Nitric oxide (NO) and nitroxyl (HNO) are closely related reactive nitrogen species with distinct biological functions, yet differentiating their chemistry remains challenging because of their high reactivity and overlapping reaction pathways. Developing simple protein-based platforms to study their reactivity is therefore of considerable interest. Rubredoxins are small, stable iron-thiolate proteins widely used as model systems for investigating metal-thiolate coordination and electron transfer. Here, we investigated the reactivity of native iron-rubredoxin (Fe-Rd) and its nickel-substituted a

Many microorganisms can degrade alkanes, using them as carbon source. The first and key step in alkane utilization is its hydroxylation, which requires a catalytic membrane-bound monooxygenase, a soluble rubredoxin and a soluble rubredoxin reductase. By comparing the phenotype of Pseudomonas aeruginosa strain TBCF10839 with an isogenic mutant that carries a plasposon within the rubredoxin reductase gene rubB (PA5349) and the complemented mutant, we report multiple, yet unknown roles of rubredoxin reductase in the physiology of P. aeruginosa beyond alkane hydroxylation. The plasposon mutant TBC

The rate constants (25 degrees , M(-1) sec(-1)), activation enthalpies (kcal/mol), and activation entropies (e.u.) for the second-order reduction of oxidized clostridial rubredoxin by Ru(NH(3))(6) (2+), V(H(2)O)(6) (2+), and Cr(H(2)O)(6) (2+) at I = 0.10 have been determined or estimated to be 9.5 x 10(4), approximately 1.4, approximately -31 (pH 6.3-7.0); 1.6 x 10(4), 0.1, -40 (pH 3.5-4.5); and 1.2 x 10(3), approximately 0, approximately -44 (pH 3.5-4.0), respectively. Ionic strength dependencies for the vanadium reaction are suggestive of a direct interaction of the reductants with the Fe(SR

Desulforedoxin (Dx), isolated from the sulfate reducing bacterium Desulfovibrio gigas, is a small homodimeric (2 x 36 amino acids) protein. Each subunit contains a high-spin iron atom tetrahedrally bound to four cysteinyl sulfur atoms, a metal center similar to that found in rubredoxin (Rd) type proteins. The simplicity of the active center in Dx and the possibility of replacing the iron by other metals make this protein an attractive case for the crystallographic analysis of metal-substituted derivatives. This study extends the relevance of Dx to the bioinorganic chemistry field and is import

Owing to the evolution of multi-drug-resistant and extremely drug-resistant Mycobacterium tuberculosis strains, there is an urgent need to develop new antituberculosis strategies to prevent TB epidemics in the industrial world. Among the potential new drug targets are two small nonheme iron-binding proteins, rubredoxin A (Rv3251c) and rubredoxin B (Rv3250c), which are believed to play a role in electron-transfer processes. Here, the solution structure and biophysical properties of one of these two proteins, rubredoxin B (Mt-RubB), determined in the zinc-substituted form are reported. The zinc-

The electron transfer protein rubredoxin from Clostridium pasteurianum contains an Fe(S-Cys)(4) active site. Mutant proteins C9G, C9A, C42G and C42A, in which cysteine ligands are replaced by non-ligating Gly or Ala residues, have been expressed in Escherichia coli. The C42A protein expresses with a Fe(III)(2)S(2) cluster in place. In contrast, the other proteins are isolated in colourless forms, although a Fe(III)(2)S(2) cluster may be assembled in the C42G protein via incubation with Fe(III)and sulfide. The four mutant proteins were isolated as stable mononuclear Hg(II)forms which were conve

Nitrosylation of the biomimetic reduced- and oxidized-form rubredoxin [Fe(SR)4]2-/1- (R = Ph, Et) in a 1:1 stoichiometry led to the formation of the extremely air- and light-sensitive mononitrosyl tris(thiolate) iron complexes (MNICs) [Fe(NO)(SR)3]- along with byproducts [SR]- or (RS)2. Transformation of [Fe(NO)(SR)3]- into dinitrosyl iron complexes (DNICs) [(RS)2Fe(NO)2]- and Roussin's red ester [Fe2(mu-SR)2(NO)4] occurs rapidly under addition of 1 equiv of NO(g) and [NO]+, respectively. Obviously, the mononitrosyl tris(thiolate) complex [Fe(NO)(SR)3]- acts as an intermediate when the biomime

Mycobacterium tuberculosis is a highly specialized human pathogen. The success of M. tuberculosis is due to its ability to replicate within host macrophages, resist host immune responses, and ultimately enter a persistent state during a latent tuberculosis infection. Understanding how M. tuberculosis adapts to and replicates in the intracellular environment of the host is crucial for the development of novel, targeted therapeutics. We report the characterization of an M. tuberculosis mutant lacking Rv3249c, a TetR transcriptional regulator. We show that Rv3249c directly represses the adjacent

