Organization of projections from the raphe nuclei to the vestibular nuclei in rats.

Previous anatomic and electrophysiological evidence suggests that serotonin modulates processing in the vestibular nuclei. This study examined the organization of projections from serotonergic raphe nuclei to the vestibular nuclei in rats. The distribution of serotonergic axons in the vestibular nuclei was visualized immunohistochemically in rat brain slices using antisera directed against the serotonin transporter. The density of serotonin transporter-immunopositive fibers is greatest in the superior vestibular nucleus and the medial vestibular nucleus, especially along the border of the four

Raphe nuclei echogenicity changes in obsessive-compulsive disorder.

Background: Dysregulation of serotonin system is hypothesized to play the main role in the etiology of obsessive-compulsive disorder (OCD). Transcranial sonography (TCS) is a helpful noninvasive and low-cost tool for the assessment of subcortical brain architectures, mainly basal nuclei, cerebellar central structures, and midbrain. In this study, an ultrasound assessment was performed for a sample of the patients with OCD and healthy control group to evaluate echogenicity of midbrain raphe nuclei (RN). Methods: A total of 35 patients with OCD and 35 healthy controls of similar age and sex ente

Divergent innervation of the olfactory bulb by distinct raphe nuclei.

The raphe nuclei provide serotonergic innervation widely in the brain, thought to mediate a variety of neuromodulatory effects. The mammalian olfactory bulb (OB) is a prominent recipient of serotonergic fibers, particularly in the glomerular layer (GL), where they are thought to gate incoming signals from the olfactory nerve. The dorsal raphe nucleus (DRN) and the median raphe nucleus (MRN) are known to densely innervate the OB. The majority of such projections are thought to terminate in the GL, but this has not been explicitly tested. We sought to investigate this using recombinant adeno-ass

Functional Connectivity of the Raphe Nuclei: Link to Tobacco Withdrawal in Smokers.

BACKGROUND: Although nicotine alters serotonergic neurochemistry, clinical trials of serotonergic medications for smoking cessation have provided mixed results. Understanding the role of serotonergic dysfunction in tobacco use disorder may advance development of novel pharmacotherapies. METHODS: Functional magnetic resonance imaging was used to measure resting-state functional connectivity of the raphe nuclei as an indicator of serotonergic function. Connectivity of the dorsal and median raphe nuclei was compared between 18 young smokers (briefly abstinent, ~40 minutes post-smoking) and 19 yo

Anorexigenic action of nitric oxide synthase inhibition in the raphe nuclei.

The present report examined the effects of midbrain raphe nuclei injections of nitric oxide synthase inhibitors NG-nitro-L-arginine methyl ester (L-NAME) and 7-nitroindazole (7-NI) on eating behavior. L-NAME (5-500 pmol) and 7-NI (2-200 pmol) were administered into either the dorsal or median raphe nucleus. Both nitric oxide synthase inhibitors decreased food intake in adult male Sprague-Dawley rats when injected into either raphe site. Further, eating elicited by dorsal and median raphe injections of the 5-HT1A agonist 8-OH-DPAT (0.8 nmol) was attenuated by L-NAME or 7-NI pretreatment. Our fi

Serotonergic projections to the ventral respiratory column from raphe nuclei in rats.

The ventral respiratory column (VRC) generates rhythmical respiration and is divided into four compartments: the Bötzinger complex (BC), pre-Bötzinger complex (PBC), rostral ventral respiratory group (rVRG), and caudal ventral respiratory group (cVRG). Serotonergic nerve fibers are densely distributed in the rostral to caudal VRC and serotonin would be one of the important modulators for the respiratory control in the VRC. In the present study, to elucidate detailed distribution of serotonergic neurons in raphe nuclei projecting to the various rostrocaudal levels of VRC, we performed combinati

Perinatal Western-style diet alters serotonergic neurons in the macaque raphe nuclei.

