The S100 calcium-binding protein A4 level is elevated in the lungs of patients with idiopathic pulmonary fibrosis.

BACKGROUND: Fibroblast dysfunction is the main pathogenic mechanism of idiopathic pulmonary fibrosis (IPF). S100 calcium-binding protein A4 (S100A4) plays critical roles in the proliferation of fibroblasts and in the development of pulmonary, hepatic, and renal fibrosis. However, the clinical implications of S100A4 in IPF have not been evaluated. METHODS AND MATERIALS: The S100A4 mRNA and protein levels were measured by real-time PCR and immunoblotting in fibroblasts from IPF patients and controls. The S100A4 level was measured by enzyme-linked immunosorbent assay in bronchoalveolar lavage fl

Repurposing of ingenol mebutate for treating human colorectal cancer by targeting S100 calcium-binding protein A4 (S100A4).

Colorectal cancer (CRC) is the world's second most common cause of cancer-related death. Novel treatments are still urgently needed. S100 calcium-binding protein A4 (S100A4) was demonstrated to be an anticancer therapeutic target. Herein, we found that higher S100A4 expression was associated with a poorer prognosis in publicly available cohorts and a Taiwanese CRC patient cohort. To identify repurposed S100A4 inhibitors, we mined the Connectivity Map (CMap) database for clinical drugs mimicking the S100A4-knockdown gene signature. Ingenol mebutate, derived from the sap of the plant Euphorbia p

Serum S100 calcium binding protein A4 improves the diagnostic accuracy of transient elastography for assessing liver fibrosis in hepatitis B.

AIM: The diagnostic performance of Fibroscan might be improved when combined with other serum fibrosis related markers. Previous study has demonstrated that S100A4 expression is associated with liver fibrosis in humans with hepatitis. This study aimed to clarify diagnostic accuracy of serum S100A4 levels for significant liver fibrosis in patients with chronic hepatitis B (CHB), and develop a combined algorithm of liver stiffness measurement (LSM) and S100A4 to predict significant liver fibrosis in CHB. METHODS: One hundred and seventy-five CHB patients who had performed liver biopsy were cons

Inositol 1,4,5-trisphosphate receptor type 3 (ITPR3) is overexpressed in cholangiocarcinoma and its expression correlates with S100 calcium-binding protein A4 (S100A4).

Cholangiocarcinoma (CCA) is the second most malignant neoplasm in the liver that arises from the biliary tree. CCA is associated with a poor prognosis, and the key players involved in its pathogenesis are still not well understood. Receptor tyrosine kinases (RTKs), such as epidermal growth factor receptor (EGFR), can mediate intracellular calcium (Ca2+) signaling pathways via inositol 1,4,5-trisphosphate (InsP3), activating inositol 1,4,5-trisphosphate receptors (ITPRs) and regulating tumor growth. ITPR isoform 3 (ITPR3) is the main intracellular Ca2+ release channel in cholangiocytes. The eff

Activity of epidermal growth factor is directly affected by S100 proteins.

Epidermal growth factor (EGF) is a therapeutically important member of a family of the growth factors that activate EGF receptors, triggering several signaling pathways vital for development, homeostasis and regeneration of many organs. Previous studies have shown the ability of S100A4, a small calcium-binding protein of the S100 family, to directly affect functional activity of EGF receptors and its ligands, including EGF. However, selectivity and functional significance of EGF-S100 interactions remain unexplored. To address this, we examined specificity of EGF for twenty-one S100 proteins us

Hepatocyte-Derived S100A4/A6/A10/A11 Proteins Promote Liver Fibrosis by Direct and Indirect Activation of HSCs.

Liver fibrosis is a chronic disease caused by various etiologies, including dysregulated interactions among hepatocytes, KCs, and HSCs. Studies have shown that the S100 protein family plays an important role in liver fibrosis, especially the S100A4/A6/A10/A11 proteins; however, the effects of hepatocyte-derived S100A4/A6/A10/A11 proteins on the crosstalk between KCs and HSCs in liver fibrosis are unknown. The aim of this study was to investigate how hepatocyte-derived S100A4/A6/A10/A11 proteins modulate KC and HSC activation during liver fibrosis. Fibrotic, HCC and ICC human liver paraffin-emb

Molecular mechanisms of FSP1-regulated ferroptosis and therapeutic implications in various cancers.

