Sampling strategies in nested case-control studies.

A stratified version of nested case-control sampling which we call "countermatching" is presented. This design uses data available for all cohort members to obtain a sample for collecting additional information in a case-control substudy. Hitherto the only stratified sampling design for such studies has involved matching of controls to cases with respect to confounding variables. However, in some situations, rather than sampling to make controls as similar as possible to cases, we might wish to make them as different as possible. This is achieved by the counter-matched design. Statistical anal

Adrenal Venous Sampling: Practical Insights and Case Studies.

Adrenal venous sampling is the procedural standard for identifying patients with primary hyperaldosteronism who may benefit from adrenalectomy. Patients with unilateral aldosterone excess are considered surgical candidates, often referred to as having surgically remediable aldosteronism. Those with bilateral aldosterone excess are typically treated with mineralocorticoid receptor agonists. In this article, the authors present 3 interesting cases that underscore the importance of data analysis. Additionally, they emphasize that primary aldosteronism represents a spectrum of aldosterone producti

Bile sampling, processing and analysis in clinical studies.

Obtaining a proper bile sample for investigative purposes is of utmost importance to obtain valid results. Bile can be collected by direct aspiration of the gallbladder, by duodenal intubation or by T-tube drainage. The optimal method of collection depends on the investigative question, as well as on the resources available to the investigator. The procedures for obtaining, processing and analyzing human bile (gallbladder and hepatic) are summarized, pointing out the disadvantages and pitfalls that may occur.

Optimal sampling of rat liver tissue for toxicogenomic studies.

Different degrees of a toxic response between and within the various lobes of the liver have been observed in rodents following treatment with acetaminophen. This study was designed to compare 2 sampling methods of the rat liver for gene-expression analysis. Ten male Fischer 344/N rats, 12-14 weeks of age, were treated with vehicle (0.5% aqueous ethyl cellulose) or acetaminophen (APAP, 1500 mg/kg) and sacrificed 24 hours following dose administration. Two representative sections were collected from the left liver lobe, stained with hematoxylin and eosin (H&E), and evaluated independently by 2

Microsampling in pediatric studies: pharmacokinetic sampling for baricitinib (Olumiant™) in global pediatric studies.

Background: Managing blood volumes in pediatric studies is challenging and should be minimized where possible. Results: A sensitive liquid chromatography with tandem mass spectrometry (LC-MS/MS) method was validated and implemented across two phase III global pediatric trials. Two 10-μl aliquots of blood were collected at each time point using the Mitra® device. Concordance between plasma and dried blood was established from older pediatric patients. Incurred sample reanalysis was performed in both studies using the second Mitra tip and acceptance was greater than 83%. Conclusion: The use of m

Implementation of web-based respondent driven sampling in epidemiological studies.

BACKGROUND: Respondent-driven sampling (RDS) is a peer chain-recruitment method for populations without a sampling frame or that are hard-to-reach. Although RDS is usually done face-to-face, the online version (WebRDS) has drawn a lot of attention as it has many potential benefits, despite this, to date there is no clear framework for its implementation. This article aims to provide guidance for researchers who want to recruit through a WebRDS. METHODS: Description of the development phase: guidance is provided addressing aspects related to the formative research, the design of the questionna

Practical recommendations for population PK studies with sampling time errors.

PURPOSE: Population pharmacokinetic (PK) data collected from routine clinical practice offers a rich source of valuable information. However, in observational population PK data, accurate time information for blood samples is often missing, resulting in measurement errors (ME) in the sampling time variable. The goal of this study was to investigate the effects on model parameters when a scheduled time is used instead of the actual blood sampling time, and to propose ME correction methods. METHODS: Simulation studies were conducted based on two major factors: the curvature in PK profiles and t

Predicting the duration of emotional experience: two experience sampling studies.

The authors present 2 studies to explain the variability in the duration of emotional experience. Participants were asked to report the duration of their fear, anger, joy, gratitude, and sadness episodes on a daily basis. Information was further collected with regard to potential predictor variables at 3 levels: trait predictors, episode predictors, and moment predictors. Discrete-time survival analyses revealed that, for all 5 emotions under study, the higher the importance of the emotion-eliciting situation and the higher the intensity of the emotion at onset, the longer the emotional experi

Momentary predictors of compliance in studies using the experience sampling method.

The influence of momentary experiences on compliance has not yet been studied extensively in diary methods such as the experience sampling method (ESM). This study investigated to what extent momentary experiences at the moment of responding (hereafter 'beep') can predict compliance in high frequency ESM protocols. Lagged-analyses were conducted using a pooled dataset of seven studies including 1,318 healthy volunteers and individuals with different mental health conditions. All studies used an ESM design of 10 beeps per day over 4 to 6 days. Overall compliance was 86% (to beeps where a subjec

Saliva as a medium for aldosterone measurement in repeated sampling studies.

