Characterization of ribulose-1,5-bisphosphate carboxylase/oxygenase carrying ribulose 1,5-bisphosphate at its regulatory sites and the mechanism of interaction of this form of the enzyme with ribulose-1,5-bisphosphate-carboxylase/oxygenase activase.

Ribulose-1,5-bisphosphate carboxylase/oxygenase [Rbu(1,5)P2CO] from plant sources shows a biphasic reaction course when assayed with more than 2 mM ribulose 1,5-bisphosphate [Rbu(1,5)P2]. In the burst, Rbu(1,5)P2CO has its substrate-binding sites occupied with Rbu(1,5)P2 for the initial few minutes, then both substrate-binding and regulatory sites are occupied by Rbu(1,5)P2 in the subsequent linear phase, at physiological concentrations of Rbu(1,5)P2 [A. Yokota (1991) J. Biochem. (Tokyo) 110, 246-252]. This study attempts the characterization of spinach Rbu(1,5)P2CO carrying Rbu(1,5)P2 at the

Effect of heat stress on the inhibition and recovery of the ribulose-1,5-bisphosphate carboxylase/oxygenase activation state.

Experiments were conducted to determine the relative contributions of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco; EC 4.1.1.39) activation state vis-a-vis Rubisco activase and metabolite levels to the inhibition of cotton (Gossypium hirsutum L.) photosynthesis by heat stress. Exposure of leaf tissue in the light to temperatures of 40 or 45 degrees C decreased the activation state of Rubisco to levels that were 65 or 10%, respectively, of the 28 degrees C control. Ribulose-1,5-bisphosphate (RuBP) levels increased in heat-stressed leaves, whereas the 3-phosphoglyceric acid pool was

Independent regulation of synthesis of form I and form II ribulose bisphosphate carboxylase-oxygenase in Rhodopseudomonas sphaeroides.

Ribulose 1,5-bisphosphate carboxylase-oxygenase (RuBPC-O) activity was greatly enhanced when Rhodopseudomonas sphaeroides was grown in a mineral salts medium supplied with 1.5% CO2 in hydrogen. Analysis of cell extracts by sodium dodecyl sulfate-polyacrylamide gel electrophoresis indicated that cells growing on 1.5% CO2 in H2 specifically accumulated RuBPC-O polypeptides. Quantitative immunological determinations revealed that accumulation of form I and form II RuBPC-O closely correlates with the increase of specific activity. However, the two enzymes appeared to be derepressed at different le

Induction of carbon monoxide dehydrogenase to facilitate redox balancing in a ribulose bisphosphate carboxylase/oxygenase-deficient mutant strain of Rhodospirillum rubrum.

A ribulose-1,5-bisphosphate carboxylase/oxygenase-deficient mutant strain (strain I-19) of Rhodospirillum rubrum was capable of growth under photoheterotrophic conditions in the absence of exogenous electron acceptors. These results suggested that alternative means of removing reducing equivalents have been acquired that allow this strain to remove reducing equivalents in the absence of a functional Calvin-Benson-Bassham reductive pentose phosphate pathway. Previously, the proton-reducing activity of the dinitrogenase complex was implicated in helping to maintain redox balance. However, since

A novel 51-kDa fragment of the large subunit of ribulose-1,5-bisphosphate carboxylase/oxygenase formed in the stroma of chloroplasts in dark-induced senescing wheat leaves.

The degradation of large subunit (LSU) of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) in wheat (Triticum aestivum L. cv. Yangmai 158) leaves was studied. A novel 51-kDa fragment was detected in leaf crude extracts and in chloroplast lysates from leaves with dark-induced senescence. Further studies showed that the 51-kDa fragment was found in the reaction solution with stroma fraction but not in that with the chloroplast membrane fraction and in the chloroplast lysates from mature wheat leaves. The reaction of producing the 51-kDa fragment was inhibited by 4-(2-aminoethyl) benzene

A Kinetic Characterization of Slow Inactivation of Ribulosebisphosphate Carboxylase during Catalysis.

The catalytic activity of ribulosebisphosphate carboxylase (Rubisco) declined as soon as catalysis was initiated by exposure to its substrate, d-ribulose-1,5-bisphosphate (ribulose-P(2)). The decline continued exponentially, with a half-time of approximately 7 minutes until, eventually, a steady state level of activity was reached which could be as low as 15% of the initial activity. The ratio of the steady state activity to the initial activity was lower at low CO(2) concentration and at low pH. The inhibitors 6-phosphogluconate and H(2)O(2) alleviated the inactivation, increasing the final/i

Effect of Aggregation and Molecular Size on the Ice Nucleation Efficiency of Proteins.

