Reversal learning and dopamine: a bayesian perspective.

Reversal learning has been studied as the process of learning to inhibit previously rewarded actions. Deficits in reversal learning have been seen after manipulations of dopamine and lesions of the orbitofrontal cortex. However, reversal learning is often studied in animals that have limited experience with reversals. As such, the animals are learning that reversals occur during data collection. We have examined a task regime in which monkeys have extensive experience with reversals and stable behavioral performance on a probabilistic two-arm bandit reversal learning task. We developed a Bayes

Neural correlates of reversal learning in frontotemporal dementia.

OBJECTIVE: Frontotemporal Dementia (FTD) is a neurodegenerative disorder that results in disinhibition and difficulty with flexible responding when provided feedback. Inflexible responding is observed early in the course of the illness and contributes to the financial and social morbidities of FTD. Reversal learning is an established cognitive paradigm that indexes flexible responding in the face of feedback signaling a change in reinforcement contingencies, with components of reversal learning associated with specific neurotransmitter systems. The objective of the study was to evaluate the ne

Lateralized nigrostriatal dopamine pathway activation promotes early reversal learning.

The dorsomedial striatum (DMS) is believed to promote action-outcome associations by integrating cortical and limbic afferents with nigrostriatal dopamine. This functional role for the DMS encompasses reinforcement learning processes traditionally ascribed to the ventral (outcome valuation) and lateral (action-selection) subdivisions of the striatum. Previous studies have shown that DMS dopamine signaling encodes actions and outcomes, often in a lateralized manner (e.g., dopamine release is greater for contralateral actions). To determine how these dynamics evolve with changing action-outcome

Relevance of Probabilistic Reversal Learning for Adolescent Drinking Trajectories.

One of the many human capabilities acquired during adolescence is the adaptivity in changing environments. In this longitudinal study, we investigated this adaptivity, as measured by probabilistic reversal learning (PReL) tasks, in N = 143 adolescents at ages 14, 16 and 18. Computational modelling and functional magnetic resonance imaging were applied to identify the neurocognitive processes underlying reversal learning and its development. Previous studies have demonstrated a correlation between heavy alcohol use and impaired reversal learning. Our hypothesis was that PReL is negatively assoc

Frontal Beta Transcranial Alternating Current Stimulation Improves Reversal Learning.

Electroencephalogram (EEG) studies suggest an association between beta (13-30 Hz) power and reversal learning performance. In search for direct evidence concerning the involvement of beta oscillations in reversal learning, transcranial alternating current stimulation (tACS) was applied in a double-blind, sham-controlled and between-subjects design. Exogenous oscillatory currents were administered bilaterally to the frontal cortex at 20 Hz with an intensity of 1 mA peak-to-peak and the effects on reward-punishment based reversal learning were evaluated in hundred-and-eight healthy volunteers. Dissociable effects of dopamine and serotonin on reversal learning.

Serotonin and dopamine are speculated to subserve motivationally opponent functions, but this hypothesis has not been directly tested. We studied the role of these neurotransmitters in probabilistic reversal learning in nearly 700 individuals as a function of two polymorphisms in the genes encoding the serotonin and dopamine transporters (SERT: 5HTTLPR plus rs25531; DAT1 3'UTR VNTR). A double dissociation was observed. The SERT polymorphism altered behavioral adaptation after losses, with increased lose-shift associated with L' homozygosity, while leaving unaffected perseveration after reversa

Distinct outcome processing in deterministic and probabilistic reversal learning.

Anticipations that fail to happen are important drivers of behavioral adaptation. Their processing appears to depend on the context. In a deterministic environment, where a stimulus unequivocally predicts the outcome, processing of absent outcomes involves the posterior orbitofrontal cortex (OFC). Failure has been linked to reality confusion with confabulations and disorientation. In a probabilistic environment, absent outcomes appear to be processed by the anterior cingulate cortex (ACC) rather than the OFC. Failure has been associated with poor decision making and schizophrenia. These data s

Acute vagus nerve stimulation enhances reversal learning in rats.

Cognitive flexibility is a prefrontal cortex-dependent neurocognitive process that enables behavioral adaptation in response to changes in environmental contingencies. Electrical vagus nerve stimulation (VNS) enhances several forms of learning and neuroplasticity, but its effects on cognitive flexibility have not been evaluated. In the current study, a within-subjects design was used to assess the effects of VNS on performance in a novel visual discrimination reversal learning task conducted in touchscreen operant chambers. The task design enabled simultaneous assessment of acute VNS both on r

Learning to reverse thermal diffusion.

Thermal diffusion, which governs heat transfer across a wide range of systems-from electronics to industrial processes-is inherently irreversible under the second law of thermodynamics, thus obscuring time-dependent information. To overcome this ill-posedness, this study introduces a physics-informed framework that centers on a novel time-reversal operator learning approach. A finite-difference-based network is first employed to robustly derive heterogeneous material properties. Crucially, the core innovation lies in the operator for thermal retrodiction. Distinct from traditional point-wise s

Reversal learning in C58 mice: Modeling higher order repetitive behavior.

Restricted, repetitive behaviors are diagnostic for autism and prevalent in other neurodevelopmental disorders. These behaviors cluster as repetitive sensory-motor behaviors and behaviors reflecting resistance to change. The C58 mouse strain is a promising model for these behaviors as it emits high rates of aberrant repetitive sensory-motor behaviors. The purpose of the present study was to extend characterization of the C58 model to resistance to change. This was done by comparing C58 to C57BL/6 mice on a reversal learning task under either a 100% or 80%/20% probabilistic reinforcement schedu

Impaired reversal learning in an animal model of anorexia nervosa.

