Urine protein excretion after Hymenoptera sting.

The urinary excretion of albumin was measured on 7 consecutive days after a bee or wasp sting in 20 healthy persons, using a semiquantitative immunochemical chromatographic procedure (Micral stick). The mean albumin excretion was 11.1 +/- 15.0 (SD) mg/l. This was not significantly different from the findings in a control group of 17 volunteers with no history of bee or wasp sting (mean 8.7 +/- 17.4 mg/l). Pathologic albumin excretion (50-100 mg/l) was found in three of the sting group, with mean 37.2 +/- 23.7 mg/l. After 2 more months the urinary albumin excretion had normalized in two of the

Toosendanin inhibits STING signaling pathway to alleviate ulcerative colitis via blocking LYN kinase and STING protein.

The stimulator of interferon genes (STING) pathway plays a critical role in inflammatory diseases like ulcerative colitis (UC), yet no STING inhibitors are clinically available. Toosendanin (TSN), a bioactive triterpenoid from fruits of Melia toosendan, has been shown to ameliorate UC, though its exact molecular targets and detailed mechanisms remain unclear. This study investigates whether TSN inhibits STING pathway activation to alleviate UC and identifies potential target proteins involved in suppressing STING signaling. Herein, we first confirmed TSN's suppression of STING-dependent genes,

African Swine Fever Virus Structural Protein p17 Inhibits cGAS-STING Signaling Pathway Through Interacting With STING.

African swine fever virus (ASFV) encodes more than 150 proteins, which establish complex interactions with the host for the benefit of the virus in order to evade the host's defenses. However, currently, there is still a lack of information regarding the roles of the viral proteins in host cells. Here, our data demonstrated that ASFV structural protein p17 exerts a negative regulatory effect on cGAS-STING signaling pathway and the STING signaling dependent anti-HSV1 and anti-VSV functions. Further, the results indicated that ASFV p17 was located in ER and Golgi apparatus, and interacted with S

Human cytomegalovirus IE86 protein aa 136-289 mediates STING degradation and blocks the cGAS-STING pathway.

We previously reported that human cytomegalovirus (HCMV) 86 kDa immediate-early 2 gene product (IE86) promotes proteasome-dependent degradation of STING. In the present study, we determined the specific residues of IE86 responsible for STING degradation using a STING-firefly luciferase fusion protein expression system for quantitative meas-urement of STING protein levels. IE86 amino acids (aa) 136-289 were sufficient to promote STING degradation and further induced down-regulation of 2'3'-cyclic GMP-AMP (cGAMP)-mediated IFN-β promoter activation. Interestingly, transactivation domains (TAD) of

Allergens in hymenoptera venom XII: how much protein is in a sting?

The protein content of insect venoms was determined using a Coomassie blue dye binding assay as well as the Lowry phenol assay. The Lowry method tended to give excessively high values with many of the venoms because of the presence of low molecular weight phenolic compounds and other interfering substances. Using the dye binding method honey bee stings were found to contain 59 +/- 7 micrograms of protein, bumblebee and carpenter bee stings from 10 to 31 micrograms, yellow jacket stings from 1.7 to 3.1 micrograms, white faced hornet stings 2.4 to 5.0 micrograms and paper wasp stings from 4.2 to

Homeostatic regulation of STING protein at the resting state by stabilizer TOLLIP.

STING (stimulator of interferon genes) is an important innate immune protein, but its homeostatic regulation at the resting state is unknown. Here, we identified TOLLIP as a stabilizer of STING through direct interaction to prevent its degradation. Tollip deficiency results in reduced STING protein in nonhematopoietic cells and tissues, and renders STING protein unstable in immune cells, leading to severely dampened STING signaling capacity. The competing degradation mechanism of resting-state STING requires IRE1α and lysosomes. TOLLIP mediates clearance of Huntington's disease-linked polyQ pr

Wiskott-Aldrich syndrome protein restricts cGAS/STING activation by dsDNA immune complexes.

Dysregulated sensing of self-nucleic acid is a leading cause of autoimmunity in multifactorial and monogenic diseases. Mutations in Wiskott-Aldrich syndrome protein (WASp), a key regulator of cytoskeletal dynamics in immune cells, cause autoimmune manifestations and increased production of type I IFNs by innate cells. Here we show that immune complexes of self-DNA and autoantibodies (DNA-ICs) contribute to elevated IFN levels via activation of the cGAS/STING pathway of cytosolic sensing. Mechanistically, lack of endosomal F-actin nucleation by WASp caused a delay in endolysosomal maturation an

Pseudorabies virus EP0 protein targets cGAS and STING for degradation through autophagy.

