As an effective non-thermal sterilization method, ultrasound remains at the level of passive bacterial death despite the initial understanding of its sterilization mechanism. Here, we present the perspective that bacteria can choose to actively enter an apoptosis-like death state in response to external ultrasonic stress. In this study, Vibrio parahaemolyticus exhibited apoptotic markers such as phosphatidylserine ectropion and activated caspases when subjected to ultrasound stress. Additionally, the accumulation of reactive oxygen species (ROS) and enhanced calcium signaling were observed. Fu

Suppression of the recA SOS response gene and reactive oxygen species (ROS) overproduction have been shown, separately, to enhance fluoroquinolone activity and lethality. Their putative synergistic impact as a strategy to potentiate the efficacy of bactericidal antimicrobial agents such as fluoroquinolones is unknown. We generated Escherichia coli mutants that exhibited a suppressed ΔrecA gene in combination with inactivated ROS detoxification system genes (ΔsodA, ΔsodB, ΔkatG, ΔkatE, and ΔahpC) or inactivated oxidative stress regulator genes (ΔoxyR and ΔrpoS) to evaluate the interplay of both

Biofilms are spatially structured microbial communities in which local physiological conditions vary across depth, potentially shaping DNA stress responses, repair pathways, and genome-plasticity-associated cellular states. However, the spatial organization of these transcriptional programs within individual bacterial biofilms remains poorly understood. Here, we re-analyzed public RAINBOW-seq spatial transcriptomic data from Escherichia coli BW25113 biofilms (GSE197541) using a curated 73-gene panel encompassing SOS response, stress response, biofilm matrix, conjugation- and competence-associa

BACKGROUND: In Staphylococcus aureus sub-populations of slow-growing cells forming small colony variants (SCVs) are associated with persistent and recurrent infections that are difficult to eradicate with antibiotic therapies. In SCVs that are resistant towards aminoglycosides, mutations have been identified in genes encoding components of the respiratory chain. Given the high frequencies of SCVs isolated clinically it is vital to understand the conditions that promote or select for SCVs.
RESULTS: In this study we have examined how exposure to sub-inhibitory concentrations of antibiotics with

Misonidazole, and RSU 1069 and 6 of its analogues are all reported to show increased cytotoxicity towards hypoxic cells compared to oxic cells. DNA is considered to be the target through which these drugs exert their cytotoxic activity. Therefore we monitored induction of the SOS response in uvrABC excinuclease proficient and deficient strains of E. coli, under oxic and hypoxic conditions, as an indirect method of assessing the activity of these drugs towards DNA in a biological system. This was done using the SOS chromotest which utilizes E. coli strains which possess a sfiA::lacZ fusion allo

Maintenance of genome integrity is critical for the survival and proliferation of bacteria. Following DNA damage, many bacteria launch the SOS response, which up-regulates many enzymes involved in DNA damage repair, as well as cell division inhibitors which coordinate DNA repair and cell division. YneA is a potent SOS response-inducible cell division inhibitor in Bacillus subtilis. It consists of a single transmembrane (TM) domain linked to an extracellular LysM domain. Previous studies suggest that YneA interferes with divisome assembly and septal peptidoglycan (sPG) synthesis, however, the u

In nucleotide excision repair DNA damage is removed through incision of the damaged strand on both sides of the lesion, followed by repair synthesis, which fills the gap using the intact strand as a template, and finally ligation. In prokaryotes the damaged base is removed in a 12-13 nucleotide (nt)-long oligomer; in eukaryotes including humans the damage is excised in a 24-32 nt-long fragment. Excision in Escherichia coli is accomplished by three proteins designated UvrA, UvrB, and UvrC. In humans, by contrast, 16 polypeptides including seven xeroderma pigmentosum (XP) proteins, the trimeric

The plasmid-mediated quinolone resistance (PMQR) genes have been shown to promote high-level bacterial resistance to fluoroquinolone antibiotics, potentially leading to clinical treatment failures. In Escherichia coli, sub-minimum inhibitory concentrations (sub-MICs) of the widely used fluoroquinolones are known to induce the SOS response. Interestingly, the expression of several PMQR qnr genes is controlled by the SOS master regulator, LexA. During the characterization of a small qnrD-plasmid carried in E. coli, we observed that the aminoglycosides become able to induce the SOS response in th

Like other Gram-negative bacteria, Vibrio cholerae, releases bacterial extracellular vesicles (BEVs), which have documented roles along the facultative human-pathogen's lifecycle. Most studies have focused on BEVs released its outermost surface under non-stressed conditions, which are mainly composed of outer membrane and periplasmic components. Herein, we comprehensively characterise stress-induced BEVs released upon exposure to the SOS response-inducing genotoxin mitomycin C or the antimicrobial emulsifier bile, which V. cholerae faces during intestinal colonisation. Compared to control BEVs

Human skin has developed elaborate defense mechanisms for combating a wide variety of potentially damaging environmental factors; principal among these is UV light. Despite these defenses, short-term damage may include painful sunburn and long-term UV damage results in both accelerated skin aging and skin cancers such as basal cell carcinoma, squamous cell carcinoma and even malignant melanoma. While UV radiation damages many cellular constituents, its most lasting effects involve DNA alteration. The following sections briefly review UV-inducible protective responses in bacteria and in skin, t

In some enterobacterial pathogens, but not in Escherichia coli, loss-of-function mutations in the ampD gene are a common route to beta-lactam antibiotic resistance. We constructed an assay system for studying mechanism(s) of enterobacterial ampD mutation using the well-developed genetics of E. coli. We integrated the Enterobacter ampRC genes into the E. coli chromosome. These cells acquire spontaneous recombination- and SOS response-independent beta-lactam resistance mutations in ampD. This chromosomal system is useful for studying mutation mechanisms that promote antibiotic resistance.

