Assessment of genetic susceptibility to ethanol intoxication in mice.

Increased use of gene manipulation in mice (e.g., targeted or random mutagenesis) has been accompanied by increased reliance on a very few rapid and simple behavioral assays, each of which aspires to model a human behavioral domain. Yet, each assay comprises multiple traits, influenced by multiple genetic factors. Motor incoordination (ataxia), a common characteristic of many neurological disorders, may reflect disordered balance, muscle strength, proprioception, and/or patterned gait. Impaired motor performance can confound interpretation of behavioral assays of learning and memory, explorati

Influence of task parameters on rotarod performance and sensitivity to ethanol in mice.

Motor performance in mice can be assessed with multiple apparatus and protocols. Use of the rotarod (a.k.a. rotorod, rota-rod, roto-rod, or accelerod) is very common, and it is often used with the apparent assumption by the experimenters that it is a straightforward and simple assay of coordination. The rotarod is sensitive to drugs that affect motor coordination, including ethanol. However, there are few systematic data assessing the range of "normal" performance in mice. There are also few data exploring optimal task parameters (e.g. the influence of different speeds of rotation). In these e

Co-administration of digoxin and trans-2-decenoic acid ethyl ester improves motor learning performance in mice.

Neural circuit formation through synaptogenesis plays a crucial role in learning, memory, and the recovery of neural function following brain dysfunction. We previously reported that administering the low-dose cardiac glycoside digoxin, which activates brain Na/K-ATPase, promotes dendritic spine formation and improves motor learning. On the other hand, brain-derived neurotrophic factor (BDNF) is also involved in axon elongation, branching, attraction, and the maturation of dendritic spines. Since trans-2-decenoic acid ethyl ester (DAEE), an ester of medium-chain fatty acid with ten carbons, ac

Ataxic gait analysis in a mouse model of the olivocerebellar degeneration.

Lurcher mutant mice represent a model of olivocerebellar degeneration. Postnatally, a complete loss of Purkinje cells and secondary reduction of granule cells and inferior olive neurons occurs. Cerebellar ataxia is among the symptoms of degeneration of the cerebellum. The aim of the work was to identify gait parameters which are changed in Lurcher mice due to cerebellar ataxia arising from functional cerebellar decortication, and to assess the correlation between gait parameters, walking speed and performance in rotarod test. We used the adult Lurcher mutant and wild type mice of the B6CBA str

Behavioral assessment of the aging mouse vestibular system.

Age related decline in balance performance is associated with deteriorating muscle strength, motor coordination and vestibular function. While a number of studies show changes in balance phenotype with age in rodents, very few isolate the vestibular contribution to balance under either normal conditions or during senescence. We use two standard behavioral tests to characterize the balance performance of mice at defined age points over the lifespan: the rotarod test and the inclined balance beam test. Importantly though, a custom built rotator is also used to stimulate the vestibular system of

A Versatile, Behavioral Method to Investigate Thyroid Hormone Effects on Cerebellar Function.

Thyroid hormone (TH) action is essential during the development of the central nervous system, including the cerebellum. In case of TH deficiency in early life such as congenital hypothyroidism, patients display neurological disorders such as cognitive retardation and motor deficits. There are various studies using mouse models with tissue- or cell-specific TH deficiency to investigate the role of TH in the cerebellum. Compared to generalized congenital hypothyroid mice, cerebellar cell-specific TH-deficient mice display milder and subtler ataxic features, making the assessment of motor functi

The detection and measurement of locomotor deficits in a transgenic mouse model of Huntington's disease are task- and protocol-dependent: influence of non-motor factors on locomotor function.

Locomotor performance of transgenic R6/2 mice carrying the Huntington's disease (HD) mutation was assessed using four different tasks, fixed speed rotarod, accelerating rotarod, Digigait and footprint test. The tasks were compared directly in age- and CAG repeat-matched R6/2 mice. Accelerating rotarod was more sensitive than fixed speed rotarod for detecting early motor deficits in R6/2 mice. The sensitivity of accelerating rotarod increased with the acceleration rate and/or the start speed from which the rod accelerated. Differences between tasks were not due to inability of R6/2 mice to main

Impaired Cognitive Performance in Mice Exposed to Prolonged Hyperoxia.

Supplementation of oxygen at concentrations significantly above environmental level for prolonged periods may lead to hyperoxia and tissue toxicity. The mammalian brain undergoes structural and functional changes during adaptation to hypoxia and hyperoxia. In this study we investigated the effect of prolonged hyperoxic exposure on cognitive and motor performance in mice. Two-month-old male mice were placed in either hyperoxic (50% O2) or normoxic conditions for 3 weeks. Cognitive function was measured using the Y-maze test. High alteration rate between the three arms of the maze is indicative

Drug-free evaluation of rat models of parkinsonism and nigral grafts using a new automated rotarod test.

A variety of tests are available for the evaluation of behavioural deficits in rat models of hemiparkinsonism; many, however, are of limited applicability or insufficiently objective. The drug-induced turning behaviour test is widely used. A disadvantage of this test is that the use of drugs may lead to misleading results. Here, we describe a drug-free rotarod test that was used to evaluate the effects of unilateral 6-hydroxydopamine lesions, nigral grafts, and subrotational doses of apomorphine. The rotarod unit was automated and interfaced to a personal computer allowing automatic recording

Quantification of early learning and movement sub-structure predictive of motor performance.