BACKGROUND: Clinical investigations indicate that anorexia nervosa (AN) is associated with impaired cognitive flexibility. Activity-based anorexia (ABA), a rodent behavioral model of AN, is characterized by compulsive wheel running associated with voluntary food restriction and progressive weight loss. The goal of this study was to test whether ABA is associated with impaired cognitive flexibility. METHODS: Female Sprague-Dawley rats were trained to perform the attentional set-shifting test (ASST) to assess cognitive flexibility, including capacity for set-shifting and reversal learning. Rats

Altered EEG markers of reward learning during abstinence in alcohol dependence: A probabilistic reversal learning study.

OBJECTIVE: This study investigated neurophysiological and behavioural adaptations in reward learning and decision making which may contribute to the development and persistence of alcohol use disorder. METHODS: 20 abstinent alcohol dependent participants (mean abstinence: 20 months, range 1-76) and 26 healthy controls completed an electroencephalography (EEG) probabilistic reversal learning paradigm. Reinforcement learning modelling, event-related potentials (ERPs), and unsupervised machine learning (tensor decomposition) were used to characterise spatiotemporal patterns of reward valuation.

Visual Discrimination, Serial Reversal, and Extinction Learning in the mdx Mouse.

Duchenne muscular dystrophy (DMD) is the most common form of muscular dystrophy and the most common neuromuscular disorder. In addition to neuromuscular consequences, some individuals with DMD experience global intellectual dysfunction and executive dysfunction of unknown mechanistic origin. The cognitive profile of the mdx mouse, the most commonly used mouse model of DMD, has been incompletely characterized and has never been assessed using the touchscreen operant conditioning paradigm. The touchscreen paradigm allows the use of protocols that are virtually identical to those used in human co

Frontal hyperconnectivity related to discounting and reversal learning in cocaine subjects.

BACKGROUND: Functional neuroimaging studies suggest that chronic cocaine use is associated with frontal lobe abnormalities. Functional connectivity (FC) alterations of cocaine-dependent individuals (CD), however, are not yet clear. This is the first study to our knowledge that examines resting FC of anterior cingulate cortex (ACC) in CD. Because ACC is known to integrate inputs from different brain regions to regulate behavior, we hypothesized that CD will have connectivity abnormalities in ACC networks. In addition, we hypothesized that abnormalities would be associated with poor performance

The neuropsychology of ventral prefrontal cortex: decision-making and reversal learning.

Converging evidence from human lesion, animal lesion, and human functional neuroimaging studies implicates overlapping neural circuitry in ventral prefrontal cortex in decision-making and reversal learning. The ascending 5-HT and dopamine neurotransmitter systems have a modulatory role in both processes. There is accumulating evidence that measures of decision-making and reversal learning may be useful as functional markers of ventral prefrontal cortex integrity in psychiatric and neurological disorders. Whilst existing measures of decision-making may have superior sensitivity, reversal learni

Restricted lesions to ventral prefrontal subareas block reversal learning but not visual discrimination learning in rats.

Previous studies have shown that extensive damage to the medial prefrontal cortex (mPFC) of rats causes reversal learning deficits. The mPFC of rats, however, consists of several subareas that are different from each other in both cytoarchitecture and neural connectivity, suggesting a functional dissociation among the mPFC subareas. In the present study, selective lesions of the mPFC of rats were made with a specially designed microknife whose intracranial placement could be controlled stereotaxically. Restricted lesions were made to each of the 3 parts of the mPFC: the anterior cingulate area

Impairments in reversal learning following short access to cocaine self-administration.

BACKGROUND: Cocaine use disorder is characterized by compulsive drug-seeking that persists long into abstinence. Work using rodent models of cocaine addiction has found evidence for reversal learning deficits 21 days after non-contingent cocaine administration and 60 days after self-administration. Here we sought to determine if a deficit in reversal learning is present 3-4 weeks after cessation of cocaine self-administration, when relapse to cocaine-seeking is robust. Conversely, we hypothesized that reversal learning training would protect against relapse, similar to other forms of environme

Impaired Activation in Cognitive Control Regions Predicts Reversal Learning in Schizophrenia.

Reinforcement learning deficits have been associated with schizophrenia (SZ). However, the pathophysiology that gives rise to these abnormalities remains unclear. To address this question, SZ patients (N = 58) and controls (CN; N = 36) completed a probabilistic reversal-learning paradigm during functional magnetic resonance imaging scanning. During the task, participants choose between 2 stimuli. Initially, 1 stimulus was frequently rewarded (80%); the other was infrequently rewarded (20%). The reward contingencies reversed periodically because the participant learned the more rewarded stimulu

A GABAergic inhibitory neural circuit regulates visual reversal learning in Drosophila.

Inflexible cognition and behavior are prominent features of prefrontal cortex damage and several neuropsychiatric disorders. The ability to flexibly adapt cognitive processing and behavior to dynamically changing environmental contingencies has been studied using the reversal learning paradigm in mammals, but the complexity of the brain circuits precludes a detailed analysis of the underlying neural mechanism. Here we study the neural circuitry mechanism supporting flexible behavior in a genetically tractable model organism, Drosophila melanogaster. Combining quantitative behavior analysis and
