Normal somatomedin and somatomedin receptors in achondroplastic dwarfism.

It has been proposed that the basic abnormality in achondroplasia may be a quantitative defect in endochondral new bone formation secondary to decreased synthesis of somatomedin (SM) or abnormal binding of SM to specific receptors. To test this hypothesis, we have measured plasma SM levels and SM receptors on circulating mononuclear cells obtained from 5 achondroplastic dwarfs and 5 age-matched controls. Plasma SM levels were 0.82 +/- 0.14 U/ml (mean +/- S.E.M.) for the achondroplastic dwarfs and 0.90 +/- 0.12 U/ml for the controls. The specific binding of 125I-SM to 50 x 10(6) mononuclear cel

Somatomedin/insulin-like growth factor tissue receptors.

There are two types of Sm/IGF receptors based on results of competitive binding experiments and investigations of receptor structure. The type I receptor preferentially interacts with IGF I rather than IGF II and interacts weakly with insulin. This receptor has a binding subunit of Mr = 130 000 which is disulphide bonded to form larger structures of Mr greater than 300 000. The type II receptor prefers IGF II to IGF I and does not interact with insulin. Its binding subunit is not linked by disulphide bonds to other membrane components (Mr = 260 000 with reduction, 220 000 without reduction). S

Characterization of the human placental receptor for basic somatomedin.

We have determined optimal conditions for the solubilization of the basic somatomedin (SM) receptor from human placental membranes and for the measurement of the binding of basic SM to the solubilized receptor. Further, we have developed conditions under which the basic SM receptor, in the presence of equivalent amounts of insulin receptor, can be selectively and specifically affinity-labeled with 125I-labeled basic SM, using the cross-linking reagent disuccinimidyl suberate (DSS). Our results with these developed methods indicate that the properties of the soluble basic SM receptor (pH optimu

Structural similarities between human receptors for somatomedin C and insulin: analysis by affinity labeling.

Human placental receptors for insulin and somatomedin C (Sm-C) were affinity labeled with [125I]insulin and [125I]Sm-C by using the bifunctional cross-linking agent disuccinimidyl suberate. Analysis by sodium dodecyl sulfate-polyacrylamide gel electrophoresis demonstrated that both labeled hormones were specifically cross-linked to three protein species with apparent molecular weights of 240 000, 310 000, and 330 000. Following disulfide bond reduction, subunits of approximately 140 000 daltons were evident. Partial reduction of disulfide bonds yielded intermediate-sized species with apparent

The Brattleboro rat: normal growth hormone secretion, decreased hepatic growth hormone receptors and low plasma somatomedin activity.

Three-month-old male Brattleboro rats with hereditary diabetes insipidus (DI) present a growth defect; Brattleboro rats were studied together with age-matched Long-Evans (LE) rats. Pituitary growth hormone (GH) content was comparable in both groups of rats. Pulsatile GH release and mean 6 h GH plasma levels did not appear significantly different in chronically catheterized DI and control animals. In parallel with the growth defect, the plasma somatomedin bioactivity was significantly lower in DI than in LE rats. The specific binding of [125I]iodo-hGH to liver microsomal membranes of DI rats wa

Density-associated loss of functional receptors for somatomedin-C/insulinlike growth factor I (SM-C/IGF-I) on cultured human fibroblast monolayers.

The mitogenic activity of somatomedin-C/insulinlike growth factor-I (SM-C/IGF-I) appears to be greatly influenced by cell culture conditions, especially the presence of other growth factors and nutrients in the culture medium. To investigate the effect of cell density on SM-C/IGF-I activity, we have evaluated SM-C/IGF-I binding and stimulation of DNA synthesis and cell replication as a function of cell density in cultured human fibroblast monolayers. At fibroblast concentrations of 2.7 X 10(5) and 1.48 X 10(6) cells per 60-mm dish, specific binding of [125I]SM-C/IGF-I per 10(6) cells was 170%

Characterization and regulation of somatomedin-C/insulin-like growth factor I (Sm-C/IGF-I) receptors on cultured pig Leydig cells. Effects of Sm-C/IGF-I on luteotropin receptors and steroidogenesis.

We have characterized somatomedin-C/insulin-like growth factor I (Sm-C/IGF-I) receptors in cultured pig Leydig cells by binding and cross-linking affinity experiments. At equilibrium the dissociation constant and the number of binding sites per cell were 1.8 +/- 0.2 X 10(-9) M and 12,200 +/- 3200 respectively. Under reducing conditions, disuccinimidyl suberate cross-linked 125I-Sm-C to one receptor complex with apparent Mr = 120,000. Continuous treatment of Leydig cells with increasing concentrations of human chorionic gonadotropin (hCG) for 48 h resulted in a dose-dependent (ED50 0.05 nM) inc

[A case of leprechaunism with disorders of insulin-like growth factor-I(IGF-I)/somatomedin C(SMC) binding protein and its receptor].

Leprechaunism is a rare genetic disorder characterized by physical abnormalities, intrauterine and postnatal growth retardation, poorly developed subcutaneous fat and muscle at birth, and early death. This patient, who was a 1.5 year-old female with typical clinical features of leprechaunism, had relatively high levels of plasma GH and IGF-I/SMC but no glucose intolerance or insulin resistance. Studies were undertaken to elucidate (1) the differences among some kinds of methods for IGF-I/SMC measurement, (2) the distribution patterns of IGF-I/SMC between two kinds of its binding protein (SMBP)

Somatomedins in aging and dementia disorders of the Alzheimer type.

