The ecophysiology of uncultured Rhodocyclus-related polyphosphate-accumulating organisms (PAO) present in three full-scale enhanced biological phosphorus removal (EBPR) activated sludge plants was studied by using microautoradiography combined with fluorescence in situ hybridization. The investigations showed that these organisms were present in all plants examined and constituted 5 to 10, 10 to 15, and 17 to 22% of the community biomass. The behavior of these bacteria generally was consistent with the biochemical models proposed for PAO, based on studies of lab-scale investigations of enriche

Chlorite dismutase is a unique heme enzyme that catalyzes the conversion of chlorite to chloride and molecular oxygen. The enzyme is highly specific for chlorite but has been known to bind several anionic and neutral ligands to the heme iron. In a pH study, the enzyme changed color from red to green in acetate buffer pH 5.0. The cause of this color change was uncovered using UV-visible and EPR spectroscopy. Chlorite dismutase in the presence of acetate showed a change of the UV-visible spectrum: a redshift and hyperchromicity of the Soret band from 391 to 404 nm and a blueshift of the charge t

Addition of diesel fuel and waste engine oil to soil was found to cause biostimulation of hydrocarbon-oxidizing microorganisms. Corynebacteria constitute a large group of hydrocarbon-oxidizing microorganisms. Addition of a liquid culture of photosynthetic bacteria to soil not only facilitates degradation of petroleum products, but also stimulates growth of hydrocarbon-oxidizing microorganisms. Combined addition of photosynthetic bacteria and compost to soil polluted with petroleum products causes even a more significant increase in the count of hydrocarbon-oxidizing bacteria and substantially

Plasmids drive horizontal gene transfer and antimicrobial resistance spread in water resource recovery facilities, yet their in situ dynamics in complex microbial communities remain poorly characterized. We apply Hi-C metagenomics to resolve plasmid-host associations across influent, activated sludge, and effluent at three facilities. We identify 944 plasmid clusters with facility- and stage-specific distributions. Plasmid host range narrows from influent to effluent, indicating that treatment selectively constrains plasmid-host associations. Persistent plasmid clusters with detectable mobilit

Fermented foods contain less chemical pollutants, such as pesticide residues, than raw materials. In this study, using Pixian broad-bean paste as a model system, the pesticide degradation during food fermentation was comprehensively elucidated through metagenomic and metabolomics analyses. As a result, β-cypermethrin (β-CY) at 5 mg/kg was almost completely degraded, with a half-life of 6.1 d. β-CY caused flavor changes in fermented products, reducing esters and increasing ketones. Metagenomic analysis revealed that β-CY promoted bacteria phyla Bacteroidota, Pseudomonadota, and enriched the gen

The genetic and biochemical basis of perchlorate-dependent H2S oxidation (PSOX) was investigated in the dissimilatory perchlorate-reducing microorganism (DPRM) Azospira suillum PS (PS). Previously, it was shown that all known DPRMs innately oxidize H2S, producing elemental sulfur (So). Although the process involving PSOX is thermodynamically favorable (ΔG°' = -206 kJ ⋅ mol-1 H2S), the underlying biochemical and genetic mechanisms are currently unknown. Interestingly, H2S is preferentially utilized over physiological electron donors such as lactate or acetate although no growth benefit is obtai

Odor is inevitably produced during sewage sludge composting, and the subsequent pollution hinders the further development of composting technologies. Third-generation high-throughput sequencing was used to analyze microbial community succession, and the correlations between odor and microbial communities were evaluated. Hydrogen sulfide (47.5-87.9 %) and ammonia (9.4-49.9 %) contributed majorly to odor emissions, accounting for 93.7-98.5 % of the emissions. Volatile sulfur compounds were mainly produced in the mesophilic and pre-thermophilic phases (43.0-83.4 %), whereas ammonia was mainly pro

The ocular surface (OS) microbiota exhibits a 12-h ultradian rhythm in healthy mice, essential for ocular homeostasis, yet its regulation in type 2 diabetes mellitus (T2DM), associated with ocular surface diseases, remains unclear. This study investigated the temporal dynamics of the OS microbiota in control (NC) and T2DM C57BL/6J mice over a 24-h cycle (ZT0, ZT6, ZT12, ZT18) using 16S rRNA sequencing and JTK_CYCLE analysis of diversity indices, OTU composition, and family-level abundance. In NC mice, a robust 12-h rhythm with distinct temporal patterns was observed. In contrast, T2DM disrupte

A membrane bioreactor (MBR) was used to treat ciprofloxacin (CIP)-contaminated artificial wastewater. The microbial community structure and the abundance of antibiotic resistant genes (ARGs) in the MBR were studied at four CIP dosages (0, 5 mg·L-1, 10 mg·L-1, and 15 mg·L-1). The results showed that Proteobacteria and Bacteroidetes remained the dominant phylum, with relative abundances of 57.5% and 12.7%, respectively, as the dosage of CIP was increased from 0 mg·L-1 to 15 mg·L-1. Rhodocyclaceae, Chitinophagaceae, and Comamonadaceae became the dominant family with abundances of 29.96%, 5.44%, a

This work evaluated the interaction of perchlorate and trichloroethene (TCE), two common co-contaminants in groundwater, during bioreduction in serum bottles containing synthetic mineral salts media and microbial consortia. TCE at concentrations up to 0.3mM did not significantly affect perchlorate reduction; however, perchlorate concentrations higher than 0.1mM made the reduction of TCE significantly slower. Perchlorate primarily inhibited the reduction of vinyl chloride (VC, a daughter product of TCE) to ethene. Mechanistic analysis showed that the inhibition was mainly because perchlorate re

