Sample Size Justification in Quantitative Research.

BACKGROUND: Sample-size justification remains a central element of quantitative research, yet classical a priori power analysis is frequently applied to designs for which it is neither required nor conceptually appropriate. When power calculations are used indiscriminately, they may misalign with the aims of studies that do not need them. This poses challenges for researchers and student nurses, particularly those without specialist training in statistics, and can lead to methodological inconsistencies across the published nursing literature. OBJECTIVES: In this paper, I aim to clarify how sa

Sample size calculation in economic evaluations.

A simulation method is presented for sample size calculation in economic evaluations. As input the method requires: the expected difference and variance of costs and effects, their correlation, the significance level (alpha) and the power of the testing method and the maximum acceptable ratio of incremental effectiveness to incremental costs. The method is illustrated with data from two trials. The first compares primary coronary angioplasty with streptokinase in the treatment of acute myocardial infarction, in the second trial, lansoprazole is compared with omeprazole in the treatment of refl

Sample size for comparing correlated concordance rates.

We consider collecting the measurements of a gold standard and two methods with error from each site of subjects. We propose an asymptotic test statistic to compare the concordance rates of two methods with the gold standard and a closed-form sample size formula. Through simulations, we show that the test statistic accurately controls the type I error in small sample sizes, and the sample size formula accurately maintains the power in various settings. The proposed test statistic and the sample size formula can be easily modified for other indices for agreement such as sensitivity and specific

Power and sample size estimation in microarray studies.

BACKGROUND: Before conducting a microarray experiment, one important issue that needs to be determined is the number of arrays required in order to have adequate power to identify differentially expressed genes. This paper discusses some crucial issues in the problem formulation, parameter specifications, and approaches that are commonly proposed for sample size estimation in microarray experiments. Common methods for sample size estimation are formulated as the minimum sample size necessary to achieve a specified sensitivity (proportion of detected truly differentially expressed genes) on ave

Sample size calculation for comparing two ROC curves.

Biomarkers are key components of personalized medicine. In this paper, we consider biomarkers taking continuous values that are associated with disease status, called case and control. The performance of such a biomarker is evaluated by the area under the curve (AUC) of its receiver operating characteristic curve. Oftentimes, two biomarkers are collected from each subject to test if one has a larger AUC than the other. We propose a simple non-parametric statistical test for comparing the performance of two biomarkers. We also present a simple sample size calculation method for this test statis

Clinical factors that influence chorionic villus sampling sample size.

PURPOSE: Our primary objective was to characterize clinical and procedural factors affecting sample size in chorionic villus sampling (CVS). METHODS: This retrospective, single-site cohort study included singleton pregnancies undergoing transabdominal (TA) and transcervical (TC) CVS between 2020 and 2023. Prenatal and maternal data were obtained from the electronic medical record. Differences by approach and sample size in relation to demographic and pregnancy characteristics were assessed. Linear regression was used to identify factors associated with yield. RESULTS: Data from 240 CVS proce

Power and sample-size estimation in human microbiome research.

Human microbiome research has become pivotal in advancing our understanding of complex diseases such as diabetes, inflammatory bowel disease, and cancer. Much of this work relies on comparing microbial communities across health and disease states, or case-control cohorts, using high-throughput metagenomic sequencing. Yet the very nature of sequencing-derived microbiome data makes robust cohort design and power-based sample-size estimation unusually difficult. Unlike other omics, microbiome profiles are compositional, sparse, and often zero inflated, properties that complicate statistical model

Precision-Based Sample Size Determination: Implications for Decision-Making.

Power-based sample size calculation is the standard approach in confirmatory clinical trials, where adequate statistical power is essential for study success. In other decision-making contexts, however, a precision-based approach to sample size determination is sometimes adopted. One such area is consumer studies that evaluate consumers' ability to appropriately select nonprescription medications. Compared with power-based designs, precision-based approaches typically require fewer study participants and can therefore reduce study costs. However, the extent of loss in statistical power has not

Sample size requirements for genetic studies on yellowfin tuna.

In population genetics, the amount of information for an analytical task is governed by the number of individuals sampled and the amount of genetic information measured on each of those individuals. In this work, we assessed the numbers of individual yellowfin tuna (Thunnus albacares) and genetic markers required for ocean-basin scale inferences. We assessed this for three distinct data analysis tasks that are often employed: testing for differences between genetic profiles; stock delineation, and; assignment of individuals to stocks. For all analytical tasks, we used real (not simulated) data

A novel modification approach for the one sample Kolmogorov-Smirnov test in large sample size.

