Photoreversible Reactions of Visual and Microbial Rhodopsins.

The review discusses experimental findings and theoretical studies of photoreversible transformations of retinal-containing proteins (rhodopsins). Understanding these processes is essential for elucidating the mechanisms of photochemical and conformational transformations of these proteins. Particular emphasis is placed on visual rhodopsins, as photoreversibility plays a crucial role in its physiological regeneration in the rhabdoms of invertebrates and the cone photoreceptors of the vertebrate retina. In recent years, interest in the mechanisms of photoreversible reactions in retinal-containi

Light-Powered Transport of Organic Anions by Microbial Rhodopsins.

Microbial rhodopsins are photoactive membrane proteins known for transporting small inorganic ions such as H+, Cl-, and Na+. Their compact structure─comprising seven transmembrane helices─has long been thought to limit their substrate range to such ions. Here, we report that several anion-pumping rhodopsins can also transport organic anions. In particular, a rhodopsin from cyanobacteria transports bulky organic anions, including those with a benzene ring, with volumes up to ∼120 Å3─five times larger than Cl-. These anions bind in the dark state and are translocated upon photoactivation, via a

[A Study on Mechanisms Underlying Proton Transport in Proton Pump-type Microbial Rhodopsins].

Microbial rhodopsins are photoreceptive membrane proteins composed of seven transmembrane α-helical apoproteins (opsin) and a covalently bound retinal chromophore. Microbial rhodopsins exhibit a cyclic photochemical reaction referred to as photocycle when illuminated. During their photocycles, these proteins perform various functions such as ions transport and photosensing. Among the various functional types of rhodopsins found to date, we have focused on the utility of proton pump-type microbial rhodopsins as optogenetic tools for optical pH control in cells or organelles. To develop effectiv

Optogenetic manipulation of neuronal and cardiomyocyte functions in zebrafish using microbial rhodopsins and adenylyl cyclases.

Even though microbial photosensitive proteins have been used for optogenetics, their use should be optimized to precisely control cell and tissue functions in vivo. We exploited GtCCR4 and KnChR, cation channelrhodopsins from algae, BeGC1, a guanylyl cyclase rhodopsin from a fungus, and photoactivated adenylyl cyclases (PACs) from cyanobacteria (OaPAC) or bacteria (bPAC), to control cell functions in zebrafish. Optical activation of GtCCR4 and KnChR in the hindbrain reticulospinal V2a neurons, which are involved in locomotion, induced swimming behavior at relatively short latencies, whereas ac

Ectopic expression of a microbial-type rhodopsin restores visual responses in mice with photoreceptor degeneration.

The death of photoreceptor cells caused by retinal degenerative diseases often results in a complete loss of retinal responses to light. We explore the feasibility of converting inner retinal neurons to photosensitive cells as a possible strategy for imparting light sensitivity to retinas lacking rods and cones. Using delivery by an adeno-associated viral vector, here, we show that long-term expression of a microbial-type rhodopsin, channelrhodopsin-2 (ChR2), can be achieved in rodent inner retinal neurons in vivo. Furthermore, we demonstrate that expression of ChR2 in surviving inner retinal

Light-harvesting by antenna-containing rhodopsins in pelagic Asgard archaea

Aquatic bacterial rhodopsin proton pumps have been recently reported to utilize hydroxylated carotenoids1,2. Here, by combining a marine chromophore extract with purified archaeal rhodopsins identified in marine metagenomes, we report on light energy transfer from diverse hydroxylated carotenoids (lutein, diatoxanthin, and fucoxanthin) to heimdallarchaeial rhodopsins (HeimdallRs)3,4 from uncultured marine planktonic members of the 'Ca. Kariarchaeaceae' ('Ca. Asgardarchaeota')5. These light-harvesting antennas absorb in the blue-light range and transfer energy to the green-light absorbing retin

Excitation-wavelength- and protonation-dependent multiple photoreaction pathways in TAT rhodopsin from femtoseconds to seconds.

