Retinal ganglion cells.

How do we perceive the beautiful and rich world around us? Light from the external environment is focused onto the retina, our visual sensory organ. Photons are captured by photoreceptors - light sensors within the retina - and converted into electrical signals. These electrical signals are transmitted to the brain through the optic nerve. If each optic nerve transmits a 'pixel' of the visual scene, then the total number of optic nerve fibers defines our biological 'resolution'. Humans possess approximately one million fibers, meaning the image sent from the retina to the brain (retinal image)

Protecting retinal ganglion cells.

Retinal ganglion cell degeneration underlies several conditions which give rise to significant visual compromise, including glaucoma, hereditary optic neuropathies, ischaemic optic neuropathies, and demyelinating disease. In this review, we discuss the emerging strategies for neuroprotection specifically in the context of glaucoma, including pharmacological neuroprotection, mesenchymal stem cells, and gene therapy approaches. We highlight potential pitfalls that need to be considered when developing these strategies and outline future directions, including the prospects for clinical trials.

Retinal ganglion cells expressing melanopsin.

The daily alternation of light and darkness resulting from Earth's rotation is one of the main environmental cues shaping the internal life of animals, orchestrating rhythms of behavior and physiologic functions. In mammals, the retina, lining the back of the eye, serves as the sole entry point for this photic information. About 25 years ago, the discovery of a novel photopigment revolutionized our understanding of how light influences the brain and body. This photopigment, highly conserved across evolution, relies on a protein named melanopsin. Research on melanopsin has revealed the presence

PANoptosis of Retinal Ganglion Cells.

PANoptosis represents a novel form of programmed cell death regulated and controlled by the PANoptosome. It encompasses the essential features of apoptosis, necroptosis, and pyroptosis and combines elements from each process. PANoptosis contributes to the development of various diseases, including bacterial and viral infections, tumors, inflammatory diseases, and neurodegenerative diseases, which offers insights into the pathological mechanisms of these diseases and potential treatments. Retinal ganglion cells (RGCs) are nerve cells located in the final segment of the retina, which belongs to

Encoding surprise by retinal ganglion cells.

The efficient coding hypothesis posits that early sensory neurons transmit maximal information about sensory stimuli, given internal constraints. A central prediction of this theory is that neurons should preferentially encode stimuli that are most surprising. Previous studies suggest this may be the case in early visual areas, where many neurons respond strongly to rare or surprising stimuli. For example, previous research showed that when presented with a rhythmic sequence of full-field flashes, many retinal ganglion cells (RGCs) respond strongly at the instance the flash sequence stops, and

Diacylglycerol Signaling in Retinal Ganglion Cells.

With impaired retinal ganglion cell (RGC) function and eventual RGC death, there is a heightened risk of experiencing glaucoma-induced blindness or other optic neuropathies. Poor RGC efficiency leads to limited transmission of visual signals between the retina and the brain by RGC axons. Increased focus on studying lipid messengers found in neurons such as endocannabinoids (eCBs) has importance due to their potential axonal pathway regenerative properties. 2-Arachidonoylglycerol (2-AG), a common eCB, is synthesized from an sn-1 hydrolysis reaction between diacylglycerol (DAG) and diacylglycero

Axonal mitophagy in retinal ganglion cells.

Neurons, exhibiting unique polarized structures, rely primarily on the mitochondrial production of ATP to maintain their hypermetabolic energy requirements. To maintain a normal energy supply, mitochondria are transported to the distal end of the axon. When mitochondria within the axon are critically damaged beyond their compensatory capacity, they are cleared via autophagosomal phagocytosis, and the degradation products are recycled to replenish energy. When the mitochondria are dysfunctional or their transport processes are blocked, axons become susceptible to degeneration triggered by energ

Glaucomatous retinal ganglion cells: death and protection.

Glaucoma is a group of diseases characterized by progressive optic nerve degeneration, with the characteristic pathological change being death of retinal ganglion cells (RGCs), which ultimately causes visual field loss and irreversible blindness. Elevated intraocular pressure (IOP) remains the most important risk factor for glaucoma, but the exact mechanism responsible for the death of RGCs is currently unknown. Neurotrophic factor deficiency, impaired mitochondrial structure and function, disrupted axonal transport, disturbed Ca2+ homeostasis, and activation of apoptotic and autophagic pathwa

Neurogenesis and Specification of Retinal Ganglion Cells.