This study aims to propose and evaluate a modified version of the One-Sample Kolmogorov-Smirnov (K-S) test that addresses its current limitations in large sample groups, with the goal of improving its accuracy and reliability in assessing normality assumptions in medical research data. In addition to the classical K-S test, a logarithmic modification was applied to reduce the impact of sample size. This modification replaces the sample size in the test calculation with a logarithmic formula (ln n2) to prevent z-values from becoming excessively small in large samples. Statistical analyses were

Power and sample size calculation of two-sample projection-based testing for sparsely observed functional data.

Projection-based testing for mean trajectory differences in two groups of irregularly and sparsely observed functional data has garnered significant attention in the literature because it accommodates a wide spectrum of group differences and (non-stationary) covariance structures. This article presents the derivation of the theoretical power function and the introduction of a comprehensive power and sample size (PASS) calculation toolkit tailored to the projection-based testing method developed by Wang1. Our approach accommodates a wide spectrum of group difference scenarios and a broad class

Blinded sample size recalculation in clinical trials incorporating historical data.

Recruiting sufficient patients within an acceptable time horizon is an issue for most clinical trials and is especially challenging in the field of rare diseases. It is therefore an attractive option to include historical data from previous (pilot) trials in the current study thus reducing the recruitment burden. In clinical trials with binary endpoint, the required sample size does not only depend on the type I error rate, the power, and the treatment group difference but additionally on the overall event rate. However, there is usually some uncertainty in the planning phase about the value o

A sample size calculation for spontaneous abortion in observational studies.

Spontaneous abortion (SAB) is an important endpoint in studying the effects of drugs or vaccines on pregnancy outcomes. Such studies are typically observational in nature. Evaluation of the risk for SAB is often conducted as part of a prospective exposure cohort study, or pregnancy registry, where women are recruited into the study at some point after recognition of pregnancy. Subjects accrued in this way are left truncated, in the sense that the time to SAB is not followed from the start of pregnancy, and the observed data is conditional on the fact that a woman has not had SAB by enrollment.

Sample size requirements for clinical trials of isolated systolic hypertension.

1. This study investigated components of blood pressure variability in elderly subjects with isolated systolic hypertension (ISH) using both ambulatory blood pressure monitoring (ABPM) and casual clinic blood pressure measurement. These were then used to determine sample size requirements for clinical trials of different designs. 2. Eleven elderly subjects not receiving antihypertensive medication were seen on four occasions at weekly intervals. On each occasion blood pressure was measured in the clinic and then for 24 h using a non-invasive ABPM device. Nested analysis of variance was used to

Bayesian Power-Based Sample Size Determination for Diagnostic Accuracy Studies.

In diagnostic accuracy studies, it is crucial to estimate the required sample size beforehand for the appropriate evaluation of diagnostic measures (e.g., sensitivity and/or specificity). In the development of pharmaceuticals and medical devices, although frequentist approaches have been used for sample size determination, Bayesian approaches have gained attention recently. We propose a Bayesian method for sample size determination using two kinds of priors (i.e., analysis prior and design prior) based on "Bayesian power." In the proposed method, an analysis prior and a design prior were intro

Flexible seamless 2-in-1 design with sample size adaptation.

The 2-in-1 design is becoming popular in oncology drug development, with the flexibility in using different endpoints at different decision time. Based on the observed interim data, sponsors can choose to seamlessly advance a small phase 2 trial to a full-scale confirmatory phase 3 trial with a pre-determined maximum sample size or remain in a phase 2 trial. While this approach may increase efficiency in drug development, it is rigid and requires a pre-specified fixed sample size. In this paper, we propose a flexible 2-in-1 design with sample size adaptation, while retaining the advantage of a

Sample size calculations for disease freedom and prevalence estimation surveys.

We developed a Bayesian approach to sample size calculations for studies designed to estimate disease prevalence that uses a hierarchical model for estimating the proportion of infected clusters (cluster-level prevalence) within a country or region. The clusters may, for instance, be villages within a region, cities within a state, or herds within a country. Our model allows for clusters with zero prevalence and for variability in prevalences among infected clusters. Moreover, uncertainty about diagnostic test accuracy and within-cluster prevalences is accounted for in the model. A predictive

Sample Size in Clinical Cardioprotection Trials Using Myocardial Salvage Index, Infarct Size, or Biochemical Markers as Endpoint.

BACKGROUND: Cardiac magnetic resonance (CMR) can quantify myocardial infarct (MI) size and myocardium at risk (MaR), enabling assessment of myocardial salvage index (MSI). We assessed how MSI impacts the number of patients needed to reach statistical power in relation to MI size alone and levels of biochemical markers in clinical cardioprotection trials and how scan day affect sample size. METHODS AND RESULTS: Controls (n=90) from the recent CHILL-MI and MITOCARE trials were included. MI size, MaR, and MSI were assessed from CMR.