Time-to-fall off an accelerating rotating rod (rotarod) is widely utilized to evaluate rodent motor performance. We reasoned that this simple outcome could be refined with additional measures explicit in the task (however inconspicuously) to examine what we call movement sub-structure. Our goal was to characterize normal variation or motor impairment more robustly than by using time-to-fall. We also hypothesized that measures (or features) early in the sub-structure could anticipate the learning expected of a mouse undergoing serial trials. Using normal untreated and baclofen-treated movement-

Behavioral Effects of Renin-Angiotensin System Modulators in a Rotenone-Induced Model of Parkinson's Disease in Wistar Albino Rats.

INTRODUCTION: Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor and non-motor dysfunction resulting from dopaminergic neuronal loss. Emerging evidence suggests a role of the renin-angiotensin system (RAS) in neurodegeneration, making it a potential therapeutic target. The present study aimed to evaluate the behavioral effects of captopril, perindopril, and losartan on motor, behavioral, and anxiety parameters in a rotenone-induced model of Parkinson's disease in Wistar albino rats. MATERIALS AND METHODS: An experimental in vivo study was conducted usi

Recovery and Regrowth After Nerve Repair: A Systematic Analysis of Four Repair Techniques.

BACKGROUND: Outcome assessments that evaluate post-transection nerve repair do not often correlate with one another. The aims of this study were twofold: to compare four nerve repair techniques with each other and incorporate both negative and positive control groups and to identify possible correlations between outcome assessments. MATERIALS AND METHODS: Sciatic nerve transection and repair was performed in Lewis rats using one of the following techniques: interrupted epineural, running epineural, grouped fascicular, epineural with absorbable type I collagen wrap, and high tension for incorp

Decreased performance in IDUA knockout mouse mimic limitations of joint function and locomotion in patients with Hurler syndrome.

BACKGROUND: Mucopolysaccharidosis type I (MPS I) is caused by the deficiency of alpha-L-iduronidase (IDUA), which is involved in the degradation of glycosaminoglycans (GAGs), such as heparan sulfate and dermatan sulfate in the lysosome. It has been reported that joint symptoms are almost universal in MPS I patients, and even in the case of attenuated disease, they are the first symptom that brings a child to medical attention. However, functional tests and biological markers have not been published for the evaluation of the limitations in joint and locomotion in animal model-mimicking MPS. ME

Murine motor and behavior functional evaluations for acute 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) intoxication.

Acute intoxication with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) induces nigrostriatal neurodegeneration that reflects Parkinson's disease (PD) pathobiology. The model is commonly used for rodent studies of PD pathogenesis and diagnostics and for developmental therapeutics. However, tests of motor function in MPTP-intoxicated mice have yielded mixed results. This unmet need reflects, in part, lesion severity, animal variability, and the overall test sensitivity and specificity. In attempts to standardize rodent motor function and behavioral tests, mice were trained on the rotarod or

Novel fused quinazolinones: further studies on the anticonvulsant activity of 1,2,9,11-tetrasubstituted-7H-thieno[2´,3´:4,5]pyrimido[6,1-b]-quinazolin-7-one and 1,3,10,12-tetrasubstituted-8H-pyrido[2´,3´:4,5]pyrimido[6,1-b]quinazolin-8-one.

BACKGROUND: Epilepsy is one of the most common neurological disorders, affecting approximately 1% of the world's population. The currently available anticonvulsants are effective in reducing the severity and number of seizures in less than 70% of patients. Moreover, their use is associated with undesirable side effects, ranging from cosmetic (gingival hyperplasia) to life-threatening (hepatotoxicity or megaloblastic anemia). Therefore, the continued search for safer and more effective anti-epileptic drugs is both urgent and necessary. A survey of the literature reveals that various derivatives

An automated rotarod method for quantitative drug-free evaluation of overall motor deficits in rat models of parkinsonism.

A variety of tests have been proposed for evaluation of motor deficits in rat models of hemiparkinsonism. The most widely used test, the "rotational behaviour" test, is based upon motor asymmetry induced by drug administration, leading to problems of sensitization, conditioning and priming. In addition, lesion-induced rotational behaviour is sometimes overcompensated by treatments (grafts) while other deficits are unaffected. However, rotational behaviour is reproducible and easy to quantify. On the contrary, most drug-free tests have proved of limited applicability because of subjective scali

Effects of buprenorphine on intracerebral collagenase-induced hematoma in Sprague-Dawley rats.

We evaluated the effects of buprenorphine (0.05 mg/kg intraperitoneally) after collagenase-induced intracerebral hemorrhage in Sprague-Dawley rats. Methods of evaluation included serum biochemistry, behavioral tests (neurologic exam and rotarod treadmill), and histopathology. Serum biochemistry parameters showed no change after surgery in controls and buprenorphine-treated animals. At 48 h after collagenase injections, the performance of treated rats on the rotarod treadmill test was not significantly different from that of untreated rats, but the neurologic exams of treated rats showed signif

Early motor deficits in mouse disease models are reliably uncovered using an automated home-cage wheel-running system: a cross-laboratory validation.

Deficits in motor function are debilitating features in disorders affecting neurological, neuromuscular and musculoskeletal systems. Although these disorders can vary greatly with respect to age of onset, symptomatic presentation, rate of progression and severity, the study of these disease models in mice is confined to the use of a small number of tests, most commonly the rotarod test. To expand the repertoire of meaningful motor function tests in mice, we tested, optimised and validated an automated home-cage-based running-wheel system, incorporating a conventional wheel with evenly spaced r
