Phenotypic and genetic characterization indicated that Hup+ bean rhizobial strains are type IIA and type IIB Rhizobium tropici. The Hup+ strain USDA 2840, which did not cluster with either of the two types of R. tropici in a restriction fragment length polymorphism analysis, had electrophoretic patterns of PCR products generated with primers for repetitive extragenic palindromic and enterobacterial repetitive intergeneric consensus sequences similar to those of three reference strains of R. tropici type IIA. The Hup+ strain USDA 2738, which clustered with the reference strain of R. tropici IIB

Bacteroid inorganic phosphorus (Pi) metabolism in the Rhizobium-legume symbiosis differs between indeterminate and determinate legume nodules. In contrast to alfalfa bacteroids, bean (Phaseolus vulgaris) bacteroids exhibit high levels of alkaline phosphatase (AP), the native reporter enzyme for the bacterial Pi stress response. 14C and 32Pi whole plant labelling techniques were used in conjunction with diagnostic mutants (lacking AP or lacking high affinity Pi transport) to assess the relative importance of the Pi stress response in Rhizobium tropici bacteroids during symbiosis. The AP- mutant

l-Fucose, a 6-deoxy-monosaccharide, is often incorporated into O-antigens in Rhizobia where it serves as an important marker for host recognition. Here, we biochemically and structurally characterize WrtF, an O-antigen polysaccharide l-fucosyltransferase from the plant endosymbiont Rhizobium tropici CIAT 899. We show that WrtF transfers l-fucose from its donor, guanosine 5'-diphospho-β-l-fucose (GDP-Fuc), to a nonreducing end glucose, creating an α-(1→4) linkage. We determined structures of WrtF at resolutions between 1.45 and 2.3 Å in the apo state, and in complex with GDP, GDP-Fuc, nonacetyl

Characterization of nine transposon-induced mutants of Rhizobium tropici with decreased salt tolerance (DST) allowed the identification of eight gene loci required for adaptation to high external NaCl. Most of the genes also were involved in adaptation to hyperosmotic media and were required to overcome the toxicity of LiCl. According to their possible functions, genes identified could be classified into three groups. The first group included two genes involved in regulation of gene expression, such as ntrY, the sensor element of the bacterial ntrY/ntrX two-component regulatory system involved

The accumulation of potentially toxic metals (PTMs) poses a threat to the cellular integrity of soil microorganisms as well as ecological stability, driving the selection of resistant microorganisms with biotechnological potential. This study aimed to evaluate the behavior of the Rhizobium sp. LBMP-C04 isolate under PTM stress, focusing on adaptive resistance mechanisms and intracellular bioaccumulation. The bacterium, which was isolated from Arachis pintoi root nodules and had previously undergone genomic characterization, was subjected to antibiotic-resistance assays, growth-curve analysis i

Phaseolus vulgaris (common bean) is nodulated by diverse Rhizobium species. Although Ecuador is recognized as one of the centers of bean diversification, its native rhizobial diversity and geographic distribution remains poorly characterized. We isolated 46 native Rhizobium strains from root nodules across four Andean provinces (Imbabura, Pichincha, Chimborazo, and Loja). Partial sequencing of the recA gene delineated nine strain clusters (R1-R9) within two major phylogenetic groups: (i) Rhizobium ecuadorense/Rhizobium leguminosarum/Rhizobium etli/Rhizobium phaseoli and (ii) R. tropici. The phylogenetic relatedness among 12 agriculturally important species in the order Rhizobiales was estimated by comparative 16S rRNA and dnaK sequence analyses. Two groups of related species were identified by neighbor-joining and maximum-parsimony analysis. One group consisted of Mesorhizobium loti and Mesorhizobium ciceri, and the other group consisted of Agrobacterium rhizogenes, Rhizobium tropici, Rhizobium etli, and Rhizobium leguminosarum. Although bootstrap support for the placement of the remaining six species varied, A. tumefaciens, Agrobacterium rubi, and Agrobacterium vitis were co

Multilocus enzyme electrophoresis (MLEE) of 99 Brucella isolates, including the type strains from all recognized species, revealed a very limited genetic diversity and supports the proposal of a monospecific genus. In MLEE-derived dendrograms, Brucella abortus and a marine Brucella sp. grouped into a single electrophoretic type related to Brucella neotomae and Brucella ovis. Brucella suis and Brucella canis formed another cluster linked to Brucella melitensis and related to Rhizobium tropici. The Brucella strains tested that were representatives of the six electrophoretic types had the same rR

A 97-kDa purified aminopeptidase N (PepN) of Brucella melitensis was previously identified to be immunogenic in humans. The B. melitensis pepN gene was cloned, expressed in Escherichia coli and purified by affinity chromatography. The recombinant PepN (rPepN) exhibited the same biochemical properties, specificity and susceptibility to inhibitors as the native PepN. rPepN was evaluated as a diagnostic antigen in an indirect enzyme-linked immunosorbent assay (ELISA) using sera from patients with acute and chronic brucellosis. The specificity of the ELISA was determined with sera from healthy don

Background/Objectives: The vaginal microbiota constitutes a highly dynamic microbial ecosystem shaped by the distinct mucosal, hormonal, and immunological environment of the female genital tract. Accumulating evidence suggests that shifts in cervical microbial composition and function may influence host-microbe interactions and contribute to gynecological disease risk. Within this framework, the present study aimed to perform an in-depth genomic characterization of the cervical microbiota in a well-defined cohort of Greek women. The primary objective was to explore the functional microbial lan

We investigated the effects of Azospirillum brasilense strains Ab-V5 and Ab-V6 in the induction of mechanisms of systemic acquired resistance (SAR) and induced system resistance (ISR) on maize (Zea mays L.) plants. Under normal growth conditions, the treatments consisted of the standard inoculation of cells at sowing, and leaf spray of cells or their metabolites at the V2.5 growth stage; under saline stress (170 mM NaCl), the treatment consisted of standard single and co-inoculation of A. brasilense and Rhizobium tropici. The main compounds in the Azospirillum metabolites were identified as in
