralstonia solanacearum.

Ralstonia solanacearum (R. solanacearum)-induced bacterial wilt of the nightshade family causes a great loss in agricultural production annually. Although there has been some efficient pesticides against R. solanacearum, inaccurate pesticide releasing according to the onset time of bacterial wilt during the use of pesticides still hinders the disease management efficiency. Herein, on the basis of the soil pH change during R. solanacearum growth, and pH sensitivity of the Schiff base structure, a pH-sensitive oxidized alginate-based double-crosslinked gel was fabricated as a pesticide carrier.

Naringenin restricts the colonization and growth of Ralstonia solanacearum in tobacco mutant KCB-1.

Bacterial wilt severely jeopardizes plant growth and causes enormous economic loss in the production of many crops, including tobacco (Nicotiana tabacum). Here, we first demonstrated that the roots of bacterial wilt-resistant tobacco mutant KCB-1 can limit the growth and reproduction of Ralstonia solanacearum. Secondly, we demonstrated that KCB-1 specifically induced an upregulation of naringenin content in root metabolites and root secretions. Further experiments showed that naringenin can disrupt the structure of R. solanacearum, inhibit the growth and reproduction of R. solanacearum, and ex

Defining the Metabolic Functions and Roles in Virulence of the rpoN1 and rpoN2 Genes in Ralstonia solanacearum GMI1000.

The alternative sigma factor RpoN is a unique regulator found among bacteria. It controls numerous processes that range from basic metabolism to more complex functions such as motility and nitrogen fixation. Our current understanding of RpoN function is largely derived from studies on prototypical bacteria such as Escherichia coli. Bacillus subtilis and Pseudomonas putida. Although the extent and necessity of RpoN-dependent functions differ radically between these model organisms, each bacterium depends on a single chromosomal rpoN gene to meet the cellular demands of RpoN regulation. The bact

Antibacterial efficacy of iron oxide and silver nanoparticles against bacterial wilt pathogen Ralstonia solanacearum.

The security of vegetable plants worldwide is threatened by bacterial wilts, one of the most infectious soil-borne bacterial plant diseases. This is caused by R. Solanacearum. Overuse of bactericides and antibiotics to combat bacterial wilt results in pesticide resistance and toxicity to beneficial living organisms. Consequently, nanoparticles are more beneficial, safe for the environment, and have strong antibacterial properties than conventional pesticides. In the present work, iron oxide nanoparticles (IONPs) and silver nanoparticles (AgNPs) were prepared by simple chemical, eco-friendly pr