BACKGROUND: Saliva is a readily available biological fluid, making it convenient in diagnosis of diseases and in multi-sampling protocols. Several salivary steroids give a useful index of free plasma levels. Increased incidence of primary aldosteronism (PA) in approximately 10% of the hypertensive population has increased interest in the mineralocorticoid aldosterone. METHODS: A biotinylated-aldosterone tracer and a commercially available antibody are used in a time-resolved fluorescence immunoassay (TR-FIA) to measure salivary aldosterone (SA). Saliva was collected in various multi-sampling

Beyond stool: sampling context and inferential scope in gut mechanistic studies.

SUMMARYHuman gut microbiome research still relies heavily on fecal sampling because stool is noninvasive, scalable, repeatable, and essential for population-level and longitudinal studies. These strengths have made stool central to human microbiome science, but fecal output is not a neutral representation of every intestinal compartment. It reflects material that has persisted through digestion, transport, microbial turnover, host absorption, and distal colonic processing. This distinction becomes important when studies move from detecting microbial associations to explaining localized activit

Dynamic sampling of liquid metal structures for theoretical studies on catalysis.

Liquid metals have recently emerged as promising catalysts that can outcompete their solid counterparts for many reactions. Although theoretical modelling is extensively used to improve solid-state catalysts, there is currently no way to capture the interactions of adsorbates with a dynamic liquid metal. We propose a new approach based on ab initio molecular dynamics sampling of an adsorbate on a liquid catalyst. Using this approach, we describe time-resolved structures for formate adsorbed on liquid Ga-In, and for all intermediates in the methanol oxidation pathway on Ga-Pt. This yields a ran

Evaluation of four sampling devices for Burkholderia pseudomallei laboratory aerosol studies.

Previous field and laboratory studies investigating airborne Burkholderia pseudomallei have used a variety of different aerosol samplers to detect and quantify concentrations of the bacteria in aerosols. However, the performance of aerosol samplers can vary in their ability to preserve the viability of collected microorganisms, depending on the resistance of the organisms to impaction, desiccation, or other stresses associated with the sampling process. Consequently, sampler selection is critical to maximizing the probability of detecting viable microorganisms in collected air samples in field

Necessity and risks of arterial blood sampling in healthy volunteer studies.

Arterial blood sampling is necessary when drugs such as the fast-acting opioid analgesic remifentanil exhibit relevant differences between arterial and venous blood concentrations. Arterial cannulation is generally considered to be clinically safe and has thus become a standard procedure in pharmacokinetic-pharmacodynamic assessments. However, rare cases of arterial occlusions have to be considered in risk-benefit assessments of arterial sampling in pharmacokinetic studies, especially when including healthy volunteers. In an actual case, arterial occlusion requiring surgical repair was caused

Experience Sampling Methodology studies of depression: the state of the art.

BACKGROUND: Experience Sampling Methodology (ESM) is ideally suited to test the predictions, and inform the development of contemporary cognitive models of depression. Yet there has been no systematic examination of ESM in depression research. METHOD: A search of databases (PsychARTICLES, PsycINFO, AMED, Ovid Medline and CINAHL) was conducted to identify studies published within the last 25 years investigating major depressive disorder (MDD) using ESM. RESULTS: Altogether, 19 studies using ESM, or comparable methodologies, with clinically depressed individuals were identified and critically

Rapid automated blood sampling system for pharmacokinetics studies of cigarette smoking.

INTRODUCTION: We developed an automated sampling system to allow multiple, discrete blood samples from a human participant to be collected rapidly and immediately following cigarette smoke exposure. We reported the details of the sampling system along with the results of a pilot study for evaluation of the system. METHODS: Components of the system include silastic tubing, solenoid pinch valves, a peristaltic pump, and a fraction collector. This system incorporates a smoking machine that allows precise delivery of cigarette smoke through a mouthpiece and intricate timing to correlate blood sam

Arterial and venous blood sampling in pharmacokinetic studies: bumetanide in rabbits.

The pharmacokinetics of bumetanide were evaluated simultaneously using both arterial and venous plasma data in 4 rabbits after a rapid 5 sec intravenous (iv) bolus dosing. Initial arterial to venous concentration ratios at 5 sec after injection were the highest with the values of 1410, 246, 4.25, and 351 for rabbits 1-4, respectively. Both curves decayed paralleling each other at the terminal phase with the higher venous levels than the arterial levels by 21.6, 48.2, 17.0, and 47.9% for rabbits 1-4, respectively. An exponential term with a negative coefficient was used to account for the short

The use of Experience Sampling Method in psychopharmacological studies: A systematic review.

The Experience Sampling Method (ESM) is increasingly used to validate and assess treatment effects. We conducted a systematic review to update, and extend, the previous review by Bos et al. (2015), by performing literature searches in PubMed, PsycInfo, and EMBASE. Search queries included every DSM-disorder, ATC-listed psychotropic medication, and multiple search-terms for ESM. Studies included medicated psychiatric patients who filled-out ambulatory prompts ≥2/day. Templates served to distil research themes. The majority of the 79 included studies involved substance use- (n = 47), mood- (n = 1