Serum levels of somatomedins were determined in apparently healthy aged individuals and dementia patients primarily with clinically suspected Alzheimer type disorder. Serum somatomedin values, determined by radioreceptorassay and radioimmunoassay, fell with advancing age in normal subjects. A significant elevation in serum somatomedins was observed in dementia patients. CSF somatomedin levels were also increased in the only two patients with suspected Alzheimer type disease examined. Since somatomedins are believed to act as anabolic hormones, it was suggested that the elevated levels represen

Hereditary forms of growth hormone deficiency and resistance.

Pituitary dwarfism is a genetically heterogeneous group of disorders associated with a variety of pathogenetic mechanisms which may involve any level of the hypothalamic-pituitary-somatomedin-peripheral tissue axis. Developmental defects of the pituitary, such as congenital absence of the pituitary, and developmental defects of the hypothalamus, such as anencephaly and holoprosencephaly, have been described. Degeneration of the hypothalamus and pituitary, as in histiocytosis X, may also result in pituitary dwarfism. In "idiopathic" forms of pituitary dwarfism, defects limited to either the hyp

Isolation of a somatomedin from plasma of rats bearing growth hormone secreting tumors.

We have purified a protein which has somatomedin-like properties from the serum of Wistar-Furth rats bearing a growth hormone producing pituitary tumor (MStT/W15). Activity was measured by a placental insulin and/or somatomedin C radioreceptor assay (SmC-RRA). The serum was initially filtered through Sephadex G-150 equilibrated with 0.1 M NH4HCO3 and 0.02% NaN3. On the G-150 column, radioreceptor insulin (RRI) and radioreceptor somatomedin C (RRSm-C) activities coincided and appeared predominantly in the 160,000 mol wt range with a minor proportion in the 50,000 mol wt range. The pooled active

Somatomedin-1 (recombinant insulin-like growth factor-1): clinical pharmacology and potential treatment of endocrine and metabolic disorders.

Insulin-like growth factor (IGF-1) is a polypeptide of 70 amino acids. The circulatory form of IGF-1 is synthesised in the liver. The metabolic activity of IGF-1 is regulated by 6 IGF-binding proteins (BPs), the most important being IGFBP-3. IGF-1 acts via its own receptor, which resembles that of insulin. It has been demonstrated that the effects of growth hormone (GH) on protein metabolism, including growth and the effect on nerve tissue versus trophic effects, are mediated by IGF-1, whereas these 2 hormones are antagonistic in their effects on insulin and some aspects of lipid metabolism. Plasma somatomedin-C levels in systemic sclerosis.

We have measured the plasma levels of somatomedin-C (SM-C) or insulin-like growth factor I (IGF-I) in 13 patients with progressive systemic sclerosis (PSS) and age and sex matched healthy controls. We found the plasma SM-C levels to be within normal limits in all the patients. Thus, if somatomedin-C plays a role in the pathogenesis of PSS, it is more likely to be at the fibroblast receptor level or in the synthetic response of fibroblasts to SM-C.

Growth hormone, somatomedin and prolactin--relationship to brain function.

In recent years it has been found that the brain of several mammals including have receptors for somatomedin both IGF-I and II and measurable hGH has been identified in human brain tissue. IGF-II has been determined in CSF. The presence of these hormones in brain tissue seems to be of developmental and functional importance as experimental studies in frogs, tadpoles and rats showed that injection of growth hormone enhanced brain growth and increased the ratio of neurons to glia. In man early initiation of hGH therapy to children with hGH (who have a less than normal head circumference) induced

Increase in placental lactogen and somatomedins after nephrectomy.

To investigate the relationship between renal agenesis and fetal growth retardation, 6 fetal sheep underwent bilateral nephrectomy and carotid artery catheterization between 105 and 125 days of gestation. Five fetal sheep were catheterized at a similar age as controls and all but one were delivered between 134 days and 147 days gestation. The weights of the fetuses did not differ significantly but the nephrectomized fetuses were shorter than their controls (P less than 0.01). Hydranencephaly was observed in two nephrectomized animals and the mean brain weight was reduced in this group (P less

[Somatomedins and their binding proteins are involved in wound healing after hypoxia of the central nervous system].

Insulin-like growth factors (IGFs) are involved in cell growth and differentiation. In muscle tissue they regulate axonal in growth and maintain the connection. They also play a role in regeneration of the peripheral nerve system. We hypothesized that IGFs might also be important factors in the recovery of central nervous tissue after traumatic damage such as perinatal asphyxia. Our group developed a rat model to mimic the resulting damage and test the changes of expression of IGF-1, -2 and several of their binding proteins. We also examined the influence of exogenous IGF-1 and -2 after asphyx

The role of the insulin‑like growth factor (IGF) axis in osteogenic and odontogenic differentiation.

The insulin-like growth factor (IGF) axis is a multicomponent molecular network which has important biological functions in the development and maintenance of differentiated tissue function(s). One of the most important functions of the IGF axis is the control of skeletal tissue metabolism by the finely tuned regulation of the process of osteogenesis. To achieve this, the IGF axis controls the activity of several cell types—osteoprogenitor cells, osteoblasts, osteocytes and osteoclasts to achieve the co-ordinated development of appropriate hard tissue structure and associated matrix deposition
