Use of Halogenated Units for the Construction of Artificial Carbohydrate Receptors.

To investigate the potential of halogen-containing building blocks in the development of artificial carbohydrate receptors, the 1,3,5-trisubstituted 2,4,6-triethylbenzene scaffold with halogenated subunits and classical hydrogen bonding sites was used as a model system. In the first studies, the influence of the presence of halogens on the binding properties of compounds bearing benzamidomethyl units was investigated, whereby the type of halogen and its ring position were varied. The question was whether the presence of halogens could lead to an increase in binding effectivity and whether this

LBL Noninvasively Peelable Biointerfacial Adhesives for Cutaneo-Inspired pH/Tactility Artificial Receptors.

Besides barrier functions, skin possesses multiple sentiences to external stimuli (e.g., temperature, force, and humidity) for human-outside interaction. Thus, skincare should be taken very seriously, especially by patients with sensory disorders. However, currently available skin-mimicking devices are always limited by so much insufficient response functions and nontunable interface behaviors so as not to realize precise health monitoring and self-defense against injury. Herein, a bioinspired cutaneous receptor-perceptual system (CRPS) patch is presented, integrating hybrid pH indicators and

4-Chloropropofol enhances chloride currents in human hyperekplexic and artificial mutated glycine receptors.

BACKGROUND: The mammalian neurological disorder hereditary hyperekplexia can be attributed to various mutations of strychnine sensitive glycine receptors. The clinical symptoms of "startle disease" predominantly occur in the newborn leading to convulsive hypertonia and an exaggerated startle response to unexpected mild stimuli. Amongst others, point mutations R271Q and R271L in the α1-subunit of strychnine sensitive glycine receptors show reduced glycine sensitivity and cause the clinical symptoms of hyperekplexia.Halogenation has been shown to be a crucial structural determinant for the poten

Assembly-formed bioorthogonal chimeric artificial receptors enable high-contrast fluorescence imaging of tumors.

Conventional receptor-targeted fluorescent probes have shown promise in tumor imaging, yet achieving a high tumor-to-normal (T/N) tissue ratio in vivo remains challenging due to limited biomarker density on tumor cell membranes. Here, we present an in situ assembly strategy of bioorthogonal-functionalized chimeric artificial receptors (BCARs) that locally constructs BCARs on tumor surfaces, which amplify fluorescence signals and enable high-contrast imaging. Rapid, selective membrane engineering under physiological conditions increases effective receptor density, enhancing fluorophore binding

Artificial receptor-facilitated solid-phase microextraction of barbiturates.

A receptor for barbiturates, N,N'-Bis-[6-(2-ethylhexanoylamino)-pyridin-2-yl]-isophthalamide, was designed to dissolve in plasticizers of poly(vinyl chloride) (PVC). Microextractions using receptor-doped films of PVC were carried out as a function of receptor concentration. The effect of the concentration of the receptor on extraction yield is considerable for barbiturates that have significant binding to the receptor but negligible for very similar molecules that do not bind to the receptor strongly. Thus, it is the receptor's ability in molecular recognition, not its generic ability as an H-

Transmembrane DNA Nanochannel-Engineered Artificial Receptors for Navigating NK Cell Immunotherapy in Solid Tumors.

Adoptive natural killer (NK) cell therapy for solid tumors faces critical challenges, including tumor antigen heterogeneity, impaired tumor infiltration, and suboptimal activation imposed by the immunosuppressive microenvironment. Here we developed an engineered nanoplatform featuring transmembrane DNA nanochannel-engineered artificial receptors (NCAR) to direct NK cells against solid tumors through two synergistic mechanisms: 1) Tumor Microenvironment (TME) Reprogramming: leveraging cholesterol-mediated insertion, NCAR incorporates into tumor membranes to disrupt phospholipid bilayers, induci

Upregulation of hepatic nuclear receptors in extremely preterm ovine fetuses undergoing artificial placenta therapy.

OBJECTIVE: Extremely preterm infants have low Nuclear Receptor (NR) expression in their developing hepatobiliary systems, as they rely on the placenta and maternal liver for compensation. NRs play a crucial role in detoxification and the elimination of both endogenous and xenobiotic substances by regulating key genes encoding specific proteins. In this study, we utilized an Artificial Placenta Therapy (APT) platform to examine the liver tissue expression of NRs of extremely preterm ovine fetuses. This fetal model, resembling a "knockout placenta," lacks placental and maternal support, while ma

The Effects of Artificial Sweeteners on Intestinal Nutrient-Sensing Receptors: Dr. Jekyll or Mr. Hyde?