Ferroptosis is an iron-dependent form of regulated cell death driven by lipid peroxidation. In addition to the canonical glutathione peroxidase 4 pathway, ferroptosis suppressor protein 1 (FSP1), also known as apoptosis-inducing factor mitochondria-associated 2, functions as a major glutathione-independent suppressor of ferroptosis. Current evidence indicates that FSP1 restrains lipid peroxidation mainly through the nicotinamide adenine dinucleotide (phosphate) [NAD(P)H]-dependent coenzyme Q10 axis. FSP1 has also been implicated in the noncanonical vitamin K cycle, which further expands its re

Network of interactions between the tumor necrosis factor superfamily members and small S100 proteins.

Tumor Necrosis Factor Superfamily (TNFSF) comprises 20 members of membrane/soluble signaling proteins regulating cell survival, cell proliferation/differentiation, and innate/adaptive immunity. Targeting signaling of TNFSF members is used clinically to treat several autoimmune and oncological diseases, and bone loss. They and their cognate receptors are in clinical trials as targets for treatment of autoimmune, inflammatory, oncological and other diseases. Recently, some representatives of S100 family of pleiotropic calcium-binding proteins were shown to interact with TNFSF members TNF and TRA

FSP1, a key ferroptosis suppressor with therapeutic potential.

Ferroptosis, a newly discovered iron-dependent regulated cell death, is characterized by aberrant iron metabolism and lipid peroxidation. Ferroptosis suppressor protein 1 (FSP1) acts as a key ferroptosis suppressor, independently of the canonical regulatory pathway, and exerts antioxidant effects by reducing coenzyme Q10 (CoQ10) and vitamin K (VK). Growing evidence regarding ferroptosis and FSP1 signaling has been published in recent years. On this basis, this review aims to give an updated and comprehensive description of FSP1, with a focus on its regulatory mechanisms, pathological roles in

Murine neonatal cardiac B cells promote cardiomyocyte proliferation and heart regeneration.

The irreversible loss of cardiomyocytes in the adult heart following cardiac injury leads to adverse cardiac remodeling and ventricular dysfunction. However, the role of B cells in cardiomyocyte proliferation and heart regeneration has not been clarified. Here, we found that the neonatal mice with B cell depletion showed markedly reduced cardiomyocyte proliferation, leading to cardiac dysfunction, fibrosis scar formation, and the complete failure of heart regeneration after apical resection. B cell depletion also significantly impaired heart regeneration and cardiac function in neonatal mice f

Hedgehog activity, epithelial-mesenchymal transitions, and biliary dysmorphogenesis in biliary atresia.

UNLABELLED: Biliary atresia (BA) is notable for marked ductular reaction and rapid development of fibrosis. Activation of the Hedgehog (Hh) pathway promotes the expansion of populations of immature epithelial cells that coexpress mesenchymal markers and may be profibrogenic. We examined the hypothesis that in BA excessive Hh activation impedes ductular morphogenesis and enhances fibrogenesis by promoting accumulation of immature ductular cells with a mesenchymal phenotype. Livers and remnant extrahepatic ducts from BA patients were evaluated by quantitative reverse-transcription polymerase cha

Contribution of S100A4-expressing fibroblasts to anti-SSA/Ro-associated atrioventricular nodal calcification and soluble S100A4 as a biomarker of clinical severity.

BACKGROUND: Fibrosis and dystrophic calcification disrupting conduction tissue architecture are histopathological lesions characterizing cardiac manifestations of neonatal lupus (cardiac-NL) associated with maternal anti-SSA/Ro antibodies. OBJECTIVES: Increased appreciation of heterogeneity in fibroblasts encourages re-examination of existing models with the consideration of multiple fibroblast subtypes (and their unique functional differences) in mind. This study addressed fibroblast heterogeneity by examining expression of α-Smooth Muscle Actin (myofibroblasts) and of S100 Calcium-Binding P

S100A4 triggeres the pyroptosis of vsmcs: association with mitochondrial damage, impaired mitophagy, and Ca2+ dysregulation.

