[Prolactin receptors and breast cancer].

A study has been carried out on 116 mammary neoplasias in which the prolactin receptors together with other clinical and hormonal parameters have been determined. A 46.3% frequency of PRLR+ has been obtained as well as a statistically significant correlation between these and blood estradiol (p less than 0.05). PRLR- have greater survival and PRLR may act as a growth factor.

Prolactin: structure, receptors, and functions.

Prolactin (PRL) is a 23-kDa protein synthesized and secreted by lactotroph cells of the anterior pituitary gland but also by other peripheral tissues. PRL binds directly to a unique transmembrane receptor (PRLR), and the JAK2/signal transducer and activator of transcription 5 (Stat5) pathway is considered the major downstream pathway for PRLR signaling. To a lesser extent, PRL may be cleaved into the shorter 16-kDa PRL, also called vasoinhibin, whose signaling is not fully known. According to rodent models of PRL signaling inactivation and the identification of human genetic alterations in PRL

Prolactin receptors and the immune system.

The existence of a physiological immunoneuroendocrine network clearly contributing to homeostasis has now been demonstrated. In this context, nervous, endocrine, and immune systems communicate with each other, using common mediators and respective receptors. An interesting aspect of this network involves the interactions between prolactin (PRL) and the immune system. Prolactin plays a significant role in regulation of the humoral and cellular immune responses in physiological as well as pathological situations, such as autoimmune diseases. This role is exerted via the existence of specific rec

Role of Prolactin Receptors in Lymphangioleiomyomatosis.

Pulmonary lymphangioleiomyomatosis (LAM) is a rare lung disease caused by mutations in the tumor suppressor genes encoding Tuberous Sclerosis Complex (TSC) 1 and TSC2. The protein product of the TSC2 gene is a well-known suppressor of the mTOR pathway. Emerging evidence suggests that the pituitary hormone prolactin (Prl) has both endocrine and paracrine modes of action. Here, we have investigated components of the Prl system in models for LAM. In a TSC2 (+/-) mouse sarcoma cell line, down-regulation of TSC2 using siRNA resulted in increased levels of the Prl receptor. In human LAM cells, the P

Prolactin receptors in human breast cancer.

Prolactin receptors have been measured in 92 human breast carcinomas. Both free and total receptors (after desaturation by MgCl2) have been looked for. Free receptors have been found in 46% of the cases, total receptors in 72%. Specific binding ranges from 0.8 to 8.0%. No correlation could be found between prolactin receptors and age, weight, menopausal status and pathological features (differentiation, histoprognostic grading, cellular density). A highly significant correlation has been found between prolactin receptors on the one hand and estradiol and progesterone receptors on the other.

Expression of prolactin receptors in rat reproductive tissues during periovulatory prolactin imbalance.

Indirect immunoperoxidase technique in combination with cytophotometry showed that prolactin imbalance in the periovulatory period is characterized by considerable changes in the expression of prolactin receptors in uterine glands and endometrial stroma (but not in the myometrium) and in the pituitary gland and hypothalamus, while receptor compartmentalization pattern remains unchanged.

Prolactin receptor expression in the gastrointestinal tract: characterization of the prolactin receptor of gastric mucosa.

There is evidence that prolactin (PRL) influences gastrointestinal function. However, the sites at which prolactin exerts these effects are not known. A monoclonal antibody was therefore generated against the rabbit mammary gland prolactin receptor (MAb 218) and used to study the distribution of the prolactin receptor in the rabbit gastrointestinal tract (GIT) by immunohistochemistry. MAb 218 is an IgG1 kappa-precipitating antibody which precipitates major affinity cross-linked mammary gland prolactin receptor subunits of molecular masses 45 and 80 kDa. It has an affinity of 0.8 x 10(9) mol/l

Human Cytomegalovirus Infection Induces High Expression of Prolactin and Prolactin Receptors in Ovarian Cancer.

