CD36 is a receptor for human sapovirus.

Human sapovirus (HuSaV) is a major cause of gastroenteritis but has long lacked a defined receptor. Here, we identify the scavenger receptor CD36 as essential for infection. Comparative transcriptomics identified CD36 as the key susceptibility factor. CRISPR knockout abolished replication, while ectopic expression restored permissiveness across diverse cells. Binding assays showed nanomolar affinity between CD36 and HuSaV particles. Infection also required the bile acid glycocholic acid, which promoted CD36 internalization and viral entry. In human intestinal organoids, CD36 knockout abrogated

Norovirus and sapovirus infections in Thailand.

Stool specimens collected between November 2002 and April 2003 from hospitalized infants with acute gastroenteritis from four distinct geographical regions in Thailand were examined for norovirus (NoV) and sapovirus (SaV) by reverse transcription-PCR and sequence analysis. Of the 80 specimens examined, we identified 11 NoV and 9 SaV single infections, and 3 NoV/SaV mixed infections. The majority of NoV strains (64%) belonged to genogroup II/ genotype 4 (GII/4; Lordsdale cluster). Other NoV strains co-circulating belonged to GII/1, GII/3, GII/6, and one new genotype cluster (GII/New). The major

[Establishment of human sapovirus culture method].

More than 40 years after the discovery of human sapovirus (HuSaV), we have established a HuSaV culture system in which HuTu80 cells derived from the human duodenum adenocarcinoma cell line are cultured together with the addition of bile acid as a supplement. In addition to being a common cell line, this system using HuTu80 cells is a versatile method because classical culture media are available, and it is easy to scale-up for culture. However, the number of culture days required to obtain sufficient viral titer, the confirmation of viral gene conservation for sample selection, and the method

CD36 is required for human sapovirus propagation.

Human sapoviruses (HuSaVs), which cause acute gastroenteritis, are highly diverse. Fifteen HuSaV genotype strains (GI.1-7, GII.1-5, -8, and GV.1-2) were efficiently propagated in the human duodenum-derived cell line HuTu80 when supplemented with conjugated bile acid (T. Oka, T.-C. Li, K. Yonemitsu, Y. Ami, et al., J Virol 98:e00639-24, 2024, https://doi.org/10.1128/jvi.00639-24). However, the host cellular factors involved in HuSaV infection and propagation remain unidentified. Using a knockout approach, we newly identified and confirmed that CD36 is a critical cellular protein for the propaga

Human Sapovirus Replication in Human Intestinal Enteroids.

Human sapoviruses (HuSaVs), like human noroviruses (HuNoV), belong to the Caliciviridae family and cause acute gastroenteritis in humans. Since their discovery in 1976, numerous attempts to grow HuSaVs in vitro were unsuccessful until 2020, when these viruses were reported to replicate in a duodenal cancer cell-derived line. Physiological cellular models allowing viral replication are essential to investigate HuSaV biology and replication mechanisms such as genetic susceptibility, restriction factors, and immune responses to infection. In this study, we demonstrate replication of two HuSaV str

A confirmation of sapovirus re-infection gastroenteritis cases with different genogroups and genetic shifts in the evolving sapovirus genotypes, 2002-2011.

Sapovirus (SaV) is an important pathogen that causes acute gastroenteritis in humans. Human SaV is highly diverse genetically and is classified into multiple genogroups and genotypes. At present, there is no clear evidence for gastroenteritis cases caused by re-infection with SaV. We found that two individuals were sequentially infected with SaVs of two different genogroups and had gastroenteritis after each infection, although in one of the subsequent cases, both SaV and norovirus were detected. We also found a genetic shift in SaVs from gastroenteritis outpatients in the same geographical lo

Sapovirus: an emerging pathogen in kidney transplant recipients?

PURPOSE: Diarrhea is an important cause of morbidity and mortality in immunocompromised patients. After including sapovirus to the viral gastroenteritis screening of our institution's laboratory, we noticed an increase in sapovirus infections among kidney transplant recipients. Therefore, we assumed former gastrointestinal tract infections with unidentified pathogens could have been caused by sapovirus. To better understand the characteristics of a sapovirus infection in a high-risk group we initiated this study. METHODS: Over a period of 6 months, all transplant recipients with diarrhea and

A nosocomial sapovirus-associated outbreak of gastroenteritis in adults.

The human caliciviruses norovirus and sapovirus are leading causes of acute, non-bacterial gastroenteritis. In contrast to norovirus, sapovirus is known to give infections mainly in infants and young children. We describe a nosocomial outbreak of gastroenteritis associated with sapovirus involving 23 adult patients and medical staff. The mean age of the patients and medical staff was 52 y and the major signs and symptoms were nausea, diarrhoea, vomiting, abdominal cramp, headache, myalgia and fever. More patients had diarrhoea (72%) than vomiting (56%) and the mean duration of symptoms was 6 d

Unmasking Sapovirus: Chronic Enteritis in an Immunocompromised Adult With Hypogammaglobulinemia.