BACKGROUND: Atherosclerotic occlusive disease of the lower extremities (ASO-LE) is associated with high rates of disability and mortality, and even after surgery, the rate of long-term lumen patency remains low. Vascular smooth muscle cells (VSMCs) are key players in atherosclerosis. The aim of this study was to investigate the action of S100 calcium-binding protein A4 (S100A4) in regulating VSMCs in ASO-LE. RESULTS: The overexpression of S100A4 in the vessel wall of ASO-LE is detected, and the expression of markers of pyroptosis is high. S100A4 causes mitochondrial damage in VSMCs, increases

Phenothiazines inhibit S100A4 function by inducing protein oligomerization.

S100A4, a member of the S100 family of Ca(2+)-binding proteins, regulates carcinoma cell motility via interactions with myosin-IIA. Numerous studies indicate that S100A4 is not simply a marker for metastatic disease, but rather has a direct role in metastatic progression. These observations suggest that S100A4 is an excellent target for therapeutic intervention. Using a unique biosensor-based assay, trifluoperazine (TFP) was identified as an inhibitor that disrupts the S100A4/myosin-IIA interaction. To examine the interaction of S100A4 with TFP, we determined the 2.3 A crystal structure of hum

Notch Blockade Specifically in Bone Marrow-Derived FSP-1-Positive Cells Ameliorates Renal Fibrosis.

BACKGROUND: The infiltration of inflammatory cells during a kidney injury stimulates myofibroblast activation leading to kidney fibrosis. Fibroblast-specific protein 1 (FSP-1) positive cells have been reported as either myofibroblasts or monocytes during tissue fibrosis. The functions of FSP-1+ cells that are associated with the development of renal fibrosis and the signaling pathways that regulate FSP-1+ cell activation have not been well defined. METHODS: In mice with unilateral ureteral obstruction (UUO), we characterized FSP-1+ cells and determined the role of the Notch signaling pathway

Tear Proteins Calcium binding protein A4 (S100A4) and Prolactin Induced Protein (PIP) are Potential Biomarkers for Thyroid Eye Disease.

There are no reliable biomarkers to predict thyroid eye disease (TED) in patients with autoimmune thyroid disease (AITD) currently. Several evidences support the involvement of the lacrimal gland in TED. The aim of our study was to quantitatively correlate the changes in tear protein profile with increasing severity of TED. Tear samples were collected from four groups of patients; AITD without TED (AITD), AITD with mild TED (mild TED), AITD with severe TED (severe TED) and normal controls. A total of 72 patients were recruited for the study. In discovery phase, isobaric tags for relative and a

A macrophage-related efferocytosis-based two-gene prognostic model for acute myeloid leukemia identified by multi-omics and machine learning.

Background Acute myeloid leukemia (AML) remains a lethal hematologic malignancy with high heterogeneity. Macrophage-mediated efferocytosis in the tumor microenvironment is implicated in immune suppression and disease progression. Methods We integrated single-cell and bulk transcriptomic data from public cohorts to identify genes associated with macrophages and efferocytosis in AML. Candidate genes were screened for prognosis using univariate Cox regression. A comprehensive machine learning framework, evaluating 117 algorithm combinations, was employed to construct a robust prognostic model. Th

Downregulation of S100 calcium-binding A4 (S100A4) ameliorates hepatic fibrosis via regulating Wnt/β-catenin signaling pathway.

S100 calcium-binding protein A4 (S100A4), a fibrosis-associated calcium-binding protein, has been implicated in fibrotic progression across multiple organs. Activation of the Wnt/β-catenin signaling pathway is a critical driver of hepatic fibrosis, yet the mechanistic role of S100A4 in this context remains poorly defined. This study investigated the regulatory role of S100A4 in hepatic fibrosis in vitro and in vivo. Hepatic stellate cells (HSCs) were treated with TGF-β to induce fibrotic activation, and S100A4 expression was silenced using shRNA. A carbon tetrachloride (CCl₄)-induced murine he
