Neuronal nicotinic acetylcholine receptors (nAChRs) are pentamers built from a variety of subunits. Some are homomeric assemblies of α subunits, others heteromeric assemblies of α and β subunits which can adopt two stoichiometries (2α:3β or 3α:2β). There is evidence for the presence of heteromeric nAChRs with the two stoichiometries in the CNS, but it has not yet been possible to identify them at a given synapse. The 2α:3β receptors are highly sensitive to agonists, whereas the 3α:2β stoichiometric variants, initially described as low sensitivity receptors, are indeed activated by low and high

A subpopulation of calbindin-immunoreactive neurons in lamina VII of the spinal cord has been identified by its location as Renshaw cells, the anatomical substrate for recurrent inhibition. The expression of calbindin (28 kDa) in these calbindin-containing rat ventral horn interneurons was studied with immunocytochemistry after sciatic nerve injuries. One week after axotomy calbindin immunoreactivity was strongly reduced on the lesioned side between levels L4 and L6, while calbindin-containing neurons and fibers were still numerous contralaterally and cranially to the lesioned levels. With the

The recurrent inhibition of alpha-motoneurons by Renshaw cells, brought about by a conditioning reflex discharge, was estimated by measuring the amplitude of a test reflex which involved only those soleus motoneurons which had already fired in response to the conditioning volley. The results obtained in 95 patients with upper motor neuron disease and in 31 normal subjects were compared. At rest, there was no evidence for a decrease in the excitability of Renshaw cells: on the contrary, recurrent inhibition, if it changed at all, was increased in 55 out of 95 patients. In most patients the chan

Motoneurons have long been considered as the final common pathway of the nervous system, transmitting the neural impulses that are transduced into action. While many studies have focussed on the inputs that motoneurons receive from local circuits within the spinal cord and from other parts of the CNS, relatively few have investigated the targets of local axonal projections from motoneurons themselves, with the notable exception of those contacting Renshaw cells or other motoneurons.Recent research has not only characterised the detailed features of the excitatory connections between motoneurons

Mechanisms underlying the development of spasticity after spinal cord injury are not understood. One spinal interneuron likely to be affected is the Renshaw cell, which acts to produce recurrent inhibition in motor neurons as well as inhibiting Ia interneurons. Descending pathways exert both excitatory and inhibitory control over Renshaw cell activity. We studied Renshaw cell activity in normal subjects and in patients with varying levels of spasticity after spinal cord injury using the conditioned H-reflex technique of Pierrot-Deseilligny and Bussel. A submaximal stimulus to the tibial nerve

In the spinal cord neuronal activity is controlled by the balance between excitatory and inhibitory neurotransmission, mediated mainly by the neurotransmitters glutamate and GABA/glycine, respectively. Alterations of this equilibrium have been associated with spinal motor neuron hyperexcitability and degeneration, which can be induced by excitotoxicity or by decreasing inhibitory neurotransmission. Here we review the ventral horn neuronal network and the possible involvement of inhibitory circuits in the mechanisms of degeneration of motor neurons characteristic of amyotrophic lateral sclerosi

By use of H-reflex technique reflex flexor muscles motoneurons recurrent inhibition was studied for man forearm muscle. It was observed the reduce of this inhibition with the muscles strain. The vibration, acting to the strained muscle, showed a disappearance of inhibition. It was marked that these features can be connected to both segmental or supraspinal action to motoneurons or Renshaw cells. The recurrent inhibition is reduced more effectively under complex vibration and muscle strain influence than under only strain factor, and it is connected to the vibration. It seems to be connected to

Ia excitation and recurrent inhibition are basic neuronal circuits in motor control in hind limb. Renshaw cells receive synaptic inputs from axon collaterals of motoneurons and inhibit motoneurons and Ia inhibitory interneurons. It is important to know properties of Ia excitation and recurrent inhibition of trunk muscle such as abdominal muscles. The abdominal muscles have many roles and change those roles for different kind of functions. Intracellular recordings were obtained from the abdominal motoneurons of the upper lumbar segments in cats anesthetized. First, dorsal roots were left intact

The aims were (1) to determine when in human postnatal development Group Ia reciprocal and Renshaw inhibition can be demonstrated; (2) to explore the relationship between the expression reciprocal inhibition and the disappearance of Group Ia excitatory reflexes between agonist and antagonist muscles. Studies were performed on 99 subjects, aged 1 day to 31 years, of whom 53 were neonates. A longitudinal study was also performed on 29 subjects recruited at birth and studied 3 monthly until 12 months of age. Reciprocal inhibitory and excitatory reflexes were recorded in the surface EMG of contrac

Spasticity is still a challenge to clinicians and scientists. The participation of Ia, Ib, II, III and IV afferents pathways, as well as those related with presynaptic inhibition. Renshaw cells modulation, and alpha and gamma motoneurons hyperexcitability, in this medical problem are reviewed. A brief overview on the more common neurophysiological tests performed during the evaluation of human spasticity is presented.