Sapovirus is a recognized cause of acute viral gastroenteritis, predominantly affecting children and older adults. While typically self-limiting in immunocompetent individuals, sapovirus can result in persistent, severe gastrointestinal illness in immunocompromised patients. Chronic sapovirus enteritis remains underrecognized and lacks standardized treatment guidelines. We report the case of a 69-year-old woman with multiple myeloma and profound hypogammaglobulinemia secondary to chimeric antigen receptor (CAR) T-cell therapy who presented with a 10-month history of chronic watery diarrhea. Septic Shock Secondary to Severe Gastroenteritis Resulting From Sapovirus Infection.

Sapovirus causes acute gastroenteritis (AGE) which manifests as severe diarrhea and vomiting. It is most often seen in, but not limited to, children and toddlers but can occur in people of all ages. It is typically more prevalent in low to middle-income countries but has also been reported in progressive countries such as the United States. Due to the universal use of reverse transcriptase-polymerase chain reaction (RT-PCR) testing, the reported incidence of sapovirus has continued to grow as the culprit agent in both AGE outbreaks and isolated cases. Its symptoms resemble what is seen with ro

Attempts to grow human noroviruses, a sapovirus, and a bovine norovirus in vitro.

Noroviruses (NoVs) and Sapoviruses (SaVs) are enteric caliciviruses that have been detected in multiple mammalian species, including humans. Currently, efficient cell culture systems have been established only for murine NoVs and porcine SaV Cowden strain. Establishment of an efficient in vitro cell culture system for other NoVs and SaVs remains challenging; however, human NoV (HuNoV) replication in 3D cultured Caco-2 cells and a clone of Caco-2 cells, C2BBe1, human enteroids and in human B cells has been reported. In this study, we tested various cells and culture conditions to grow HuNoVs an

A foodborne outbreak of sapovirus linked to catered box lunches in Japan.

Sapovirus (SaV) is a common cause of acute viral gastroenteritis worldwide, and SaV outbreaks have become more frequent in recent years. In January 2010, an outbreak of acute gastroenteritis due to SaV occurred in Aichi, Gifu and Mie Prefectures, Japan. The illness was strongly associated with eating a delivered box lunch prepared by one catering company. In total, 655 (17.1 %) of 3827 individuals developed gastroenteritic symptoms. SaV was detected in seven of the nine people who became ill and in seven of the 52 food handlers at the catering company, but all the tested samples were negative

Propagation of human sapovirus GII.7 strain in HuTu80 subcloned cells.

Human sapoviruses (HuSaVs), which cause acute diarrhea worldwide, are genetically diverse and divided into four genogroups (GI, GII, GIV, and GV). These were further subdivided into 18 genotypes (GI.1-7, GII.1-8, GIV.1, GV.1, and -2). Among them, only one strain has been reported worldwide for GII.7 to date. This strain was detected during a fecal pathogen screening of outpatients with gastroenteritis in Kumamoto Prefecture, Japan in 2008. We established a virus stock panel of diverse HuSaV strains as bioresources for further virological characterization studies and successfully propagated mul

Porcine Sapovirus Protease Controls the Innate Immune Response and Targets TBK1.

Human sapoviruses (HuSaVs) and noroviruses are considered the leading cause of acute gastroenteritis worldwide. While extensive research has focused on noroviruses, our understanding of sapoviruses (SaVs) and their interactions with the host's immune response remains limited. HuSaVs have been challenging to propagate in vitro, making the porcine sapovirus (PSaV) Cowden strain a valuable model for studying SaV pathogenesis. In this study we show, for the first time, that PSaV Cowden strain has mechanisms to evade the host's innate immune response. The virus 3C-like protease (NS6) inhibits type

Human sapovirus propagation in human cell lines supplemented with bile acids.

Human sapoviruses (HuSaVs) cause acute gastroenteritis similar to human noroviruses. Although HuSaVs were discovered four decades ago, no HuSaV has been grown in vitro, which has significantly impeded the understanding of viral biology and the development of antiviral strategies. In this study, we identified two susceptible human cell lines, that originated from testis and duodenum, that support HuSaV replication and found that replication requires bile acids. HuSaVs replicated more efficiently in the duodenum cell line, and viral RNA levels increased up to ∼6 log10-fold. We also detected doub

Global distribution of sporadic sapovirus infections: A systematic review and meta-analysis.

Acute gastroenteritis (AGE), characterized by diarrhea and vomiting, is an important cause of global mortality, accounting for 9% of all deaths in children under five years of age. Since the reduction of rotavirus in countries that have included rotavirus vaccines in their national immunization programs, other viruses such as norovirus and sapovirus have emerged as more common causes of AGE. Due to widespread use of real-time RT-PCR testing, sapovirus has been increasingly reported as the etiologic agent in both AGE outbreaks and sporadic AGE cases. We aimed to assess the role of sapovirus as