Macrophyte floating roots are considered as hotspots for methylmercury (MeHg) production in aquatic ecosystems through microbial activity. Nevertheless, very little is known about periphyton bacterial communities and mercury (Hg) methylators in such ecological niches. The ability to methylate inorganic Hg is broadly distributed among prokaryotes; however, sulfate-reducers have been reported to be the most important MeHg producers in macrophyte floating roots. In the present work, the periphyton bacterial communities colonizing Ludwigia sp. floating roots were investigated through molecular met

The performance of photo-sequencing batch reactor (PSBR) in removing multiple antibiotics and nutrients from aquaculture wastewater as well as the antibiotic resistance genes (ARGs) proliferation were firstly investigated during the long-term experiments. The operational conditions (i.e. light intensity, light time, aeration and solid retention time) were optimised to realise the simultaneous removal of antibiotics and nutrients. It was found that, compared with traditional SBR, PSBR has similar nutrient removal rate and a 30% higher antibiotics removal rate due to the corporation of microalga

UNLABELLED: Reactive chlorine species (RCS) defense mechanisms are important for bacterial fitness in diverse environments. In addition to the anthropogenic use of RCS in the form of bleach, these compounds are also produced naturally through photochemical reactions of natural organic matter and in vivo by the mammalian immune system in response to invading microorganisms. To gain insight into bacterial RCS defense mechanisms, we investigated Azospira suillum strain PS, which produces periplasmic RCS as an intermediate of perchlorate respiration. Our studies identified an RCS response involvin

INTRODUCTION: Oral lichen planus (OLP) is a chronic inflammatory condition often associated with desquamative gingivitis (DG). The oral microbiome's role in OLP and DG (OLP-DG) is gaining recognition, but prior 16S rRNA studies lacked taxonomic resolution. This study introduced shotgun metagenomic sequencing to thoroughly compare the supragingival and subgingival plaque microbiomes of individuals with and without OLP-DG.
METHODS: Twenty-seven participants (9 OLP-DG, 18 non-OLP) were recruited. Supra- and subgingival plaque samples were collected separately. Genomic DNA was analyzed using shot

Asthma, a chronic bronchial disorder prevalent in children/adolescents, is exacerbated under environmental conditions like dust storms. The current study investigated heavy metal levels, airborne bacteria, and serum IL-4/IL-8 in asthmatics during before/after dust storms in Iraq's Al-Anbar, Baghdad, and Kirkuk provinces. Airborne heavy metals were quantified by ICP-MS, serum cytokines by ELISA, and bacterial communities via metagenomics. Statistical analysis was performed using GraphPad Prism (p < 0.05 significant). ICP-MS revealed considerably elevated post-storm concentrations of As, Ag, B,

Soil microbial fuel cells (MFCs) are a sustainable technology that degrades organic pollutants while generating electricity. However, there have been no detailed studies of the mechanisms of pollutant degradation in soil MFCs. In this study, the effects of external resistance and electrode effectiveness on atrazine and hexachlorobenzene (HCB) degradation were evaluated, the performance of soil MFCs in the degradation of these pollutants under different soil redox conditions was assessed, and the associated microorganisms in the anode were investigated. With an external resistance of 20Ω, the d

Sulfur-based denitrification offers unique advantages in nitrogen removal in low-carbon environments. Despite its importance, the prevalence, controlling factors, and microbial mediators of sulfur-based denitrification in aquifers remain largely unknown. By integrating multiple stable isotopes and genome-resolved metagenomics, we investigated the coupling of nitrogen and sulfur cycling along the groundwater flowpath from unconfined to confined alluvial-lacustrine aquifers in the Poyang Lake Plain, China. Isotope (i.e., δ15N-NO3-, δ18O-NO3-, δ34S-SO42-, δ18O-SO42-, δ18O-H2O, δ13C-DIC, and δ13C-

Sulfides accumulation during anaerobic digestion negatively affects biogas quality, process stability, and infrastructure integrity, making efficient desulfurization strategies essential. This study investigated sulfur-oxidizing bacterial communities associated with anaerobic digesters and a Thiopaq® bioreactor at the Marrakech wastewater treatment plant. High-throughput 16S rRNA gene sequencing revealed a marked ecological contrast between the two systems. While the Thiopaq® bioreactor was largely dominated by a single Thioalkalibacteraceae lineage, anaerobic sludge environments harbored a ph

Adjuvants are substances that enhance adaptive immune response to antigen. Development of a safe and effective immunostimulant adjuvant is essential for the efficacy of a vaccine to protect against infectious pathogens. Purple non-sulfur photosynthetic bacteria exhibited nontoxic natural lipid A variants that are distinct in their chemical structures from that of the Escherichia coli-type lipid A. In this study, the adjuvant efficacy of attenuated lipid A variants and their corresponding lipopolysaccharides (LPSs), derived from purple photosynthetic bacteria (Rhodocyclus tenuis and Rhodobacter

Aerobic methane (CH4) oxidation coupled to denitrification (AME-D) is a promising wastewater treatment process for CH4 utilization and nitrogen removal. However, it is unclear which CH4-derived carbons are suitable for the AME-D process and how these organics are metabolized. In this study, metagenomics coupled with a thermodynamic model were used to explore the microorganisms and their metabolic mechanisms in an AME-D membrane biofilm reactor (MBfR) with high nitrogen removal efficiency. Results revealed that the aerobic methanotrophs of Methylomonas with the CH4-based fermentation potential
