Chapter 9 Infections caused by rubella, reoviridae, retro, Norwalk and ronaviruses.

This chapter focuses on infections caused by rubella, reoviridae, retro, Norwalk and coronaviruses. High incidence of acute gastroenteritis caused by rotaviruses calls for prophylactic and therapeutic measures. Although no vaccine is presently available, it seems likely that vaccines will be developed in the next few years. There are also several rotavirus enzymes useful as targets for antiviral drugs. However, no antiviral drugs have shown therapeutic effects against rotavirus infections. The newly discovered human retrovirus (HTLV) has not yet been investigated in such detail as to predict t

Genus Coltivirus (family Reoviridae): genomic and morphologic characterization of Old World and New World viruses.

We report a genomic and morphologic study of the European Eyach (EYA) virus (genus Coltivirus, family Reoviridae) and a comparative analysis with the American Colorado tick fever (CTF) virus (the type species of the genus). The previously established, but distant, antigenic relationship between these viruses was strengthened by genetic findings (presence of cognate genes, amino acid identity between 55 and 88%, similar conserved terminal motifs, suspected read-through phenomenon in segment 9 of both viruses) and by indistinguishable ultramicroscopic morphologies. Moreover, putative constitutiv

Genome sequencing and phylogenetic analysis of Banna virus (genus Seadornavirus, family Reoviridae) isolated from Culicoides.

In an investigation of blood-sucking insects and arboviruses, a virus (YN12243) was isolated from Culicoides samples collected in the Sino-Burmese border region of Yunnan Province, China. The virus caused cytopathic effect (CPE) in C6/36 cells and passaged stably. Polyacrylamide gel analysis showed that the genome of YN12243 was composed of 12 segments of double-stranded RNA (dsRNA), with a distribution pattern of 6-6. The nucleotide and amino acid sequences of the coding region (1‒12 segments) were 17,803 bp and 5,925 amino acids in length, respectively. The phylogenetic analysis of VP1 prote

Complete nucleotide sequences of genome segments 1 and 3 of Rosellinia anti-rot virus in the family Reoviridae.

The nucleotide sequences of genome segments 1 and 3 of Rosellinia anti-rot virus (RArV) from a hypovirulent isolate, W370, of the plant pathogen Rosellinia necatrix were determined. The complete nucleotide sequence of the genome segment 1 encoded a putative RNA-dependent RNA polymerase (RDRP). The deduced amino acid sequence of RDRP of RArV showed 29% identity with RDRPs of Colorado tick fever virus (CTFV) and European Eyach virus (EYAV) in the genus Coltivirus, and identities of 23-21% with members of the genera Fijivirus and Cypovirus. Both RArV and the Coltivirus member might have originate

Isolation of Eyach virus (Reoviridae, Colorado tick fever group) from Ixodes ricinus and I. ventalloi ticks in France.

During the spring of 1981, two strains of an identical virus were isolated in Mayenne, France, from Ixodes ricinus L. and Ixodes ventalloi Gil Collado, parasiting a wild rabbit (Oryctolagus cuniculus L.). Virological and serological results indicated that these strains belong to the Colorado tick fever serogroup and are indistinguishable from Eyach virus, a tick-borne virus previously described from West Germany. Study of dsRNA PAGE profiles confirmed these conclusions. The morphology and morphogenesis of French isolates were studied in infected suckling mouse brains showing some peculiar feat

Sequence determination and analysis of the full-length genome of colorado tick fever virus, the type species of genus Coltivirus (Family Reoviridae).

The Colorado tick fever virus (CTFV) is the type species of genus Coltivirus, family Reoviridae. Its genome consisting of 12 segments of dsRNA was completely sequenced. It was found to be 29,174 nucleotides long (the longest of all Reoviridae genomes characterized to date). Conserved sequences at the 5' end (SACUUUUGY) and at the 3' end (WUGCAGUS) of the 12 segments were identified. The analysis of the putative proteins deduced from the nucleotide sequences permitted to identify functional motifs. In particular, the VP1 was identified unambiguously as the viral RNA dependent RNA pylmerase (RDR

Complete sequence determination and genetic analysis of Banna virus and Kadipiro virus: proposal for assignment to a new genus (Seadornavirus) within the family Reoviridae.

Arboviruses with genomes composed of 12 segments of double-stranded (ds) RNA have previously been classified as members or probable members of the genus Coltivirus within the family REOVIRIDAE: A number of these viruses have been isolated in North America and Europe and are serologically and genetically related to Colorado tick fever virus, the Coltivirus type species. These isolates constitute subgroup A of the coltiviruses. The complete genome sequences are now presented of two Asian arboviruses, Kadipiro virus (KDV) and Banna virus (BAV), which are currently classified as subgroup B coltivi

Complete genome sequence of Mycoreovirus-1/Cp9B21, a member of a novel genus within the family Reoviridae, isolated from the chestnut blight fungus Cryphonectria parasitica.

Mycoreovirus 1 (MYRV-1) is the type species of the newly described genus Mycoreovirus of the large virus family Reoviridae. The virus was isolated from a hypovirulent strain (9B21) of the chestnut blight fungus, Cryphonectria parasitica. A previous study showed that double-shelled particles introduced to fungal spheroplasts resulted in stably infected colonies. Of the 11 double-stranded RNA genomic segments (S1-S11), the three largest (S1-S3) were sequenced previously and shown to have moderate levels of similarity to the homologous segments of mammal-pathogenic coltiviruses (Eyach virus and C

Nucleotide sequences of double-stranded RNA segments from a hypovirulent strain of the white root rot fungus Rosellinia necatrix: possibility of the first member of the Reoviridae from fungus.