One of the potential biomarkers for ovarian cancer patients is high serum level of prolactin (PRL), which is a growth factor that may promote tumor cell growth. The prolactin receptor (PRLR) and human cytomegalovirus (HCMV) proteins are frequently detected in ovarian tumor tissue specimens, but the potential impact of HCMV infection on the PRL system have so far not been investigated. In this study, HCMV's effects on PRL and PRLR expression were assessed in infected ovarian cancer cells (SKOV3) by PCR and Western blot techniques. The levels of both PRL and PRLR transcripts as well as the corre

[Progress in prolactin receptor research].

Prolactin (PRL) is secreted by lactotrophs in the anterior pituitary and some extra-pituitary tissues such as breast, lacrimal gland, uterus, thymus and spleen, etc. Since PRL is closely related to growth hormone (GH) and placental lactogens (PL), it has been broadly accepted that PRL, GH and PL are resulted from the duplication of an ancestral gene. PRL regulates hundreds of biological functions by endocrine, paracrine and autocrine manners. Prolactin initiates its effects by binding to its receptor (PRLR). PRLR belongs to the class I cytokine receptor superfamily. Up to now, three membrane--

Prolactin and prolactin receptor expression in cervical intraepithelial neoplasia and cancer.

Prolactin receptor (PRLR) overexpression could play a role in tumorigenesis. The aim of this study was to determine prolactin (PRL) and PRLR expression in biopsies from patients with precursor lesions and uterine cervical cancer. PRLR expression was analyzed in 63 paraffin-embedded biopsies of uterine cervical tissue. In total, eleven low-grade squamous intraepithelial lesions (LSIL), 23 high-grade squamous intraepithelial lesions (HSIL), 21 uterine cervical cancers (UCC) and 8 normal epithelium (NE) were examined using immunoperoxidase staining and Western blot analysis. Additionally, PRL exp

Opiates, prolactin, and the dopamine receptor.

The administration of a dopamine antagonist, chlorpromazine, and two opiates, morphine and methadone, resulted in a significant rise in serum PRL within 90-150 min. Prior administration of dopamine receptor agonists (apomorphine, levodopa, aand bromocriptine) blocked this effect. In contrast, cyproheptadine, a serotonin antagonist, did not. We suggest that the opiates induce hyperprolactinemia in man via dopamine receptor blockade.

Effect of oxidizing agents on rabbit mammary prolactin receptors.

Treatment of rabbit mammary membrane-bound and solubilized prolactin receptors with the oxidizing agents N-chlorobenzene sulfonamide and N-bromosuccinimide resulted in total inactivation of the ability of the receptor to bind prolactin. A similar inactivation was obtained with 2-hydroxy-5-nitrobenzylbromide. The results suggest the possible importance of tryptophan in maintaining the activity of the prolactin receptor.

Cellular distribution of prolactin receptors in human digestive tissues.

In the present study we analyzed the expression of prolactin receptors (PRLR) in human digestive tissues by immunohistochemistry. PRLR immunoreactivity was primarily localized in the cytoplasm. However, in some organs (liver and salivary glands) a nuclear positivity was also found. The liver was used as control and showed a diffuse immunostaining in the parenchymal cells. In the gastrointestinal tract, PRLR immunoreactivity was observed in the mucosa, muscularis layer, and nervous plexuses. The more intense immunostaining in the mucosa of the different segments was as follows: esophagus, super

Prolactin receptor antagonism in mouse anterior pituitary: effects on cell turnover and prolactin receptor expression.

Since anterior pituitary expresses prolactin receptors, prolactin secreted by lactotropes could exert autocrine or paracrine actions on anterior pituitary cells. In fact, it has been observed that prolactin inhibits its own expression by lactotropes. Our hypothesis is that prolactin participates in the control of anterior pituitary cell turnover. In the present study, we explored the action of prolactin on proliferation and apoptosis of anterior pituitary cells and its effect on the expression of the prolactin receptor. To determine the activity of endogenous prolactin, we evaluated the effect

Hypothalamo-pituitary regulation of hepatic prolactin receptors in the rat.

