Isolation of a ranavirus from a gecko (Uroplatus fimbriatus).

A virus was isolated from the liver and stomach of a leaf-tailed gecko (Uroplatus fimbriatus) with granulomatous lesions on the tongue and hepatitis. The virus was identified as an iridovirus on the basis of morphology by electron microscopy, restriction endonuclease assay, and sequencing of a large portion of the major capsid protein gene. Comparative analysis revealed that this isolate is related to frog virus 3, the type species of the genus Ranavirus.

Evaluation and application of andrographolide against largemouth bass ranavirus.

It is well-established that Largemouth bass ranavirus (LMBRaV) induces viral ulcerative syndrome, a highly transmissible and lethal viral disease afflicting largemouth bass aquaculture. Given the urgent need for available prevention and control methods, this study evaluated the antiviral activities of fifteen naturally occurring compounds derived from herbal medicines against LMBRaV using an in vitro cell model. Andrographolide (Andro) was one of the substances that was evaluated and showed highly effective anti-LMBRaV activity of a 99.2 % maximal inhibition rate of at 20 mg/L dosage. Mechanis

Antiviral effects of rhein on largemouth bass ranavirus (LMBRaV).

Largemouth bass ranavirus (LMBRaV), also known as largemouth bass virus (LMBV), is a highly damaging viral pathogen that causes widespread mortality in cultured largemouth bass and poses a significant threat to the farming industry in China. However, there is a paucity of effective preventive methods for this disease by LMBRaV. Research has established that herbal extracts possess antiviral activity, potentially exerting a controlling effect on viruses in aquaculture. Therefore, the identification of simple and structurally safe herbal extract immunomodulators is critical for largemouth bass h

First Detection of Ranavirus Infection in Threatened Brazilian Anurans.

Amphibians are the most threatened vertebrate group worldwide, with emerging infectious diseases playing an important role in their ongoing declines. Although the amphibian chytrid fungus Batrachochytrium dendrobatidis (Bd) has received considerable attention, viruses of the genus Ranavirus (Rv) remain less explored, particularly in South America. In this study, we report the first detection of Rv in two threatened amphibian species from Brazil. Specifically, we found infected tadpoles of the redbelly toads Melanophryniscus admirabilis (1.4% prevalence) and Melanophryniscus biancae (24% preval

Serological survey of Australian native reptiles for exposure to ranavirus.

Ranaviruses have been isolated from many ectothermic vertebrates, and serological surveys of both amphibians and reptiles have shown the presence of ranaviral antibodies in a proportion of these populations. An enzyme-linked immunosorbent assay (ELISA) was developed to measure serum antibodies against ranavirus in Australian reptiles. The ELISA was validated with serum from challenge trials with Bohle iridovirus (BIV) in 6 reptilian species. A preliminary sero-survey of northern Queensland riparian reptile fauna (saw-shelled turtles Myuchelys latisternum, Krefft's river turtles Emydura macquar

Ranavirus infection does not reduce heat tolerance in a larval amphibian.

Extreme heat events and emerging infectious diseases negatively impact wildlife populations, but the interacting effects of infection and host heat tolerance remain understudied. The few studies covering this subject have demonstrated that pathogens lower the heat tolerance of their hosts, which places infected hosts at a greater risk experiencing lethal heat stress. Here, we studied how ranavirus infection influenced heat tolerance in larval wood frogs (Lithobates sylvaticus). In line with similar studies, we predicted the elevated costs of ranavirus infection would lower heat tolerance, meas

Environmental Drivers of Ranavirus Infection in Anurans of the Brazilian Atlantic Forest.

Anthropogenic habitat alteration makes amphibians particularly susceptible to pathogens such as Ranavirus (Rv). Despite severe fragmentation, Brazil's Atlantic Forest remains understudied regarding Rv distribution and impacts. We fill these gaps by mapping Rv across six amphibian communities from areas with varying habitat alteration to identify key conservation areas. We investigated whether susceptibility varies among hosts considering their reproductive modes and habitats. We screened ethanol-preserved livers from specimens previously tested for Batrachochytrium dendrobatidis (Bd) to detect

TLR5-Mediated Reactivation of Quiescent Ranavirus FV3 in Xenopus Peritoneal Macrophages.

Ranaviruses such as frog virus 3 (FV3) are large double-stranded DNA (dsDNA) viruses causing emerging infectious diseases leading to extensive morbidity and mortality of amphibians and other ectothermic vertebrates worldwide. Among the hosts of FV3, some are highly susceptible, whereas others are resistant and asymptomatic carriers that can take part in disseminating the infectious virus. To date, the mechanisms involved in the processes of FV3 viral persistence associated with subclinical infection transitioning to lethal outbreaks remain unknown. Investigation in Xenopus laevis has revealed

Palmatine inhibits largemouth bass ranavirus replication and enhances host antiviral defense.

