High affinity receptors for atrial natriuretic factor in PC12 cells.

Specific receptors for atrial natriuretic factor (ANF) are characterized on PC12 cells (rat pheochromocytoma cell line). Radioiodinated synthetic ANF (Rat, 8-33) bound to a single class of high affinity binding sites with the Kd value of 6.7 X 10(-10) M. The Bmax value was 29 fmol/10(5) cells and receptor density was calculated as 194,000 +/- 20,000/cell. Photoaffinity labelling of ANF receptor specifically labelled two protein bands with apparent m.wt of 70,000 and 130,000. When the cells were incubated with the labelled ligand at 37 degrees C the ligand was internalized. The rate of internal

Identification and solubilization of atrial natriuretic factor receptors in human placenta.

Specific high affinity 125I-atrial natriuretic factor binding sites have been identified in human placental membranes. Using the nonionic detergent, Triton X-100, these binding sites were quantitatively solubilized and retained binding activity. In the solubilized preparation, the macromolecular component that binds atrial natriuretic factor is a 160,000 dalton protein as shown by covalently cross-linking it to 125I-atrial natriuretic factor with the bifunctional chemical crosslinker, disuccinimidyl suberate, followed by gel electrophoresis and autoradiography. On Sephadex G-200 gel filtration

Photoaffinity labeling of atrial natriuretic factor receptors of rat kidney cortex plasma membranes.

Synthetic rat atrial natriuretic factor (ANF) was derivatized with the N-hydroxysuccinimide ester of [125I]iodoazidosalicylic acid to yield a radioactive photoaffinity probe. Incubation of purified plasma membranes from rat kidney cortex with this photoaffinity probe resulted in the specific labeling of a 140-kDa glycoprotein. The photoaffinity labeling of this protein was inhibited by ANF but not by reduced and alkylated ANF nor by other unrelated peptides. A 140-kDa band was also specifically labeled in liver plasma membranes but not in adipocyte plasma membranes. These observations suggest

Atrial natriuretic factor stimulates exocrine pancreatic secretion in the rat through NPR-C receptors.

The urethral closure mechanism under a stress condition induced by sneezing was investigated in urethane-anesthetized female rats. During sneezing, while the responses measured by microtip transducer catheters in the proximal and middle parts of the urethra increased, the response in the proximal urethra was almost negligible when the bladder response was subtracted from the urethral response or when the abdomen was opened. In contrast, the response in the middle urethra during sneezing was still observed after subtracting the bladder response or after opening the abdomen. These responses in t

Identification of the receptor for atrial natriuretic factor on cultured vascular cells.

Binding experiments with 125I-atrial natriuretic factor (ANF) followed by covalent attachment with disuccimidyl suberate show that the peptide binds predominantly to a protein of apparent molecular mass of 66,000 daltons on the cell surface of cultured bovine aortic smooth muscle cells. A minor protein species of 180,000 Mr is also visualized after cross-linking. Endothelial cells, however, whose ANF binding parameters differ substantially from smooth muscle cells, also appear to have qualitatively identical 125I-ANF binding proteins. The identity of these putative proteins, as the ANF recepto

Receptor sites for atrial natriuretic factors in brain and associated structures: an overview.

1. Recent data have clearly shown the existence of specific receptor binding sites for atrial natriuretic factors (ANF) or polypeptides in mammalian brain tissues. 2. Ligand selectivity pattern and coupling to cGMP production suggest that brain ANF sites are similar to high-affinity/low-capacity sites found in various peripheral tissues (kidney, adrenal gland, blood vessels). These brain ANF sites possibly are of the B-ANP subtype. 3. High densities of ANF binding sites are found especially in areas of the central nervous system associated with the control of various cardiovascular parameters

Experimental glaucoma significantly decreases atrial natriuretic factor (ANF) receptors in the ciliary processes of the rabbit eye.

ANF binding sites were analysed in the ciliary processes of rabbits with unilateral experimental glaucoma which had been induced by injecting alpha-chymotrypsin into the posterior chamber of the right eyes. The intraocular pressure (IOP) of glaucomatous eyes was significantly greater (28.4 +/- 4 mmHg) than that of normotensive control eyes (13.1 +/- 1.4 mmHg, mean +/- S.E.M., n = 23, P less than 0.05). ANF concentrations in aqueous humour and the ciliary processes were significantly higher in glaucomatous eyes (91 +/- 2 pg ml-1 and 30.4 +/- 4.2 pg g-1 wet weight) than in normal eyes (3.1 +/- 2

Localization and characterization of specific receptors for atrial natriuretic factor in the ciliary processes of the eye.

By light and electron microscope radioautography in vivo, competitive binding sites for 125I-Arg 101-Tyr 126 atrial natriuretic factor were localized mostly on the "pigmented" epithelium of the rat ciliary process. Further investigation using isolated ciliary processes from rabbits demonstrated the presence of specific receptors for 125I-atrial natriuretic factor. In addition, synthetic atrial natriuretic factor inhibited basal and stimulated adenylate cyclase activity. These results demonstrate for the first time the presence of specific receptors for atrial natriuretic factor in the ciliary

The release of atrial natriuretic factor induced by central carbachol injection may be mediated by muscarinic M1 receptors.