INTRODUCTION: The neurotransmitter serotonin is a key regulator of neurotransmission, mood, and behavior and is essential in neurodevelopment. Dysfunction in this important neurotransmitter system is connected to behavioral disorders such as depression and anxiety. We have previously shown that the developing serotonin system is sensitive to perinatal exposure to Western-style diet (WSD). METHODS: To advance our hypothesis that perinatal WSD has a long-term impact on the serotonergic system, we designed a fluorescent immunohistochemistry experiment using antibodies against tryptophan hydroxyl

Dorsal raphe nuclei integrate allostatic information evoked by depletion-induced sodium ingestion.

Structures of the lamina terminalis (LT) sense and integrate information reflecting the state of body water and sodium content. Output from the LT projects into a neural network that regulates body fluid balance. Serotonin (5-HT) and the dorsal raphe nuclei (DRN) have been implicated in the inhibitory control of salt intake (i.e., sodium appetite). Signals arriving from the LT evoked by fluid depletion-induced sodium ingestion interact with this inhibitory serotonergic system. We investigated the role of neurons along the LT that directly project to the DRN. We analyzed the pattern of immunore

Raphe nuclei in cognition: A network-level framework for behavioral-state control.

Adaptive behavior requires organisms to maintain, update, and switch between behavioral states as environmental conditions change. The raphe nuclei (RN), best known as a major source of forebrain serotonin, have long been linked to arousal, inhibition, patience, affect, and flexibility, but these functions have often been considered separately. Here, we argue that recent advances in circuit tracing, cell-type-specific manipulation, high-resolution neuroimaging and neurostimulation, and computational modeling support a more integrated view in which RN are embedded within distributed cortico-sub

Differential effects of neonatal hypoxic-ischemic brain injury on brainstem serotonergic raphe nuclei.

Serotonergic fibres have a pervasive innervation of hypoxic-ischemic (HI)-affected areas in the neonatal brain and serotonin (5-HT) is pivotal in numerous neurobehaviours that match many HI-induced deficits. However, little is known about how neonatal HI affects the serotonergic system. We therefore examined whether neonatal HI can alter numbers of serotonergic raphe neurons in specific sub-divisions of the midbrain and brainstem since these nuclei are the primary sources of serotonin throughout the central nervous system (CNS). We utilised an established neonatal HI model in the postnatal day

Immobilization stress elevates tryptophan hydroxylase mRNA and protein in the rat raphe nuclei.

BACKGROUND: Stress triggers adaptive and maladaptive changes in the central nervous system, including activation of the hypothalamic-pituitary-adrenal axis, and can trigger mood disorders and posttraumatic stress disorder. We examined the effect of immobilization stress (IMO) on gene expression of tryptophan hydroxylase (TPH), the rate-limiting enzyme in serotonin biosynthesis, and the role of cortisol in that response. METHODS: Regular and adrenalectomized Sprague-Dawley rats were exposed to various repetitions of IMO. Tryptophan hydroxylase messenger ribonucleic acid (mRNA) was determined b

Brainstem raphe nuclei and their serotonergic neurons: An integrative review of distribution, function and projection.

Serotonergic neurons in the raphe nuclei of the brainstem play a central role in a wide range of physiological and psychological processes. These neurons form intricate neuroregulatory networks via extensive ascending and descending projections to multiple brain regions. They are critically involved in integrating and refining neural signal transmission to sustain normal physiological homeostasis, and they play pivotal roles in higher-order cerebral functions such as emotional processing, cognitive decision-making, and behavioral modulation. Functional dysregulation of serotonergic networks is

The ascending serotonergic system in the hamster: comparison with projections of the dorsal and median raphe nuclei.

The ascending serotonergic projections are derived largely from the midbrain median and dorsal raphe nuclei, and contribute to the regulation of many behavioral and physiological systems. Serotonergic innervation of the hamster circadian system has been shown to be substantially different from earlier results obtained with other methods and species. The present study was conducted to determine whether similar differences are observed in other brain regions. Ascending projections from the hamster dorsal or median raphe were identified using an anterograde tracer, Phaseolus vulgans leucoagglutin

Pressor effects of electrical stimulation of the dorsal and median raphe nuclei in anesthetized rats.

