The genus Ruscus (Asparagaceae family) is native to the Mediterranean, Southern and Western Europe and is represented by perennial, rhizomatous, and evergreen shrubs. Among the approximately seven species spread throughout Europe up to Iran, Ruscus aculeatus L. (butcher's broom) is the most widely distributed and appreciated. This review provides an overview of the traditional use of Ruscus spp., the current knowledge of the chemistry of this genus, and the pharmacological studies carried out on Ruscus spp. extracts. The underground parts of Ruscus plants are a source of steroidal saponins tha

CONTEXT: Chronic orthostatic hypotension (OH) is frequently a severely debilitating disease that affects large groups of the population with autonomic insufficiency--the elderly; patients with diabetes, Parkinson's disease, and chronic fatigue syndrome; and anyone on drugs that affect the autonomic nervous system. Unfortunately, even though more than 60 medications are currently being used to treat OH, none of them is particularly or consistently effective. Ruscus aculeatus, a phytotherapeutic agent that is well known in Europe, may, however, change this. Its vasoconstrictive and venotonic pro

Ruscus hypoglossum L. is an evergreen ornamental species valued as cut foliage, but its commercial production is constrained by slow vegetative propagation and limited availability of uniform planting material. This study aimed to develop an efficient in vitro propagation protocol for R. hypoglossum by evaluating the effects of different cytokinins and spermidine on shoot organogenesis and subsequent rooting. Thidiazuron (TDZ), zeatin, kinetin, and spermidine were applied at 0.5 and 1.0 mg L-1 to rhizome explants derived from in vitro-grown seedlings. Explant length, shoot and cladode number,

Ruscus aculeatus L. (butcher's broom) is traditionally used in the management of chronic venous disorders; however, the molecular mechanisms underlying its vascular activity remain insufficiently characterized. This study investigated the effects of an 80% ethanolic rhizome extract on human cerebral microvascular endothelial hCMEC/D3 cells under normoxic and hypoxic conditions. The extract was phytochemically characterized using ultra-high-performance liquid chromatography-diode array detection-high-resolution mass spectrometry (UHPLC-DAD-HRMS), while its biological effects were evaluated by a

Alzheimer's disease is characterized by the accumulation of cerebral amyloid beta (Aβ) peptides, making the inhibition of Aβ production a key therapeutic strategy. We screened a plant extract library for Aβ42 inhibitors using human neural progenitor cells and identified a Ruscus aculeatus extract as a potential candidate. Although rhizome extracts of R. aculeatus exhibit vasotonic, and anti-inflammatory properties, their effects on amyloid-β precursor protein (APP) processing have not been investigated. We aimed to elucidate how R. aculeatus extract influences Aβ production and its underlying

The growing need for sustainable alternatives to antibiotics in aquaculture has increased interest in plant-derived antimicrobial compounds. This study characterized ethanol and methanol extracts from the cladodes and aerial parts of butcher's broom (Ruscus aculeatus L.) by GC-MS and screened, as a preliminary in vitro assessment, their antibacterial activity against Aeromonas hydrophila, Lactococcus garvieae, Vibrio anguillarum, Edwardsiella tarda, and Yersinia ruckeri. The extracts showed distinct chemical profiles. Phytol showed the highest relative chromatographic abundance in the ethanol

The endothelium, once considered merely a vascular lining responsible for selective permeability to water and electrolytes, is now recognised as a key regulator of vascular tone through the release of mediators such as oxylipins, nitric oxide, and hyperpolarizing factors. This in vitro study investigated the biological activity of Vesvein, a natural formulation containing Diosmin/Hesperidin, Ruscus aculeatus, Bromelain, and Ananas comosus, on intestinal and endothelial cells. Vesvein enhanced intestinal cell viability and preserved barrier integrity, as demonstrated by increased tight junction

The aim of this study is to evaluate the polyphenolic and flavonoid contents in the leaves extracts of Ruscus hypophyllum. Antioxidant activity was estimated by α,α-diphenyl-β-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulfonic acid (ABTS) assays. The anticoagulant activity of Ruscus extracts was evaluated in vitro, using the prothrombin time (PT) and a PTT-activated partial thromboplastin time tests. The antibacterial activity was tested against large number of important medically and resistant bacteria by the broth dilution method. In this study, ethyl acetate and chlor

BACKGROUND: Chronic venous disease and lymphoedema frequently coexist, leading to significant symptom burden and impaired quality of life (QoL). Phlebotonic agents, such as diosmin, Ruscus aculeatus, Melilotus officinalis and Vitis vinifera, have demonstrated complementary anti-inflammatory, venotonic and lymphokinetic effects. However, real-world evidence on their combined use remains limited.
METHODS: The VIVEMA Stasis study was a retrospective observational analysis conducted in a real-world clinical setting. Adult patients with lower limb venous and/or lymphatic oedema received a 30-day t

A detailed phytochemical investigation of rhizomes and roots of Ruscus aculeatus led to the isolation of fifteen pure compounds, belonging to the furostanol and spirostanol glycoside classes. Among them, one novel furostanol saponin (1) was identified, alongside twelve known furostanol (2-8) and spirostanol (11-15) saponins and two sulphated glycosides (9-10). Structural elucidation of all isolated compounds was accomplished through comprehensive 1D and 2D NMR spectroscopic analyses, complemented by mass spectrometry. The CHCl3 extract of Ruscus aculeatus and its major metabolite, the spirosta

Chronic venous disease (CVD) causes substantial symptom burden and healthcare cost, driven not only by venous hypertension but by a persistent molecular cascade of endothelial activation, glycocalyx degradation, and inflammation within the vein wall. Venoactive drugs (VADs) - a heterogeneous class of flavonoid, synthetic, animal-derived, and plant-derived compounds - target this cascade and are recommended by international guidelines as adjuncts to compression therapy, endovenous intervention, and wound care in venous leg ulcer (VLU) management. This review synthesizes the pharmacological rati

This paper continues our review of scientifically evaluated plant extracts in dermatology. After plants effective against dermatophytes, botanicals with anti-edema effects in chronic venous insufficiency are discussed. There is good evidence from randomized clinical studies that plant extracts from grape vine leaves (Vitis vinifera), horse chestnut (Aesculus hippocastanum), sea pine (Pinus maritima) and butcher's broom (Ruscus aculeatus) can reduce edema in chronic venous insufficiency. Plant extracts from witch hazel (Hamamelis virginiana), green tea (Camellia sinensis), the fern Polypodium l

Compression therapy is the gold standard treatment for venous leg ulcers (VLUs); however, with adjunctive pharmacological therapies and poor patient adherence using compressive dressings, clinicians are looking to find the advantage in treating VLUs. This literature review focuses on the efficacy of pharmacological agents, quality of life using agents in addition to compression therapy, and cost effectiveness to indicate the best outcomes for pharmacological treatment of VLUs. The following available venotonic, hemorheologic, and fibrinolytic agents were reviewed for oral management in treatin

Ruscogenin (RUS) is a steroidal sapogenin found in Ruscus aculeatus and Ophiopogon japonicus with several pharmacological activities. In the work reported herein, a novel method termed competitive fluorescence-linked immunosorbent assay (cFLISA) based on monoclonal antibodies (mAbs) coupled with quantum dots (QDs) was developed for the quick and sensitive determination of RUS in biological samples. The mAbs against RUS were conjugated with CdSe/ZnS QDs by the crossing-linking reagents and an indirect cFLISA method was developed. There was a good linear relationship between inhibition efficienc
