The neuropeptide galanin (GAL) is widely distributed within intrinsic and extrinsic sources supplying the eye. It is involved in regulation of the vascular tone, thus important for ocular homeostasis. Since the presence/distribution of its receptors is unknown, we here screen for the presence of the various GAL receptors in the human eye. Meeting the Helsinki-Declaration, human eyes (n = 6; 45-83 years of age, of both sex, post mortem time 10-19 h) were obtained from the cornea bank and prepared for immunohistochemistry against GAL receptors 1-3 (GALR1-GALR3). Over-expressing cell assays serve

BACKGROUND: Allergic rhinitis (AR) is caused by allergic reaction to allergens such as pollen. Galanin (GAL), a neuropeptide that regulates inflammatory processes, is widely expressed in the central and peripheral nervous systems. Although neuropeptides are implicated in arthritis and chemically induced ileitis, their roles in AR remain unclear.
METHODS: We developed a murine model of AR and generated control, systemic sensitization, mild AR, and severe AR groups. We examined GAL and GAL receptor (GALR) mRNA and protein levels and localization patterns in each group using reverse transcriptio

Galanin-like peptide (GALP) was isolated from porcine hypothalamus in 1999 on the basis of its ability to activate galanin receptors in vitro. Extensive studies carried out since the discovery of GALP contributed to the significant progress in our knowledge regarding this neuropeptide. GALP is synthesized mainly in the neurons of the hypothalamic arcuate nucleus and in the pituicytes of the posterior pituitary. The effects of GALP are mediated by well-characterized G-protein coupled galanin receptor subtypes. The fact that GALP shares homology only with 13 amino acids of the galanin sequence s

Galanin, a 29-aminoacid peptide (30 in humans), is widely distributed in the nervous and endocrine systems and exerts its actions via three G-protein-coupled receptors, GalR1-3. The galanin system has, among others, been associated with tumorigenesis. Our objective was to assess the expression of galanin and its receptors in pituitary tumors. Transcript levels of galanin and galanin receptors 1-3 (GalR1-3) were measured using quantitative real-time PCR (q-PCR) in pituitary tumors, surgically removed from thirteen patients, and twelve post mortem pituitaries. Galanin, GalR1 and GalR2 mRNA, but

Galanin-like peptide (GALP) is a 60 amino acid neuropeptide originally discovered from porcine hypothalamus, and is involved in the regulation of food intake in mammals. Since its discovery, GALP and its receptors (GALR1 and GALR2) have been characterized in mammals, but no publications are available on GALP in fish and other non-mammals. The present study aimed to characterize brain and intestinal GALP and its receptors using immunohistochemistry in a teleost, the goldfish (Carassius auratus), and to study its effects on feeding behavior. Immunostaining of brain sections shows the presence of

The neuropeptide galanin (GAL) has recognized physiological actions in the nervous system and other tissues, but there is no documented evidence of GAL influencing normal or pathological bone metabolism. GAL expression, however, is upregulated in central and peripheral nerves following axotomy and is known to influence neural regeneration. Thus, severance of skeletal-associated nerves during fracture could similarly increase local GAL concentrations and thereby influence fracture healing. The initial aim of this study was therefore to identify the presence of GAL in normal bone and/or fracture

Although the tuberomammillary nucleus (TM) is well defined in terms of anatomy and neurochemistry, little is known about its function in nociceptive modulation. There was an abundance of galanin-immunoreactive fibers in the TM, and galanin has been implicated in pain processing. The present study assessed the role of galanin in the modulation of nociception in the TM of rats. Intra-TM injection of galanin dose-dependently increased the hindpaw withdrawal latency of rats to a noxious thermal stimulus, indicating an antinociceptive role of galanin in the TM. The antinociceptive effect of galanin

This study examined the effects of specific antagonists to kappa- and mu-opioid receptors on the feeding induced by injecting galanin into the lateral cerebral ventricle (LCV). Galanin injected into the lateral cerebral ventricle of sated rats stimulated the consumption of high-fat diet when compared to controls injected with saline vehicle. The mu-opioid receptor antagonist, CTOP, completely abolished galanin-induced feeding in sated rats whereas the kappa-opioid receptor antagonist, nor-BNI, had no effect on galanin-induced feeding. Neither CTOP nor nor-BNI alone produced any change in food

The mechanisms of protective action of the neuropeptide galanin and its N-terminal fragments against myocardial ischaemia/reperfusion (I/R) injury remain obscure. The aim of this work was to study effects of a novel peptide agonist of galanin receptors [βAla14, His15]-galanin (2-15) (G1) and the full-length galanin (G2) on energy and antioxidant status of the heart with acute infarction. The peptides were synthesized by the automatic solid phase method using Fmoc technology. Their structure was identified by 1 H-NMR spectroscopy and MALDI-TOF mass spectrometry. Experiments were performed on an

The present study was performed to explore the role of galanin and galanin receptor 2 in nociceptive modulation in anterior cingulate cortex (ACC) of normal rats and rats with mononeuropathy. Intra-ACC injection of galanin induced significant increases in hindpaw withdrawal latencies (HWLs) to thermal and mechanical stimulations in both normal rats and rats with mononeuropathy, the increased HWLs were attenuated significantly by intra-ACC injection of galanin receptor 2 antagonist M871, indicating an involvement of galanin receptor 2 in nociceptive modulation in ACC. Interestingly, the galanin