Exposure of E. coli AB1157 (a K-12 wild-type strain) to very large, daily X-ray doses has produced mutant strains resistant to both X-rays and UV photons. Results reported here are with KS0(160), the most resistant strain isolated thus far. Relative to its parent, KS0(160) is about 2.3 x more resistant to X-rays and about 2.2 x more resistant to ultraviolet photons (ratios of sensitivities in air). Two other characteristics of KS0(160) make its responses to X-rays different from those of AB1157: KS0(160) has an oxygen enhancement ratio of only 1.8 compared with 2.7 for its parent; glycerol red

The suppression of the SOS response has been shown to enhance the in vitro activity of quinolones. Furthermore, Dam-dependent base methylation has an impact on susceptibility to other antimicrobials affecting DNA synthesis. Here, we investigated the interplay between these two processes, alone and in combination, in terms of antimicrobial activity. A genetic strategy was used employing single- and double-gene mutants for the SOS response (recA gene) and the Dam methylation system (dam gene) in isogenic models of Escherichia coli both susceptible and resistant to quinolones. Regarding the bacte

During DNA replication, forks may encounter unrepaired lesions that hamper DNA synthesis. Cells have universal strategies to promote damage bypass allowing cells to survive. DNA damage tolerance can be performed upon template switch or by specialized DNA polymerases, known as translesion (TLS) polymerases. Human cells count on more than eleven TLS polymerases and this work reviews the functions of some of these enzymes: Rev1, Pol η, Pol ι, Pol κ, Pol θ and Pol ζ. The mechanisms of damage bypass vary according to the lesion, as well as to the TLS polymerases available, and may occur directly at

Samples of variously-sized total suspended particles in the air of one sampling site of TaiYuan city were collected. The samples were extracted with acid and a simulated lung fluid (SLF) respectively. Both extracts mainly consisted of metallic elements. Using SOS chromotest as a means to test the gentoxity of the extracts and five metallic compounds, namely Cr6+, Ni2+, Pb2+, Mn2+ and Cd2+ were tested first. All the compounds tested could induce the SOS response to various extent, showing that the method was sensitive to metallic compounds. For the extracts of air particles, both the extracts o

OBJECTIVE: Prophages, as an integral part of bacterial genomes, play a pivotal role in regulating host virulence and shaping bacterial evolution. Exposure to antibiotics, particularly those that cause DNA damage, can trigger a shift from lysogeny to the lytic cycle, i.e., initiate prophage induction.
METHODS: This review compares the effects of various antibiotic classes on prophage induction, with particular emphasis on the associated clinical consequences.
RESULTS: This process can lead to undesirable consequences, such as increased toxin production, enhanced transduction frequency, and mo

Antibiotics inhibit essential bacterial processes, resulting in arrest of growth and, in some cases, cell death. Many antibiotics are also reported to trigger endogenous production of reactive oxygen species (ROS), which damage DNA, leading to induction of the mutagenic SOS response associated with the emergence of drug resistance. However, the type of DNA damage that arises and how this triggers the SOS response are largely unclear. We found that several different classes of antibiotic triggered dose-dependent induction of the SOS response in Staphylococcus aureus, indicative of DNA damage, i

The host's immune system plays a key role in modulating growth of pathogens and the intestinal microbiota in the gut. In particular, inflammatory bowel disorders and pathogen infections induce shifts of the resident commensal microbiota which can result in overgrowth of Enterobacteriaceae ("inflammation-inflicted blooms"). Here, we investigated competition of the human pathogenic Salmonella enterica serovar Typhimurium strain SL1344 (S. Tm) and commensal E. coli in inflammation-inflicted blooms. S. Tm produces colicin Ib (ColIb), which is a narrow-spectrum protein toxin active against related

8,5'-Cyclopurines, making up an important class of ionizing radiation-induced tandem DNA damage, are repaired only by nucleotide excision repair (NER). They accumulate in NER-impaired cells, as in Cockayne syndrome group B and certain Xeroderma Pigmentosum patients. A plasmid containing (5'S)-8,5'-cyclo-2'-deoxyguanosine (S-cdG) was replicated in Escherichia coli with specific DNA polymerase knockouts. Viability was <1% in the wild-type strain, which increased to 5.5% with SOS. Viability decreased further in a pol II(-) strain, whereas it increased considerably in a pol IV(-) strain. Remarkabl
