Biochemical studies on muscarinic receptors.

Muscarinic receptors have been characterized in smooth muscle and brain by the binding of reversible (e.g. atropine, quinuclidinylbenzylate) or irreversible (benzilylcholine or propylbenzilylcholine mustards) ligands. There is a close correlation between affinity constants derived from binding experiments and the affinities of muscarinic ligands for these sites obtained in pharmacological experiments on smooth muscle. Whereas atropine shows a single high affinity binding component (in subcellular preparations) several other ligands (QNB, ACh, oxotremorine) show multiple affinity binding. This

Muscarinic receptors: what we know.

An understanding of muscarinic receptors is tantamount to an understanding of overactive bladder. The M(3) muscarinic receptor subtype is responsible for detrusor smooth muscle contraction and it exerts an exocrine function in the salivary glands. Alterations in the receptor's response to acetylcholine as a result of injury may lead to hypersensitivity and overactivity. The M(2) receptor subtype, which is mainly responsible for cardiac function, is the muscarinic receptor of highest proportion in the detrusor. M(2) also may play a role in detrusor contraction in injury and pathologic states. M

On the muscarinic receptors in the urinary bladder and the putative subclassification of muscarinic receptors.

The muscarinic cholinergic receptors in the urinary bladders of man, guinea pig, rat and rabbit were studied by means of a receptor binding technique, with l-quinuclidinyl [phenyl 4-3H]benzilate, (-)3H-QNB, as radioligand. The potential role of the receptors in the supersensitivity of the rat bladder to muscarinic agonists, following parasympathetic denervation, hypertrophy and urinary diversion, was also investigated. In addition, the binding of various unlabelled antimuscarinic drugs in the guinea pig bladder was compared to that in other tissues in order to study the putative muscarinic rec

CNS muscarinic receptors and muscarinic receptor agonists in Alzheimer disease treatment.

This review explores the main aspects that form the basis of the cholinergic-oriented treatment of Alzheimer disease. Muscarinic acetylcholine receptor subtypes in the brain and periphery are discussed. It includes a new and updated overview of the involvement of muscarinic receptors in Alzheimer disease and the recent development of new and highly selective M1 muscarinic receptor agonists with disease-modifying potential. Activation of the M1 muscarinic receptor is a rational therapeutic strategy for the treatment of schizophrenia and Alzheimer disease, as this receptor plays a pivotal role i

Targeting muscarinic receptors for treating schizophrenia.

Schizophrenia is a chronic, disabling and potentially fatal psychiatric syndrome characterized by three primary symptom domains: positive, negative, and cognitive symptoms, for which current dopamine D2 receptor antagonists provide only partial benefit and are limited by significant side effects. Muscarinic acetylcholine receptors (mAChRs), broadly expressed across cortical, striatal, and midbrain circuits, have emerged as promising targets for next-generation therapies. Among these, M1 and M4 receptor subtypes play key roles in regulating glutamatergic and dopaminergic transmission. Clinical

Muscarinic receptors in the ocular surface.

PURPOSE OF REVIEW: A wide distribution of the eye's muscarinic receptor system has been found and several roles for the muscarinic system in the eye proposed, although functional consequences of muscarinic receptor activation are not always fully characterized. The present paper reviews current knowledge about the presence of muscarinic receptors in the ocular surface and the implication of alterations on their expression and/or functioning in eye diseases. RECENT FINDINGS: Several authors have described alterations in muscarinic receptor subtype expression in the eye under pathological condi

Evidence of paired M2 muscarinic receptors.

Binding assays involving various antagonists, including N-[3H] methylscopolamine, [3H]quinuclidinyl benzilate, AFDX-116, pirenzepine, and propylbenzilylcholine mustard, disclosed only a single population of M2 muscarinic receptors in membranes from the rat "brainstem" (medulla, pons, and colliculi). However, competition curves between N-[3H]methylscopolamine and various agonists, including oxotremorine, cis-dioxolane, and acetylethylcholine mustard, showed approximately equal numbers of guanine nucleotide-sensitive high affinity (H) sites and guanine nucleotide-insensitive low affinity (L) sit

Targeting muscarinic receptors to treat schizophrenia.

Schizophrenia is a severe neuropsychiatric disorder characterized by a diverse range of symptoms that can have profound impacts on the lives of patients. Currently available antipsychotics target dopamine receptors, and while they are useful for ameliorating the positive symptoms of the disorder, this approach often does not significantly improve negative and cognitive symptoms. Excitingly, preclinical and clinical research suggests that targeting specific muscarinic acetylcholine receptor subtypes could provide more comprehensive symptomatic relief with the potential to ameliorate numerous sy

Muscarinic acetylcholine receptors and airway diseases.

Parasympathetic nerves provide the dominant autonomic innervation of the airways. Release of acetylcholine from parasympathetic nerves activates postjunctional muscarinic receptors present on airway smooth muscle, submucosal glands, and blood vessels to cause bronchoconstriction, mucus secretion, and vasodilatation, respectively. Acetylcholine also feeds back onto prejunctional muscarinic receptors to enhance or inhibit further acetylcholine release. In asthma and chronic obstructive pulmonary disease, bronchoconstriction and mucus secretion is increased and the airways are hyperresponsive to

Muscarinic receptors in diabetic rat prostate.

To investigate the effects of experimentally-induced diabetes on prostatic muscarinic cholinergic receptors, the binding characteristics of [3H]quinuclidinyl benzilate ([3H]QNB) to prostatic membrane particulates were examined in four groups of rats: control, diabetic, diabetic insulin treated, and diabetic myo-inositol treated. Diabetes was induced by i.v. injection of streptozotocin (STZ), 65 mg/kg. Diabetic and diabetic myo-inositol-treated rats had hyperglycemia, hypoinsulinemia, glucosuria, polydipsia, and polyuria as well as significantly smaller prostates and lower body weights compared

Muscarinic acetylcholine receptors in the urinary tract.

