Interstitial retinol-binding protein (IRBP) is a soluble glycoprotein present between the retina and pigmented epithelium, which may function to shuttle vitamin A derivatives between these tissues. While previous studies have shown that the retina is solely responsible for IRBP synthesis, the specific retinal cell(s) in which this occurs has not been established. Since the carbohydrate moiety of IRBP contains fucose, the authors have analyzed the sites of incorporation of 3H-fucose in the human retina in vitro, using autoradiography. Following a 30-min pulse incubation, all retinal layers exhi

BACKGROUND: Idiopathic interstitial nephritis (IIN) is common in the UK Indo-Asian population. Lack of systemic involvement and unremarkable urinalysis on stick testing suggest that it may underlie some cases of end-stage renal failure of undetermined cause. If IIN is diagnosed early, prompt initiation of treatment can improve long-term outcome.
AIMS: To investigate whether urinary retinol binding protein (RBP) is elevated more commonly than urinary albumin in IIN, and might be useful in the early detection of renal disease in Indo-Asian patients.
DESIGN: Preliminary observational study

Interphotoreceptor retinoid binding protein (IRBP), the major soluble protein component of the interphotoreceptor matrix, is believed to participate in the visual cycle by transporting retinoids between retinal pigment epithelium and photoreceptor cells in the eye. IRBP can associate with several chemical and isomeric forms of retinoids but displays the highest affinity towards the retinoids that are important in the visual cycle, 11-cis-retinal and all-trans-retinol. It was previously reported that IRBP can associate with 2 mol of all-trans-retinol or 2 mol of 11-cis-retinal per mol of protei

Interphotoreceptor retinoid-binding protein (IRBP) is an ocular protein which is believed to participate in the visual cycle by mediating transport of retinoids between pigment epithelium and photoreceptor cells. The molecular mechanism underlying the ability of IRBP to target particular retinoids to the specific cells that are their sites of action and metabolism is not completely clear, and little information is available regarding the structure of the protein's multiple ligand-binding sites. IRBP possesses two retinoid-binding sites, and it was reported that binding of the visual chromophor

Vitamin A, an unsaturated 20 carbon cyclic alcohol, has a variety of physiological functions including a role in vision, reproduction, growth and maintenance of epithelial and bone structures. The main sources of vitamin A are from preformed vitamin A in animal foods or from -carotene in green plants. Carotene is cleaved in the intestinal mucosa to retinol, which is transported in chylomicrons mainly to the liver which is the major storage site of vitamin A, where stores are mainly of retinyl palmitate. Utilization of vitamin A appears to be a highly regulated process; retinol is transported i

Autoimmune mechanisms are thought to be involved in the pathogenesis of the chorioretinal changes in ocular onchocerciasis. The humoral autoimmune response was determined by measuring serum levels of autoantibodies, directed against human S-antigen and interphotoreceptor retinoid binding protein (IRBP) using an enzyme immunoassay. The cell-mediated immune response to these antigens and a crude retinal extract was investigated by means of a two-step migration inhibition factor assay. Patients with onchocerciasis (n = 50) were subdivided into three groups: 1. without ocular involvement (n = 10),

Interstitial retinol-binding protein (IRBP), cellular retinol-binding protein (CRBP), and cellular retinal-binding protein (CRALBP) were localized in developing normal and RCS rat retinas. IRBP is found in the presumptive interphotoreceptor space in normal and RCS rats on P1. It reaches adult levels on P18. In the RCS rat, the level of IRBP decreases after P18 until there is only a small amount of staining on P38. CRBP and CRALBP are localized in the retinal pigment epithelium from P1 to adult. CRALBP is also found in the ciliary body pigment epithelium and in the outer epithelium of the iris

We aimed to characterize the vascular phenotypes of an experimental autoimmune retinal uveitis (EAU) model induced by interphotoreceptor retinoid-binding protein (IRBP) using multimodal imaging techniques. We systemically administered IRBP or vehicle to adult C57BL/6 mice. Fundus photography, optical coherence tomography (OCT), in vivo live confocal imaging using different tracers, OCT angiography (OCTA), and electroretinography (ERG) were performed after IRBP immunization. Hematoxylin and eosin and immunofluorescence staining were performed to characterize the immune response and vascular per

Intracellular binding proteins have been identified and isolated from B cells by their ability to bind to the synthetic peptide (1169-1191), the major immunodominant epitope of bovine interphotoreceptor retinoid-binding protein (IRBP) coupled to cyanogen bromide activated Sepharose 4B. After SDS-PAGE, two discrete protein bands of approximately 72 and 74 kDa, were found to be present in B cells of naive Lewis rats as well as in EBV transformed B cells from a human patient with ocular Behçet's disease. Enhanced expression of these peptide-binding proteins was achieved by incubating the cells wi

Interphotoreceptor retinoid-binding protein (IRBP) was studied using immunochemical and immunocytochemical techniques in retinae of mice with allelic combinations at the rd and rds loci at different stages of development and degeneration. Until postnatal day 7 (P7), IRBP is located intracellularly in developing retinae of the different genotypes. Thereafter, IRBP is present mainly in the interphotoreceptor matrix. As previously noted, cell death is slowest in the heterozygous +/+,rds/+ mutant with loss increasing in order in +/+,rds/rds, rd/rd, rds/rds and rd/rd,+/+ animals. The IRBP content o

