Chapter 9 Infections caused by rubella, reoviridae, retro, Norwalk and ronaviruses.

This chapter focuses on infections caused by rubella, reoviridae, retro, Norwalk and coronaviruses. High incidence of acute gastroenteritis caused by rotaviruses calls for prophylactic and therapeutic measures. Although no vaccine is presently available, it seems likely that vaccines will be developed in the next few years. There are also several rotavirus enzymes useful as targets for antiviral drugs. However, no antiviral drugs have shown therapeutic effects against rotavirus infections. The newly discovered human retrovirus (HTLV) has not yet been investigated in such detail as to predict t

Effective management of Mud crab Reovirus (MCRV) infection in mud crab, Scylla serrata (Forskal, 1775) culture, using an individual compartmentalized system.

Mud crab (Scylla serrata) aquaculture is a critical contributor to coastal economies and export markets in India. However, the sector faces significant challenges from Mud Crab Reovirus (MCRV), a highly pathogenic virus reported to cause mass mortalities and severe economic losses. This study assessed the performance of different rearing systems-open pen, cage-in-pen, and external cage systems-focusing on their efficacy in managing MCRV transmission, mortality, and production outcomes. The open pen system exhibited the highest mortality rate (73 ± 4.57 %) and MCRV infection rate (89.04 ± 3.04

Teratogenic bluetongue and related orbivirus infections in pregnant ruminant livestock: timing and pathogen genetics are critical.

Congenital infections of domestic animals with viruses in several families, including Bunyaviridae, Flaviridae, Parvoviridae, and Reoviridae, are the cause of naturally occurring teratogenic central nervous system and/or musculoskeletal defects (arthrogryposis) in domestic animals. Congenital infections of ruminant livestock with bluetongue virus (BTV) and some related members of the genus Orbivirus (family Reoviridae) have clearly shown the critical role of gestational age at infection in determining outcome. Specifically, fetuses infected prior to mid-gestation that survive congenital BTV in

A reovirus of the fungus Cryphonectria parasitica that is infectious as particles and related to the coltivirus genus of animal pathogens.

RNA viruses of filamentous fungi fall into two broad categories, those that contain double-stranded RNA (dsRNA) genomes in rigid particles and those that are more closely related to positive-sense, single-stranded RNA viruses with dsRNA replicative intermediates found within lipid vesicles. Effective infectivity systems have been described for the latter, using RNA transcripts, but not for the former. We report the characterization of a reovirus from Cryphonectria parasitica, the filamentous fungus that causes chestnut blight disease. The virus substantially reduces the virulence of the fungus

Sclerosing and obstructive cholangiopathy in biliary atresia: mechanisms and association with biliary innate immunity.

Biliary atresia (BA) is histologically characterized by a progressive, sclerosing cholangitis and the obstruction of extrahepatic bile ducts. In terms of the etiology and pathogenesis of BA, several viral infections consisting of dsRNA, including Reoviridae, have been implicated. Human biliary epithelial cells (BECs) possess an innate immune system consisting of Toll-like receptors (TLRs). BECs have negative regulatory mechanisms of TLR tolerance to avoid an excessive inflammatory response to lipopolysaccharide (LPS), a TLR4 ligand; however, they lack the tolerance to poly(I:C) (a synthetic an

Experimental infections of feline reovirus serotype 2 isolates.

Pathogenicity of feline reovirus serotype 2 isolates, Nos. 114 and 140 strains recovered from the feces of cats with diarrhea, was experimentally determined. Kittens orally inoculated with No. 114 strain did not show any clinical signs. Fecal consistency of kittens inoculated with No. 140 strain became rather diarrheic on the 3rd and 4th days after inoculation, but any other clinical abnormalities were not observed. Virus was excreted for the first five to six days after inoculation. Seroconversion was observed in the sera of all the inoculated kittens. Virus excretion and seroconversion were

Evidence for viral infection as a causative factor of human biliary atresia.

OBJECTIVES: To explore the evidence for viral infections triggering human biliary atresia (BA) by reviewing archival original articles that analyzed human samples via polymerase chain reaction (PCR) experiments, considering the recent experimental trend of extensive use of rotaviral BA animal models. METHODS: A PubMed search retrieved original articles that reported the results of PCR experiments for detecting viral DNA or RNA in patient samples as proof of past infection. Search terms included the often-debated DNA or RNA viruses and BA. Special focus was directed toward PCR analyses that ta

Avian reoviruses: propagation, quantification, and storage.

Avian reoviruses (ARVs) are pathogens that cause significant morbidity among commercial poultry. ARVs are prototypic representatives of non-enveloped viruses that can cause cell-cell fusion. They belong to the Reoviridae family, which contains many highly pathogenic viruses. ARVs are ubiquitous in commercial poultry and are frequently isolated from the gastrointestinal and respiratory tracts of chickens with acute infections. The virus causes a range of disease states in chicken, including viral arthritis/tenosynovitis, gastroenteritis, hepatitis, myocarditis, "pale bird syndrome," runting-stu

Neuroimaging features of arboviral infections in the Americas.

Arboviruses are zoonotic RNA viruses maintained in nature in cycles that involve arthropod vectors. The arboviruses that cause disease in humans are members of the Bunyaviridae, Togaviridae, Flaviviridae, and Reoviridae families. These viral species have geographically and climatically restricted distributions due to particular ecological and vector features. The main emerging arboviruses in the Americas are dengue, zika, yellow fever (Flaviviridae), and chikungunya (Togaviridae). All of these viruses can be transmitted by the Aedes aegypti and Aedes albopictus mosquitoes. Although not commonl

Protective Efficacy of Vaccines Against Grass Carp Hemorrhagic Disease: A Systematic Review and Meta-Analysis.