The number of specific binding sites for 125I-labeled human prolactin (hPrl) in crude membrane fractions from rat liver is sexually differentiated. Livers from female rats specifically bound much more of added [125I]hPrl than male liver membrane fractions which bound little, if any, Prl. Hypophysectomy of female animals eliminated hepatic binding of Prl; the effect was reversed by a pituitary implant under the kidney capsule. Treatment of male rats with estradiol valerate induced hepatic hPrl receptors, but only in the presence of an intact pituitary. Anterior hypothalamic deafferentation at t

Sex-specific Regulation of Prolactin Secretion by Pituitary Bradykinin Receptors.

Sex differences in the control of prolactin secretion are well documented. Sex-related differences in intrapituitary factors regulating lactotroph function have recently attracted attention. Sex differences in prolactinoma development are well documented in clinic, prolactinomas being more frequent in women but more aggressive in men, for poorly understood reasons. Kallikrein, the enzyme releasing kinins has been found in the pituitary, but there is no information on pituitary kinin receptors and their function. In the present work, we characterized pituitary bradykinin receptors (BRs) at the

Dopaminergic receptors in human prolactin-secreting adenomas: a quantitative study.

Dopaminergic receptors were observed on the membranes of 26 human PRL-secreting adenomas. Two binding sites were found for [3H]domperidone, a selective dopamine antagonist, with a Kd1 of 0.29 +/- 0.06 nM and a Kd2 of 4.19 +/- 0.7 nM (n = 5). Bmax1 and Bmax2 (maximum numbers of binding sites, first and second sites, respectively) varied from one adenoma to another. When considering Bmax1, two different categories of PRL-secreting adenomas were distinguished, one with a concentration of receptor over 250 fmol/mg protein and others with lower concentrations of receptor (< 250 fmol/mg protein). Variant Prolactin Receptor in Agalactia and Hyperprolactinemia.

A loss-of-function variant in the gene encoding the prolactin receptor ( PRLR) was reported previously in a woman with persistent postpartum galactorrhea; however, this paradoxical phenotype is not completely understood. Here we describe a 35-year-old woman who presented with idiopathic hyperprolactinemia that was associated with a complete lack of lactation after each of her two deliveries. She is a compound heterozygote for loss-of-function variants of PRLR. Her unaffected parents are heterozygotes. These findings are consistent with previous work showing that mice deficient in functional Pr

Modulation of prolactin receptors in the rat hypothalamus in response to changes in serum concentration of endogenous prolactin or to ovine prolactin administration.

Specific binding of 125I-labeled rat prolactin (125I-rat PRL) to hypothalamic membranes was studied in Sprague-Dawley rats after ovine PRL administration and in relation to rat PRL serum variations induced by ectopic pituitary implants or by drugs which stimulate (domperidone) or inhibit (bromocriptine) PRL release. Repeated treatments with ovine PRL markedly increased specific binding values of 125I-rat PRL to hypothalamic membranes of female rats. Repeated treatments with domperidone also increased specific PRL binding in the hypothalamus. This effect was associated with an increase in PRL s

Mutant prolactin receptor and familial hyperprolactinemia.

Hyperprolactinemia that is not associated with gestation or the puerperium is usually due to tumors in the anterior pituitary gland and occurs occasionally in hereditary multiple endocrine neoplasia syndromes. Here, we report data from three sisters with hyperprolactinemia, two of whom presented with oligomenorrhea and one with infertility. These symptoms were not associated with pituitary tumors or multiple endocrine neoplasia but were due to a heterozygous mutation in the prolactin receptor gene, PRLR, resulting in an amino acid change from histidine to arginine at codon 188 (His188Arg).