PURPOSE: Myopia is a complex disorder with etiology involving an interplay between several genetic and environmental factors. Interphotoreceptor retinoid-binding protein (IRBP) is found in the subretinal space and is crucial in the visual cycle. The interphotoreceptor retinoid-binding protein knockout mouse (IRBP KO) was established as a model system to understand myopia and retinal degeneration. The current study investigated genes associated with myopia, retinal homeostasis, and inflammation in IRBP KO.
METHODS: RNA from retinas of congenic IRBP KO and wild-type C57BL/6J (WT) mice at postna

In closing, molecular biology has helped us in the study of uveitis, in defining the parts of the IRBP molecule that cause EAU/EAP. Simple northern and Southern blotting experiments can answer significant questions in the study of gene structure and expression and in relating these to genetic eye diseases. Molecular biology has provided answers to several long-standing cell biology question such as: where is IRBP synthesized, where is the gene expressed, and how is the IRBP polypeptide processed? In considering how mother nature invented the IRBP gene, the gene structure suggests some interest

Interleukin-2 (IL-2) was injected intraperitoneally into Lewis rats immunized with interphotoreceptor retinoid-binding protein (IRBP) to study its effect on the development of experimental autoimmune uveoretinitis (EAU) and experimental autoimmune pinealitis (EAP) and on the immune response. It was found that the EAU and the EAP were significantly intensified by IL-2, and the peak of serum anti-IRBP antibody appeared on the 7th day after immunization while that in the controls on the 26th day. These results, together with those previously documented that the development, severity and subsidenc

Lewis rats were immunized with varied doses of interphotoreceptor retinoid-binding protein (IRBP), a potent uveitogenic antigen. Experimental autoimmune uveoretinitis (EAU) and experimental autoimmune pinealitis (EAP) were induced with 25 micrograms and 50 micrograms of IRBP respectively. The EAU and EAP models provide useful means for investigating the pathogenetic mechanism of human uveoretinitis and its involvement with the nervous system.

Retinol-binding protein 3 (RBP3) is a photoreceptor-derived glyco-lipoprotein that plays a central role in retinoid trafficking between photoreceptors and the retinal pigment epithelium. Beyond its canonical role in the visual cycle, emerging evidence suggests that RBP3 may participate in retinal metabolic regulation, neurovascular protection, inflammatory modulation, and disease-related immune responses. This review critically summarizes current evidence regarding RBP3 in diabetic retinopathy, retinal degeneration, pathological myopia, primary congenital glaucoma, and uveitis. Among these con

Multiple functions for Interphotoreceptor Retinoid-Binding Protein (IRBP) may explain its localization in the retina, vitreous and pineal gland and association with retinitis pigmentosa and myopia. We have been engaged in uncovering the structure-function relationships of this interesting protein long thought to bind visual-cycle retinoids and fatty acids in the subretinal space. Although hydrophobic domains capable of binding such ligands have now been found, we ask what other structural domains might be present that could predict new functions? Interestingly, IRBP possesses a fold similar to

The aim of this work was to establish a modified pre-diagnostic polymerase chain reaction (PCR) protocol using a single primer set that enables successful amplification of a highly conserved mammalian sequence in order to determine overall sample DNA quality for multiple mammalian species that inhabit areas endemic for leishmaniasis. The gene encoding interphotoreceptor retinoid-binding protein (IRBP), but not other conserved genes, was efficiently amplified in DNA samples from tail skin, ear skin, bone marrow, liver and spleen from all of the species tested. In tissue samples that were PCR-po

Rhodopsin regeneration requires attachment between the retinal pigment epithelium (RPE) and rod outer segments; however, in experimentally induced retinal detachment, rhodopsin regeneration can be restored partially upon addition of IRBP (interphotoreceptor retinoid binding protein). The mi(vit)/mi(vit) (vitiligo) mutant mouse, a model of slowly progressing photoreceptor cell degeneration, has a marked elevation of IRBP at 4 weeks as well as progressive detachment of the retina. The purpose of this study was to determine whether this mutant is capable of regenerating rhodopsin within a few hou

The mechanisms by which pathogenic T cells enter the immune-privileged retina at the onset of uveitis remain unclear. To identify regulatory targets that promote retinal inflammation, we established an experimental autoimmune uveoretinitis (EAU) model by immunizing C57BL/6J mice with IRBP651-670 and dynamically evaluated retinal inflammation after induction; fundus lesions were prominent, with the first 14 days representing the initiation phase, Days 14-28 the amplification phase, Days 28-35 the peak phase, and Days 35-42 the regression phase. We characterized retinal cellular alterations from

The knockout (KO) of the adiponectin receptor 1 (AdipoR1) gene causes retinal degeneration. Here we report that ADIPOR1 protein is primarily found in the eye and brain with little expression in other tissues. Further analysis of AdipoR1 KO mice revealed that these animals exhibit early visual system abnormalities and are depleted of RHODOPSIN prior to pronounced photoreceptor death. A KO of AdipoR1 post-development either in photoreceptors or the retinal pigment epithelium (RPE) resulted in decreased expression of retinal proteins, establishing a role for ADIPOR1 in supporting vision in adulth
