Rewiring methanol metabolism in Komagataella phaffii through implementation and evolution of a heterologous RuMP cycle.

Biotechnology holds great potential for sustainable industrial production, with methanol emerging as a promising alternative carbon source due to its availability, cost-effectiveness, and high degree of reduction. Natural methylotrophic microorganisms, such as Komagataella phaffii, are well-suited for methanol-based processes. However, the native xylulose monophosphate (XuMP) cycle in K. phaffii is less energy-efficient than the bacterial ribulose monophosphate (RuMP) cycle, which requires fewer ATP molecules for methanol assimilation. In this study, we introduced the bacterial RuMP cycle as t

Awakening of the RuMP cycle for partial methylotrophy in the thermophile Parageobacillus thermoglucosidasius.

Given sustainability and scalability concerns of using sugar feedstocks for microbial bioproduction of bulk chemicals, widening the feedstock range for microbial cell factories is of high interest. Methanol is a one-carbon alcohol that stands out as an alternative feedstock for the bioproduction of chemicals, as it is electron-rich, water-miscible and can be produced from several renewable resources. Bioconversion of methanol into products under thermophilic conditions (>50 °C) could be highly advantageous for industrial biotechnology. Although progress is being made with natural, thermophilic

[Microbial synthesis of deuterium labelled L-phenylalanine with different levels of isotopic enrichment by facultative methylotrophic bacterium Brevibacterium methylicum with RMP assimilation of carbon].

The preparative microbial synthesis of amino acids labelled with stable isotopes, including deuterium ( 2 H), suitable for biomedical applications by methylotrophic bacteria was studied using L-phenylalanine as example. This amino acid is secreted by Gram-negative aerobic facultative methylotrophic bacteria Brevibacterium methylicum, assimilating methanol via ribulose-5-monophosphate (RMP) cycle of assimilation of carbon, The data on adaptation of L-phenylalanine secreted by methylotrophic bacterium В. methylicum to the maximal concentration of deuterium in the growth medium with 98% 2 Н 2 O a

Development of nucleic acid isolation by non-silica-based nanoparticles and real-time PCR kit for edible vegetable oil traceability.

For efficient extraction of amplifiable DNA from edible vegetable oils, we developed a novel DNA extraction approach based on the non-silica-based dipolar nanocomposites. The nanoparticle comprises a hydrophilic polymethyl methacrylate core with abundant capillaries, hydrophilic vesicles decorated with molecules having DNA affinity and a coating hydrophobic polystyrene layer. The nanoparticles are soluble in oil, adsorb the DNA from the aqueous phase and gave a high DNA recovery ratio. All DNA extracts from fully refined vegetable oil soybean, peanut, rapeseed, and cottonseed oils, including t

Crystal Structure, Mutations, and Catalytic Properties of 3-Hexulose-6-phosphate Synthase from Pyrococcus horikoshii.

3-Hexulose-6-phosphate synthase, a key enzyme in the ribulose monophosphate pathway, plays a central role in formaldehyde assimilation and detoxification, offering great potential for third-generation green biomanufacturing. In this study, we determined the crystal structure of HPS from Pyrococcus horikoshii at a 2.64 Å resolution. Combined approaches, including molecular docking, multiple sequence alignment, and alanine scanning, identified critical catalytic residues. Two variants, T136C and V186W, exhibited over 6-fold higher activity than the wild type. Molecular dynamics simulations indic

Structural modeling and docking studies of ribose 5-phosphate isomerase from Leishmania major and Homo sapiens: a comparative analysis for Leishmaniasis treatment.

Leishmaniases are caused by protozoa of the genus Leishmania and are considered the second-highest cause of death worldwide by parasitic infection. The drugs available for treatment in humans are becoming ineffective mainly due to parasite resistance; therefore, it is extremely important to develop a new chemotherapy against these parasites. A crucial aspect of drug design development is the identification and characterization of novel molecular targets. In this work, through an in silico comparative analysis between the genomes of Leishmania major and Homo sapiens, the enzyme ribose 5-phospha

Structure of mediator of RhoA-dependent invasion (MRDI) explains its dual function as a metabolic enzyme and a mediator of cell invasion.

Metastatic melanoma is among the most intractable cancers to treat; patients show resistance to therapy and limited survival time. A critical step in the development of metastatic melanoma is the acquisition of invasion and transition from thin to thick tumors on the skin, followed by invasion to lymph nodes. Prior studies have shown that metastatic melanoma is associated with dysregulation of RhoA and enhanced expression of a protein named "mediator of RhoA-dependent invasion (MRDI)". Importantly, MRDI is a "moonlighting" enzyme, with two distinct functions in melanoma cells. First, MRDI acts

Disclosing the essentiality of ribose-5-phosphate isomerase B in Trypanosomatids.

Ribose-5-phosphate isomerase (RPI) belongs to the non-oxidative branch of the pentose phosphate pathway, catalysing the inter-conversion of D-ribose-5-phosphate and D-ribulose-5-phosphate. Trypanosomatids encode a type B RPI, whereas humans have a structurally unrelated type A, making RPIB worthy of exploration as a potential drug target. Null mutant generation in Leishmania infantum was only possible when an episomal copy of RPIB gene was provided, and the latter was retained both in vitro and in vivo in the absence of drug pressure. This suggests the gene is essential for parasite survival.