Galanin (Gal), a bioactive neuropeptide, is widely distributed throughout the central nervous system and has diverse modulatory effects. To understand the central effect of this training-stimulatory peptide on insulin sensitivity, its antagonist M35 was injected into the cerebral ventricle in type 2 diabetic rats. A treadmill running of the rats was used to stimulate circulating Gal secretion and central Gal mRNA expression. The results showed that M35 significantly decreased glucose infusion rates in euglycemic-hyperinsulinemic clamp tests as well as 2-deoxy-[(3) H]D-glucose uptake and peroxi

High affinity binding sites for galanin are identified and characterized in membranes from a hamster pancreatic beta-cell tumor. Using the radioiodinated peptide [125I] galanin, interaction of the peptide with pancreatic membranes is shown to be saturable, reversible, and time, temperature, membrane protein concentration, pH, and ionic strength dependent. In optimized equilibrium conditions of binding (90 min at 10 C), native galanin competitively inhibits the binding of [125I]galanin in a dose-dependent manner (from 10(-11)-10(-8) M); half-maximal inhibition is induced by 1 nM peptide. Identifying non-addictive opioid medications is a high priority in medical sciences, but μ-opioid receptors mediate both the analgesic and addictive effects of opioids. We found a significant pharmacodynamic difference between morphine and methadone that is determined entirely by heteromerization of μ-opioid receptors with galanin Gal1 receptors, rendering a profound decrease in the potency of methadone. This was explained by methadone's weaker proficiency to activate the dopaminergic system as compared to morphine and predicted a dissociation of therapeutic versus euphoric effects of methadon

Uterine inflammation is a very common and serious pathology in domestic animals, the development and progression of which often result from disturbed myometrial contractility. We investigated the effect of inflammation on the protein expression of galanin (GAL) receptor subtypes (GALR)1 and GALR2 in myometrium and their role in the contractile amplitude and frequency of an inflamed gilt uterus. The gilts of the E. coli and SAL groups received E. coli suspension or saline in their uteri, respectively, and only laparotomy was performed (CON group). Eight days later, the E. coli group developed s

Galanin was first identified 30 years ago as a "classic neuropeptide," with actions primarily as a modulator of neurotransmission in the brain and peripheral nervous system. Other structurally-related peptides-galanin-like peptide and alarin-with diverse biologic actions in brain and other tissues have since been identified, although, unlike galanin, their cognate receptors are currently unknown. Over the last two decades, in addition to many neuronal actions, a number of nonneuronal actions of galanin and other galanin family peptides have been described. These include actions associated with

The neuropeptide galanin receptor 3 (GALR3) is a class A G protein-coupled receptor (GPCR) broadly expressed in the nervous system, including the retina. GALR3 is involved in the modulation of immune and inflammatory responses. Tight control of these processes is critical for maintaining homeostasis in the retina and is required to sustain vision. Here, we investigated the role of GALR3 in retina pathologies triggered by bright light and P23H mutation in the rhodopsin (RHO) gene, associated with the activation of oxidative stress and inflammatory responses. We used a multiphase approach involv

Galanin is a 29 amino acid neuropeptide that is widely distributed in the nervous system and acts by binding to three G protein-coupled receptors (GalR1, GalR2 and GalR3). In the literature, the presence of galanin has been reported in nerve fibers innervating the rat carotid body; however, direct evidence of the different galanin receptor subtypes expressed in carotid body cells has yet to be provided. In the present study, we investigated the presence and location of the three galanin receptor subtypes in 12 rat carotid bodies through real-time polymerase chain reaction (PCR) and immunohisto

Using in situ hybridization the regulation of mRNA encoding the galanin receptor R1 was investigated in the mutant Brattelboro (diabetes insipidus) rat. We here report an increase of the galanin receptor R1 mRNA levels in the hypothalamic supraoptic and paraventricular nuclei of the mutant strains. The increase seemed to be confined to magnocellular neurons, since no changes were detected in galanin receptor R1 mRNA levels in the extra-hypothalamic nucleus of the olfactory tract. The results confirm that osmotic stimulation induces up-regulation of galanin receptor R1 mRNA levels. This may inc

A large number of neuroanatomical, neurophysiologic, and neurochemical mechanisms are thought to contribute to the development and maintenance of neuropathic pain. However, mechanisms responsible for neuropathic pain have not been completely delineated. It has been demonstrated that neuropeptide galanin (Gal) is upregulated after injury in the dorsal root ganglion (DRG) and spinal dorsal horn (SDH) where it plays a predominantly antinociceptive role. In the present study, sciatic nerve-pinch injury rat model was used to determine the effects of exogenous Gal on the expression of the Gal and it

Recent studies have proposed that heteromers of µ-opioid receptors (MORs) and galanin Gal1 receptors (Gal1Rs) localized in the mesencephalon mediate the dopaminergic effects of opioids. The present study reports converging evidence, using a peptide-interfering approach combined with biophysical and biochemical techniques, including total internal reflection fluorescence microscopy, for a predominant homodimeric structure of MOR and Gal1R when expressed individually, and for their preference to form functional heterotetramers when co-expressed. Results show that a heteromerization-dependent cha