Electrical stimulation (1--50 Hz, 100--500 muA, 0.3 msec pulse duration) of the dorsal or median raphe nucleus in anesthetized rats produces a transient increase in arterial blood pressure. Heart rate and respiratory rate do not appear to change systematically after stimulation. Transections rostral or caudal to the raphe nuclei or electrolytic lesions in the area of the nucleus linearis, pars caudalis abolishes the pressor response to stimulation. Depletion of brain serotonin with p-chlorophenylalanine significantly attenuates the pressor effect of both dorsal and median raphe stimulation. Th

Raphe Nuclei Echogenicity and Diameter of Third Ventricle in Schizophrenia Measured by Transcranial Sonography.

INTRODUCTION: Serotonergic system hyperactivity at 5-HT2A receptors on glutamate neurons in the cerebral cortex is one of the pathways that is theoretically linked to psychosis. In addition to neurotransmitter dysfunction, volumetric studies have revealed the loss of cortical gray matter and ventricular enlargement in patients with schizophrenia, although there is no case-control research on patients with schizophrenia to evaluate echogenicity of raphe nuclei (RN) or diameter of the third ventricle (DTV). To address these issues, the present study assessed midbrain RN, as the main source of br

Depressive disorder may be associated with raphe nuclei lesions in patients with brainstem infarction.

BACKGROUND: Depression is a common symptom after stroke, but its neural substrates remain unclear. The ascending serotonergic system originates from the raphe nuclei in the brainstem. We hypothesized that depressive disorder due to brainstem infarction is associated with damage to the raphe nuclei. METHODS: We prospectively enrolled 19 patients who had the first-ever acute isolated brainstem infarction in an observational cross-sectional study. All patients were evaluated by using the Montgomery Åsberg Depression Rating Scale (MADRS), the clinician-rated version of Apathy Evaluation Scale (AE

Serotonin neurons of the caudal raphe nuclei contribute to sympathetic recovery following hypotensive hemorrhage.

Serotonin is thought to contribute to the syncopal-like response that develops during severe blood loss by inhibiting presympathetic neurons of the rostroventrolateral medulla (RVLM). Here, we tested whether serotonin cells activated during hypotensive hemorrhage, i.e., express the protein product of the immediate early gene c-Fos, are critical for the normal sympathetic response to blood loss in unanesthetized rats. Serotonin-immunoreactive cells of the raphe obscurus and raphe magnus, parapyramidal cells of the B3 region, subependymal cells of the ventral parapyramidal region, and cells of t

GABAergic MicroRNA-34a in the Dorsal Raphe Nuclei Contributes to Circulating MicroRNA-34a Levels.

Brain microRNAs (miRs) are expressed in a cell-specific manner and are implicated in several pathologies. Despite circulating miRs (c-miRs) being increasingly recognized as biomarkers of brain disorders, few studies have directly linked specific brain regions or cell types to plasma miR levels under physiological conditions. In the dorsal raphe nuclei (DRN), mir-34a is selectively expressed in GABAergic neurons. This expression increases following prolonged stimuli that challenge DRN activity, like repeated stress or chronic SSRI administration. Here, we investigate whether plasma levels of mi

Afferent projections to the rat nuclei raphe magnus, raphe pallidus and reticularis gigantocellularis pars alpha demonstrated by iontophoretic application of choleratoxin (subunit b).

The aim of the present study was to identify the specific afferent projections to the rostral and caudal nucleus raphe magnus, the gigantocellular reticular nucleus pars alpha and the rostral nucleus raphe pallidus. For this purpose, small iontophoretic injections of the sensitive retrograde tracer choleratoxin (subunit b) were made in each of these structures. In agreement with previous retrograde studies, after all injection sites, a substantial to large number of labeled neurons were observed in the dorsal hypothalamic area and dorsolateral and ventrolateral parts of the periaqueductal gray