The cGAS-STING axis is a central DNA-sensing pathway that initiates antiviral immune responses, yet the mechanisms by which viruses evade this defense system remain incompletely understood. Here, we identify the early protein EP0 of pseudorabies virus (PRV), a member of the Alphaherpesvirinae subfamily, as a viral virulence factor. Mechanistically, PRV EP0 suppresses cGAS-STING-mediated innate immunity by promoting the degradation of both cGAS and STING via the autophagy-lysosome pathway, thereby inhibiting downstream phosphorylation of TBK1 and IRF3. This degradation is strictly dependent on

Mutant huntingtin protein induces MLH1 degradation, DNA hyperexcision, and cGAS-STING-dependent apoptosis.

Huntington's disease (HD) is an inherited neurodegenerative disorder caused by an expanded CAG repeat in the huntingtin (HTT) gene. The repeat-expanded HTT encodes a mutated HTT (mHTT), which is known to induce DNA double-strand breaks (DSBs), activation of the cGAS-STING pathway, and apoptosis in HD. However, the mechanism by which mHTT triggers these events is unknown. Here, we show that HTT interacts with both exonuclease 1 (Exo1) and MutLα (MLH1-PMS2), a negative regulator of Exo1. While the HTT-Exo1 interaction suppresses the Exo1-catalyzed DNA end resection during DSB repair, the HTT-Mut

Pseudorabies virus tegument protein UL13 recruits RNF5 to inhibit STING-mediated antiviral immunity.

Pseudorabies virus (PRV) has evolved various immune evasion mechanisms that target host antiviral immune responses. However, it is unclear whether and how PRV encoded proteins modulate the cGAS-STING axis for immune evasion. Here, we show that PRV tegument protein UL13 inhibits STING-mediated antiviral signaling via regulation of STING stability. Mechanistically, UL13 interacts with the CDN domain of STING and recruits the E3 ligase RING-finger protein 5 (RNF5) to promote K27-/K29-linked ubiquitination and degradation of STING. Consequently, deficiency of RNF5 enhances host antiviral immune re

STING-ExositeDB: An AI-assisted curated database of protein exosites for drug discovery.

MOTIVATION: Protein exosites are secondary binding sites that recruit macromolecular partners and control molecular recognition across protein families. Despite their therapeutic potential demonstrated by approved drugs such as bivalirudin, systematic characterization remains limited by fragmented literature and inconsistent reporting. Existing databases lack exosite-specific curation, residue-level contact mapping, and structure-function integration. RESULTS: We present ExositeDB, a curated database of protein exosites built through AI-assisted curation combining large language model extract

Polyelectrolyte nanoparticles enable intracellular delivery of STING protein fragments for ovarian cancer immunotherapy.

Therapeutic activation of stimulator of interferon genes (STING) signaling shows promise as a method of inflaming the tumor microenvironment to generate an anti-cancer immune response. Frequent loss of STING expression in many human cancers renders small-molecule agonists ineffective in cancer cells. We previously demonstrated that delivering a fragment of the STING protein, dubbed STINGΔTM, to the cytosol can bypass this challenge by directly activating downstream signaling molecules in the cytosol; however, this strategy requires an effective cytosolic protein delivery vehicle to be therapeu

Protein Nanovaccines Co-Delivering STING Agonists for Enhanced Dendritic Cell Activation and Antitumor Immunity.

Cancer nanovaccines offer great promise for eliciting antitumor immune responses, yet challenges persist in achieving efficient antigen delivery, robust immune activation, and formulation simplicity suitable for practical application. Here, we present a generalizable vaccine design strategy based on protein cross-linking combined with innate immune stimulation via the STING agonist MSA-2. This approach enables the construction of self-adjuvanted protein nanovaccines that unify tumor antigen stabilization, lymph node delivery, and immune activation within a single nanosystem. Monkeypox virus protein OPG188 antagonizes cGAS-STING antiviral signaling pathway to mediate immune evasion.