Twelve double-stranded (ds) RNA segments were detected from a hypovirulent strain W370 of the white root rot fungus Rosellinia necatrix. The estimated molecular weights ranged from 0.41 x 10(6) to 2.95 x 10(6). Full length cDNA clones for eight segments were obtained. Northern blot analysis suggested that each segment was genetically unique. The nucleotide sequences of eight full length dsRNA segments were determined. One long open reading frame was found in each segment. Conserved sequences at the 5'-end (5'-ACAAUUU-3') and at the 3'-end (5'-UGCAGAC-3') were identified in all eight segments.

Effective management of Mud crab Reovirus (MCRV) infection in mud crab, Scylla serrata (Forskal, 1775) culture, using an individual compartmentalized system.

Mud crab (Scylla serrata) aquaculture is a critical contributor to coastal economies and export markets in India. However, the sector faces significant challenges from Mud Crab Reovirus (MCRV), a highly pathogenic virus reported to cause mass mortalities and severe economic losses. This study assessed the performance of different rearing systems-open pen, cage-in-pen, and external cage systems-focusing on their efficacy in managing MCRV transmission, mortality, and production outcomes. The open pen system exhibited the highest mortality rate (73 ± 4.57 %) and MCRV infection rate (89.04 ± 3.04

A reovirus of the fungus Cryphonectria parasitica that is infectious as particles and related to the coltivirus genus of animal pathogens.

RNA viruses of filamentous fungi fall into two broad categories, those that contain double-stranded RNA (dsRNA) genomes in rigid particles and those that are more closely related to positive-sense, single-stranded RNA viruses with dsRNA replicative intermediates found within lipid vesicles. Effective infectivity systems have been described for the latter, using RNA transcripts, but not for the former. We report the characterization of a reovirus from Cryphonectria parasitica, the filamentous fungus that causes chestnut blight disease. The virus substantially reduces the virulence of the fungus

Experimental infections of feline reovirus serotype 2 isolates.

Pathogenicity of feline reovirus serotype 2 isolates, Nos. 114 and 140 strains recovered from the feces of cats with diarrhea, was experimentally determined. Kittens orally inoculated with No. 114 strain did not show any clinical signs. Fecal consistency of kittens inoculated with No. 140 strain became rather diarrheic on the 3rd and 4th days after inoculation, but any other clinical abnormalities were not observed. Virus was excreted for the first five to six days after inoculation. Seroconversion was observed in the sera of all the inoculated kittens. Virus excretion and seroconversion were

Evidence for viral infection as a causative factor of human biliary atresia.

OBJECTIVES: To explore the evidence for viral infections triggering human biliary atresia (BA) by reviewing archival original articles that analyzed human samples via polymerase chain reaction (PCR) experiments, considering the recent experimental trend of extensive use of rotaviral BA animal models. METHODS: A PubMed search retrieved original articles that reported the results of PCR experiments for detecting viral DNA or RNA in patient samples as proof of past infection. Search terms included the often-debated DNA or RNA viruses and BA. Special focus was directed toward PCR analyses that ta

Sclerosing and obstructive cholangiopathy in biliary atresia: mechanisms and association with biliary innate immunity.

Biliary atresia (BA) is histologically characterized by a progressive, sclerosing cholangitis and the obstruction of extrahepatic bile ducts. In terms of the etiology and pathogenesis of BA, several viral infections consisting of dsRNA, including Reoviridae, have been implicated. Human biliary epithelial cells (BECs) possess an innate immune system consisting of Toll-like receptors (TLRs). BECs have negative regulatory mechanisms of TLR tolerance to avoid an excessive inflammatory response to lipopolysaccharide (LPS), a TLR4 ligand; however, they lack the tolerance to poly(I:C) (a synthetic an

Teratogenic bluetongue and related orbivirus infections in pregnant ruminant livestock: timing and pathogen genetics are critical.

Congenital infections of domestic animals with viruses in several families, including Bunyaviridae, Flaviridae, Parvoviridae, and Reoviridae, are the cause of naturally occurring teratogenic central nervous system and/or musculoskeletal defects (arthrogryposis) in domestic animals. Congenital infections of ruminant livestock with bluetongue virus (BTV) and some related members of the genus Orbivirus (family Reoviridae) have clearly shown the critical role of gestational age at infection in determining outcome. Specifically, fetuses infected prior to mid-gestation that survive congenital BTV in

Protective Efficacy of Vaccines Against Grass Carp Hemorrhagic Disease: A Systematic Review and Meta-Analysis.

Grass carp hemorrhagic disease, mainly caused by grass carp reovirus (GCRV), is a major threat to grass carp aquaculture, and vaccination is considered a sustainable strategy for disease prevention and control. However, the protective efficacy of GCRV vaccines and the factors influencing it have not been systematically synthesised. In this study, literature on GCRV vaccines was retrieved from CNKI, ScienceDirect, and Web of Science in accordance with the PRISMA 2020 statement, covering database inception to March 10, 2026, followed by meta-analysis. The Intestinal Eukaryotic Virome in Healthy and Diarrhoeic Neonatal Piglets.

Neonatal porcine diarrhoea of uncertain aetiology has been reported from a number of European countries. The aim of the present study was to use viral metagenomics to examine a potential viral involvement in this diarrhoea and to describe the intestinal virome with focus on eukaryotic viruses. Samples from the distal jejunum of 50 diarrhoeic and 19 healthy piglets from 10 affected herds were analysed. The viral fraction of the samples was isolated and nucleic acids (RNA and DNA fractions) were subjected to sequence independent amplification. Samples from diarrhoeic piglets from the same herds