We investigated the effect of selective muscarinic antagonists on atrial natriuretic factor (ANF) release induced by intracerebroventricular (i.c.v) injection of carbachol in the rat. The muscarinic antagonists were given by i.c.v. injection 1 min before carbachol, 1 micrograms/rat. 4-Diphenylacetoxy-N-methylpiperidine methiodide (4-DAMP), a rather selective M1 and M3 receptor antagonist, was the most potent inhibitor of carbachol-induced ANF release, its ID50 being 0.18 nmol/rat. Pirenzepine, a selective M1 antagonist, also potently inhibited the effect of carbachol, its ID50 being 2.74 nmol/

Glucocorticoids inhibit D1B, but not D2, receptor-mediated effects on hypothalamic atrial natriuretic factor neurons.

Recent evidence suggests that ANF neurons of the hypothalamus are dopamine sensitive, and the catecholamine may exert a direct stimulatory or inhibitory effect on the neurons mediated through D1 or D2 receptors, respectively, in a manner related to the differential dopamine binding sensitivity of the two receptor subtypes. Employing well characterized ANF RIA and colorimetric Northern blot analysis with synthetic oligonucleotide probes complementary to pro-ANF messenger RNA (mRNA), we report here the effect of dexamethasone (DM), a potent synthetic glucocorticoid, on DA-stimulated ANF neurons

Bradykinin type 2 receptor deficiency alters vascular endothelial growth factor and atrial natriuretic peptide levels in early-stage diabetic retinopathy.

Diabetic retinopathy is a leading cause of visual impairment among working-age adults. Given the multiple pathophysiologic functions of bradykinin and its links to vascular endothelial growth factor (VEGF) and atrial natriuretic peptide (ANP) signaling, we hypothesized their interconnected involvement in the development of retinal vascular permeability in early-stage diabetic retinopathy. Diabetic mice, bradykinin type 2 receptor (B2R) knock-out diabetic mice, and their non-diabetic controls underwent magnetic resonance imaging, fluorescein angiography, vascular permeability measurements, reti

[Vasopressin, the antidiuretic hormone].

Vasopressin is actively involved in the regulation of blood pressure to the same degree as catecholamines and the renin angiotensin aldosterone system are, especially in stressful situations. Vasopressin induces and increase in blood pressure when mechanisms buffering its potent vasoconstrictor effect are altered. Vasopressin binds to specific membrane receptors classified into two main types. The V1 receptors found in blood vessels, platelets and hepatocytes are linked to two intra-cellular messengers, namely 1,2 diacylglycerol and 1,4,5 inositol triphosphate which stimulate protein kinase C

Atrial natriuretic factor significantly contributes to the mineralocorticoid escape phenomenon. Evidence for a guanylate cyclase-mediated pathway.

The mechanism underlying the mineralocorticoid escape phenomenon remains unknown. To assess the possible contribution of natriuretic peptides to mineralocorticoid escape, rats were injected with 5 mg deoxycorticosterone acetate for 3 d. Plasma atrial natriuretic factor (ANF) rose to twice basal levels and atrial ANF content decreased significantly by 24 h of treatment. This coincided with renal escape and with a significant increase in urinary cGMP excretion. Plasma ANF remained elevated and atrial ANF content continued to decline by 48 and 72 h while atrial ANF mRNA levels increased significa

[N-terminal atrial natriuretic peptides].

Atrial myocytes synthesise atrial natriuretic factor prohormone consisting of 126 amino acids (ANP1-126) which is subsequently processed to several fragments. Atrial natriuretic factor (ANF, ANP99-126) originating from the C-terminal portion of prohormone is a best described atrial peptide. However, several peptides originating from the N-terminus of this precursor also circulate and produce significant diuresis, natriuresis and vasodilatation. These are: long acting natriuretic peptide (ANP1-30), vessel dilator (ANP31-67) and kaliuretic peptide (ANP79-98). ANP1-98 and ANP68-98 also circulate.

Neutral metalloendopeptidase inhibitors as ANF potentiators: sites and mechanisms of action.

Inhibition of the enzyme neutral metalloendopeptidase potentiates responses to atrial natriuretic factor and elicits reductions of blood pressure in desoxycorticosterone acetate sodium hypertensive rats. The present study evaluated the role of atrial natriuretic factor and bradykinin in the antihypertensive response to neutral metalloendopeptidase inhibition through the use of antibodies and antagonists, respectively. In addition, the pharmacokinetic mechanism by which neutral metalloendopeptidase inhibition interferes with atrial natriuretic factor metabolism was explored. The antihypertensiv

Atrial natriuretic peptide and renal dopaminergic system: a positive friendly relationship?

Sodium metabolism by the kidney is accomplished by an intricate interaction between signals from extrarenal and intrarenal sources and between antinatriuretic and natriuretic factors. Renal dopamine plays a central role in this interactive network. The natriuretic hormones, such as the atrial natriuretic peptide, mediate some of their effects by affecting the renal dopaminergic system. Renal dopaminergic tonus can be modulated at different steps of dopamine metabolism (synthesis, uptake, release, catabolism, and receptor sensitization) which can be regulated by the atrial natriuretic peptide.
