Amyotrophic lateral sclerosis: alterations in neurotransmitter receptors.

Loss of motor neurons is the primary pathological hallmark of amyotrophic lateral sclerosis. Drug and neurotransmitter receptors are neuronal markers and can be indicators of neuronal connectivity. Knowledge of alterations in receptors in amyotrophic lateral sclerosis should contribute to our understanding of normal spinal cord neurotransmitter systems as well as of the pathophysiology of amyotrophic lateral sclerosis. We therefore used a sensitive, light microscopic in vitro labeling receptor autoradiographic technique to map and quantitate muscarinic cholinergic, glycinergic, and benzodiazep

Possible involvement of neuropeptide and neurotransmitter receptors in Adenomyosis.

BACKGROUND: Accumulating data indicate that sensory nerve derived neuropeptides such as substance P and calcitonin gene related-protein (CGRP) can accelerate the progression of endometriosis via their respective receptors, so can agonists to their respective receptors receptor 1 (NK1R), receptor activity modifying protein 1 (RAMP-1) and calcitonin receptor-like receptor (CRLR). Adrenergic β2 receptor (ADRB2) agonists also can facilitate lesional progression. In contrast, women with endometriosis appear to have depressed vagal activity, concordant with reduced expression of α7 nicotinic acetylc

Developmental profiles of neurotransmitter receptors in respiratory motor nuclei.

We discuss the time course of postnatal development of selected neurotransmitter receptors in motoneurons that innervate respiratory pump and accessory respiratory muscles, with emphasis on other than classic respiratory signals as important regulatory factors. Functions of those brainstem motoneurons that innervate the pharynx and larynx change more dramatically during early postnatal development than those of spinal respiratory motoneurons. Possibly in relation to this difference, the time course of postnatal expression of distinct receptors for serotonin differ between the hypoglossal (XII)

Neurotransmitter receptors in the life and death of oligodendrocytes.

Oligodendrocytes are crucial to the function of the mammalian brain: they increase the action potential conduction speed for a given axon diameter and thus facilitate the rapid flow of information between different brain areas. The proliferation and differentiation of developing oligodendrocytes, and their myelination of axons, are partly controlled by neurotransmitters. In addition, in models of conditions like stroke, periventricular leukomalacia leading to cerebral palsy, spinal cord injury and multiple sclerosis, oligodendrocytes are damaged by glutamate and, contrary to dogma, it has rece

Astrocytic neurotransmitter receptors (astro‑glioreceptors) and their role in neuroplasticity.

Astrocytes express a set of neurotransmitter receptors (glioreceptors) that enable them to regulate synaptic transmission and neuroplasticity, and to function as integral partners in synaptic signaling and the modification of neural circuits. This review presents the current understanding of how glioreceptors on astrocytes (astro‑gliorecptors) mediate bidirectional communication between neurons and glia across major neurotransmitter systems. The review focuses on receptors for glutamate, GABA, acetylcholine, monoamines, neuropeptides, opioids, and purines. Through these receptors, astrocytes c

Altered expression of neurotransmitter receptors and neuromediators in vernal keratoconjunctivitis.

BACKGROUND: There is growing evidence that autonomic innervation is involved in the pathogenesis of mucus hypersecretion, goblet cell hyperplasia, and conjunctival hyperreactivity. OBJECTIVE: To determine the expression of neurotransmitters and neurotransmitter receptors in vernal keratoconjunctivitis (VKC) tissues to evaluate whether neurogenic inflammation plays a role in this ocular atopic-related disorder. METHODS: Biopsy specimens of upper tarsal conjunctiva from 8 VKC patients with active inflammation and from 4 healthy subjects were processed for immunohistochemistry using anti-M1, an

Autoantibodies against four kinds of neurotransmitter receptors in psychiatric disorders.

There is a hypothesis that autoimmune abnormalities in neurotransmitter receptors might cause some psychiatric disorders. Using a sensitive radioligand assay, we detected serum autoantibodies to recombinant human muscarinic cholinergic receptor 1 (CHRM1, 34.4%), mu-opioid receptor (OPRM1, 13.1%), 5-hydroxytryptamine receptor 1A (HTR1A, 7.4%), and dopamine receptor D2 (DRD2, 4.9%) in 122 psychiatric patients. Positive antibodies to CHRM1 were found in 34.1%, 34.9%, 33.3%, and 9.1% of patients with schizophrenic disorders (n=44), mood disorders (n=63), other psychiatric disorders (n=15) and auto

Glycine receptors support excitatory neurotransmitter release in developing mouse visual cortex.

Glycine receptors (GlyRs) are found in most areas of the brain, and their dysfunction can cause severe neurological disorders. While traditionally thought of as inhibitory receptors, presynaptic-acting GlyRs (preGlyRs) can also facilitate glutamate release under certain circumstances, although the underlying molecular mechanisms are unknown. In the current study, we sought to better understand the role of GlyRs in the facilitation of excitatory neurotransmitter release in mouse visual cortex. Using whole-cell recordings, we found that preGlyRs facilitate glutamate release in developing, but no

Activation of membrane receptors by neurotransmitter released from temperature-sensitive hydrogels.