TAT rhodopsin is a member of the microbial rhodopsin family, a large group of seven-transmembrane photoreceptive proteins that bind an all-trans-retinal chromophore. While the Schiff-base linkage between the retinal and the protein moiety is protonated in most microbial rhodopsins, it is partially deprotonated in TAT rhodopsin, leading to the co-existence of a visible-absorbing protonated state and a UV-absorbing deprotonated state under physiological conditions. Here, we investigated the photoreaction dynamics of TAT rhodopsin in both the visible-absorbing protonated and UV-absorbing deproton

Voltage Imaging with Engineered Proton-Pumping Rhodopsins: Insights from the Proton Transfer Pathway.

Voltage imaging using genetically encoded voltage indicators (GEVIs) has taken the field of neuroscience by storm in the past decade. Its ability to create subcellular and network level readouts of electrical dynamics depends critically on the kinetics of the response to voltage of the indicator used. Engineered microbial rhodopsins form a GEVI subclass known for their high voltage sensitivity and fast response kinetics. Here we review the essential aspects of microbial rhodopsin photocycles that are critical to understanding the mechanisms of voltage sensitivity in these proteins and link the

Structural inhomogeneity and inter-chromophore electronic/protein-mediated coupling in the Gloeobacter rhodopsin-canthaxanthin complex: Insights from resonance Raman spectroscopy.

Microbial rhodopsins are photoreceptive membrane proteins that utilize retinal as a chromophore to absorb light and drive diverse photochemical functions. Recent studies have revealed that some microbial rhodopsins bind carotenoids in addition to retinal. The bound carotenoids may function as light-harvesting antennas in vivo by absorbing light in spectral regions where retinal absorbs less efficiently and transferring the excitation energy to the retinal chromophore. Here, we employed resonance Raman spectroscopy with excitation wavelengths from 440 to 600 nm to investigate the retinal and ca

FTIR Spectroscopy of a Viral Rhodopsin, OLPVR2.

Microbial rhodopsins in giant viruses are classified into two major classes: VR1 and VR2. Organic Lake Phycodnavirus Rhodopsin 2 (OLPVR2) belongs to the VR2 family, while OLPVR1 is a viral channelrhodopsin. OLPVR2 is known to form a pentamer, and it was proposed that OLPVR2 is a light-gated ion channel, whose ion pathway is not inside the monomer but in the pentameric center. OLPVR2 possesses a water-containing pentagonal cluster structure in the retinal Schiff base region, which is very similar to that of the OLPVR1 and bacteriorhodopsin (BR), a light-driven proton pump. Photochemistry of Acetabularia rhodopsin II from a marine plant, Acetabularia acetabulum.

Acetabularia rhodopsins are the first microbial rhodopsins discovered in a marine plant organism, Acetabularia acetabulum. Previously, we expressed Acetabularia rhodopsin II (ARII) by a cell-free system from one of two opsin genes in A. acetabulum cDNA and showed that ARII is a light-driven proton pump [Wada, T., et al. (2011) J. Mol. Biol. 411, 986-998]. In this study, the photochemistry of ARII was examined using the flash-photolysis technique, and data were analyzed using a sequential irreversible model. Five photochemically defined intermediates (P(i)) were sufficient to simulate the data.

Evidence for the acquisition of a proteorhodopsin-like rhodopsin by a chrysophyte-infecting giant virus.

Chrysophytes are nanoflagellate protists widespread in aquatic ecosystems with diverse trophic roles as primary producers and bacterivores. Molecular evidence suggests that chrysophytes are commonly infected by giant viruses, but isolates of such virus-host systems have not been reported. Here, we describe the first cultivated chrysophyte-infecting virus, Chrysophyceae Clade H virus SA1 (ChrysoHV), isolated along with its phago-mixotrophic host alga from surface waters in the tropical North Pacific Ocean. The ChrysoHV capsid (290 ± 40 nm diameter) is associated with a loose, sac-like membrane

Advantages of human opsins in optogenetic visual restoration.

Optogenetic vision restoration has progressed from proof-of-concept to early clinical testing, yet most programmes rely on microbial channels that demand high irradiance and offer limited adaptation. This review synthesizes preclinical and clinical evidence comparing microbial actuators with human opsins (rhodopsin, cone opsins, melanopsin) and outlines vector and safety considerations for translation. Human opsins activate G-protein-coupled cascades, providing intrinsic signal amplification and operation at room-light levels (∼1011-1012 photons⋅cm-2⋅s-1), in contrast to the ≥1015 photons⋅cm-2

Mechanism of the Irreversible Transition from Pentamer to Monomer at pH 2 in a Blue Proteorhodopsin.