The design of protein-metal complexes is rapidly advancing, with applications spanning catalysis, sensing, and bioremediation. We report a comprehensive investigation of METPsc1, a Miniaturized Electron Transfer Protein, in complex with cadmium. This study elucidates the impact of metal coordination on protein folding and structural dynamics across temperatures from 100 K to 300 K. Our findings reveal that METPsc1, composed of two similar halves stabilized by intramolecular hydrogen bonds, exhibits a unique "clothespin-like" recoil mechanism. This allows it to adapt to metal ions of varying ra

Proton transfer is essential in all multi-electron transformations. Identifying the proton donor in such processes is crucial to understanding the catalytic mechanism and optimizing activity through pKa matching, which can enhance proton transfer rates. However, aqueous solutions offer multiple proton sources, which confounds clear identification of the catalytically relevant proton donor. In this work, we utilize a model hydrogenase, nickel-substituted rubredoxin (NiRd), which is capable of electrocatalytic hydrogen evolution. Prior mechanistic studies suggested a proton transfer process was

Herein, we introduce an operationally simple approach for the direct covalent modification of metalloenzymes via Au(III)-mediated S-arylation chemistry. Using a model class of rubredoxin metalloenzymes containing Fe or Ni tetrathiolate active site, we demonstrate site-selective installation of diverse abiotic functional groups by targeting a single solvent-exposed cysteine residue distal to the metal center. The bioconjugation proceeds under mild aqueous conditions, rapidly, and in quantitative yields, while preserving the structural and functional integrity of the enzyme. Detailed structural

Spirochetes are suspected to be linked to the genesis of neurological diseases, including neurosyphillis or neurodegeneration (ND). Impaired iron homeostasis has been implicated in loss of function in several enzymes requiring iron as a cofactor, formation of toxic oxidative species, inflammation and elevated production of beta-amyloid proteins. This review proposes to discuss the link that may exist between the involvement of Treponema spp. in the genesis or worsening of ND, and iron dyshomeostasis. Proteins secreted by Treponema can act directly on iron metabolism, with hemin binding ability

J-proteins are obligate cochaperones of Hsp70s and stimulate their ATPase activity via the J-domain. Although the functions of J-proteins have been well understood in the context of Hsp70s, their additional co-evolved "physiological functions" are still elusive. We report here the solution structure and mechanism of novel iron-mediated functional roles of human Dph4, a type III J-protein playing a vital role in diphthamide biosynthesis and normal development. The NMR structure of Dph4 reveals two domains: a conserved J-domain and a CSL-domain connected via a flexible linker-helix. The linker-h

Tuberculosis (TB) is an infectious disease that affects humans and animals. The pathogens that cause TB belong to the Mycobacterium tuberculosis complex (MTBC), with M. tuberculosis and Mycobacterium bovis as the main representatives of human- and animal-adapted strains, respectively. One key genetic regulator of the MTBC members is the PhoPR system, which controls many processes, including the stress response, lipid metabolism and pathogenesis, among others. Previous studies identified a key G71I substitution in the M. bovis PhoR orthologue relative to M. tuberculosis PhoR and suggested that

When individual titratable sites in a molecule interact with each other, their pH titration can be considerably more complex than that of an independent site described by the classical Henderson-Hasselbalch equation. We propose a novel framework that decomposes any complex titration behavior into simple standard components. The approach maps the set of N interacting sites in the molecule onto a set of N independent, noninteracting quasi-sites, each characterized by a pK'(a) value. The titration curve of an individual site in the molecule is a weighted sum of Henderson-Hasselbalch curves corres

Ferruginous conditions prevailed through Earth's early oceans history, yet our understanding of biogeochemical cycles in anoxic iron-rich, sulfate-poor sediments remains elusive in terms of redox processes and organic matter remineralization. Using comprehensive geochemistry, cell counts, and metagenomic data, we investigated the taxonomic and functional distribution of the microbial subsurface biosphere in Lake Towuti, a stratified ferruginous analogue. Below the zone in which pore water becomes depleted in electron acceptors, cell densities exponentially decreased while microbial assemblages

D-Altronate hydratase and D-mannonate hydratase belong to a class of Fe2+-requiring enzymes, but the function of iron in these enzymes is largely unknown. Methods are described for the convenient preparation of both these hydratases from Escherichia coli and studies related to metal activation are presented. The enzymes are inactive in the absence of a bivalent metal and a reducing agent such as dithiothreitol. Fe2+ at low concentrations activates the enzymes efficiently, but inhibits them over 2 mM. Furthermore Mn2+ is also capable of activating aldonic acid hydratases and appears to be a con