Aerosol acts as ice-nucleating particles (INPs) by catalyzing the formation of ice crystals in clouds at temperatures above the homogeneous nucleation threshold (-38 °C). In this study, we show that the immersion mode ice nucleation efficiency of the environmentally relevant protein, ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO), occurs at temperatures between -6.8 and -31.6 °C. Further, we suggest that this range is controlled by the RuBisCO concentration and protein aggregation. The warmest median nucleation temperature (-7.9 ± 0.8 °C) was associated with the highest concentratio

Chemoautotrophic growth of hydrogen-uptake-positive strains of Rhizobium japonicum.

Recently reported research from this laboratory has demonstrated the autotrophic growth of certain hydrogen-uptake-positive strains of Rhizobium japonicum and defined minimal conditions for such growth. Ribulose 1,5-bisphosphate carboxylase has been detected in autotrophically growing cells, but at low specific activity. Moreover, growth rates were low, and growth ceased at low cell densities. We report here improved autotrophic growth rates of R. japonicum SR through the use of a modified mineral salts/vitamins medium and a programmed increase in oxygen tension as autotrophic growth proceeds.

Rhizobium japonicum mutant strains unable to grow chemoautotrophically with H2.

Rhizobium japonicum strain SR grows chemoautotrophically on a mineral salts medium when incubated in an H2- and CO2-containing atmosphere. Mutant strains unable to grow or that grow very poorly chemoautotrophically with H2 have been isolated from strain SR. The mutant isolation procedure involved mutagenesis with ethyl methane sulfonate, penicillin selection under chemoautotrophic growth conditions, and plating of the survivors onto medium containing carbon. The resulting colonies were replica plated onto medium that did not contain carbon, and the plates were incubated in an H2- and CO2-conta

DNA-Specific DAPI Staining of the Pyrenoid Matrix During its Fission in Dunaliella salina (Dunal) Teodoresco (Chlorophyta).

The algal pyrenoid is a naked phase-separated liquid compartment inside the chloroplast consisting predominantly of densely packaged Rubisco and most often transversed by a system of lipid membranes. The pyrenoid participates in carbon-concentrating mechanisms of algae. During the cell division, the daughter cells of algae acquire the pyrenoids via their assembly or fission, the mechanisms of which are not fully understood. We suppose that the chloroplast nucleoid scaffolds the new pyrenoid like the cyanobacterial nucleoid positions carboxysomes before the cell division. This work was aimed at

Hornwort pyrenoids: a terrestrial exception with engineering lessons.

Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) underpins nearly all primary production yet remains a slow, error-prone enzyme because it catalyzes both carboxylation and oxygenation, the latter initiating photorespiration and reducing net carbon gain. Many organisms mitigate these limitations not by improving Rubisco selectivity for CO2 directly, but by modifying its local environment using CO₂-concentrating mechanisms (CCMs). In algae, a prominent biophysical CCM strategy is the pyrenoid: a phase-separated, Rubisco-rich condensate coupled to bicarbonate transport, local carbonic an

Synthetic biology approaches to improve Rubisco carboxylation efficiency in C3 Plants: Direct and Indirect Strategies.

Food security remains a pressing issue due to the growing global population and climate change, including the global warming along with increased atmospheric CO2 levels, which can negatively impact C3 crop yields. A major limitation in C3 plants is the inefficiency of Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) due to its low carboxylation activity and competing oxygenase activity. Improving Rubisco efficiency in C3 plants is thus essential for improving photosynthetic performance. Recent advances in synthetic biology have introduced promising strategies to overcome these limitat

Rubisco-Centric Strategies for Carbon Conservation in Synthetic Biology.

The escalating global climate crisis urgently demands biomanufacturing technologies with higher carbon efficiency. Ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco), the central enzyme catalyzing carbon dioxide fixation in the Calvin-Benson cycle, exhibits low efficiency, thereby limiting its industrial application. This review summarizes fundamental Rubisco research, laying the foundation for its efficiency-enhancing engineering and biomanufacturing applications. We first outline the discovery, systematic classification, and molecular evolution of Rubisco, and then elucidate the struc

Pyrenoid-based CO2-concentrating mechanisms across diverse lineages: convergent features and their applications for engineering.