The consumption of artificial and low-calorie sweeteners (ASs, LCSs) is an important component of the Western diet. ASs play a role in the pathogenesis of metabolic syndrome, dysbiosis, inflammatory bowel diseases (IBDs), and various inflammatory conditions. Intestinal nutrient-sensing receptors act as a crosstalk between dietary components, the gut microbiota, and the regulation of immune, endocrinological, and neurological responses. This narrative review aimed to summarize the possible effects of ASs and LCSs on intestinal nutrient-sensing receptors and their related functions. Based on the

Extraction of radio-labelled xanthine derivatives by artificial receptors: deep insight into the association behaviour.

Association constants for the interaction of almost insoluble substrates with triphenylene ketal-based receptors in toluene have been determined by means of an extraction method employing the corresponding radio-labelled substrates. Flexible and more polar receptors tend to aggregate and exhibit inferior extraction qualities. Binding constants in toluene were found to be in the range 10(5)-10(7) m(-1), which is significantly higher than in dichloromethane. X-ray analyses indicate the direct participation of a water molecule in the binding process, which may account for the surprisingly small e

Enhanced extraction of phenobarbital from serum with a designed artificial receptor.

The primary goal of this work was to determine whether artificial receptors that function on the basis of molecular recognition have analytical capabilities. As an example of such a receptor, we have chosen one directed toward barbiturates. Chloroform enriched with this artificial receptor (1 mM) can extract more than 90% of the phenobarbital from a 20 microM phenobarbital solution in human control serum using a volume ratio (organic/serum) as small as 0.5. In the absence of this receptor, the volume ratio must be greater than 10 to achieve similar extraction efficiencies. In addition to volum

The Plasminogen Activator System, Glucocorticoid, and Mineralocorticoid Receptors in the Primate Endometrium During Artificial Menstrual Cycles.

As a key mechanism in fibrinolysis and tissue remodeling, the plasminogen activator system has been suggested in the process of endometrial shedding and tissue remodeling. Previous studies have explored the role of estrogen, progesterone, and androgen receptors as well as elements of the renin-angiotensin-aldosterone system in shaping the morphology of the endometrium. This study investigates the distribution and concentrations of the mineralocorticoid receptor, glucocorticoid receptor, tissue plasminogen activator, urokinase plasminogen activator, and plasminogen activator inhibitor-1 within

CAR-T cell signaling dynamics, rational design principles and artificial intelligence for next-generation chimeric antigen receptors.

Chimeric antigen receptor (CAR) T cell therapy has transformed the treatment of hematologic malignancies, yet its efficacy in solid tumors and durability across broader application remain limited. A central challenge lies in how CAR signaling is initiated, amplified, and regulated over time. Unlike the native T cell receptor (TCR), CARs are synthetic, modular receptors whose signaling output is dictated by the composition and spatial organization of their extracellular, transmembrane, and intracellular domains. Emerging evidence suggests that CAR signaling requirements are not static: insuffic

Multi-threshold synaptic transistor for an artificial sensory-pain integrated receptor.

Pain is a simple yet effective self-defense mechanism in humans that enables rapid responses to harmful stimuli. The inability to sense pain increases the risk of injury due to delayed reactions to such stimuli. However, current prostheses often struggle to differentiate between sensory and pain-like signals and require complex systems to achieve this functionality. In this study, we developed an artificial receptor capable of detecting both sensory and pain-like signals with the threshold discrimination and memory functions intrinsically embedded in the transistor. This functionality was real

Biologic activity of antigen receptors artificially incorporated onto B lymphocytes.

We describe a method for incorporating monoclonal antibody molecules onto viable murine lymphocytes and summarize the biologic activity of these artificial receptors on B cells. Mouse spleen cells incubated overnight with palmitate conjugates of a monoclonal anti-DNP IgA (protein 315) in the presence of deoxycholic acid incorporate about 50,000 antibody molecules per cell. When concentrations of deoxycholate and palmitoyl-protein 315 are carefully controlled, this labeling procedure does not affect the viability or the normal functions of the receptor-decorated cells. The incorporated antibody

Intracellular artificial DNA receptor-loaded gelated cells for aflatoxin theranostics.

