Systematics of Mepraia (Hemiptera-Reduviidae): cytogenetic and molecular variation.

The haematophagous insects of the subfamily Triatominae (Hemiptera-Reduviidae) have great epidemiological importance as vectors of Trypanosoma cruzi, the causative agent of Chagas disease. Mepraia was originally described as a monotypic genus comprised of Mepraia spinolai, distributed along coastal areas of northern Chile (from Region I to the Metropolitan Region). Recently, some M. spinolai populations have been ranked as a new species named Mepraia gajardoi. Several populations along the distribution range of the genus were sampled, and genetic differentiation was studied based upon the anal

The use of aggregation signals by Triatoma brasiliensis (Heteroptera: Reduviidae).

Triatoma brasiliensis is considered the main vector of Chagas disease in the semiarid zones of North-eastern Brazil. In this work, we describe the spatial pattern of defecation in T. brasiliensis and the spatial relationship of faeces with the shelters of these bugs. Afterwards, we analyse the attraction response of T. brasiliensis larvae to potential odour cues present in their dry faeces, to substances present in their cuticule (footprints) and to the dry faeces of Triatoma pseudomaculata. We show that faecal spots are deposited in a larger density inside the shelter than in the remaining ar

A baited trap for kissing bugs (Hemiptera: Reduviidae: Triatominae).

Chagas disease is a key vector-borne disease. This illness is caused by Trypanosoma cruzi Chagas, which is transmitted by triatomine bugs. Largely, the control of this disease relies on reducing such contact. We optimized the performance of a box trap in laboratory conditions to capture four triatomine species: Triatoma pallidipennis (Stål), Triatoma infestans Klug, Triatoma phyllosoma (Burmeister), and Rhodnius prolixus Stål. We varied four components for a box trap: material, color, height, and bait attractants. All species were captured more in corrugated cardboard traps than in other trap

Identification of trypanosomes isolated from reduviidae from north Chile.

Examination of 54 Triatoma infestans from a village near the European Southern Observatory La Silla in Chile and of 9 Triatoma spinolai from the territory of the observatory showed that 10 T. infestans were infected with trypanosomatids. Mice were infected with in vitro cultures initiated with five different trypanosomatid isolates and treated with the immunosuppressive drug cyclophosphamide to increase the parasitemia of the flagellates. Evidence of the presence of T. cruzi was provided by a comparative biometrical analysis of blood trypomastigotes and the occurrence of intracellular amastigo

Copulation and mate guarding behavior in Triatoma brasiliensis (Hemiptera: Reduviidae).

The hematophagous bug Triatoma brasiliensis (Neiva) is an important vector of Chagas disease in Brazil. We describe the mating behavior of this species in a comparative manner and analyze the effect of time elapsed since the first imaginal bloodmeal on motivation of males to mate, on female mate-rejections, and on overall mating frequency. We also evaluate whether females of this species copulate with several males in short time intervals, and we examine whether the duration of copulation may vary in the presence of other males compared with what is observed with isolated pairs. Age affected t

Markers for the population genetics studies of Triatoma sordida (Hemiptera: Reduviidae).

BACKGROUND: Triatoma sordida, a vector of Trypanosoma cruzi, is native of Brazil, Bolivia, Paraguay, Argentina, and Uruguay, and occurs primarily in peridomiciles. Currently, it is the species most frequently captured by the Chagas Disease Control Program in Brazil. For this reason, population genetic studies attract great interest, as they can provide further information about the dispersal and household invasion processes of this species. In the absence of suitable markers, the objective of this study was to test the cross amplification of microsatellite primers. FINDINGS: 23 primers were t

An Integrative Sialomic Analysis Reveals Molecules From Triatoma sordida (Hemiptera: Reduviidae).

Triatomines have evolved salivary glands that produce versatile molecules with various biological functions, including those leading their interactions with vertebrate hosts' hemostatic and immunological systems. Here, using high-throughput transcriptomics and proteomics, we report the first sialome study on the synanthropic triatomine Triatoma sordida. As a result, 57,645,372 reads were assembled into 26,670 coding sequences (CDS). From these, a total of 16,683 were successfully annotated. The sialotranscriptomic profile shows Lipocalin as the most abundant protein family within putative secr

Isoenzymes detect variation in populations of Triatoma brasiliensis (Hemiptera: Reduviidae: Triatominae).

Triatoma brasiliensis is one of the most important vectors of Chagas disease in the semiarid zone of the northeast of Brazil. Intraspecific morphological and behavioural variation has been reported for different populations. Results for four distinct populations using eight isoenzymes are reported here. The literature describes three subspecies: T. brasiliensis brasiliensis Neiva, 1911; T. brasiliensis melanica Neiva & Lent, 1941 and T. brasiliensis macromelasoma Galvão, 1956. These subspecies differ mainly in their cuticle colour pattern and were regarded as synonyms by Lent and Wygodzinsky (

Laboratory maintenance and care of Rhodnius prolixus (Hemiptera: Reduviidae) and other Triatominae.

Triatomines (Hemiptera: Reduviidae) are hematophagous insects that transmit Trypanosoma cruzi, the etiological agent for Chagas disease, to humans and other mammals. As medically important vectors, species such as Rhodnius prolixus (Hemiptera: Reduviidae) have long been used as a model organism for physiological studies. Laboratory rearing of triatomines is needed to support vector and parasite research. Many environmental conditions, such as suitable housing containers, light source and duration, temperature, humidity, and density, must be addressed when adapting triatomines from a natural ha

Life-History Data of a Mexican Population of Triatoma nitida (Hemiptera: Reduviidae).