Engineering pyrenoid-based CO2-concentrating mechanisms (pCCMs) into crop plants is a promising strategy to boost photosynthetic performance, enhance yield potential, and improve resilience to future climates. Achieving this goal requires a deeper understanding of the molecular principles that enable pyrenoids to elevate CO2 around Rubisco. Although pyrenoids span a remarkable diversity of forms across algae and hornworts, they consistently exhibit 3 core features: a condensed Rubisco matrix, specialized membranes that deliver inorganic carbon, and a diffusion barrier that restricts CO2 leakag

Architectural and biochemical advances in understanding microbial CO2 concentrating mechanisms.

CO2 concentrating mechanisms (CCMs) are ubiquitous in phototrophs. They are found in the micro as well as the macro world. Different domains of life have developed strikingly different approaches to concentrate inorganic carbon around Rubisco, the most prevalent enzyme on earth. While prokaryotes have developed carboxysomes, icosahedral shell protein cages, microbial eukaryotic phototrophs such as green algae, on the other hand, have developed pyrenoids, liquid-liquid phase separated organelles. In contrast to carboxysomes and pyrenoids, which represent biophysical CCMs, some vascular plants (

Rubisco functional integrity requires a dedicated three-step maturation pathway.

Proteins generally begin with methionine, which is often removed to expose the second residue. RuBP carboxylase/oxygenase (Rubisco), the most abundant enzyme on Earth and a major carbon and nitrogen reservoir, escapes this paradigm. Its catalytic subunit, RbcL, undergoes unusual amino terminal maturation, including atypical initiator methionine-serine cleavage and proline acetylation, a modification not seen in other biological systems. We define a specialized pathway of two sequential aminopeptidases and an amino terminal acetyltransferase, dedicated to RbcL, that executes processing. This ma

First glimpses into the molecular kaleidoscope of pyrenoid diversity.

A significant proportion of global photosynthetic carbon fixation relies on the pyrenoid, a biomolecular condensate found in the chloroplast of most unicellular algae, where the CO2-fixing enzyme Rubisco is exposed to saturating concentrations of the gas. In this review, we highlight recent advances in our understanding of the molecular basis of diverse pyrenoids. Phase separation of phylogenetically distant Rubiscos is mediated by convergently evolved linker proteins, with an emerging theme of pyrenoid condensation being organized via Rubisco-binding motifs. To minimize CO2 leakage out of the

Rubisco kinetic diversity and effectiveness of CO2-concentrating mechanisms in aquatic photosynthetic organisms.

Aquatic photoautotrophs experience strong physicochemical constraints on inorganic carbon acquisition due to low CO2 and O2 diffusion in water producing a strong reduction in the gas conductance. These limitations have driven the widespread evolution of CO2-concentrating mechanisms (CCMs), which enhance CO2 availability around Rubisco and mitigate its catalytic inefficiencies. A compilation of the Rubisco kinetics and apparent photosynthetic CO2 affinities (1/Km CO2) for a wide range of taxa shows that, despite the structural diversity of CCMs, there is an evolutive convergence among cyanobact

From seawater to pyrenoid: the molecular architecture of the diatom CO2-concentrating mechanism.

Diatoms are the most successful group of algae and their photosynthesis accounts for up to 20% of annual global CO2 fixation. A key factor to their ecological success is the operation of an efficient biophysical CO2-concentrating mechanism (CCM) that enriches CO2 for fixation by ribulose 1,5-bisphosphate carboxylase/oxygenase (Rubisco) in a specialized microcompartment called the pyrenoid. The diatom CCM is comprised of the following components, functional modules, and processes: (i) acquisition of external dissolved inorganic carbon (DIC); (ii) translocation of internal DIC to the chloroplast

The pyrenoid at a glance.

Ribulose-1,5-bisphosphate carboxylase-oxygenase (Rubisco) is the central CO2-fixing enzyme of the biosphere. It is present in virtually all photosynthetic organisms, including cyanobacteria, algae and land plants. Rubisco catalyses the carboxylation step of the Calvin-Benson-Bassham (CBB) cycle that underpins the light-independent 'dark' reactions of oxygenic photosynthesis. However, Rubisco exhibits a low catalytic rate and wasteful oxygenation activity, a constraint rooted in its ancient evolutionary origin. To overcome this specificity limitation, many photosynthetic lineages have evolved C
