Relative biological effectiveness of 31 meV thermal neutrons in peripheral blood lymphocytes.

The reported relative biological effectiveness (RBE) for thermal neutrons has a large range (5-51, for cytogenetic endpoints), which can confound radiation protection decision-making. To determine whether thermal neutron spectra can influence RBE, the RBE of reactor-derived thermal neutrons of average energy 31 meV was evaluated in human peripheral blood lymphocytes using two classical DNA double-strand break endpoints: the dicentric chromosome assay (DCA) and the cytokinesis-block micronucleus assay. Dose-response curves for 41 to 408 mGy revealed a preference for linear regression. Maximum R

NRG Oncology White Paper on the Relative Biological Effectiveness in Proton Therapy.

This position paper, led by the NRG Oncology Particle Therapy Work Group, focuses on the concept of relative biologic effect (RBE) in clinical proton therapy (PT), with the goal of providing recommendations for the next-generation clinical trials with PT on the best practice of investigating and using RBE, which could deviate from the current standard proton RBE value of 1.1 relative to photons. In part 1, current clinical utilization and practice are reviewed, giving the context and history of RBE. Evidence for variation in RBE is presented along with the concept of linear energy transfer (LE

Interlaced proton grid therapy - Linear energy transfer and relative biological effectiveness distributions.

PURPOSE: Interlaced beams have previously been proposed for delivering proton grid therapy. This study aims to assess dose-averaged LET (LETd) and RBE-weighted dose (DRBE) distributions of such beam geometries, and compare them with conventional intensity modulated proton therapy (IMPT). METHODS: IMPT plans and four different interlaced proton grid therapy plans were generated for five patient cases (esophagus, lung, liver, prostate, anus). The constant RBE = 1.1 was assumed for optimization. The LETd was subsequently Monte Carlo calculated for each plan and used as input for two LET-dependen

Microdosimetric characterization of proton relative biological effectiveness in pancreatic ductal adenocarcinoma cell lines.

PURPOSE: Pancreatic ductal adenocarcinoma (PDAC) is among the most radioresistant malignancies. Proton therapy assumes a uniform relative biological effectiveness (RBE) of 1.1 regardless of radiation quality. We characterized proton RBE as a function of dose-mean lineal energy (ȳD ) in PDAC cell lines. METHODS: Proton RBE was assessed in Panc-1 and COLO 357 FG cells using a high-throughput 96-well irradiation platform with an 81.4 MeV spot-scanning proton beam. Cells were irradiated at 12 discrete depths along the pristine Bragg curve, spanning ȳD values of 1.9 to 20.0 keV/μm. Dose was varied

Increased relative biological effectiveness and periventricular radiosensitivity in proton therapy of glioma patients.

PURPOSE: Currently, there is an intense debate on variations in intra-cerebral radiosensitivity and relative biological effectiveness (RBE) in proton therapy of primary brain tumours. Here, both effects were retrospectively investigated using late radiation-induced brain injuries (RIBI) observed in follow-up after proton therapy of patients with diagnosed glioma. METHODS: In total, 42 WHO grade 2-3 glioma patients out of a consecutive patient cohort having received (adjuvant) proton radio(chemo)therapy between 2014 and 2017 were eligible for analysis. RIBI lesions (symptomatic or clinically a

Disruption of SLX4-MUS81 Function Increases the Relative Biological Effectiveness of Proton Radiation.

PURPOSE: Clinical proton beam therapy has been based on the use of a generic relative biological effectiveness (RBE) of ∼1.1. However, emerging data have suggested that Fanconi anemia (FA) and homologous recombination pathway defects can lead to a variable RBE, at least in vitro. We investigated the role of SLX4 (FANCP), which acts as a docking platform for the assembly of multiple structure-specific endonucleases, in the response to proton irradiation. METHODS AND MATERIALS: Isogenic cell pairs for the study of SLX4, XPF/ERCC1, MUS81, and SLX1 were irradiated at the mid-spread-out Bragg peak

Evaluation of relative biological effectiveness for diseases of the circulatory system based on microdosimetry.

In the next decade, the International Commission on Radiological Protection (ICRP) will issue the next set of general recommendations, for which evaluation of relative biological effectiveness (RBE) for various types of tissue reactions would be needed. ICRP has recently classified diseases of the circulatory system (DCS) as a tissue reaction, but has not recommended RBE for DCS. We therefore evaluated the mean and uncertainty of RBE for DCS by applying a microdosimetric kinetic model specialized for RBE estimation of tissue reactions. For this purpose, we analyzed several RBE data for DCS det

Reanalysis of Mechanism-Inspired Relative Biological Effectiveness Models Based on Track-structure Monte Carlo Simulation.

PURPOSE: Models predicting relative biological effectiveness (RBE) are essential for proton and heavy ion radiotherapy. The Microdosimetric Kinetic Model (MKM) and Local Effect Model (LEM) are two primary classes of mechanism-inspired RBE models. Rigorous model evaluations should extend beyond their ability to reproduce known experimental data. This study systematically examines the core principles of these models by establishing specific test conditions designed to assess their fundamental predictive accuracy and robustness. METHODS: Track-structure Monte Carlo (TSMC) simulations were used t

Relative biological effectiveness and neural stem cell fate in carbon ion-irradiated human brain organoids.

