Drug screening of rhodanine derivatives for antibacterial activity.

Introduction: Bacteriological infections are a major risk to human health. These include all hospital and public-acquired infections. In drug discovery, rhodanines are privileged heterocyclic frameworks. Their derivatives possess strong anti-bacterial activity and some of them have shown potent activity against multidrug-resistant pathogens, both under in vitro and in vivo conditions. To treat multi-drug resistant pathogens, the development of novel potent drugs, with superior anti-bacterial efficacy, is paramount. One avenue which shows promise is the design and development of novel rhodanine

Rhodanine derivatives as selective protease inhibitors against bacterial toxins.

In this study, we analyzed a series of rhodanine derivatives, as potential inhibitors of bacterial toxins, namely the proteases anthrax lethal factor and the botulinum neurotoxin type A. Conducting an extensive structure-activity relationship study on rhodanine derivatives, we profiled their selectivity against the two bacterial toxins and two related human metalloproteases using in vitro assays. In addition, we examined initial in vitro ADME-Tox properties of selected compounds and their ability to protect lethal factor-induced cell death of macrophages. These data allowed the selection of on

Rhodanine derivatives as potent anti-HIV and anti-HSV microbicides.

Although highly active antiretroviral therapies (HAART) remarkably increased life expectancy of HIV positive people, the rate of novel HIV-1 infections worldwide still represent a major concern. In this context, pre-exposure prophylaxis (PrEP) approaches such as vaginal microbicide gels topically releasing antiretroviral drugs, showed to have a striking impact in limiting HIV-1 spread. Nevertheless, the co-presence of other genital infections, particularly those due to HSV-1 or 2, constitute a serious drawback that strongly limits the efficacy of PrEP approaches. For this reason, combinations

Exploring the rhodanine universe: Design and synthesis of fluorescent rhodanine-based derivatives as anti-fibrillar and anti-oligomer agents against α-synuclein and 2N4R tau.

Tau and α-synuclein (α-syn) are prone-to-aggregate proteins that can be responsible for pathological lesions found in the brains of Alzheimer's disease (AD), Lewy body dementia (LBD), and Parkinson's disease (PD) patients. The early-stage oligomers and protofibrils of tau are believed to be strongly linked to human cognitive impairment while the toxic α-syn oligomers are associated with behavioral motor deficits. Therefore, concurrent targeting of both proteinaceous aggregates and oligomers are very challenging. Herein, rhodanine-based compounds were designed and synthesized to target the fibr

Arylidene and amino spacer-linked rhodanine-quinoline hybrids as upgraded antimicrobial agents.

Antibiotic resistance associated with various microorganisms such as Gram-positive, Gram-negative, fungal strains, and multidrug-resistant tuberculosis increases the risk of healthcare survival. Preliminary therapeutics becoming ineffective that might lead to noteworthy mortality presents a crucial challenge for the scientific community. Hence, there is an urgent need to develop hybrid compounds as antimicrobial agents by combining two or more bioactive heterocyclic moieties into a single molecular framework with fewer side effects and a unique mode of action. This review highlights the recent

Rhodanine-Inspired Development of Dual-Action Mycobacterium tuberculosis Protein Tyrosine Phosphatase B Inhibitors.

We developed a pharmacophore fusion strategy to design dual-action Mycobacterium tuberculosis protein tyrosine phosphatase B (MptpB) inhibitors. This involved integrating rhodanine-3-acetic acid with the nitroimidazooxazine/oxazole core of antitubercular nitroimidazoles. The resulting compounds showed potent MptpB inhibition and direct antituberculosis activity. Docking studies indicated a conserved binding mode, with the rhodanine moiety in the P1 pocket and the nitroimidazole core in the P2 pocket. Compound 4 was most promising, exhibiting strong MptpB inhibition (IC50: 0.19 μM), good antitu

Structural insights into 3-[5-(chlorobenzylidene)rhodanine]propionic acid isomers with antibacterial activity.

The rhodanine derivatives are a group of compounds known for their pharmacological action. Four new crystal structures of 3-[5-(chlorobenzylidene)rhodanine]propionic acid isomers, characterized by X-ray diffraction analysis and theoretical calculations, are reported. 3-[5-(2-Chlorobenzylidene)-4-oxo-2-sulfanylidene-1,3-thiazoliden-3-yl]propanoic acid, C13H10ClNO3S2, 1, crystallizes in the space group P21/c, with one molecule in the asymmetric unit. The other compounds, namely, 3-[5-(3-chlorobenzylidene)-4-oxo-2-sulfanylidene-1,3-thiazoliden-3-yl]propanoic acid, C13H10ClNO3S2, 2 and its polymor

A conceptual review of rhodanine: current applications of antiviral drugs, anticancer and antimicrobial activities.

