In vitro activity of chemicals and commercial products against Saprolegnia parasitica and Saprolegnia delica strains.

Oomycetes of the genus Saprolegnia are responsible for severe economic losses in freshwater aquaculture. Following the ban of malachite green in food fish production, the demand for new treatments pushes towards the selection of more safe and environment-friendly products. In the present work, in vitro activity of ten chemicals and three commercial products was tested on different strains of Saprolegnia, using malachite green as reference compound. The compounds were screened in agar and in water to assess the minimum inhibitory concentration (MIC) and the minimum lethal concentration (MLC), r

Anti-saprolegnia potency of some plant extracts against Saprolegnia diclina, the causative agent of saprolengiasis.

Saprolegnosis of fresh water fishes caused by Saprolegnia diclina often results in serious economic losses to fish hatcheries. Despite the proven efficiency of malachite green as a potential fungicide in prevention and control of fish saprolegnosis, there is a strong debate about its safety aspects in use since it was documented to be responsible for many carcinogenic and teratogenic attributes. Bioactivity of four ethanolic plant extracts were assessed to attain a natural alternative to the traditional fungicide currently used in saprolegnosis control. Ethanolic extracts of Punica granatum an

Growth and sporulation differences of genetically distinct Saprolegnia parasitica strains in vitro.

Saprolegnia species are major oomycete pathogens of freshwater fishes, causing significant economic and ecological losses in aquaculture and wild populations. Most saprolegniosis disease outbreaks have been attributed to single or very few genetic clones, but potential differences among strains and the covariation of their virulence and transmission traits have remained largely unresolved. In this study, we investigated differences in laboratory growth and sporulation among 9 pathogenic S. parasitica isolates originating from infected salmonids from 6 aquaculture facilities and 1 wild waterbod

Saprolegnia species isolated from rainbow trout: molecular identification, specific PCR and drug sensitivity.

Saprolegniosis is a common infectious disease caused by members of the genus Saprolegnia. This genus contains several species, some of which can cause severe infection in vulnerable fish such as rainbow trout Oncorhynchus mykiss. This study reports the isolation and identification of 4 Saprolegnia species, namely S. parasitica, S. diclina, S. aenigmatica and S. asterophora, from a rainbow trout farm. The isolates were identified by amplification and sequencing of the internal transcribed spacer region. Among the isolates, S. parasitica and S. diclina were more common. S. diclina was found only

Effects of Saprolegnia parasitica on pathological damage and metabolism of Epithelioma papulosum cyprini cell.

Saprolegniasis is a common fungal disease in aquaculture. It will form white flocculent hyphae on the skin of fish, and the hyphae may grow inward and penetrate into muscle tissue, which will reduce the immunity of the body and eventually lead to death. However, there are still some gaps in the mechanism of the fish body surface against the invasion of Saprolegnia. This study explored the defense mechanism of Epithelioma papulosum cyprini cell (EPC) in the process of Saprolegnia parasitica infection from the perspective of pathogenic bacteria and host cells, so as to provide a theoretical basi

Chitin synthases from Saprolegnia are involved in tip growth and represent a potential target for anti-oomycete drugs.

Oomycetes represent some of the most devastating plant and animal pathogens. Typical examples are Phytophthora infestans, which causes potato and tomato late blight, and Saprolegnia parasitica, responsible for fish diseases. Despite the economical and environmental importance of oomycete diseases, their control is difficult, particularly in the aquaculture industry. Carbohydrate synthases are vital for hyphal growth and represent interesting targets for tackling the pathogens. The existence of 2 different chitin synthase genes (SmChs1 and SmChs2) in Saprolegnia monoica was demonstrated using b

Characterizing the Mechanisms of Metalaxyl, Bronopol and Copper Sulfate against Saprolegnia parasitica Using Modern Transcriptomics.

Saprolegniasis, which is caused by Saprolegnia parasitica, leads to considerable economic losses. Recently, we showed that metalaxyl, bronopol and copper sulfate are good antimicrobial agents for aquaculture. In the current study, the efficacies of metalaxyl, bronopol and copper sulfate are evaluated by in vitro antimicrobial experiments, and the mechanism of action of these three antimicrobials on S. parasitica is explored using transcriptome technology. Finally, the potential target genes of antimicrobials on S. parasitica are identified by protein-protein interaction network analysis. Coppe

Therapeutic potential of intraperitoneal miconazole nitrate against Saprolegnia parasitica in Labeo rohita under feeding refusal conditions.

Fungal infections caused by Saprolegnia parasitica are a major concern in freshwater aquaculture, particularly during advanced stages of disease when affected fish often exhibit feeding refusal, rendering oral medication ineffective. This study investigates the therapeutic potential of intraperitoneally administered miconazole nitrate (MCZ) in Labeo rohita fingerlings under such critical conditions. The 96-h LD50 of MCZ was determined to be 0.20 ± 0.13 mg kg-1 body weight (BW). Three sub-lethal doses (0.01, 0.02, and 0.04 mg MCZ kg-1 BW) were evaluated alongside a DMSO control over eight time

Infections may select for filial cannibalism by impacting egg survival in interactions with water salinity and egg density.

In aquatic environments, externally developing eggs are in constant contact with the surrounding water, highlighting the significance of water parameters and pathogens for egg survival. In this study we tested the impact of water salinity, egg density and infection potential of the environment on egg viability in the sand goby (Pomatoschistus minutus), a small fish that exhibits paternal egg care and has a marine origin, but which in the Baltic Sea lives in brackish water. To manipulate the infection potential of the environment, we added either a Saprolegnia infection vector into UV-filtered

Reprint of: The diversity of oomycetes on crayfish: Morphological vs. molecular identification of cultures obtained while isolating the crayfish plague pathogen.

Numerous oomycetes colonise the crayfish cuticle, the best known being the crayfish plague pathogen Aphanomyces astaci. Although other oomycetes associated with crayfish complicate the isolation and molecular detection of A. astaci, their diversity is little known. To improve this knowledge, we analysed 95 oomycete isolates obtained during attempts to isolate A. astaci from crayfish presumably infected by this pathogen. We characterized the isolates morphologically and by sequencing of the nuclear internal transcribed spacer (ITS) region. We identified 13 taxa by molecular analysis. Ten of the