BACKGROUND AND PURPOSE: Heavy ion therapy offers a promising approach for treating brain tumors due to the high precision of ion beams and biological effectiveness. However, the mechanisms underlying contrast-enhanced brain lesions, which occur more frequently after ions than conventional X-rays, remain to be elucidated. MATERIALS AND METHODS: Cerebral organoids were generated from human embryonic stem cells and exposed to high-energy carbon-ions at early (day 20) and late (day 80) developmental stages. Analyses of gene expression were performed using RT-PCR analysis, and protein expression w

Equivalent relative biological effectiveness for cell survival and micronuclei formation: insights from a biophysical approach.

BACKGROUND: Micronuclei (MN), which are chromosome fragments, are formed after exposure to ionizing radiation. Radiation-induced MN is currently used as a quantitative indicator of the chromosomal aberrations detectable at a relatively early phase (e.g., within one cell-cycle progression). Meanwhile, the MN formation assay is also used to evaluate radiosensitivity (e.g., cell-killing). As such, the technique to assay the MN formation has been followed with increasing interest. However, the meaning of MN and the corresponding cellular responses remains uncertain. PURPOSE: This study presents a

Relative biological effectiveness of radon: An in vitro study using chromosome aberrations as a biomarker.

PURPOSE: The need to determine a reliable relative biological effectiveness (RBE) value for alpha exposures has become important as reports remain controversial. Although a radiation-weighting factor of 20 has been designated for alpha particles, uncertainty exists on realistic value of the RBE of alpha radiation. The aim of this study was to estimate RBE values for radon using chromosome aberrations as the endpoint in respect to various dose rates of gamma radiation. MATERIALS AND METHODS: Human blood samples were exposed ex vivo to different doses of radon ranging from 0.0011-0.008 Gy. Bloo

Relative biological effectiveness of neutrons: induction of malformations in a mouse strain with genetic predisposition.

Female hybrid mice (C57BL/6J x HLG) were exposed to 0.5 Gy of cyclotron neutrons (mean energy 5.8 MeV) after mating with HLG males when embryos were at the early zygote stage. Uterine contents were examined on gestation day 19 for prenatal mortality and malformed development. The results were compared to those obtained after irradiation of zygotes with 1.0 and 1.7 Gy X rays. The number of malformed fetuses was greater than for the unirradiated controls. Gastroschisis was the main observation, but exencephaly was also observed. The neutron relative biological effectiveness (RBE) for the inducti

The Abridged Microdosimetric Distribution Methodology for flexible and efficient relative biological effectiveness calculations in ion radiotherapy.

Objective. Current clinical approaches for calculating the relative biological effectiveness (RBE) in ion beam therapy rely on pre-computed lookup tables tied to a specific RBE model and cell line. While practical, this framework limits flexibility, as a subsequent RBE recalculation with a different biological model or cell line requires re-running the radiation transport simulations. This work systematically benchmarks a computationally efficient method that allows accurate and flexible RBE calculations in ion beam therapy.Approach.The Abridged Microdosimetric Distribution Methodology (AMDM)

Reference radiation selection is confirmed as a significant source of relative biological effectiveness variation for neutrons.

PURPOSE: To confirm that the selection of a reference radiation affects the magnitude and range of the relative biological effectiveness (RBE) evaluations of neutron test radiation. Particular attention was paid to the published thermal neutron RBE dataset that is highly variable, with values ranging from 5.4-51.1. MATERIALS AND METHODS: This involved a dual approach of 1) reaffirming dicentric chromosome assay (DCA) dose-response curve differences for 60Co, 137Cs, and 250 kVp X-rays, and 2) recalculating maximum RBE at minimal doses (RBEM) for our previously reported neutron data, accompanie

Protons Show Greater Relative Biological Effectiveness for Mammary Tumorigenesis with Higher ERα- and HER2-Positive Tumors Relative to γ-rays in APCMin/+ Mice.

PURPOSE: Exposure to ionizing radiation increases risk of breast cancer. Although proton radiation is encountered in outer space and in medicine, we do not fully understand breast cancer risks from protons owing to limited in vivo data. The purpose of this study was to comparatively assess the effects of γ-rays and protons on mammary tumorigenesis in APCMin/+ mice. METHODS AND MATERIALS: Female APCMin/+ mice were exposed to 1 GeV protons (1.88 or 4.71 Gy) and 137Cs γ-rays (2 or 5 Gy). Assessing the Relative Biological Effectiveness of Neutrons across Organs of Varying Depth among the Atomic Bomb Survivors.

When assessing radiation-related risk among the atomic bomb survivors, choices in modeling approach can have an important impact on the results, which are then used to inform radiation protection standards throughout the world. The atomic bombings of Hiroshima and Nagasaki produced a mixed-field radiation exposure from two sources: neutrons and gamma rays. Neutrons are more densely ionizing and cause greater biological damage per unit absorbed dose, resulting in greater relative biological effectiveness (RBE) than gamma rays. To account for this, a combined weighted dose is typically calculate