1. The effects of D-alpha-aminoadipate (DalphaAA), D-alpha-aminosuberate (DalphaAS) and other excitatory amino acid antagonists have been compared on the excitatory responses of neurones of the cat spinal cord to acetylcholine, a range of glutamate-related amino acids and stimulation of appropriate excitatory synaptic pathways. The ionophoretic technique was used for administration of excitants and antagonists. 2. DalphaAA and DalphaAS had little or no effect on acetylcholine-induced excitation of Renshaw cells. Responses of either Renshaw cells or dorsal horn neurones in the spinal cord to ex

Spinal recurrent inhibition (RI), mediated by Renshaw cells, exhibits a highly conserved proximal-to-distal gradient across mammals: it is robust in proximal motoneuron (MN) pools but absent in MNs innervating both extrinsic and intrinsic digit muscles. Although this organizational pattern has been recognised for more than six decades, its mechanistic basis has remained unexplained. The widely accepted view that the gradient reflects the evolutionary emergence of monosynaptic corticomotoneuronal (CM) projections is challenged by the cat, which lacks monosynaptic CM input to digit MNs yet exhib

This study examined whether activation of group I metabotropic glutamate receptors (mGluRs) could modulate synaptic inhibition of spinal motoneurons in the neonatal rat isolated spinal cord. Recurrent inhibitory postsynaptic potentials (IPSPs) generated by Renshaw cells were evoked via antidromic stimulation of motor axon collaterals and recorded intracellularly from lumbar motoneurons. The selective agonist of group I mGluRs DHPG (5 micromol L-1) depressed the recurrent IPSP, an effect prevented by the selective antagonist AIDA (500 micromol L-1). The depression by DHPG was use-independent an

We describe a 35-year-old woman who presented with the syndrome of painful hand-moving fingers on the right side. Eight months later, she developed similar finger movements and hand discomfort on the left side. She had a history of hand trauma and recurrent shoulder dislocation on the right side. Kinesiologic electromyography suggested a common central oscillator for finger movements in both hands. Electrophysiological assessment of spinal alpha motor neuron excitability, reciprocal inhibition, and Renshaw cell inhibition failed to show any abnormalities. Somatosensory evoked potential test sh

Most neuronal heteromeric nicotinic receptors seem able to adopt two different stochiometries depending on the ratio of α and β subunits. In recombinant receptors these two stoichiometries have been associated with different affinities to ACh, but it is not known which stoichiometry is present at nicotinic synapses in the nervous system. One possible clue to this identification is the speed of decay of the synaptic currents. In many ionotropic receptors this speed has been linked to the dissociation rate of the transmitter, which is itself related to its affinity. On this basis we propose that

In many neurological conditions, early-stage neural circuit adaptation preserves relatively normal behavior. In some diseases, spinal motoneurons progressively degenerate yet movement remains initially preserved. This study investigates whether these neurons and associated microcircuits adapt in a mouse model of progressive motoneuron degeneration. Using a combination of in vitro and in vivo electrophysiology and super-resolution microscopy, we find that, early in the disease, neurotransmission in a key pre-motor circuit, the recurrent inhibition mediated by Renshaw cells, is reduced by half d

Nicotinic acetylcholine receptors (nAChRs) play crucial roles in various human disorders, with the α7, α4, α6, and α3-containing nAChR subtypes extensively studied in relation to conditions such as Alzheimer's disease, Parkinson's disease, nicotine dependence, mood disorders, and stress disorders. In contrast, the α2-nAChR subunit has received less attention due to its more restricted expression and the scarcity of specific agonists and antagonists for studying its function. Nevertheless, recent research has shed light on the unique expression pattern of the Chrna2 gene, which encodes the α2-n

Three types of inhibition in the spinal cord were studied during barbiturate withdrawal. Cats were made physically dependent on pentobarbital using a standard technique for chronic treatment developed in this laboratory. All animals treated this way developed severe withdrawal signs including spontaneous grand mal type convulsions, upon abrupt drug withdrawal. Animals were prepared for electrophysiological studies 48 hours after the last dose of chronic pentobarbital treatment, near peak intensity of the withdrawal syndrome. Under halothane anesthesia, the neuraxis was sectioned at C1 and both

Monoamines can modulate the output of a variety of invertebrate and vertebrate networks, including the spinal cord networks that control walking. Here we examined the multiple changes in the output of locomotor networks induced by dopamine (DA). We found that DA can depress the activation of locomotor networks in the neonatal mouse spinal cord following ventral root stimulation. By examining disinhibited rhythms, where the Renshaw cell pathway was blocked, we found that DA depresses a putative recurrent excitatory pathway that projects onto rhythm-generating circuitry of the spinal cord. This
