Rhizoctonia cerealis (Rc) as a major pathogen causing sharp eyespot (SE) seriously affects wheat production worldwide. We collected 1683 wheat plants exhibiting SE symptoms from 30 locations in China over 7 years. We next constructed high-quality genome maps by de novo sequencing of five representative Rc isolates and established an atlas of genomic variation by resequencing 207 isolates. We identified a key effector, RcPL3, conferring Rc pathogenicity. RcPL3 increased the protein accumulation of histone deacetylase TaHDA9 in wheat cytoplasm to promote Rc infection. TaHDA9 inhibited the E3 ubi

Rhizoctonia solani AG1-IA is a polyphagous necrotrophic fungal pathogen that causes sheath blight disease in rice. Efforts are being made to identify pathogenicity-associated genes in R. solani and modulate them to develop a disease control strategy. Here, we investigated the roles of some predicted pathogenicity-associated genes of R. solani that have previously been reported to be upregulated during infection in rice. The tobacco rattle virus-based host-induced gene silencing of the selected pathogenicity-associated genes revealed that silencing of Rs_MEP1, a zinc-containing Peptidase_M43 do

The concern regarding the emergence of phytopathogens strains which are resistant to conventional agrochemicals has given support to the search for alternatives on the use of chemical pesticides in agriculture. In this context, microorganisms are considered as promising sources of useful natural compounds and actinobacteria are particularly relevant since they are known to produce several bioactive metabolites. The objective of this work was to investigate the production of secondary metabolites with antifungal activity by a strain of the actinobacteria Streptomyces lunalinharesii (A54A) under

Rhizoctonia solani AG3-TB is a destructive necrotrophic pathogen causing tobacco target spot disease. WRKY70 acts as a key regulator integrating salicylic acid (SA) and jasmonic acid (JA) signals in plant immunity. Our previous study found that NtWRKY70 in Nicotiana tabacum L. is significantly induced by R. solani AG3-TB infection; however, its functional mechanism remains elusive. In this study, bioinformatics analysis showed that NtWRKY70 is a nuclear localized protein with conserved domains and transcriptional autoactivation activity. Further investigations using virus-induced gene silencin

Rice sheath blight, caused by Rhizoctonia solani, has spread worldwide and causes serious economic losses. It is critically important to investigate alternative fungicides in light of the emergence of fungicide resistance and chemical residues. This study evaluated the natural substance α-pinene's activity. The R. solani strain's EC50 values for Taxodium 'Zhongshanshan' essential oil suppression of mycelial growth ranged from 201.99 to 1,523.37 μg/ml, and the EC50 value of α-pinene against R. solani was 111.31 μg/ml. Cell membrane permeability rose considerably after α-pinene treatment, althou

Dihydrofolate reductase (DHFR) is an essential enzyme in the folate pathway and has been recognized as a well-known target for antibacterial and antifungal drugs. We discovered eight compounds from the ZINC database using virtual screening to inhibit Rhizoctonia solani (R. solani), a fungal pathogen in crops. These compounds were evaluated with in vitro assays for enzymatic and antifungal activity. Among these, compound Hit8 is the most active R. solani DHFR inhibitor, with the IC50 of 10.2 μM. The selectivity of inhibition is 22.3 against human DHFR with the IC50 of 227.7 μM. Moreover, Hit8 h

The current study was designed to evaluate the antifungal properties of Datura metel L. against Rizoctonia solani Kuhn. To achieve this objective, six concentrations of leaves & stem methanol extract of D. metel viz. 1%, 1.5%, 2%, 2.5%, 3% & 3.5% were tested against R. solani in vitro. Leaf extract of D. metel was found more effective as its 3.5% concentration caused 75% retardation in test fungal growth as compared to the stem extract. D. metel methanolic leaf extract was fractioned between n-butanol, n-hexane, chloroform & ethyl acetate & bioactivities of isolated fractions were tested again

The antifungal properties and mechanism of three types of chitosan against the rice sheath blight pathogen, Rhizoctonia solani, were evaluated. Each chitosan had strong antifungal activity against R. solani and protected rice seedlings from sheath blight, in particular, two types of acid-soluble chitosan caused a 60-91 % inhibition in mycelial growth, 31-84 % inhibition of disease incidence, and 66-91 % inhibition in lesion length. The mechanism of chitosan in protection of rice from R. solani pathogen was attributed to direct destruction of the mycelium, evidenced by scanning and transmission

Three isolates of Pseudomonas aeruginosa were used for seed treatment of rice; all showed plant growth promoting activity and induced systemic resistance in rice against Rhizoctonia solani G5 and increased seed yield. Production of salicylic acid (Sal) by P. aeruginosa both in vitro and in vivo was quantified with high performance liquid chromatography. All three isolates produced more Sal in King's B broth than in induced roots. Using a split root system, more Sal accumulated in root tissues of bacterized site than in distant roots on the opposite site of the root system after 1 d, but this d

The pulsed electric field (PEF) of μs duration can induce electroporation by causing permanent damage to the membrane, leading to cell death. The microbe was treated by a homemade PEF generator instrument. The sterilization effect of PEF on the Rhizoctonia solani was observed by scanning electron microscope (SEM) and transmission electron microscope (TEM), and the leakage of the intracellular contents was measured with a conductometer and an ultraviolet spectrophotometer. The increases in the electrical conductivity and the optical density (OD) value indicated that the cell membrane was damage

Rhizoctonia solani is a soil-borne necrotrophic fungus that causes sheath blight in grasses. The basal resistance of compatible interactions between R. solani and rice is known to be modulated by some WRKY transcription factors (TFs). However, genes and defense responses involved in incompatible interaction with R. solani remain unexplored, because no such interactions are known in any host plants. Recently, we demonstrated that Bd3-1, an accession of the model grass Brachypodium distachyon, is resistant to R. solani and, upon inoculation with the fungus, undergoes rapid induction of genes res

During 2010-2012, a total of 120 isolates of Rhizoctonia cerealis were collected from wheat with symptoms of sharp eyespot in four provinces (Henan, Shandong, Anhui and Jiangsu) in China. All the isolates were determined for baseline sensitivity to thifluzamide, a succinate dehydrogenase inhibitor (SDHI) with strong antifungal activity. The sampled pathogenic populations, never exposed to SDHIs, had similar sensitivity to trifluzamide (0.025-0.359 µg/ml) in the four regions and over the two years. The baseline sensitivity was distributed as a skewed unimodal curve with a mean EC50 value (effec

A nanomicrocapsule system was constructed through the polymerization of tannic acid (TA) and emulsifier OP-10 (OP-10), followed by the chelation of iron ions, to develop a safe and effective method for controlling Rhizoctonia solani in agriculture. The encapsulated active component is a rosin-based triazole derivative (RTD) previously synthesized by our research group (RTD@OP10-TA-Fe). The encapsulation efficiency of the nanomicrocapsules is 82.39%, with an effective compound loading capacity of 96.49%. Through the encapsulation of the RTD via nanomicrocapsules, we improved its water solubilit