Grass carp hemorrhagic disease, mainly caused by grass carp reovirus (GCRV), is a major threat to grass carp aquaculture, and vaccination is considered a sustainable strategy for disease prevention and control. However, the protective efficacy of GCRV vaccines and the factors influencing it have not been systematically synthesised. In this study, literature on GCRV vaccines was retrieved from CNKI, ScienceDirect, and Web of Science in accordance with the PRISMA 2020 statement, covering database inception to March 10, 2026, followed by meta-analysis. After screening, 44 studies were included, o

Type II Grass Carp Reovirus Rapidly Invades Grass Carp (Ctenopharyngodon idella) via Nostril-Olfactory System-Brain Axis, Gill, and Skin on Head.

Type II grass carp reovirus (GCRV-II) with high pathogenicity and infectivity causes severe hemorrhagic disease, which leads to extensive death in the grass carp and black carp aquaculture. However, the early invasion portal remains unclear. In this study, we explored the invasion portal, time, and pathway of GCRV-II by immersion infection in grass carp. Through the detection of the infected grass carp external body surface tissues, most of them could be detected to carry GCRV-II within 45 min except for the skin covered by scales. Further shortening the duration of infection, we proved that G

Innate immune response to double-stranded RNA in biliary epithelial cells is associated with the pathogenesis of biliary atresia.

UNLABELLED: Infections of Reoviridae consisting of a double-stranded RNA (dsRNA) genome are a possible cause of biliary atresia (BA). The aim of the present study is to clarify the pathophysiological function of dsRNA viruses in the pathogenesis of BA. The expression of dsRNA pattern-recognizing receptors, Toll-like receptor 3 (TLR3), retinoic acid inducible gene I (RIG-I), melanoma differentiation-associated gene-5 (MDA-5), and dsRNA-activated protein kinase R (PKR) was constitutively detected in cultured human biliary epithelial cells (BECs). Stimulation with polyinosinic-polycytidylic acid

Isolation, Identification, and Genomic Analysis of a Novel Reovirus from Healthy Grass Carp and Its Dynamic Proliferation In Vitro and In Vivo.

A new grass carp reovirus (GCRV), healthy grass carp reovirus (HGCRV), was isolated from grass carp in 2019. Its complete genome sequence was determined and contained 11 dsRNAs with a total size of 23,688 bp and 57.2 mol% G+C content, encoding 12 proteins. All segments had conserved 5' and 3' termini. Sequence comparisons showed that HGCRV was closely related to GCRV-873 (GCRV-I; 69.57-96.71% protein sequence identity) but shared only 22.65-45.85% and 23.37-43.39% identities with GCRV-HZ08 and Hubei grass carp disease reovirus (HGDRV), respectively. Occurrence of reovirus infection in Ontario.

Reovirus Type 2 was isolated from three patients aged 5, 9 and 10 years. The etiological role of Reovirus in one case was confirmed by demonstration of antibodies in convalescent serum and none in acute serum. Symptoms in this case were suggestive of infectious mononucleosis. In the second case, rhinitis and non-purulent otitis were preceded by two waves of fever, abdominal pain and emesis. Acute serum was not available but convalescent serum had a high titre to a Reovirus, thus supporting a Reovirus etiology. Symptoms in the third case were fever, intermittent headache, neck stiffness, abdomi

Transferrin Receptor 1-Associated Iron Accumulation and Oxidative Stress Provides a Way for Grass Carp to Fight against Reovirus Infection.

Iron is an essential element, closely linked with host immune responses. Nevertheless, the relationship between iron metabolism and virus infection is still unclear in aquatic vertebrates. To address this issue, we employed grass carp (Ctenopharyngodon idella) and its lethal virus, grass carp reovirus (GCRV), a double-strand RNA virus, as models. Our results demonstrate that GCRV infection increases the iron content and alters the expression of iron metabolism-related genes both in vivo and in vitro. Of note, the expression of C. idella transferrin receptor 1 (CiTfR1) rather than transferrin i

The Intestinal Eukaryotic Virome in Healthy and Diarrhoeic Neonatal Piglets.

Neonatal porcine diarrhoea of uncertain aetiology has been reported from a number of European countries. The aim of the present study was to use viral metagenomics to examine a potential viral involvement in this diarrhoea and to describe the intestinal virome with focus on eukaryotic viruses. Samples from the distal jejunum of 50 diarrhoeic and 19 healthy piglets from 10 affected herds were analysed. The viral fraction of the samples was isolated and nucleic acids (RNA and DNA fractions) were subjected to sequence independent amplification. Samples from diarrhoeic piglets from the same herds

Analysis of GCRV Pathogenesis and Therapeutic Measures Through Proteomic and Metabolomic Investigations in GCRV-Infected Tissues of Grass Carp (Ctenopharyngodon idella).

Hemorrhagic disease caused by grass carp reovirus (GCRV) infection is a major problem affecting the grass carp aquaculture industry. Therefore, inhibiting the spread of GCRV infection is of great economic significance. Herein, we sequenced five tissues (gill, liver, intestine, kidney, and muscle) from grass carp before and after GCRV infection using data-independent acquisition proteomic and untargeted metabolomic technologies, and quantitatively identified 10,808 proteins and 4040 metabolites.