Proteorhodopsin (PR) is a light-driven proton pump found in marine bacteria, and thousands of PRs are classified as blue-absorbing PRs (BPR; λmax ∼ 490 nm) and green-absorbing PRs (GPR; λmax ∼ 525 nm). We previously converted BPR into GPR using an anomalous pH effect, which was achieved by an irreversible process at around pH 2. Recent size-exclusion chromatography (SEC) and atomic force microscopy (AFM) analyses of BPR from Vibrio califitulae (VcBPR) revealed the anomalous pH effect owing to the irreversible transition from pentamer to monomer. Different pKa values of the Schiff base counteri

Functional Insertion of the Light-Induced Ion Pump KR2 into Block Copolymer Membranes.

Light-driven membrane proteins are attractive functional elements for biohybrid membranes, as they enable direct conversion of light energy into ion gradients. While microbial rhodopsins have been extensively studied in lipid bilayers, their controlled incorporation into synthetic polymer membranes remains challenging. Here, we investigate the reconstitution of the light-driven proton and sodium pump Krokinobacter eikastus rhodopsin 2 (KR2) into amphiphilic block copolymer membranes using a mild detergent-assisted strategy. KR2 and a C-terminal GFP fusion variant were incorporated into both so

A proteorhodopsin-based biohybrid light-powering pH sensor.

The biohybrid sensor is an emerging technique for multi-functional detection that utilizes the instinctive responses or interactions of biomolecules. We develop a biohybrid pH sensor by taking advantage of the pH-dependent photoelectric characteristics of proteorhodopsin (pR). The transient absorption kinetics study indicates that the photoelectric behavior of pR is attributed to the varying lifetime of the M intermediate at different environmental pH values. This pR-based biohybrid light-powering sensor with microfluidic design can achieve real-time pH detection with quick response and high s

A light-gated cation channel with high reactivity to weak light.

The cryptophyte algae, Guillardia theta, possesses 46 genes that are homologous to microbial rhodopsins. Five of them are functionally light-gated cation channelrhodopsins (GtCCR1-5) that are phylogenetically distinct from chlorophyte channelrhodopsins (ChRs) such as ChR2 from Chlamydomonas reinhardtii. In this study, we report the ion channel properties of these five CCRs and compared them with ChR2 and other ChRs widely used in optogenetics. We revealed that light sensitivity varied among GtCCR1-5, in which GtCCR1-3 exhibited an apparent EC50 of 0.21-1.16 mW/mm2, similar to that of ChR2, whe

The proteorhodopsins of the dinoflagellate Oxyrrhis marina: ultrastructure and localization by immunofluorescence light microscopy and immunoelectron microscopy.

At least 7 proteorhodopsin sequences of Oxyrrhis marina were recently proven in bands obtained by sucrose density gradient centrifugation, and MS analyses revealed that the bands consisted almost of pure, native proteorhodopsins (Rhiel et al. 2020). The proteorhodopsin fractions, i.e., bands B2, B3, and B4 were subjected to transmission electron microscopy. Negative staining revealed that band B2 consisted most likely of monomeric/oligomeric proteorhodopsins with particle dimensions of about 6 nm. Negative staining, freeze-fracture, and cryo-transmission electron microscopy revealed that bands

A tin oxide transparent electrode provides the means for rapid time-resolved pH measurements: application to photoinduced proton transfer of bacteriorhodopsin and proteorhodopsin.

An electrochemical cell was previously reported in which bacteriorhodopsin (BR, purple membrane) was adsorbed on the surface of a transparent SnO(2) electrode, and illumination resulted in potential or current changes (Koyama et al., Science 265:762-765, 1994; Robertson and Lukashev, Biophys. J. 68:1507-1517, 1995; Koyama et al., Photochem. Photobiol. 68:400-406, 1998). In this paper, we concluded that pH changes caused by proton transfer by the deposited BR or proteorhodopsin (PR) films lead to the flash-induced potential change in the SnO(2) electrode. Thus, the signals originate from BR and