Aflatoxin B1 (AFB1) is one of the most common mycotoxins, which causes severe hepatic damage and even liver cancer. However, the development of specific tools for AFB1 diagnosis and detoxification remains elusive. Herein, we develop a DNA receptor-loaded gelated cell (DR-GC)-based adsorbent for specifically capturing and detecting AFB1, both in vitro and in vivo. In this system, fragile living cells are transformed into robust GCs using an intracellular hydrogel technique, and then loaded with aptamer-based artificial DNA receptors inside cells via a polymer-assisted strategy. These intracellu

Artificial receptor-attached amphiphilic copolymer for barbiturate binding in aqueous media.

A water-soluble self-associating amphiphilic copolymer was employed to provide a microenvironment for the solvation of a hydrogen-bonding barbiturate artificial receptor, to facilitate molecular recognition in water. The receptor-attached amphiphilic polymer (RP) was synthesized through random copolymerization of 3% (mol) barbiturate receptor-monomer, 70% (mol) 3-sulfopropyl methacrylate, and 27% (mol) n-dodecyl acrylate. Difference UV spectra of pH 6.5 aqueous solutions of phenobarbital and receptor-polymer (RP) gave peaks and valleys at 272 and 301 nm respectively, consistent with binding ch

Explanation of the pseudohypoaldosteronism (PHA)-stress syndrome with an artificial aldosterone receptor model.

A receptor for aldosterone was studied in the cytosol of rectal mucosa of two sisters (M.A., M.B.) with the clinical manifestations of pseudohypoaldosteronism (PHA). Compared to age matched controls the patients showed a decreased affinity for aldosterone (M.A. Kd1: 0.18 nM, Kd2: 4.55 nM; Nmax1: 0.185 fmol/mg cytosol protein (CP), Nmax2: 3.12 fmol/mg CP, respectively). In an attempt to find an explanation for the phenomenon of stress-induced electrolyte imbalance in PHA patients an experimental set up was designed, using aldosterone antibody material as artificial aldosterone receptor. Cell death of olfactory receptor neurons in a rat with nasosinusitis infected artificially with Staphylococcus.

Nasosinusitis is a common cause of acquired hyposmia or anosmia. To study the apoptotic death of olfactory receptor neurons in nasosinusitis, we made an inflammation model in rat infected with Staphylococcus. The histochemical changes in olfactory epithelium were examined using antibodies against protein gene product 9.5 (PGP 9.5), single-strand DNA (ssDNA), Bcl-2 and Bax that might be involved in the apoptosis of olfactory receptor neurons. The thickness of olfactory epithelium and the number of ssDNA-labeled cells were evaluated in each post-treatment group and the results were analyzed by t

Artificial intelligence-based structural modeling of PSTOL1 receptor kinases in rice and sorghum.

Originally identified in rice (Oryza sativa OsPSTOL1), PHOSPHORUS-STARVATION TOLERANCE 1 (PSTOL1) proteins are essential determinants of phosphorus efficiency in several plant species, including Sorghum bicolor (SbPSTOL1). In contrast to OsPSTOL1, SbPSTOL1 homologs exhibit a transmembrane receptor-like kinase (RLK) architecture with polymorphic extracellular domains. We employed an AI-driven computational pipeline to investigate the structure and conformational dynamics of SbPSTOL1 proteins. Monomeric predictions using multiple deep-learning platforms yielded structures that were inconsistent

Harnessing artificial intelligence to identify Bufalin as a molecular glue degrader of estrogen receptor alpha.

Target identification in natural products plays a critical role in the development of innovative drugs. Bufalin, a compound derived from traditional medicines, has shown promising anti-cancer activity; however, its precise molecular mechanism of action remains unclear. Here, we employ artificial intelligence, molecular docking, and molecular dynamics simulations to elucidate the molecular mechanism of Bufalin. Using an integrated multi-predictive strategy, we identify CYP17A1, ESR1, mTOR, AR, and PRKCD as the potential targets of Bufalin. Subsequent validation via surface plasmon resonance, bi