Across all species, retinal ganglion cells (RGCs) are the first retinal neurons generated during development, followed by the other retinal cell types. How are retinal progenitor cells (RPCs) able to produce these cell types in a specific and timely order? Here, we will review the different models of retinal neurogenesis proposed over the last decades as well as the extrinsic and intrinsic factors controlling it. We will then focus on the molecular mechanisms, especially the cascade of transcription factors that regulate, more specifically, RGC fate. We will also comment on the recent discover

Changes in ganglion cells during retinal degeneration.

Inherited retinal degeneration such as retinitis pigmentosa (RP) is associated with photoreceptor loss and concomitant morphological and functional changes in the inner retina. It is not known whether these changes are associated with changes in the density and distribution of synaptic inputs to retinal ganglion cells (RGCs). We quantified changes in ganglion cell density in rd1 and age-matched C57BL/6J-(wildtype, WT) mice using the immunocytochemical marker, RBPMS. Our data revealed that following complete loss of photoreceptors, (∼3months of age), there was a reduction in ganglion cell densi

Intrinsic survival mechanisms for retinal ganglion cells.

Retinal ganglion cells (RGCs) undergo programmed cell death (apoptosis) after axonal injury. This cell death is mediated by several mechanisms, including deprivation of neurotrophic factors, alterations in gene expression, and production of reactive oxygen species. However, death of RGCs is delayed after axonal injury, and a significant number survive even after several days. This suggests that RGC death is not an immediate result of axonal injury, and that other pro-survival factors may play a role. While we and other researchers have focused on the mechanisms of cell death after axonal injur

Trim9 regulates the directional differentiation of retinal Müller cells to retinal ganglion cells.

OBJECTIVES: Glaucoma is a leading cause of irreversible blindness, and effective therapies to reverse the visual system damage caused by glaucoma are still lacking. Recently, the stem cell therapy enable the repair and regeneration of the damaged retinal neurons, but challenges regarding the source of stem cells remain. This study aims to investigate a protocol that allows the dedifferentiation of Müller cells into retinal stem cells, following by directed differentiation into retinal ganglion cells with high efficiency, and to provide a new method of cellular acquisition for retinal stem cell

Microtubule dynamics in adult retinal ganglion cells and dorsal root ganglion neurons.

INTRODUCTION: While axon regeneration is very limited in the adult central nervous system (CNS) in vivo, this is not the case in the peripheral nervous system (PNS). Indeed, both CNS and PNS neurons can regenerate in vitro although to varying degrees. Given the role of microtubule stabilization in promoting regeneration, we have examined microtubule polymerization during the regeneration of two types of adult neurons in vitro, retinal ganglion cells (RGCs) from the CNS and dorsal root ganglion (DRG) neurons from the PNS. METHODS: In order to compare microtubule dynamics between these cell typ

Intrinsic electrical diversity of PV+ retinal ganglion cells.

Mouse retinal ganglion cells (RGCs) transform visual signals into spiking outputs and are known as a heterogeneous cell population. We combined electrophysiological recordings and anatomical analyses to characterize parvalbumin-positive (PV+) RGC subtypes, including ON-OFF direction selective (OODS), ON transient small receptive field (ON tr SmRF), OFF transient medium receptive field (OFF tr MeRF), and the OFF transient small receptive field (OFF tr SmRF) RGC. Intrinsic properties such as input resistance, maximum depolarization rate, and maximum rebound firing rate distinguished five functio

Gene Gun DiOlistic Labelling of Retinal Ganglion Cells.

Gene gun DiOlistic labelling enables the detailed visualization of retinal ganglion cells (RGCs) dendritic structure. Since the level of labelling is independent of cellular health, it is useful for the characterization of neuronal structure in degenerating neurons where expressed reporters may be inadequate. The method uses compressed helium gas to fire tungsten or gold microparticles coated in carbocyanine dyes (DiD, DiI, DiO) into flat mounted retinas. Here we describe the methods to optimize labelling and ensure a high yield of adequately labelled cells, with a focus on retinal ganglion ce

Retinal ganglion cells in Alzheimer's disease and aging.

Optic nerve and retinal ganglion cell (GC) degeneration are possible explanations for the poor visual function reported in patients with Alzheimer's disease (AD). We investigated whether GC loss could be attributed to AD compared with control subjects by measuring the spatial density of GC (cells/mm2) with methods previously used to analyze the GC distribution of young normal retinas. Retinas from 4 autopsy-confirmed, severely demented patients with AD and 4 age- and sex-matched control subjects (ages, 66-86 yr for both groups) without history of dementing or ocular disease were prepared as un