Largemouth bass ranavirus (LMBV), a member of the family Iridoviridae, is a major viral pathogen in largemouth bass aquaculture. Here, we investigated the antiviral activity of palmatine, a plant-derived isoquinoline alkaloid, against LMBV infection in vitro and in vivo. Palmatine showed no detectable cytotoxicity in epithelioma papulosum cyprini (EPC) cells at concentrations up to 100 μg/mL and inhibited LMBV replication in a dose-dependent manner, with an IC50 of 21.53 μg/mL. At 100 μg/mL, palmatine reduced viral gene expression by more than 90%, markedly attenuated cytopathic effects (CPE),

Immunoglobulin M response in largemouth bass (Micropterus salmoides) following ranavirus infection.

Immunoglobulin M (IgM) and IgM+ B cells are key components of the humoral immune system, providing defense against pathogen invasion. While the role of IgM in the systemic and mucosal immune responses of fish to parasites and bacteria has been partially investigated, its function in viral infections remains underexplored. This study successfully developed a largemouth bass (Micropterus salmoides) model for ranavirus immersion infection. Our findings revealed that viral infection caused significant pathological changes in the gill and head kidney tissues, along with a marked upregulation of ada

Establishment of RPA-CRISPR/Cas12a Detection Methods for Rapid Largemouth Bass Ranavirus Surveillance.

The high mortality associated with largemouth bass ranavirus (LMBRaV) disease requires rapid and sensitive diagnosis methods to prevent virus spread and subsequent outbreak. The recombinase polymerase amplification combined with the CRISPR/Cas12a system (RPA-CRISPR/Cas12a) assay and RPA-CRISPR/Cas12a, coupled with lateral flow dipstick (RPA-CRISPR/Cas12a-LFD) assay for LMBRaV, were established in this study for low viral load surveillance. The major capsid protein (mcp) gene is highly conserved and is widely used as the target sequence in LMBRaV detection. First, based on the mcp gene sequence

FATAL RANAVIRUS INFECTION IN A GROUP OF ZOO-HOUSED MELLER'S CHAMELEONS (TRIOCEROS MELLERI).

A group of five juvenile Meller's chameleons (Trioceros melleri) experienced 100% mortality over a period of 1 mo due to ranavirus infection. The index case was found dead without premonitory signs. The three subsequent cases presented with nonspecific clinical signs (lethargy, decreased appetite, ocular discharge) and were ultimately euthanatized. The final case died after initially presenting with skin lesions. Postmortem examination revealed thin body condition in all five animals and mild coelomic effusion and petechiae affecting the tongue and kidneys of one animal. Microscopically, all a

Microplastics selectively modify ranavirus-driven physiological disruption and gut microbiome restructuring in amphibians.

Amphibians often face overlapping infectious and environmental stressors that interact with non-equivalent magnitudes. We investigated the combined effects of microplastics (MPs) and ranavirus (RV) on the physiology and gut microbiome of the white tree frog (Litoria caerulea). Using a multiblock approach, we integrated diverse endpoints including corticosterone (CORT), body condition, serum biochemistry, antioxidant activity, and gut microbiome profiles. RV was the major driver of host variation, significantly disrupting body condition, protein homeostasis, bioenergetic budget, and renal/hepat

Ranavirus-associated mass mortality in wild amphibians, the Netherlands, 2010: a first report.

In 2010, a mass die-off of over 1000 wild water frogs (Pelophylax spp.) and at least 10 common newts (Lissotriton vulgaris) occurred in a pond in The Netherlands. Haemorrhagic disease with hepatomegaly and splenomegaly was evident. Microscopically, multiple organs presented cells with multifocal intracytoplasmic inclusion bodies, in which ranavirus-like particles were demonstrated ultrastructurally. All specimens examined tested positive for ranavirus by PCR. The sequence obtained showed a 100% identity with the one deposited for common midwife toad virus (CMTV). This is the first report of ra

Case report: ranavirus infections in captive eastern box turtles (Terrapene carolina carolina) in Japan.

Ranaviruses are broad host-range pathogens that cause fatal infections in ectothermic vertebrates, including fish, amphibians, and reptiles and are considered emerging infectious diseases. This report describes a case of ranavirus infection identified at a box turtle breeding facility in Japan. Of 12 box turtles, representing three species and housed in a mixed-species outdoor pen, all six eastern box turtles (Terrapene carolina carolina) exhibited clinical signs, and three died. Severe clinical signs in four turtles included nasal and oral discharge and palpebral edema, while two showed only

Fusing a TurboID tag with the Andrias davidianus ranavirus 2L reduced virus adsorption efficiency.

Andrias davidianus ranavirus (ADRV) is a member of the genus ranavirus (family Iridoviridae). ADRV 2L is an envelope protein that could be essential in viral infection. In the present study, the function of ADRV 2L was investigated by fusion with the biotin ligase TurboID tag. A recombinant ADRV with a V5-TurboID tag fused in the N-terminal of 2L (ADRVT-2L) and a recombinant ADRV expressing V5-TurboID (ADRVT) were constructed, respectively. Infection of the recombinant viruses and wild-type ADRV (ADRVWT) in the Chinese giant salamander thymus cell line (GSTC) showed that ADRVT-2L had reduced c