This study reports the third collection of Triatoma nitida Usinger in Mexico, with a brief description of the collection area and an investigation of parameters related to its vectorial capacity. Whether a triatomine (Hemiptera: Reduviidae) species is a primary or secondary vector is determined by factors that include vectorial capacity, anthropophilic habits, geographic distribution, and capacity to invade and colonize human dwellings. However, when the primary vectors are removed, secondary vectors, such as T. nitida, can become important transmitters of Trypanosoma cruzi Chagas to humans. T

Mitochondrial genomes of three kissing bugs (Reduviidae: Triatominae) and their phylogenetic implications.

The reduviid subfamily Triatominae, also called kissing bugs, are vectors of Chagas disease, which is one of the most seriously neglected tropical parasitic diseases. Only three complete mitochondrial genomes of kissing bugs from the genus Triatoma have been sequenced to date. To better understand the diversity of mitochondrial genomes and the evolution of kissing bugs, mitochondrial genomes of three kissing bugs, Triatoma migrans, Panstrongylus rufotuberculatus, and Rhodnius pictipes, were sequenced using next-generation sequencing and a comparative mitochondrial genomic analysis of three gen

Morphology of the terminal abdominal segments in females of Triatominae (Insecta: Hemiptera: Reduviidae).

Triatominae (Insecta: Hemiptera: Reduviidae) includes 151 extant species that are all potential vectors of Chagas disease. The relationships among these species have been recently studied based on molecular and cytogenetical approaches, and although the morphology of these insects is fairly described, wide comparative studies and cladistics analysis based on this feature are lacking. A comparative study of the terminal abdominal segments in females of 26 species of Triatominae was performed in order to evaluate their importance for the distinction of genera and species, and obtain characters t

We examined the degree of reproductive isolation in four populations of Triatoma mexicana Herrich-Schaeffer from Mexico along with fertility and the segregation of morphological characteristics in two generations of offspring from crosses between these populations. The percentage of couples with (fertile) offspring was high among all sets of crosses between cohorts from Peñamiller, Meztitlán, and Orizabita. It was low in crosses involving a cohort from Tierra Blanca, mainly in crosses with Meztitlán. Among sets of crosses involving Tierra Blanca specimens, whole first-generation (F1) individua

Clock Gene Period in the Chagas Disease Vector Triatoma infestans (Hemiptera: Reduviidae).

To contribute to a better understanding of the molecular bases of the circadian biological rhythms in Chagas disease vectors, in this work we identified functional domains in the sequences of the clock protein PERIOD (PER) in Rhodnius prolixus and Triatoma infestans and analyzed the expression of the PER gene at mRNA level in T. infestans. The PER protein sequences comparison among these species and those from other insects revealed that the most similar regions are the PAS domains and the most variable is the COOH-terminal. On the other hand, the per gene expression in nervous tissue of adult

A New Record of the Introduced Species Triatoma infestans (Hemiptera: Reduviidae) in Mexico.

Here, we report a new record of Triatoma infestans (Klug) in Mexico after 50 years and provide a brief description of the discovery area. Fifty-nine specimens (71.2% adults) of the introduced species were collected from the peridomestic areas of a single house in the port of Manzanillo in the state of Colima, Mexico. Thirty-one specimens (52.5%) were collected from the exterior walls of the house and were apparently attracted to light. The other specimens (47.5%) were associated with chickens. No specimen was infected with Trypanosoma cruzi Chagas, the causative agent of Chagas disease, possib

Dynamics of thermopreference in the Chagas disease vector Panstrongylus megistus (Hemiptera: Reduviidae).

The thermopreference of resting P. megistus (Burmeister, 1834) adults was studied in a temperature gradient. We also determined the thermopreference for oviposition and ecdysis. Thermopreference of resting individuals depended on the feeding state of the insects. Results demonstrated an initial resting preference of approximately 290 degrees C, which gradually changed to 26-27 degrees C with increasing starvation. A daily variation in thermopreference was also observed with this species. The insects showed a daily maximum preference at the beginning of the dark phase, and a minimum preference

Modulation of murine lymphocyte responsiveness by the saliva of Rhodnius prolixus (Hemiptera: Reduviidae).

Incorporation of 3H-thymidine by splenic lymphocytes was studied in CBA mice that were bitten by the triatomine bug Rhodnius prolixus (Stål) 14, 7, 4, and 2 d before lymphocyte isolation, respectively. In bitten mice, both spontaneous and mitogen-induced proliferative responses were reduced. The most pronounced effect was observed 4 d after exposure when lymphocytes were significantly suppressed in proliferative response to the mitogens concanavalin A, phytohemagglutinin, and bacterial lipopolysaccharide. The maximum inhibition caused an 85% reduction of proliferation and was observed in phyto

New discoveries of sylvatic Triatoma infestans (Hemiptera: Reduviidae) throughout the Bolivian Chaco.

Sylvatic populations of Triatoma infestans might be involved in the recolonization of human dwellings. We report here the discoveries of new T. infestans sylvatic foci in the Bolivian Chaco. Eighty-one triatomines were caught, 38 of which were identified as T. infestans. Triatoma sordida and Panstrongylus geniculatus were the other species collected. One T. infestans and one T. sordida were infected with Trypanosoma cruzi TcI; one T. infestans was infected with TcII. These discoveries add to the debate on the geographic distribution of sylvatic T. infestans populations, the geographic origin o
