Extracts of leaves and fruits of Sapindus saponaria L. were orally administered to rats and the parameters of gastric secretion (volume, pH and acidity) were evaluated after pylorus ligature. The inhibitory effect of the extracts on lesions induced by stress was compared to that of cimetidine. The volume and concentration of hydrochloric acid were reduced after oral administration of the extracts. A significant reduction of the lesion index was observed in the acute assays. No significant alteration in body or organ weight was detected in animals treated orally for 30 days with the extracts. T

Saponins are a large group of compounds, produced mostly by plants as a side product of their metabolic activity. These compounds have attracted much attention over the years mostly because of their surface activity and antibacterial, anti-inflammatory and antifungal properties. On the other hand, most of the hitherto research has concerned the action of saponins against microbial cells as a whole. Therefore, knowing the possible interaction of saponins with biomembrane, we decided to check in-vitro the influence of saponin-rich extract of Saponaria officinalis on spheroplasts of two Candida s

The presence of aldehyde groups at C-23 and C-24 of the triterpen aglycon moiety was disclosed in 1H NMR spectra of both the Riedel de Haen saponin (R) (delta 9.336) and Quillaja saponaria QuilA saponin (delta 9.348). The sign of the C-28 acylated linked moiety (delta 176) was present in both saponins, while the delta 171 at C-28 (carboxy group) corresponding to the deacylated saponin, was only detected in the QuilA preparation, indicating 50% of hydrolysis of the ester moiety, probably due to the storage in aqueous solution. The normoterpen moiety was present in both saponins (signals at delt

QS-21 is a triterpenoid saponin adjuvant component widely used in human vaccines, but its commercial supply is limited because it is sourced almost exclusively from the bark of mature Quillaja saponaria trees in Chile. In this study, we report the identification, isolation, as well as structural and immunological characterization of QS-21-Rhamnose (QS-21-Rha), a naturally occurring structural variant that predominates in the leaves and twigs of young Q. saponaria shrubs cultivated in California. Analytical profiling showed that QS-21-Rha represents more than 95% of QS-21 variants in the aerial

Leishmaniasis is a neglected tropical disease (NTDs), endemic in 88 countries that affect more than 12 million people. Current drugs are limited due to their toxicity, development of biological resistance, length of treatment and high cost. Thus, the search for new effective and less toxic treatments is an urgent need. In this study, we report the synthesis of 3 new amide derivatives of hederagenin (22-24) with yields between 70% and 90%, along with 57 other derivatives of hederagenin (1-21, 25-60) carrying different groups at C-28 previously reported by our group, and the results of their in

Immunotoxins were prepared by conjugating saporin, a ribosome-inactivating protein from Saponaria officinalis, to a monoclonal antibody against the Thy1.1 antigen, or to its F(ab')2 fragment. The immunotoxins were eight- to 16-fold more toxic to mice than free saporin. Injection of the immunotoxins induced necrosis of the liver and spleen, whereas free saporin caused necrosis of the epithelium of the kidney tubules. The cytoplasm of the hepatic parenchymal cells was affected by the immunotoxins, lesions being apparent in the rough endoplasmic reticulum and, later, in the mitochondria. These ch

Piceatannol is a natural stilbene polyphenol with promising bioactivities, but its poor water solubility and low bioavailability limit industrial applications. In this study, a novel UDP-dependent glycosyltransferase, SoUGT, was identified from Saponaria officinalis for regioselective glycosylation of piceatannol. SoUGT belongs to the UGT72 subfamily and contains the conserved PSPG (Plant Secondary Product Glycosyltransferase) motif typical of plant O-glycosyltransferases. Molecular docking analysis predicted several residues, including Tyr143, Val120, Ala121, and Val123, that may contribute t

In the present work, the properties of ZnO nanoparticles obtained using an eco-friendly synthesis (biomediated methods in microwave irradiation) were studied. Saponaria officinalis extracts were used as both reducing and capping agents in the green nanochemistry synthesis of ZnO. Inorganic zinc oxide nanopowders were successfully prepared by a modified hydrothermal method and plant extract-mediated method. The influence of microwave irradiation was studied in both cases. The size, composition, crystallinity and morphology of inorganic nanoparticles (NPs) were investigated using dynamic light s

Ultraviolet B (UVB) irradiation mainly affects biological tissues by inducing an increase in reactive oxygen species (ROS) production which leads to deleterious outcomes for the skin, including pain and inflammation. As a protective strategy, many studies have focused on the use of natural products. The aim of this study was to investigate the effects of Aloe saponaria on nociceptive, inflammatory, and oxidative parameters in a model of UVB-induced sunburn in adult male Wistar rats. Sunburned animals were topically treated with vehicle (base cream), 1% silver sulfadiazine (positive control) or

Quillaja: is an abundant source of triterpenic saponins, glycosides with immunostimulant properties, currently employed as vaccine adjuvants. Although current research and commercial applications have focused on the most abundant saponins in Quillaja extracts (QS-7, QS-17, QS-18 and QS-21), the vast chemical variety in Quillaja saponins remains largely unexplored. The discovery of new saponins suited as vaccine adjuvants would open the door to a wider supply of adjuvants in the future. This paper describes the isolation and chemical characterization of Saponin FR, a new saponin fraction purifi

Background/Objectives: Ribosome-inactivating proteins (RIPs) are plant-derived enzymes with potent cytotoxic activity, widely studied as anticancer agents, particularly as toxic payloads in immunoconjugates. Despite numerous encouraging results reported, their clinical application has been limited by their immunogenicity. RIPs from edible plants have been proposed as potentially more suitable candidates due to their possible improved tolerability. However, this aspect still requires validation in vivo in animal models. This study investigated the cytotoxic activity, mechanisms of action and tr