Innate immune evasion is critical for productive viral replication. Activation of the cGAS-STING antiviral signaling pathway and its downstream effector genes plays a pivotal role in restricting viral replication during early DNA virus infection. Through a comprehensive genomic screen of monkeypox virus (MPXV), we identified three viral genes, OPG147, OPG188, and OPG200, whose expression potently suppresses cGAS-STING pathway activation. Notably, the N-terminal domain of the Poxin protein encoded by OPG188 exhibits nuclease activity and cleaves the cyclic dinucleotide second messenger 2'3'-cGA

RING finger protein 13 protects against nonalcoholic steatohepatitis by targeting STING-relayed signaling pathways.

Nonalcoholic fatty liver disease (NAFLD) is the most common liver disorder worldwide. Recent studies show that innate immunity-related signaling pathways fuel NAFLD progression. This study aims to identify potent regulators of innate immunity during NAFLD progression. To this end, a phenotype-based high-content screening is performed, and RING finger protein 13 (RNF13) is identified as an effective inhibitor of lipid accumulation in vitro. In vivo gain- and loss-of-function assays are conducted to investigate the role of RNF13 in NAFLD. Transcriptome sequencing and immunoprecipitation-mass spe

Brucella abortus Cyclic Dinucleotides Trigger STING-Dependent Unfolded Protein Response That Favors Bacterial Replication.

Brucella abortus is a facultative intracellular bacterium that causes brucellosis, a prevalent zoonosis that leads to abortion and infertility in cattle, and undulant fever, debilitating arthritis, endocarditis, and meningitis in humans. Signaling pathways triggered by B. abortus involves stimulator of IFN genes (STING), which leads to production of type I IFNs. In this study, we evaluated the pathway linking the unfolded protein response (UPR) and the endoplasmic reticulum-resident transmembrane molecule STING, during B. abortus infection. We demonstrated that B. abortus infection induces the

Ring-finger protein 5 protects against diabetic kidney disease by targeting and degrading STING.

The stimulator of interferon genes (STING) has been recognized as a pivotal modulator in the pathological progression of diabetic kidney disease (DKD). Ring-finger protein 5 (RNF5) is an important modulator of STING and is implicated in various disease processes. Nevertheless, the function of RNF5/STING axis in the context of DKD has yet to be investigated. This research was undertaken to explore whether RNF5 regulates the progression of DKD through modulation of the STING pathway, as well as to clarify the mechanisms involved. We found that RNF5 levels were significantly reduced in high gluco

SADS-CoV nucleocapsid protein antagonizes DAD1-mediated STING activation to prevent cytosolic DNA sensing.

The STING pathway is pivotal in defense against RNA viruses; however, its involvement in swine acute diarrhea syndrome coronavirus (SADS-CoV) infection remains unclear. This study reveals a dual mechanism where in SADS-CoV triggers STING activation via nuclear envelope rupture but subsequently evades immunity through its nucleocapsid (N) protein. Mechanistically, this process involves chromatin leakage that activates the cGAS-STING pathway, triggering interferon responses. The endoplasmic reticulum-resident protein defender against cell death 1 (DAD1) plays a key role in promoting STING phosph

Pseudorabies virus tegument protein US2 antagonizes antiviral innate immunity by targeting cGAS-STING signaling pathway.

BACKGROUND: The cGAS-STING axis-mediated type I interferon pathway is a crucial strategy for host defense against DNA virus infection. Numerous evasion strategies developed by the pseudorabies virus (PRV) counteract host antiviral immunity. To what extent PRV-encoded proteins evade the cGAS-STING signaling pathway is unknown. METHODS: Using US2 stably expressing cell lines and US2-deficient PRV model, we revealed that the PRV tegument protein US2 reduces STING protein stability and downregulates STING-mediated antiviral signaling. RESULTS: To promote K48-linked ubiquitination and STING degra

Varicella-Zoster Virus ORF39 Transmembrane Protein Suppresses Interferon-Beta Promoter Activation by Interacting with STING.

Varicella-Zoster virus (VZV) causes varicella in primary infection of children and zoster during reactivation in adults. Type I interferon (IFN) signaling suppresses VZV growth, and stimulator of interferon genes (STING) plays an important role in anti-VZV responses by regulating type I IFN signaling. VZV-encoded proteins are shown to inhibit STING-mediated activation of the IFN-β promoter. However, the mechanisms by which VZV regulates STING-mediated signaling pathways are largely unknown. In this study, we demonstrate that the transmembrane protein encoded by VZV open reading frame (ORF) 39