Rhodanines are accepted as advantaged heterocycles in medicinal chemistry as one of the 4-thiazolidinones subtypes. The aim of this paper is to analyze the features of rhodanine and its application in pharmacy and medicine. Some of the properties of rhodanine such as antiviral, anticancer, antimicrobial, and drug discovery have recently been reported. Although there are still vague points in the structure and mechanism of polymerization of this substance, there is a significant increase in the use of rhodanine in medicine. In this review paper, it can be said that we have provided a general ov

Deciphering the Role of Rhodanine Flanked Non-Fullerene Acceptor Molecules for Efficient Organic Photovoltaics.

In recent years, extensive research has been conducted with the aim of developing non-fullerene acceptors as they have a promising ability to drive the development of cost-effective and highly efficient organic solar cells (OSCs). By harnessing the potential of rhodanine-flanked non-fullerene acceptors (NFAs), we proposed eight new A-D-A type NFAs (SBA1-SBA8) through precise end-cap modifications on both sides of the bridging-core unit. We performed various advanced quantum chemical analyses to unveil these designed materials' potential and compared them with the synthetic reference molecule (

Correlation of hepatic copper levels, rhodanine scores and histological diagnosis in archived canine liver samples.

The liver is the main storage site for copper. Excess copper accumulation, however, is a risk factor for the development of chronic hepatitis in dogs. Mass spectrometry or rhodanine staining are frequently used methods to assess copper levels in the liver. The association was studied between analytic hepatic copper levels and rhodanine scores in archived canine formalin-fixed-paraffinembedded liver sections from 2014 to 2021 with various diagnoses. Thirty-six (N = 36) liver samples with analytic interpretation of toxic (n = 12), high normal (n = 17), and normal (n = 7) copper levels were selec

Structure activity relationship studies on rhodanines and derived enethiol inhibitors of metallo-β-lactamases.

Metallo-β-lactamases (MBLs) enable bacterial resistance to almost all classes of β-lactam antibiotics. We report studies on enethiol containing MBL inhibitors, which were prepared by rhodanine hydrolysis. The enethiols inhibit MBLs from different subclasses. Crystallographic analyses reveal that the enethiol sulphur displaces the di-Zn(II) ion bridging 'hydrolytic' water. In some, but not all, cases biophysical analyses provide evidence that rhodanine/enethiol inhibition involves formation of a ternary MBL enethiol rhodanine complex. The results demonstrate how low molecular weight active site

Rhodanine and 2,4-dioxothiazolidine-based amides: synthesis and evaluation as dual-action cholinesterase inhibitors and antioxidants.

This study presents the synthesis and biological evaluation of substituted 2-(2,4-dioxothiazolidin-3-yl)-N-phenylacetamides and their thioxo analogues as potential multitarget agents for neurodegenerative diseases. A diverse library of amides was designed to explore structure-activity relationships and synthesized using two complementary approaches: a carbodiimide-mediated coupling and a microwave-assisted procedure. The compounds were screened for acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibition and antioxidant activity. Several derivatives displayed micromolar dual inh

Design, synthesis, and structure-activity studies of new rhodanine derivatives as carbonic anhydrase II, IX inhibitors.

Rhodanine and its derivatives are an important class of heterocycles with diverse biological properties, including anticancer, antibacterial, and anti-mycobacterial activities. In the present work, four series of new Rhodanine derivatives were synthesized and evaluated for their inhibitory activity against carbonic anhydrase I, II, IX, and XII isoforms. Interestingly, the tested compounds exhibited good inhibitory activity against the cytosolic isoform human carbonic anhydrase (hCA) II and tumor-associated hCA IX. Synthesis, Crystal Structures, Lipophilic Properties and Antimicrobial Activity of 5-Pyridylmethylidene-3-rhodanine-carboxyalkyl Acids Derivatives.

The constant increase in the resistance of pathogenic bacteria to the commonly used drugs so far makes it necessary to search for new substances with antibacterial activity. Taking up this challenge, we obtained a series of rhodanine-3-carboxyalkyl acid derivatives containing 2- or 3- or 4-pyridinyl moiety at the C-5 position. These compounds were tested for their antibacterial and antifungal activities. They showed activity against Gram-positive bacteria while they were inactive against Gram-negative bacteria and yeast. In order to explain the relationship between the activity of the compound