Muscarinic receptors comprise five cloned subtypes, encoded by five distinct genes, which correspond to pharmacologically defined receptors (M(1)-M(5)). They belong to the family of G-protein-coupled receptors and couple differentially to the G-proteins. Preferentially, the inhibitory muscarinic M(2) and M(4) receptors couple to G(i/o), whereas the excitatory muscarinic M(1), M(3), and M(5) receptors preferentially couple to G(q/11). In general, muscarinic M(1), M(3), and M(5) receptors increase intracellular calcium by mobilizing phosphoinositides that generate inositol 1,4,5-trisphosphate (I

Allosteric binding sites on muscarinic acetylcholine receptors.

In this issue of Molecular Pharmacology, Tränkle et al. (p. 1597) present new findings regarding the existence of a second allosteric site on the M2 muscarinic acetylcholine receptor (M2 mAChR). The M2 mAChR is a prototypic class A G protein-coupled receptor (GPCR) that has proven to be a very useful model system to study the molecular mechanisms involved in the binding of allosteric GPCR ligands. Previous studies have identified several allosteric muscarinic ligands, including the acetylcholinesterase inhibitor tacrine and the bis-pyridinium derivative 4,4'-bis-[(2,6-dichloro-benzyloxy-imino)

Functional muscarinic receptors in cultured skeletal muscle.

We studied the influence of muscarinic and nicotinic stimulation on both phosphoinositide metabolism and intracellular calcium levels in rat skeletal muscle primary cultures. Both nicotine and muscarine induced an increase in cytosolic calcium measured by fluo 3 fluorescence in confocal microscopy. The mass of inositol (1,4,5)trisphosphate measured by radioreceptor assay rose 2- to 3.5-fold upon carbachol, nicotine, or muscarine stimulation. The muscarine effect was mimicked by oxotremorine-M; pirenzepine prevented the muscarine-induced inositol (1,4,5)trisphosphate increase, whereas 4-dipheny

Muscarinic cholinergic receptors in developing rat lung.

Muscarinic cholinergic receptors were identified and partially characterized in crude membrane fractions of rat lung and trachea from day 17 of gestation to adulthood using (-)-[3H]quinuclidinyl benzilate (QNB). (-)-[3H]QNB binding to rat lung membrane was characteristic of muscarinic cholinergic receptor sites. Binding capacity of muscarinic receptors sites was relatively low in rat lung compared to that in other tissues. The number of (-)-[3H]QNB-binding sites (binding capacity) decreased progressively and significantly from 79 +/- 8 fmol X mg-1 protein on days 17-18 of gestation to 21 +/- 3

The Novel Functions of M4 Muscarinic Receptors.

The role of M4 muscarinic receptors in locomotor activity regulation remains controversial, with conflicting findings hampering our understanding of movement disorders. This uncertainty is further complicated by the unexplored relationship between M4 receptors and biological rhythms, as well as potential sex-specific effects that may explain previous inconsistent results. Through systematic investigation of locomotor functions in biological rhythm paradigms, we discovered that M4 muscarinic receptors significantly modulate locomotor activity rhythms via brain pacemaker mechanisms. Notably, thi

Affinity of muscarinic receptor antagonists for three putative muscarinic receptor binding sites.

1. A range of muscarinic receptor antagonists were examined for affinity at the M1 muscarinic binding site, present in rat cerebrocortical membranes and the M2 muscarinic binding sites of rat cardiac and submaxillary gland membranes. 2. The results obtained were consistent with the presence of three classes of muscarinic binding site. 3. Both the M1 binding site, labelled by [3H]-pirenzepine ([3H]-Pir) in rat cerebrocortical membranes, and the M2 gland binding site, labelled by [3H]-N-methyl scopolamine ([3H]-NMS) in rat submaxillary gland membranes, displayed higher affinity for pirenzepine,

Intramolecular interactions in muscarinic acetylcholine receptors studied with chimeric m2/m5 receptors.

Current models of the three-dimensional structures of muscarinic acetylcholine receptors and other G protein-coupled receptors are based primarily on high-resolution electron diffraction data obtained with bacteriorhodopsin, the molecular structure of which is characterized by the presence of seven alpha-helical transmembrane domains (TM I-VII). However, bacteriorhodopsin does not couple to G proteins and its primary sequence lacks a series of amino acids that are conserved among virtually all G protein-coupled receptors. Therefore, it remains to be shown experimentally whether the molecular s

Ethanol inhibits neuritogenesis induced by astrocyte muscarinic receptors.

In utero alcohol exposure can lead to fetal alcohol spectrum disorders, characterized by cognitive and behavioral deficits. In vivo and in vitro studies have shown that ethanol alters neuronal development. We have recently shown that stimulation of M(3) muscarinic receptors in astrocytes increases the synthesis and release of fibronectin, laminin, and plasminogen activator inhibitor-1, causing neurite outgrowth in hippocampal neurons. As M(3) muscarinic receptor signaling in astroglial cells is strongly inhibited by ethanol, we hypothesized that ethanol may also inhibit neuritogenesis in hippo

Positive and negative allosteric interactions on muscarinic receptors.

It was previously found that alcuronium increases the binding of [3H]methyl-N-scopolamine to cardiac muscarinic receptors by a positive allosteric action while its effect on the binding of [3H]quinuclidinyl benzilate is negative. The, features of the antagonist's molecule which decide whether its allosteric interaction with alcuronium is positive or negative are not known.