The present paper describes the design, construction and testing of a temperature-sensitive N-isopropylacrylamide hydrogel device for studying the controlled presentation of gamma-aminobutyric acid (GABA) to GABA(C) membrane receptors expressed in Xenopus laevis oocytes. Upon temperature lowering, the GABA-loaded hydrogel positioned near the surface of the GABA(C)-expressing oocyte elicits a membrane current response resembling that induced by superfusion of the oocyte with free GABA. The response to cooling is not observed when GABA is omitted from the hydrogel loading solution. In addition,

Neurotransmitter receptors in the proliferative zones of the developing primate occipital lobe.

Film autoradiography was used to investigate the expression of several neurotransmitter receptor subtypes in the transient ventricular and subventricular proliferative zones of the developing occipital lobe in two groups of macaque monkey fetuses. The first group of fetuses were between 60 and 93 days after conception (E60-E93), when the ventricular and subventricular zones of the monkey occipital lobe produce neurons destined for the visual cortex. In the second group, fetuses were between E107 and E128, after generation of cortical neurons has ceased. In the E60-E93 group of fetuses, ventric

The excitatory neurotransmitter glutamate and its receptors: the culprits behind neuropathic pain?

Human understanding of the nature of pain has undergone a profound shift from mechanical reductionism to a multidimensional, integrative approach. Among the many types of chronic pain, neuropathic pain (NP) has long been a key focus of research in the fields of neurobiology and pain medicine, owing to its high prevalence and complex mechanisms. Its core pathological feature is an abnormal increase in neuron excitability caused by peripheral and central sensitization. As the most important excitatory neurotransmitter in the nervous system, glutamate plays an irreplaceable and central role in si

Hallucinogenic and neuroleptic drug interactions with potential neurotransmitter receptors in parasitic nematodes.

Receptors potentially involved in neurotransmitting have been characterised in the muscle tissue and in whole worms of the nematodes Ascaris suum and Onchocerca volvulus, respectively. Binding studies revealed a high affinity for LSD with apparent KD values of 94 nM for A. suum and 120 nM for O. volvulus, whereas those of the neuroleptics haloperidol, spiperone and mianserin were found to be in the micromolar range. A variety of neurotransmitter antagonists, known to bind with high affinities either to mammalian D1/2 or to 5-HT1/2 receptors, were tested for their ability to bind to the nematod

Neurotransmitter receptor alterations in hepatic encephalopathy: a review.

Hepatic encephalopathy (HE), a complex neuropsychiatric syndrome with symptoms ranging from subtle neuropsychiatric and motor disturbances to deep coma and death, is thought to be a clinical manifestation of a low-grade cerebral oedema associated with an altered neuron-astrocyte crosstalk and exacerbated by hyperammonemia and oxidative stress. These events are tightly coupled with alterations in neurotransmission, either in a causal or a causative manner, resulting in a net increase of inhibitory neurotransmission. Therefore, research focussed mainly on the potential role of γ-aminobutyric aci

Evidence for glutamate as the olfactory receptor cell neurotransmitter.

1. Synaptic transmission between olfactory receptor neurons and mitral/tufted cells was examined using a whole-cell recording technique in a hemisected preparation of the turtle olfactory bulb. To determine the olfactory receptor neuron transmitter, we isolated components of the synaptic response of mitral/tufted cells to olfactory nerve stimulation using postsynaptic receptor antagonists. 2. Low-intensity stimulation of the olfactory nerve evoked monosynaptic excitatory postsynaptic potentials in mitral/tufted cells that consisted of a rapid and prolonged depolarization with little contributi

Fentanyl but not Morphine Interacts with Nonopioid Recombinant Human Neurotransmitter Receptors and Transporters.

Synthetic opioids, including fentanyl and its analogs, have therapeutic efficacy in analgesia and anesthesia. However, their illicit use in the United States has increased and contributed to the number one cause of death for adults 18-50 years old. Fentanyl and the heroin metabolite morphine induce respiratory depression that can be treated with the μ opioid receptor (MOR) antagonist naloxone. With higher or more rapid dosing, fentanyl, more than morphine, causes chest wall rigidity and can also induce rapid onset laryngospasm. Because non-MORs could mediate differing clinical manifestations,

Daily Profiles of Neuropeptides, Catecholamines, and Neurotransmitter Receptors in the Chicken Pineal Gland.

The avian pineal gland is one of three central biological clocks that contain all the components of a circadian system: a photoreceptive input, oscillator, and rhythmically secreted melatonin (MEL) as an effector. The biosynthesis of MEL is regulated by the neurotransmitters noradrenaline (NA), vasoactive intestinal peptide (VIP), and pituitary adenylate cyclase-activating polypeptide (PACAP). The aim of the present study was to characterize the daily profile of neurotransmitters and their receptors in the pineal gland of male Hy-Line chickens housed under controlled light (12:12 light:dark) c
